Bioethanol derived from lignocellulosic biomass has the potential to replace gasoline. Cellulose is naturally recalcitrant to enzymatic attack, and it also surrounded by the matrix of xylan and lignin, which enhances the recalcitrance. Therefore, lignocellulosic materials must be pretreated to make the cellulose easily degraded into sugars and further fermented to ethanol. In this work, hydrothermal pretreatment on corn stover at $195^{\circ}C$ for 15 min with and without lower concentration of formic acid was compared in terms of sugar recoveries and ethanol fermentation. For pretreatment with formic acid, the overall glucan recovery was 89% and pretreatment without formic acid yielded the recovery of 94%. Compared with glucan, xylan was more sensitive to the pretreatment condition. The lowest xylan recovery of 55% was obtained after pretreatment with formic acid and the highest of 75% found following pretreatment without formic acid. Toxicity tests of liquor parts showed that there were no inhibitions found for both pretreatment conditions. After simultaneous saccharification and fermentation (SSF) of the pretreated corn stover with Baker's yeast, the highest ethanol yield of 76.5% of the theoretical was observed from corn stover pretreated at $195^{\circ}C$ for 15 min with formic acid.
Various recovery methods were investigated to maximize hemicellulose recovery from lignocellulosic material hydrolyzed by pure water. The pretreatment conditions of water hydrolysis were $170\~180^{\circ}C$ and 1 hour of reaction time. The percentage of hemicellulose solubilized increased as the temperature increased from 170 to $180^{\circ}C$. However, significant decomposition of sugar was observed at temperature of $180^{\circ}C$. From the results of water hydrolysis, the total amount of glucan in solid residue and liquid hydrolyzate was close to the total glucan in the original biomass. For hemicellulose, however, there was a significant difference between both contents. To prove this difference, various recovery methods were proposed. From the total sugar accountability (sugar in liquid + sugar in solid), it was confirmed that hemicellulose recovery in the hydrolyzate was increased if the product including both hydrolyzate and solid residue was physically stimulated by such as heating and ultrasound irradiation. This indicated that, in commercial scale processes that much bigger substrate sizes are used and a sufficient amount of leaching solvent can not be used after pretreatment, a significant amount of oligomers could be trapped in the solid matrix.

This study was conducted to investigate the $\beta$-glucan contents and their characteristics of winter cereals according to particle sizes and milling recoveries. Sieved fractions differed in their average contents of $\beta$-glucans, and the coarse fraction had higher contents of $\beta$-glucan than finely milled fractions. In all winter cereals, the $\beta$-glucan contents of raw flours were higher than those of their brans, and the highest $\beta$-glucan contents of every cereals were observed at 100 mesh > or 100-140 mesh fractions except the Chalssalbori fractions which showed the higest $\beta$-glucan contents (12.9%) at 140-200 mesh fraction. As compared with the $\beta$-glucan content of Chalbori among the various milling recoveries, the $\beta$-glucan was distributed more evenly throughout the endosperm but $\beta$-glucan content in bran of Chalbori was only 1.5%. However, $\beta$-glucan content of Chalssalbori (hull-less waxy barley) was the highest in the subaleurone region (8.2%) and declined slightly toward inner layers of grain. This results suggest that $\beta$-glucan distribution between high (Chalbori) and low $\beta$-glucan barley (Chalssalbori) may explain the difference in milling performance of barley. On the other hand, $\beta$-glucan contents of two rye varieties (Chilbohomil, Chunchoohomil) were lower than those of two waxy barley varieties, and the higest $\beta$-glucan contents were observed at the 60% milling recoveries. In all winter cereals, the L-values (lightness) of raw flours were higher than those of brans. And the L-values of barley varieties were higher than those of oat and rye varieties. As the particle sizes and milling recovery ratios were decreased, the L-value were increased. The a-values (redness) in brans of every winter cereals were higher than those of every particle size flours and every milling ratio fractions, and this tendency was observed in the b-values (yellowness) of every particle size of cereal flours. The L and b-value of barley, the b-value of oat, and L, a, b-value of rye have the significant relationship with the $\beta$-glucan contents, respectively. This results represent the fact that $\beta$-glucans affected the color of the flours and pounded grains of winter cereals.

A two-step process was investigated for pretreatment and fractionation of rice straw. The two-step fractionation process involves first, soaking rice straw in aqueous ammonia (SAA) in a batch reactor to recover lignin-rich hydrolysate. This is followed by a second-step treatment in a fixed-bed flow-through column reactor to recover xylo-oligomer-rich hydrolysate. The remaining glucan-rich solid cake is then subjected to an enzymatic process. In the first variant, SAA treatment in the first step dissolves lignin at moderate temperature (60 and $80^{\circ}C$), while in the second step, hot-water treatment is used for xylan removal at higher temperatures ($150{\sim}210^{\circ}C$). Under optimal conditions ($190^{\circ}C$ reaction temperature, 30 min reaction time, 5.0 ml/min flow rate, and 2.3 MPa reaction pressure), the SAA-hot-water fractionation removed 79.2% of the lignin and 63.4% of the xylan. In the second variant, SAA was followed by treatment with dilute sulfuric acid. With this process, optimal treatment conditions for effective fractionation of xylo-oligomer were found to be $80^{\circ}C$, 12 h reaction time, solid-to-liquid ratio of 1:12 in the first step; and 5.0 ml $H_2SO_4/min$, $170^{\circ}C$, and 2.3 MPa in the second step. After this two-step fractionation process, 85.4% lignin removal and 78.9% xylan removal (26.8% xylan recovery) were achieved. Use of the optimized second variant of the two-step fractionation process (SAA and $H_2SO_4$) resulted in enhanced enzymatic digestibility of the treated solid (99% glucan digestibility) with 15 FPU (filter paper unit) of CTec2 (cellulase)/g-glucan of enzyme loading, which was higher than 92% in the two-step fractionation process (SAA and hot-water).
DNA fragment being able to restore in vitro activity of ${\beta}-1,3-glucan$ synthase was cloned by transformation of the Saccharomyces cerevisiae LP353 mutant strain with genomic library constructed in the YCp50. For the selection of transformants which showed no detectable phenotype linked to recovery of the defect in ${\beta}-1,3-glucan$ synthase activity, the colony autoradiography was succesfully applied. The restriction map of the cloned DNA fragment, which is 8.5-kb in length, was constructed. Both the YEplac195 and the YCp50 carrying the 8.5-kb fragment increased ${\beta}-1,3-glucan$ synthase activity of LP353 by two fold. Neither the YEplac195 nor the YCp50 carrying the 8.5-kb DNA fragment, however, complemented the temperature-dependent osmotic sensitivity which is another distinctive phenotype of LP353. Subcloning experiments indicated that a functional region was located in 4.8-kb BglII-KpnI fragment. The 4.8-kb fragment was also able to increase the level of ${\beta}-1,3-glucan$ content in cell wall as well as the resistance of cells to cell wall lytic enzyme, ${\beta}-1,3-glucanase$. The growth rate of the LP353 with 4.8-kb fragment was almost same as that of wild type strain in liquid medium with 1.2 M sorbitol at nonpermissive temperature. Taken these results together, the 4.8-kb fragment seemed to contain the BGS2 gene for ${\beta}-1,3-glucan$ synthase activity in yeast S. cerevisiae.

In this study, the efficiency of M/F(micro filtration) system was investigated about the wastewater generated from the production process of ${\beta}-glucan$. M/F membrane used the pellicon 2 cassette filter module of millipore(USA) for the operation of M/F plant system. Flux was rised as operation pressure increased, and decreased with the operation time. As concentration ratio increased, the recovery of ${\beta}-glucan$, which was remaind in retentate was effective. As the fermentation solution of ${\beta}-glucan$ reused, the conversion ratio was 42.5%, and the status of fermentation was stable. Based on these results, we suggested that permeate was applicable as water reuse in cleaner production technology.

The aim of this study was to investigate the method validation for the determination of eleutherosides (B and E) and ${\beta}$-glucan in Acanthopanax (A.) koreanum. This medicinal plant reportedly mainly included eleutherosides which exhibit the pharmacological effects, and ${\beta}$-glucan substantially enhances the function of the immune system by activating macrophages. The specificity, linearity, precision, accuracy, limit of detection (LOD, S/N=3), and limit of quantification (LOQ, S/N=10) were measured by HPLC and enzymatic methods. Our results showed that the coefficient of calibration correlation ($R^2$) for eleutheroside B and E were 0.9997 and 0.9999, respectively. The limits of detection (LOD) for eleutheroside B and E were $0.050{\mu}g/mL$ and $0.025{\mu}g/mL$, respectively. The recovery rate of eleutheroside B and E were revealed in the high range of 100.66~110.04% and 94.26~111.62%, respectively. The inter-day precision of eleutheroside B and E in the root and stem in A. koreanum were 1.4~5.0% and 1.1~2.5%, respectively. The intra-day precision of eleutheroside B and E in the root and stem in A. koreanum were 2.8~2.9% and 0.4~1.1%, respectively. Furthermore, the inter-day and intra-day precision of ${\beta}$-glucan in the stem, leaf, and fruit of A. koreanum were 1.32~5.67% and 8.01~11.76%, respectively. In conclusion, the methods were validated for the detection of eleutherosides and ${\beta}$-glucan in A. koreanum.
Polysaccharide-based dressings have increasingly become viable alternatives to somewhat less biocompatible and often problematic cotton or viscose gauzes traditionally used for wound dressings. Polysaccharide which is particularly abundunt in Agaricus blazei Murill is known as the bioactive materials. Polysaccharide extracted from Agaricus blazei Murill enhances the recovery of the damaged epidermal tissue and the production of epithelial cell growth factors which are necessary to heal the burn and wound on the epidermal tissue. In this study, the biopolymer delivery system of polysaccharide extracted from Agaricus blazei Murill into epidermal tissue was elucidated using the skin permeating enhancer in vitro. The enhancing effects of various penetration enhancers and some other enhancers on the permeation of ${\beta}-glucan$ were evaluated using Franz diffusion cell. Permeation of polysaccharide in nomal and burn skin was compared. Polysaccharide was studied for the burn and wound healing activity in the epidermal tissue on rats.
Cyclodextrin glucanotransferase (EC 2.4.1.19 : 1,4-$alpha$-glucan 4-$alpha$-D-(1,4-glucano) transferase, cyclizing; CGTase) was recovered by starch adsorption. The adsorption and desorption of CGTase to starch was studied as a function of pH, temperature, and starch type. The optimal pH, temperature, and starch for adsorption were, 8.0, $4^{circ}C$, and 1% (w/v) corn starch, respectively, per 205 U/mL enzyme activity in the presence of 25% (w/v) ammonium sulfate. The maximum adsorption ratio was 95%. On the other hand, the optimal pH, temperature, and starch type for desorption were 8.0 (tris-buffer), $50^{circ}C$, and oxidized starch, respectively. The maximum desorption ratio was 98% by tris-buffer solution at pH 8.0. The efficiency of adsorption and desorption were affected slightly by the removal of cells from the fermentation broth.

Landsat TM 영상을 이용, 명암차가 높은 산악 지역에 적용해왔던 알고리즘을 개선하여 비교적 명암차가 낮고 넓게 분포하는 충적층 지역의 선구조를 추출하는 알고리즘을 개발하였다. 수치지형모델(OEM)에 대하여 Local Enhancement 를 이용해서 평탄한 지역을 선정하여 이로부터 충적층을 추출하였다. Zevenbergen & Thorno's Method를 3×3 moving windowing을 통해서 최대 경사방향과 경사를 구해서 충적층을 지나는 선구조 요소를 추출하고 다시 Hough 변환을 이용해서 1차 선구조를 추출하였다 이를 이용하여 충적층의 직각방향의 지형단면의 경사를 유추해서 spline 보간법을 이용해 단면의 최저점을 구하고 이 구해진 점들을 다시 Hough 변환을 이용해서 최종 선구조를 추출하였다. 본 연구에서 사용한 알고리즘은 기존 알고리즘에서 사용된 소창문보다 훨씬 큰 충적층이 분포하는 지역의 지형 경사가 수렴하는 부분에 선구조가 뚜렷이 나타남을 볼 수 있다. 최대경사방향과 경사를 구해서 얻어진 1 차선구조와 최종 선구조에서 선구조 방향이 다소 차이를 보인다. 1 차 선구조의 수직방향 지형단면의 자료를 이용함에 있어, 지형 단면의 시작정과 끝지점을 임의적으로 결정하는 것이 아니라, 충적층을 가로질러 최고점까지 또는 다음 충적층이 나을 때까지의 자료를 이용해서 보간법을 사용하였고, 충적층의 넓이에 따라 보간할 자료량의 차이에 의한 오차가 발생할 수 있다. 넓은 충적층에서 선구조가 잘 추출되는 반면에 좁은 충적층이 분포하거나 계곡에 해당하는 지역에l서는 경사수렴부와 일치하지 않는 선구조가 추출되었다. 이는 향후 계속적으로 연구해서 보완되어야 할 것으로 사료된다.페클 잡영 제거 정도에 있어 다른 필터들과 큰 차이가 없지만 경계선보존지수는 다른 필터들에 비하여 가장 우수함을 확인할 수 있었다.rbon 탐식효율을 조사한 결과 B, D 및 E 분획에서 유의적인 효과를 나타내었다. 이상의 결과를 종합해볼 때, ${\beta}$-glucan은 고용량일 때 직접적으로 또는 $IFN-{\gamma}$ 존재시에는 저용량에서도 복강 큰 포식세로를 활성화시킬 뿐 아니라, 탐식효율도 높임으로써 면역기능을 증진 시키는 것으로 나타났고, 그 효과는 crude ${\beta}$-glucan의 추출조건에 따라 달라지는 것을 알 수 있었다.eveloped.
