Effect of Dietary Protein Levels on the Manifestation of Gramoxone Toxicity in Rat Liver (Gramoxone이 단백질 level에 따라 흰쥐 간에 미치는 독성에 관한 연구)

  • Effects of dietary protein levels on the manifestations of the toxicity of gramoxone, a bipyridine herbicide, in the liver of rats were investigated. The addition of gramoxone, with regard to the body weight and feed efficiency ratio of rats, had a move dramatic effect on animals fed a low or intermediate protein diet than for those similarly treated among rats fed a relatively high protein diet. Lipid content in the rat liver tended to increase with the addition of gramoxone into each protein diet, with the exception of the high protein-gramoxone diet. The addition of gramoxone tended to increase hepatic TBA value significantly in rats, especially among those fed the low protein-gramoxone diet or the control-gramoxone diet. Significant morphological changes, including fat changes of hepatic cells and increases in the number of Kupffer cells, were found both in rats fed the low protein diet and those fed any of the gramoxone-treated diets. fat changes within hepatic cells were found to be especially severe in rats fed the low protein-gramoxone diet. Distributions of glycogen in rat liver appeared to increase in rats fed any of the diets to which gramoxone had been added.

Alleviating Effects of Vitamin C on the Gramoxone Toxicity in the Total Lipid Contents, Lipid Peroxidation and Protein Patterns of Rat Liver (흰쥐 간 지질함량, 과산화지질 및 단백질양상에 미치는 제초제 Gramoxone 독성에 대한 비타민 C의 완화효과)

  • This study was made to determine alleviating effects of vitamin C(Vt C) on the gramoxone toxicity in rats with respect to the growth gain, feed effeciency ratio, total lipid contents, lipid peroxication and protein patterns in rat liver. Growth gain, feed efficiency ratio and liver weight in the gramoxone group were decreased significantly as compared with the control group, while those values obtained in the gramoxone-Vt C group were increased as compared with the gramoxone group. Lipid contents and TBA value of rat liver of the gramoxone group were increased significantly as compared with the control group. However, those values of the gramoxone+Vt C group were decreased as compared with the gramoxone group. No significant differences were observed in the liver protein patterns between the gramoxone and the control group. The contents of liver protein were decreased significantly in the gramoxone group fed for 2 weeks as compared with the control group. The change of the liver protein patterns, such as the decrease of high molecular weight protein and the increase of low molecular weight protein were observed in the gramoxone+Vt C group.

  • The behavior of glycogen and histological changes of hepatic tissues in the liver of rats, aged 6 to 7 weeks, fed 18% casein diet under control, gramoxone and gramoxone + vitamin C(Vt. C) diets has been investigated in a comined histropathological and histochemical studies. Cloudy swelling and fat changes of hepatic cells were observed in the gramoxone group with the duration of feeding time. Fat changes of hepatic cells were observed more obviously than cloudy welling, especially in the hepatic cells of periportal area. The number of Kupffer's cells increased signifciantly in the gramoxone group fed for 4weeks. The cloudy swelling and fat changes decreased obviously in the gramxone + Vt. C group. Glycogen content of heaptic cells tended to increase slightly in the gramoxone group as compared with the control group . moreover, glycogen depositons were higher in the hepatic cells where fat changes were obvious. It seems to be that Vt.C alleviating effects on the gramoxone toxicity in the patterns of glycogen distribution and histological structure of heaptic tissues.

  • The effect of vitamin C on th egramoxone toxicity in the duodenal globlet cells of rats were investigated suing histochemical methods. Rats in control, gramoxone and gramoxone+vitmain C (Vt.C) group, aged 6 to 7 weeks, were fed 18% casein diet. In th egramoxone group, neutral and acid mucins of the globlet cells in villi and crypts of duodenum tended to decrease as compared with the control group. And the globle cells secreting nonsuphated mucins tended to increase in number, being usually accompanied by a decrease of the globlet cells secreting suphated mucins which are prominent in the duodenal mucosa of control group. However, the goblet cells secreting nonsuphated mucins tended to increase in the gramoxone + Vt.C group. Morphological changes of the globlet cells in the gramoxone group were noted vacuolation and demolition of globlet cells, while those changes were not significant in the gramoxone +Vt. C group. It seems to be that Vt. C has alleviating effects on the gramoxone toxicity in secretion and production of the duodenal goblet cells.

  • Physiological toxic and mutagenic effects of gramoxone in Drosophila melanogaster were invetigated. Gramoxone was highly toxic on the development, resulting in of lowering the viability and in prolongation of the developmental times. Adults treated with gramoxone during the developmental stages caused a lowering of the productivity and a little chinge in protein quantity. But the effect on the sex-linked lethal mutagenesis was found to be negative. The order of mortality causing ado리t stoa형e feeding to gramoxone in the D. melanogaster complex was like this ; D. mauritiana, D. sechellia, D. simulans and D. melanogasteu Two species of the D. yakuba complex were alike. Those results were more or less correlation with speciation of the D. melanogaster subgroup.

  • This study was undertaken to evaluate the effect of legume reseeding on legume establishment, dry matter yield, quality and botanical composition of grasses in timothy(Ph1eum prarense L.) dominated pasture at alpine area. Establishment number and percentage of legume in timothy dominated pasture were increased with spring, red clover(Tr$olium pratense L.) and gramoxone $treatment Dry matter yield of pasture was increased with fall and grazing after overseeding, but, there waq no difference in legume species. Crude protein content(%) of grasses at the first cutting was increased with red clover and paraquat (gramoxone) treatment. Based on the results, it is suggested that spring in seeding time, red clover in legume species and oveneeding after gramoxone treatment are good for effective renovation of timothy dominated pasture at alpine area.

  • Adequate regrowth control of weeds and shrubs has been one of the important problem for pasture establishment by overseeding on reclaimed forest hilly land. This experiment was carried out on sloping shrub land near Anseong, Gyeonggido to evaluate the effects of herbicide application on growth control of native weeds, shrubs and trees. We evaluated 63 different weeds and wood species in the trials, which are grown common in korean forest. Buthidazol and 4 other herbicides (sodium chlorate, glyphosate, U-46 and gramoxone) were sprayed each at two application rates during the period of the most intensive growth, on 20 July 45 days before overseeding of pasture species. Glyphosate and sodium chlorate gave excellant control of growth of all species. However. Uncus effusus, Carex heterostachya, Polygonatum humile, Rubus corchorifolius, Lonicera coeralea and Rhododendron mucronulatum were classified as a tolerant weed and wood species to herbicides. Herbicide buthidazol showed also successive control of regrowth of weeds and wood species. But buthidazol was evaluated as a none available herbicide for use in pasture establishment. because o! their long persist of toxic herbicide residue in soils after treatments. Herbicide U-46 and gramoxone had almost no effect on regrowth of shrubs and tree species

Hepatopotective Effects of Black Rice on Superoxide Anion Radicals in HepG2 Cells

  • Cyanidin 3-glucoside (C3G) isolated from black rice was investigated for hepatoprotective effects in HepG2 cells under oxidative stress. When an increase in the production of reactive oxygen species (ROS) was induced by gramoxone, cell viability was drastically decreased by 42%. However, in the presence of C3G, no hepatocytic damage was observed in HepG2 cells treated with gramoxone. C3G was found to manifest a stronger scavenging effect (91%) on superoxide anion radical ($O_2\;^{.-}$) than any of the other natural and synthetic antioxidants. Results suggest that C3G from black rice possesses hepatoprotective effects in vitro, which may be, at least in part, due to $O_2\;^{.-}$ scavenging.
