Meat Species Identification using Loop-mediated Isothermal Amplification Assay Targeting Species-specific Mitochondrial DNA

  • Meat source fraud and adulteration scandals have led to consumer demands for accurate meat identification methods. Nucleotide amplification assays have been proposed as an alternative method to protein-based assays for meat identification. In this study, we designed Loop-mediated isothermal amplification (LAMP) assays targeting species-specific mitochondrial DNA to identify and discriminate eight meat species; cattle, pig, horse, goat, sheep, chicken, duck, and turkey. The LAMP primer sets were designed and the target genes were discriminated according to their unique annealing temperature generated by annealing curve analysis. Their unique annealing temperatures were found to be $85.56{\pm}0.07^{\circ}C$ for cattle, $84.96{\pm}0.08^{\circ}C$ for pig, and $85.99{\pm}0.05^{\circ}C$ for horse in the BSE-LAMP set (Bos taurus, Sus scrofa domesticus and Equus caballus); $84.91{\pm}0.11^{\circ}C$ for goat and $83.90{\pm}0.11^{\circ}C$ for sheep in the CO-LAMP set (Capra hircus and Ovis aries); and $86.31{\pm}0.23^{\circ}C$ for chicken, $88.66{\pm}0.12^{\circ}C$ for duck, and $84.49{\pm}0.08^{\circ}C$ for turkey in the GAM-LAMP set (Gallus gallus, Anas platyrhynchos and Meleagris gallopavo). No cross-reactivity was observed in each set. The limits of detection (LODs) of the LAMP assays in raw and cooked meat were determined from $10pg/{\mu}L$ to $100fg/{\mu}L$ levels, and LODs in raw and cooked meat admixtures were determined from 0.01% to 0.0001% levels. The assays were performed within 30 min and showed greater sensitivity than that of the PCR assays. These novel LAMP assays provide a simple, rapid, accurate, and sensitive technology for discrimination of eight meat species.

육성 암컷 흑염소 방목 야초지에서 사료가치, 사초 건물 수량 및 방목 강도의 계절별 변화에 관한 연구 (Changes in Feed Value, Forage Productivity, and Grazing Intensity at Native Pasture Grazed by Growing Korean Native Female Goat(Capra hircus coreanae))

  • 본 연구는 야초지에서 흑염소 방목사육 시 사초생산성, 사료가치, 적정 방목강도 규명 및 가축생산성 분석을 통하여 야초지의 이용성을 평가하고 흑염소 방목 사육에 대한 기초적인 자료를 제공하고자 실시하였다. 공시축은 평균체중 14kg 전후의 4개월 령 육성기 암컷 흑염소 10두로 방목실험을 실시하였다. 방목 시 보충사료는 체중의 1.5%로 추가 급여 하였으며, 물은 자유 급수 할 수 있도록 하였다. 야초지의 건물함량은 봄에서 가을로 갈수록 건물함량이 증가하는 보고와 비슷하게 6월과 9월에 각각 $24.9{\pm}0.56%$, $24.4{\pm}0.89%$로 유의하게 높았다(p<0.05). 조단백질 함량은 평균함량 11%~12%를 유지하여 처리구간 유의성은 보이지 않았다. 야초지의 사료가치는 일정수준으로 영양소 함량이 유지되었으나 계절별로 생산되는 야초의 종류가 다르기 때문에 차이가 존재하였다. 방목 시 사초생산성은 야초의 경우 대부분 C-4형 식물이기 때문에 기온이 높은 여름철인 6월부터 8월까지 생산성의 증가한 것으로 보이며 8월의 경우 유의하게 높았다(p<0.05). 8월 이후 생산성이 급격히 감소하여 봄철과 가을철에 급격하게 생산성이 저하되는 야초지의 특성을 보였다. 방목 초지의 월별 생산성과 흑염소의 채식량을 기준으로 분석을 통해 월별 방목 강도를 설정하였다. 계산된 방목 강도는 8월에 55두/ha 로 가장 높았으며, 사초생산성이 떨어지는 시기인 9월에 21두/ha로 가장 낮았다. 평균 방목강도는 37두/ha로 이를 기준으로 보충사료의 양을 조절하여 방목 흑염소에게 부족하거나 과잉된 영양소 공급의 균형을 맞출 필요가 있을 것으로 사료된다. 흑염소 생산성의 계절별 변화는 증체량이 80.2 g/d 이었다. 방목구에서 안정적인 증체의 결과는 방목 후 추가로 농후사료를 보충급여 함으로써 흑염소의 영양소 요구량을 충족시켰기 때문이며, 방목사육 시 사초생산성이 떨어지는 시기에 적절한 방목강도 조절과 필요 영양수준에 맞춘 보충사료를 급여 한다면 가축의 생산성을 더 높일 수 있을 것으로 판단되며 야초지의 효율적인 활용기술 및 흑염소 방목기술의 추가적인 연구를 통하여 유기축산 및 산지생태축산 실현에 기여할 수 있을 것으로 사료된다.

유기사료급여가 흑염소의 사료섭취량, 영양소소화율 및 질소 축적에 미치는 영향 (Effects of Organic Feed Supplementation on Feed Intake, Nutrient Digestibility, and Nitrogen Retention in Korean Native Goats(Capra hircus))

  • This trial was carried out to determine effects of organic feeds in comparison to conventional diet on feed intake, digestibility, and nitrogen retention in Korean native goats. Twelve Korean native goats were allotted to treatments in four groups of three goats and then they were housed in separate metabolism cages for 21 days. Treatments included conventional diet (A) as a control group and three organic feed groups (B: organic rice straw, C: organic nee leaves, D: organic mixture of rice straw and tree leaves). The A treatment, conventional diet, consisted of common rice straw and commercial concentrates at a proportion of 60 and 40%, respectively. All ingredients of organic feeds treatments were organically produced-agricultural products without any application of chemical fertilizer and pesticide. Four experimental diets were formulated to have the same ratio of forage to concentrate and similar contents for protein and carbohydrate across treatments and they were offered to goats ad libitum. Feed intake, apparent nutrient digestibility and nitrogen retention were investigated. For chemical compositions of experimental diets, all nutrients except crude ash and ether extract were not significantly different across treatments as we expected. Crude ash content was highest in the A treatment (P<0.05), however, it was not significantly different among organic feeds treatments. Ether extract content was higher (P<0.05) in C and D treatments than in A and B. Even if dry matter intakes for organic feeds treatments were not significantly different among them, they were significantly higher (P<0.05) compared with conventional diet. Fecal excreta were not significantly different across treatments, resulting in significantly higher digestible dry matter (g/day) in treatments of organic feeds (P<0.01). Average daily gain (ADG) and feed efficiency (FE) were more increased (P<0.01) in treatments of organic feeds compared with conventional diet. Digestibilities for most of nutrients except NFC had the same trend as ADG and FE, however, NFC digestibilities for C and D treatments were significantly lower (P<0.01) than those of A and B. Nitrogen intakes for organic feeds treatments were significantly higher (P<0.001) than conventional diet, with no difference among organic feeds treatments. Fecal nitrogen loss was higher (P<0.05) for C and D treatments than for A and B. Retained nitrogen contents were significantly higher (P<0.05) for organic feeds treatments than for conventional diet, but nitrogen retention rate did not show any difference across treatments. The results showed that organic feed supplementation more improved feed intake, digestibility and nitrogen retention in comparison with conventional diet, and thus they could be concluded that organic feeds might contribute to animal performance and a safer production of animal product.

Effects of diet and castration on fatty acid composition and volatile compounds in the meat of Korean native black goats

  • Objective: This study determined the effects of dietary treatments and castration on meat quality, fatty acids (FAs) profiles, and volatile compounds in Korean native black goats (KNBG, Capra hircus coreanae), including the relationship between the population of rumen microbiomes and meat FA profiles. Methods: Twenty-four KNBG (48.6±1.4 kg) were randomly allocated to one of four treatments arranged into a 2×2 factorial structure. The factors were dietary forage to concentrate ratio (high forage [HF, 80:20] and low forage [LF, 20:80]), and a castration treatment (castration [CA] vs non-castration [NCA]). Results: Among meat quality traits, the CA group exhibited a higher percentage of crude fat and water holding capacity (p<0.05). The profiles of the saturated fatty acid (SFA) in meat sample derived from CA KNBG showed a significantly lower percentage compared to NCA individuals, due to the lower proportion of C14:0 and C18:0. Feeding a high-forage diet to KNBG increased the formation of C18:1n7, C18:3n3, C20:1n9, C22:4n6 in meat, and polyunsaturated fatty acid (PUFA) profiles (p<0.05). Consequently, the n6:n3 ratio declined (p<0.05). There was an interaction between dietary treatment and castration for formation of C20:5n3 (p<0.05), while C18:1n9, C22:6n3, monounsaturated fatty acid (MUFA) and the MUFA:SFA ratio were influenced by both diet and castration (p<0.05). Nine volatile compounds were identified and were strongly influenced by both dietary treatments, castration (p<0.05), and their interaction. In addition, principal component analysis (PCA) revealed distinctly different odor patterns in the NCA goats fed LF diets. Spearman correlation analysis showed a high correlation between rumen bacteria and meat PUFAs. Conclusion: These results suggest the essential effects of the rumen microbial population for the synthesis of meat FAs and volatile compounds in KNBG meat, where dietary intake and castration also contribute substantially.

에너지 및 단백질 공급 수준에 따른 비유 중기 유산양의 유생산성에 미치는 영향 (Effects of Different Dietary Energy and Protein Levels on Milk Production in Saanen Dairy Goats in Mid Lactation)

  • 본 시험은 산양유 생산을 위해 유기사료를 이용하여 유산양의 적정 에너지 및 단백질 공급 수준과 조농비율을 결정하기 위하여 비유중기 Saanen종 유산양 4두($57.5{\pm}10.8\;kg$ BW)를 이용하여 $4{\times}4$ 라틴방각법으로 실시하였다. 대사에너지 공급수준과 단백질 공급수준을 10.59 (T1), 10.14 (T2), 9.45 (T3) 및 8.75 (T4) MJ/kg과 11.90 (T1), 12.73 (T2), 15.19 (T3) 및 16.60% (T4)로 달리하였으며, 조농비율은 49:51 (T1), 59:41 (T2), 74:26 (T3) 및 90:10 (T4)의 네 처리구로 나누었다. 사료 섭취량 및 체중은 모든 처리구 사이의 유의한 차이가 없었다. 유량은 T3구에서 가장 높았으며, T2 및 T4구와 비교하여 유의하게 높게 나타났다 (p<0.05). 유지방함량과 생산량은 T4구에서 다른 처리구와 비교하여 가장 낮았다 (p<0.05). 유단백질과 유당 생산량은 T1 (61.29 및 70.95 g/일)과 T3 (60.52 및 75.82 g/일)구에서 T2 (50.49 및 59.64 g/일)와 T4 (52.49 및 59.82 g/일)구와 비교하여 유의하게 높게 나타났다. 결과적으로 유산양의 유생산성을 극대화시키기 위해서 에너지 공급수준은 최소 9 MJ/kg 이상과 단백질 공급수준은 최소 15% 이상을 공급해 주어야할 것으로 기대된다.
