Spectroscopic and Electrochemical Detection of Thrombin/5'-SH or 3'-SH Aptamer Immobilized on (porous) Gold Substrates

  • Thrombin is a serine protease that catalyzes the conversion of soluble fibrinogen to insoluble fibrin, and thus induces physiological and pathological blood coagulation. Therefore, it is important to detect thrombin in blood serum for purposes of diagnosis. To achieve this goal, it has been suggested that a 15-mer aptamer strongly binds with thrombin to form a G-quartet structure of the aptamer. Generally, 5'-end thiol-functionalized aptamer has been used as an anti-thrombin binder. Herein, we evaluate the possibility of utilizing a 3'-SH aptasensor for thrombin detection using SPR spectroscopy, and compare the enhancement of the electrochemical signal of the thrombin-aptamer bound on a porous gold substrate. Although the two aptamers have similar configurations, in SPR analysis, the 3'-SH aptamer was a effective aptasensor as well as 5'-SH aptamer. Results from electrochemical analysis showed that the porous gold substrate acted as a good substrate for an aptasensor and demonstrated 5-fold enhancement of current change, as compared to gold thin film.

  • In this paper, we introduce an intelligent robotic arc welding system which exploits sensors like as LVS (Laser Vision Sensor), Hall effect sensor, voltmeter and so on. The use of industrial robot is saturated because of its own limitation, and one of the major limitations is that industrial robot cannot recognize the environment. Lately, sensor-based environmental awareness research of the industrial robot is performed actively to overcome such limitation, and it can expand application field and improve productivity. We classify the sensor-based intelligent arc welding robot system by the goal and the sensing data. The goals can be categorized into detection of a welding start point, tracking of a welding line and correction of a torch deformation. The Sensing data can be categorized into welding data (i.e. current, voltage and short circuit detection) and displacement data (i.e. distance, position). This paper covers not only the explanation of the each category but also its advantage and limitation.

  • This paper addresses solutions to the challenges of carrier phase integer ambiguity resolution and cycle slip detection/identification, for maintaining high accuracy of an integrated GPS/Pseudolite/INS system. Such a hybrid positioning and navigation system is an augmentation of standard GPS/INS systems in localized areas. To achieve the goal of high accuracy, the carrier phase measurements with correctly estimated integer ambiguities must be utilized to update the system integration filter's states. The occurrence of a cycle slip that is undetected is, however, can significantly degrade the filter's performance. This contribution presents an effective approach to increase the reliability and speed of integer ambiguity resolution through using pseudolite and INS measurements, with special emphasis on reducing the ambiguity search space. In addition, an algorithm which can effectively detect and correct the cycle slips is described as well. The algorithm utilizes additional position information provided by the INS, and applies a statistical technique known as the cumulative-sum (CUSUM) test that is very sensitive to abrupt changes of mean values. Results of simulation studies and field tests indicate that the algorithms are performed pretty well, so that the accuracy and performance of the integrated system can be maintained, even if cycle slips exist in the raw GPS measurements.

Non-invasive acceleration-based methodology for damage detection and assessment of water distribution system

  • This paper presents the results of a pilot study and verification of a concept of a novel methodology for damage detection and assessment of water distribution system. The unique feature of the proposed noninvasive methodology is the use of accelerometers installed on the pipe surface, instead of pressure sensors that are traditionally installed invasively. Experimental observations show that a sharp change in pressure is always accompanied by a sharp change of pipe surface acceleration at the corresponding locations along the pipe length. Therefore, water pressure-monitoring can be transformed into acceleration-monitoring of the pipe surface. The latter is a significantly more economical alternative due to the use of less expensive sensors such as MEMS (Micro-Electro-Mechanical Systems) or other acceleration sensors. In this scenario, monitoring is made for Maximum Pipe Acceleration Gradient (MPAG) rather than Maximum Water Head Gradient (MWHG). This paper presents the results of a small-scale laboratory experiment that serves as the proof of concept of the proposed technology. The ultimate goal of this study is to improve upon the existing SCADA (Supervisory Control And Data Acquisition) by integrating the proposed non-invasive monitoring techniques to ultimately develop the next generation SCADA system for water distribution systems.

The Robust Artillery Locating Radar Deployment Model Against Enemy' s Attack Scenarios (적 공격시나리오 기반 대포병 표적탐지레이더 배치모형)

  • The ROK Army must detect the enemy's location and the type of artillery weapon to respond effectively at wartime. This paper proposes a radar positioning model by applying a scenario-based robust optimization method i.e., binary integer programming. The model consists of the different types of radar, its available quantity and specification. Input data is a combination of target, weapon types and enemy position in enemy's attack scenarios. In this scenario, as the components increase by one unit, the total number increases exponentially, making it difficult to use all scenarios. Therefore, we use partial scenarios to see if they produce results similar to those of the total scenario, and then apply them to case studies. The goal of this model is to deploy an artillery locating radar that maximizes the detection probability at a given candidate site, based on the probability of all possible attack scenarios at an expected enemy artillery position. The results of various experiments including real case study show the appropriateness and practicality of our proposed model. In addition, the validity of the model is reviewed by comparing the case study results with the detection rate of the currently available radar deployment positions of Corps. We are looking forward to enhance Korea Artillery force combat capability through our research.

A Study on the Fracture Detection of Multi-Point-Tool (다인공구의 파손검출에 관한 연구)

  • In modern industry the requirement of automation of manufacturing process increases so that unmanned system has been popular as an ultimate goal of modern manufacturing process. In unmanned manufacturing process the tool fracture is a very serious problem because it results in the damage of workpieces and can stop the operation of whole manufa- turing system. In this study, image processing technique is used to detect the fracture of insert tip of face milling using multi-point-tool. In order to acquire the image information of fracture shape of rotation insert tip. We set up the optical system using a light beam chopper. In this system we can reduce the image degradation generated from stopped image of rotating insert tip using image restoration technique. We calculated the mean square error to diagnose the condition of tool fracture, and determind the criteria of tool fracture using experimental and staticstical method. From the results of this study we've developed non- contact detection technique of tool fracture using image processing method and proposed the fracture direction of automation and unmanned system considering the optimal time of tool change milling.

Pulse pile-up correction by auto-regression on linear operations (ARLO) method: A comparison with integration-based algorithms

  • Radiation detection at high count rate suffers from pulse pile-up, where the counting data and energy information of the system are affected by the overlapping of the system output pulses. There exist various pile-up correction strategies to recover the true information of the pulses, among which pulse-tail extrapolation is a well-known method focused on in this study. Present work aims to use a mono-exponential model for extrapolating the pileup-distorted trailing edge of a pulse, to provide a reference line for calculating the true amplitude of its subsequent overlapping pulse. To this goal, the auto-regression on linear operations (ARLO) method is examined and compared with two integration-based methods (the Foss and the Matheson methods), as well as the non-linear least squares (NLS) method. Despite a higher sensitivity to noise, the ARLO method was able to provide a simple, non-iterative solution with a performance over 400 times faster than the NLS algorithm, according to the analysis of a high count rate set of experimental pulses from a NaI(Tl) detection system. Foss and Matheson methods also provided solutions reasonably faster than NLS (but not surpassing ARLO), performing exactly the same as each other with results very close to NLS, benefiting from their non-iterative nature.

Deforestation Patterns Analysis of the Baekdudaegan Mountain Range (백두대간지역의 산림훼손경향 분석)

  • The Baekdudaegan Mountain Range is a backbone of the Korean Peninsula which carries special spiritual and sentimental signatures for Koreans as well as significant ecological values for diverse organisms. However, in spite of importance of this region, the forests of Baekdudaegan have been damaged in a variety of human activities by being used as highland vegetable grower, lumber region, grass land, and bare land, and are still undergoing destruction. The existing researches had determined the details of the damage through on-site and recent observations. Such methods cannot provide quantitative and integrated analysis therefore could not be utilized as objective data for the ecological conservation of Baekdudaegan forests. The goal of this study is to quantitatively analyze the forest damage in the Baekdudaegan preservation region through land cover categorization and change detection techniques by using satellite images, which are 1980s, and 1990s Landsat TM, and 2000s Landsat ETM+. The analysis was executed by detecting land cover changed areas from forest to others and analyzing changed areas' spatial patterns. Through the change detection analysis based on land cover classification, we found out that the deforested areas were approximately three times larger after the 1990s than from the 1980s to the 1990s. These areas were related to various topographical and spatial elements, altitude, slope, the distance form road, and water system, etc. This study has the significance as quantitative and integrated analysis about the Baekdudaegan preservation region since 1980s. These results could actually be utilized as basic data for forest conservation policies and the management of the Baekdudaegan preservation region.

MAGE Gene Expression in Bronchial Washing Fluid in Suspected Parenchymal Lung Cancer (주변부 폐암에서 기관지세척액을 이용한 MAGE유전자검사법의 임상적 유용성)

  • Background: The main goal of this study was to evaluate the diagnostic efficacy of reverse transcription-nested polymerase chain reaction (RT-nested PCR) in bronchial washing fluid with MAGE A1-6 common primers for the detection of lung cancers invisible by bronchoscopy. Methods: To determine the expression of MAGE A1-6 gene in 189 lung cancers diagnosed by conventional fluoroscopy-guided lung biopsy and 89 cancer-free controls, RT-nested PCR was performed in bronchial washing specimens. We analyzed MAGE A1-6 RT-nested PCR data according to tumor histology, stage, size, and compared them with cytological data. Results: 189 patients (111 cases in adenocarcinoma, 47 cases in squamous cell carcinoma, 22 cases in small cell lung carcinoma, and 9 cases in other cancers) and 89 benign patients were investigated. The expression of MAGE was performed by nested RT-PCR using common MAGE primer. Among 189 cancer patients, the expression rate of MAGE was 49.2%, and the positive predictive value was 89.4%. However, the expression rate of MAGE in patients with benign lesions was 12.4%. In peripheral lung cancer, the positive rate of MAGE expression was 57.4% in squamous cell carcinoma, 44.1% in adenocarcinoma and 59.1% in small cell lung cancer. Whereas the expression rate of bronchial washing cytology in peripheral lung cancer was 9.0% (p=0.011). Conclusion: MAGE RT-PCR in bronchial washing fluid gave us promising data for the detection of peripheral lung cancer. It could be a useful method for selecting diagnostic tools for peripheral lesions.

The Comparative Study on Performance Analysis of Windows 7 and Ubuntu Applying Open Source IDS/IPS Suricata (오픈소스 IDS/IPS Suricata를 적용한 Windows7과 Ubuntu 성능 비교 분석)

  • Nowadays, It is undeniable that the threat of network security is growing as time flows due to worldwide development of wire/wireless, various Internet platform and sophisticated hacking techniques. The amount of traffics that Network security solution has to handle is increasing and recently many occurrence of explosive traffic attacks from PulseWave are being observed which has many similar characteristics to New DDos. Medium and small sized firms abroad have developed and distributed Snort and Suricata that are based on open-source Intrusion Detection System(IDS) / Intrusion Prevention System (IPS). The goal of this study is to compare between Windows7 by applying suicata 4.0.0 32bit version and Ubuntu 16.04.3 LTS by applying suicata 4.0.0 version which is an open source Intrusion Detection System / Intrusion Protection System that uses multi threads method. This experiment's environment was set as followed C1100 server model of Dell, Intel Xeon CPU L5520 2.27GHz*2 with 8 cores and 16 threads, 72GB of RAM, Samsung SSD 250GB*4 of HDD which was set on RAID0. According to the result, Suricata in Ubuntu is superior to Suricata in Windows7 in performance and this result indicates that Ubuntu's performance is far advanced than Windows7. This meaningful result is derived because Ubuntu that applied Suricata used multi core CPU and RAM more effectively.
