Enzymatic Hydrolysis of Crystalline Chitin in an Agitated Bead Reaction System and Its Reaction Characteristics

  • Native crystalline chitin was hydrolyzed in an agitated bead reaction system using crude chitinase excreted from Aspergillus fumigatus JC-19. The reaction was enhanced significantly, and the concentration and yield of reducing sugar after 48 hours were measured to be 35.42 g/I (w/v) and 0.64, respectively, around 1.86 times higher than those of the conventional system that was carried out without glass beads. The effect of reaction conditions, such as the amounts of chitin, chitinase and glass beads, and the size of glass bead, were examined. Ball milled chitin was also hydrolyzed in the agitated bead reaction system, the conversion yield and reaction rate of ball milled chitin for 24 hours increased up to 0.87 and 48.02 g/I, respectively. Chitinase showed relatively high stability in the agitated bead reaction system, particularly in the presence of enzyme stabilizer, $Ca^{++}$, which played a critical role in preventing the deactivation of chitinase by the physical impact of glass beads. The variations of the structural features of chitin during the reaction were followed by SEM and X-ray diffraction, and the enhanced hydrolysis reaction was caused by both the fragmentation of chitin particles and the destruction of the crystalline structure owing to the synergic effects of the attrition of glass beads and the hydrolytic action of chitinase.

  • The effect of photocatalyst-carrying media porosity on the photocatalytic removal efficiency of malodor and VOC of waste air was evaluated when the photocatalytic removal efficiency of porous silica-based media was compared with that of glass bead as control. The amount of photocatalyst coated on the surface of porous silica-based media was observed to be $1,716.3{\mu}g/cm^2$, which was 250% as much as that of nonporous glass bead (control) of $670{\mu}g/cm^2$. The removal efficiencies of hydrogen sulfide and toluene in case of porous silica-based media were observed to be 22% and 82%, respectively, while the removal efficiencies of hydrogen sulfide and toluene in case of nonporous glass bead media were observed to be 19% and 53%, respectively. Therefore, the removal efficiencies of hydrogen sulfide and toluene increased by 16% and 55%, respectively, when the removal efficiencies of porous silica-based media were compared with those of nonporous glass bead media. Thus the increment ratio of the removal efficiency of toluene was observed to be 3.4 times higher than that of hydrogen sulfide.

  • To improve the fire vulnerability of the organic insulation sandwich panel core, which is the main culprit of the large-scale fire disaster, an experiment was conducted to examine the thermal conductivity properties of the core material mixed with the organic insulation material EPS Bead and the inorganic insulation material glass wool. As the Additional ratio of glass wool increased, the thermal conductivity decreased, and it was determined that the replacement of glass wool of 3% or more had little effect on the decrease in thermal conductivity. In addition, it can be seen that the most ideal thermal conductivity is exhibited when 1% Replacement ratio of EPS and 3% glass wool are added. The core material of such organic and inorganic insulation materials is judged to be a core material that can compensate for the fire vulnerability of existing insulation materials. Therefore, in order to determine whether it is used as a core material for sandwich panels, additional studies such as fire resistance experiments and sound absorption experiments are needed in the future.

Manufacturing of Flexible Patterned Cover Film for Solar Cell by Solution Coating (용액 코팅을 이용한 태양전지용 고분자 유연 패턴필름 제조)

  • The flexible patterned cover film was made by a simple solution coating process using polymethyl methacrylate (PMMA) solution with glass beads. The effect of patterned cover film on the efficiency of solar cell has been investigated. It was found that the relative solar cell efficiency increased up to 3.4% with the incident light angle between $0-90^{\circ}$ by the sphere shape of glass bead coated on the film surface. This was understood that the loss of transmittance and scattering due to the light directional dependency on solar cell were minimized because the light entered glass beads normal to its surface regardless of incident light angle. The maximum relative solar cell efficiency was achieved when glass bead shape on the film is hemisphere and the relative efficiency increased with increasing the amount of glass bead on the film surface. However, too much glass beads on the film surface resulted in the lower relative solar cell efficiency due to the lowering of transmittance as well as the occurring of light interference.

  • Kyungsan nursery complex which has a vast area for the production of various species of fruit tree stocks is in a high demand of virus-free saplings. Apple tree stocks, the most important products, urgently need more rapid and reliable viral diagnosis. In this study, a bead beater was tested because of convenience in dealing with large number of samples. Also, industrial glass bead abrasive (0.4 mm in diameter) at very low cost was used in a disposable way. For bead beater-aided RNA extraction from apple stem tissues, the guanidine thiocyanate method was confirmed to be very reliable. Silca membrane filter tube in connection to vacuum filtering device was strongly suggested for simplifying RNA capture and washing steps. Apple virus detection was confirmed by RT-PCR.

Effect of Attachment Substrate Size on the Growth of a Benthic Microalgae Nitzschia sp. in Culture Condition (실내 배양시 부착기질 크기에 따른 저서성 미세조류 Nitzschia sp.의 성장 특성)

  • To understand the effect of attachment substrate on the growth of benthic microalgae, we experimentally investigated the growth of benthic microalgae Nitzschia sp. (Jinhae Bay strain) with additions of glass beads in different sizes. The glass beads used in this study are 0.09-0.15 mm (G1), 0.25-0.50 mm (G2), 0.75-1.00 mm (G3) and 1.25-1.65 mm (G4). No addition of glass beads used as controls. Highest specific growth rate (0.37/day) and maximum cell density ($9,232{\pm}840$ cells/mL) of Nitzschia sp. showed at the smallest glass beads (G1), and the specific growth rate and maximum cell density were decreasing with increasing size of glass beads (specific growth rate and maximum cell density of G4 was 0.24/day and $6,397{\pm}524$ cells/mL, respectively). Moreover, specific growth rate of the control experiment (0.23/day) was significantly lower than their of G1 to G3 experiment. The results indicated that the attachment substrate for benthic microalgae as Nitzschia sp. is important factor which affecting the growth rate as well as cell density. Therefore, the physiological experiment of benthic microalgae seems to be necessary to preliminary experiment, which is addition or not of the attachment suitable substrate and the grain size for the target species of benthic microalgae.

  • For the purpose of drying high moisture rough rice effectively, from the view point of pre-drying, some basic experiments of fluidized bed drying of rough rice were carried out. The minimum fluidization velocities $(U_{mf})$ for both rough rice and glass bead were analyzed to find out fluidizing characteristics. The main results obtained were as follows ; 1) Minimum fluidization velocity of rough rice and glass bead were 2.01m/s and 4.07m/s, respectively, when using the distributor with $16\%$ opening ratio. 2) $U_{mf}$ calculated by Shirai's empirical equation and that calculated by Wen's modified equation were inconsistent with experimental data, while $U_{mf}$ calculated by Ergun's equation was consistent with the experimental data. 3) The following equations, on the basis of Leva's equation, were obtained. $$C_{mf}=1.19\times10^{-4}(Re_p)^{-0.0318)\;(rough rice)$$ $$C_{mf}=1.02imes10^{-4}(Re_p)^{-0.0047)\;(glass bead)$$

Optimization of Ultrasonic Soil Washing Processes Using Aluminum Foil Erosion Tests (알루미늄 호일 부식 실험을 이용한 초음파 토양 세척 공정 개발의 기초 연구)

  • The physical effect induced by acoustic cavitation was investigated to accumulate basic data for the design of ultrasonic soil washing processes using aluminum foil erosion tests. A square aluminum foil was placed on the glass beads in the pyrex vessel submerged in the sonoreactor equipped with a 36 kHz ultrasound transducer module at the bottom. Cavitational erosion of foils was quantitatively analyzed for various glass bead diameter conditions (1, 2, and 4 mm), glass bead height conditions (5, 10, 15, and 20 mm), and water height conditions (5, 10, 15, and 20 mm). It was found that aluminum foil erosion significantly increased as the glass bead diameter increased and water height over the glass bead increased due to less attenuation of ultrasound and the optimization of sound field for cavitation. Moreover mechanical mixing was suggested to move constantly particles to the bottom area where the acoustic cavitation occurs most violently. It was because aluminium foil erosion by ultrasound transmitted through glass beads was relatively too weak.

  • Background: The production of glass beads in Ghana is greatly impacted by the ingenuity of Ghanaian women. Preliminary investigations revealed the lack of interest on the part of women due to poor working conditions as a result of the influence of culture-specific silence. Therefore, the study investigated the poor working conditions faced by these industrious women with the ultimate goal of suggesting ways they can be empowered. Methods: A phenomenological study was conducted in two indigenous glass bead communities in Ghana. Data were solicited via direct observations, personal interviews and focus group discussions. Twenty-six purposively sampled respondents were recruited for the study. Data from the study were analyzed using Interpretative Phenomenological Analysis. Results: The results confirmed that the elderly women glass bead makers are much influenced by the Ghanaian culture of silence. This prevents the women from speaking about the challenges they are facing in their work. Also, the women are silenced because of the fear of losing their jobs as well as the reluctance of their male managers to remedy the challenges they encounter in the course of work. This has resulted in poor remuneration, lack of insurance packages for workers, certification, and absence of personal protective tools for the women. Conclusion: The study tasks the government of Ghana, the Legal Advocacy for Women in Africa (LAWA), the Fair Wages and Salaries Commission in Ghana, the Ghana Trade Union as well as the Local Government Workers' Union to empower the women to sustain the glass bead industry in Ghana.

The Preparation of Glass Beads (Glass beads 제조에 관하여)

  • We produced the glass beads using a spherical vessel which is rotable with the mix ture of releasing agent and glass powder in a electric furnace (apparatus 1), a tubing apparatus of electric furnace which can be gravitated a cullet (apparatus 2) and a tubing apparatus which is dispersible glass powders with the flame of propane gas (apparatus 3). The substrates which are Korean sodium silicates glass 1, 2, boro silicates glass and lead silicates glass are used and the size of cullets is 60-300 mesh. In the results of experimental apparatus, the preferable temperature of apparatus 1 is 880$^{\circ}C$, apparatus 2 is 980$^{\circ}C$ and apparatus 3 is 1100$^{\circ}C$. However, the method of apparatus 3 is more effective than the methods of apparatus 1 and 2 in view of treating time and rate of adhesion.
