Effects of Cultured Wild-Ginseng Root and Xylitol on Fermentation of Kimchi

  • This study evaluates the effects of cultured wild ginseng root (0.05%, 0.1% v/w) and xylitol in kimchi. The fermented characteristics of kimchi were investigated during 28 days of fermentation at $4^{\circ}C$. The pH value in the sample with the cultured wild ginseng root was higher than that of control group. The total acidity in the sugar groups (SG groups) was higher than that of xylitol groups (XG groups). Comparing total bacterial count, XG groups were lower than SG groups, regardless of the additional ratio of the cultured wild ginseng root. Reducing sugar of XG groups decreased more slowly than SG groups for seven days; glucose and fructose of XG groups were lower than the control group. DPPH radical scavenging activity was higher in groups with cultured wild ginseng root than in control. In the result of sensory evaluation, XG groups were more preferred than other groups. In conclusion, our results indicate that cultured wild-ginseng root and xylitol have a positive effect on the quality of kimchi, such as antimicrobial and antioxidant functions.

PLANT REGENERATION THROUGH SOMATIC EMBRYOGENESIS IN ROOT-DERIVED CALLUS OF GINSENG

  • Callus culture was initiated from explants of mature root tissues of ginseng (Panax ginseng C. A. Meyer) on MS medium enriched with 2, 4-D. The aging callus produced numerious embryoids in the same medium. Reculture of these embryoids in the media (1/2 MS or B5) supplemented with benzyladenine and gibberellic acid resulted in profuse plantlet regeneration.

인삼 모상근 유도를 위한 최적 조건 (The Optimum Conditions for Induction of Ginseng Hairy Roots)

  • 인삼 뿌리 절편으로 부터 모상근 유기를 위한 최적 조건을 확립하고자 Agrobacterium rhirogenes와 인삼 뿌리 절편의 항생제 내성 조사 및 최적의 모상근 유도 배지를 조사하기 위하여 수행하였다. NaOCl로 인삼 뿌리를 멸균하였을 때, 오염 정도가 감소하면서 조직의 손상이 일어나지 않는 NaOCl의 농도는 7% NaOCl에서 15-20분, 9% NaOCl에서 5분으로 나타났다. 인삼근은 년수가 증가할수록 오염 정도가 심하였으며, 특히 6년근중 표피가 있는 처리구는 오염 정도가 매우 높았다. Agrobacterium의 성장억제를 위한 항생제는 tetracycline이 가장 효과적이었으며, 30mg/L 이상의 농도에서 균의 성장이 억제되었다. 하지만 30mg/L tetracycline에서 인삼 조직이 고사하였으며, cefotaxime(500mg/L), carbenicillin(500mg/L)에서 균의 성장을 완전히 억제하였으며, 조직의 손상이 일어나지 않았다. 3년근 인삼에서 모상근 유도을 위한 배지로는 1/2MS 배지에 500mg/L의 cefotaxime이 첨가된 배지가 가장 좋았으며, 인삼 뿌리 절편에 Agrobacterium을 발라주는 것 보다는 균과 공동배양할때가 절편이 좋았다. Agrobacterium접종 2주 후부터 callus가 유기되기 시작한 후, 다시 2주 후에 모상근이 유도되었다. 유도된 hairy roots는 PCR에 의하여 rol C유전자를 조사함으로서 형질전환체임을 확인하였다.

Enhanced Secondary Metabolite Biosynthesis by Abiotic Elicitor in Transformed Plant Root System

  • Plants generally produce secondary metabolites in nature as a defense mechanism against pathogenic and insect attack. In this study, we applied several abiotic elicitors in order to enhance growth and ginseng saponin biosynthesis in the hairy roots of P. ginseng. Generally, elicitor treatments were found to inhibit the growth of the hairy roots, although simultaneously enhancing ginseng saponin biosynthesis. The addition of selenium at inoculum time did not significantly affect ginseng saponin biosynthesis. However, when 0.5 mM selenium was added as an elicitor after 21 days of culture, ginseng saponin content and productivity increased to about 1.31 and 1.33 times control levels, respectively. These results suggest that processing time for the generation of ginseng saponin in a hairy root culture can be reduced via the application of an elicitor.

인삼근부병의 기구와 방제에 대한 생화학적 연구 (Biochemical Study on the Mechanism and Protection of Ginseng Root-Rot Disease)

  • In this study it was found that the enzyme activities of diseased ginseng contributed greatly on the development of its root-rot disease. The 4i sensed ginseng showed higher activities of $\alpha$-amylase, p amylase, invertase, catalase, and cellulase than those of fresh one. The increased enzyme activities of diseased ginseng were originated in those from infected pathogens, which showed a proportional relationship between enzyme activities and root-rot power of them. The increases of enzyme activities during incubation of inoculated ginseng could be considerably depressed by controlling culture environments as to temperature below 4$^{\circ}C$, pH 8-9, and relative humidity about 60%, Some metal ions and organic reagents also inhibited the enzyme activities of diseased ginseng. But their inhibitory effects were not so great that they might be used to protect the disease.

Adventitious Root Development and Ginsenoside Production in Panax ginseng, Panax quinquefolium and Panax japonicum

  • This work was carried out to establish adventitious root culture system in three Panax species (wild-grown P. ginseng, P. quinquefolium, and P. japonicum) to analyze their ginsenoside productivity. Adventitious roots were induced directly from segments of seedlings after cultured on MS(Murashige andSkoog 1962) solid medium containing 3.0 mg/l IBA. Omission of $NH_4NO_3$ from the medium greatly enhanced both the frequency of adventitious root formation and number of roots per explants in all the three Panax species. However, elongation of post-induced adventitious roots was enhanced on medium with $NH_4NO_3$. Two-step culture protocol: $NH_4NO_3$-free medium for first two weeks of culture, followed by $NH_4NO_3$ containing medium for further 4 weeks, greatly enhanced the fresh weight increase of adventitious roots in all the three ginseng species. The fresh weight of adventitious roots was high in P. quinquefolium and low in P. ginseng, followed by P. japonioum regardless of the composition of medium. Pattern and content of ginsenosides in adventitious roots differed among the three Panax species. Total ginsenoside content of adventitious roots in P. quinquefolium, P. ginseng, and p. japonicum was 8.03, 15.7 and 1.2 mg/g dry weight, respectively. Among the three speices, adventitious roots in P. quinquefolium produced hig-hamount of ginsenosides. The pattern of ginsenoside fractions between P. ginseng and P. quinquefolium was similar but the amount of ginsenoside differed between the two, While, in P japonicum, total ginsenoside content was very low and some ginsenosides such as ginsenoside Rb2 and Rf were not detected. Conclusively, we demonstrate that same culture condition was required for induction and elongation of adventitious roots of three ginseng species but growth of adventitious roots and their ginsenoside production were different among them.

THE EFFECTS OF DIFFERENT SHADING OF MULCHING ON YIELD OF ROOT AND QUALITY IN PANAX GINSENG

  • This experiment was on the purpose to study the effects of different shading of Mulching treatments on the quality and yield of ginseng root. This experiment were conducted at Mei-Feng for one year, from July, 1972 to July, 1973. The variety been used was introduced Korea Panax ginsvng. Three different Shading of Mulching treatments have been studied. The results were summerized as follows: 1. The growth of ginseng plant is good under around 4,300 Lux of light intensity. Fig . showed the shadow treatment of straw had a better effect than that of black or grey plastic film. The differences between treatments were significant. 2. The adequate soil temperature for ginseng culture was in the range of $16-18^{\circ}C$. Fig 2. showed that there were significant differences among treatments, of which the straw shadow treatment had the best effect. 3. The growth of ginseng plant was greatly affected wth various shadow treatments. Fig 1. showed both straw and black plastic film treatments had a better effects on growth of stem, leaf area and leaf numbers. 4. Fig. 2. 3. 4. 5 indicated there were distingished differences among all treatments. The straw and black plastics film mulching treatments had a better effects on root length, root diameter, root weight and leaf weight than the grey plastic film. 5. The amount of plant alkaloids and panacene content had related to the shadow treatment, as showed in Fig. 6 and 7 that straw shadow treatment had greatly increased the procuction of plant alkaloids and panacene content. 6. The quality and yield of roots of ginseng greatly affected by different shading of mulching treatments.

세포배양에 의한 고려인삼 성분의 생산 연구(II) (Production of Ginseng Saponins with Cell Culture(II))

  • Tissue culture of the roots of Panax ginseng was carried out to enhance the production of ginseng callus as well as to increase its contents of ginsenosides. A long cylinder type callus mass was formed when cultured IK callus by rotary shaking culture method, the growth ratio of the callus being 7.71 which was approximately 4 fold higher than those obtained by other culture methods. Ginsenosides $Rg_1$, Re and $Rb_1$ could be detected from the callus mass by TLC, however, their total contents were revealed to be approximately 9% compared to that of the fresh ginseng root equivalent by HPLC analysis,.

Effect of Growth Conditions on Saponin Content and Ginsenoside Pattern of Panax ginseng

  • For the elucidation of significance of saponin as quality criterion of ginseng ginsenoside content(GC) and ginsenoside pattern similarity(GPS) by simple correlation were investigated in relation to red ginseng quality factors, age, plant part, harvest season, mineral nutrition, soil physical characteristics, growth light and temperature, shading material, growth location, physiological disease and crop stand through survey of ginseng plantstions, field experiments, water culture and phytotron experiments. Effect of tissue culture was also reviewed. GC was negatively correlated with good quality of red ·ginseng and positively with bad quality. Age did not show any consistency with GC but GPS was less with the increase of age difference. GPS was less or not significant between taproot that is lowest in GC and epidermis highest, and significant between leaf and taproot. Harvest season marked with the lowest GC and Pattern was also different. Nutrient imbalance, the increase of hazardous soil nutrient and physical condition to growth increased GC, but GPS was little different. The higher the growth lights intensity and temperature the higher the GC but GPS was little changed. Root rust increased GC, but root scab decreased it. Sponge-like and inside cavity phenomena increased GC. Ginsenoside pattern of cultured tissues and rootlet showed great variation. These results strongly indicate that there are optimum saponin content and ginsenoside pattern and that these are accomplished under the optimum growth condition.
