Optimal fertilizer application for Panax notoginseng and effect of soil water on root rot disease and saponin contents

  • Background: Blind and excessive application of fertilizers was found during the cultivation of Panax notoginseng in fields, as well as increase in root rot disease incidence. Methods: Both "3414" application and orthogonal test designs were performed at Shilin county, Yunnan province, China, for NPK (nitrogen, phosphorus, and potassium) and mineral fertilizers, respectively. The data were used to construct the one-, two-, and three-factor quadratic regression models. The effect of fertilizer deficiency on root yield loss was also analyzed to confirm the result predicted by these models. A pot culture experiment was performed to observe the incidence rate of root rot disease and to obtain the best range in which the highest yield of root and saponins could be realized. Results: The best application strategy for NPK fertilizer was $0kg/667m^2$, $17.01kg/667m^2$, and $56.87kg/667m^2$, respectively, which can produce the highest root yield of 1,861.90 g (dried root of 100 plants). For mineral fertilizers, calcium and magnesium fertilizers had a significant and positive effect on root yield and the content of four active saponins, respectively. The severity of root rot disease increased with the increase in soil moisture. The best range of soil moisture varied from 0.56 FC (field capacity of water) to 0.59 FC, when the highest yield of root and saponins could be realized as well as the lower incidence rate of root disease. Conclusion: These results indicate that the amount of nitrogen fertilizer used in these fields is excessive and that of potassium fertilizer is deficient. Higher soil moisture is an important factor that increases the severity of the root rot disease.

Growth and Ginsenoside Content of One Year Old Ginseng Seedlings in Hydroponic Culture over a Range of Days after Transplanting (수경재배 시 1년생 묘삼 이식 후 경과일수에 따른 인삼의 생육 및 Ginsenoside 함량)

  • Background: Ginseng produced by hydroponics can be cultivated without using agricultural chemicals; thus, it can be used as a raw materials for functional foods, medicines, and cosmetics. This study aimed to determine the optimal harvesting time to obtain the highest levels of ginsenoside and ginseng, as this was not previously unknown. Methods and Results: One-year-old organic ginseng seedlings were transplanted and cultivated using hydroponics for 150 days in a venlo-type greenhouse, using ginseng nursery bed soil and a nutrient solution ($NO_3{^-}-N$; 6.165, P; 3.525, K; 5.625, Ca; 4.365, Mg; 5.085, S; $5.31mEq/{\ell}$). Ginsenoside content and fresh and dry weights were higher at 120 days after transplanting than at 30, 60, 90, and 150 days. Total ginsenoside content was 11.86 times higher in the leaf and stem than in the root at 120 days after transplanting. Ginsenosides F1, F2, F3, and F5 were detected in ginseng leaves and stems. These chemical compounds are known to be effective in altering skin properties, including whitening, anti-inflammation, and anti-aging. Conclusions: Optimal harvesting time for ginseng cultivated using hydroponics was 120 days after transplanting when the biomass and ginsenoside content were highest.

Ginsenosides Production through in vitro Culture of Adventitous Roots Induced from Panax ginseng "Chunpoong" (인삼 천풍의 부정근 배양을 통한 Ginsenosides 생산)

  • We have investigated the optimal conditions for the growth and ginsenoside production in adventitious root of “Chunpoong” (Panax ginseng C.A. Meyer). Ginseng adventitous roots induced from the embryo of “Chunpoong” were cultured in various plant media supplemented with several growth hormones. The best growth rate and high ginsenoside contents were obtained in B5 medium supplemented with IBA (2 mg/L) and kinetin (0.1 mg/L). The supplement of 2.5 mM KH$_2$PO$_4$ was good for high growth rate of the adventitious roots, but the accumulation of ginsenosides was increased by reducing the KH$_2$PO$_4$ concentration to 1.25 mM. We have established the effective liquid culture system for the optimal growth and ginsenoside production of “Chunpoong” adventitious roots.

Effects of Petroleum Ether Extract of Ginseng Root on Some Enzyme Activity in Human Colon Cancer Cells (고려인삼중 지용성 성분이 인체암 세포의 수종 효소활성에 미치는 영향.)

  • This study was devised to observed the growth inhibition and change of disaccharidase activities of human colon cancer cells cultured in medium containing the ginseng extract. Three species of human colon cancer cell lines, HRT-18, HCT-48 and HT-29, were used for the experiment. The activities of sucrease, lactase, maltase and trehalase in the cancer cells were determined. The results obtained are summarized as follows; 1. The doubling times of the HRT-18, HT-29 and HCT-48 were about 20,22 and 24 hours, respectively. 2. The growth rates of the HRT-18 and HCT-48 in culture medium containing the ginseng extract were inhibited gradually according to increase of the concentration of ginseng extract and extension of the incubation time. 3. The activities of disaccharidase in HRT-18 and HCT-48 cultured in the medium containing the ginseng extract were increased compared with control group as follows;

THE EFFECT OF KOREAN RED GINSENG SAPONIN ON THE ALKALINE PHOSPHATASE ACTIVITY OF RAT OSTEOBLASTIC CELL(ROS17/2.8) IN CULTURE (한국 홍삼사포닌이 배양중인 쥐 조골세포의 염기성 인산분해효소 활성도에 미치는 영향)

  • Using the Korean red ginseng saponin, which is known to world-wide and thd effects of it have been investigated by many reserachers for years. Ginseng saponin, one of the major components of Korea ginseng root, has many various biologic effects, such as cytotoxic effect, tumorcidal activity, protein biosynthesis and membrane modifying effect. The purpose of this study was to evaluate effects of ginseng saponin on the alkaline phosphatase activity of ROS cells in culture. After ROS cells were seeded into a 96-well plate, 96-well plate cultured until confluence was obtained. To evaluate cytotoxic effect of total saponin in cultured ROS cells, the plates were added to each total saponin concentration (0-1mg/ml). After 48hr., cells were counted by stain with 0.2% trypan blue at randomly selected field microscopically. Also, to evaluate alkaline phosphatase(ALP) activity of total saponin in cultured ROS cell, the plate was added to each total saponin concentration (0-1mg/ml) and ALP activity was assayed. To evaluate time-course of ALP activity, $31.25{\mu}g/ml$ of saponin added to 96-well plate. After culture of 6, 12, 24 and 48hr., ALP activity test was performed. To evaluate effect of cycloheximide in ALP activity, 96-well plate was added to saponin and cycloheximide. In control group, the plate was added saponin only. The results were as follows. 1. After the various concentration of total saponin was added in the medium, 500 and $1000{\mu}g/ml$ of total saponin showed cytotoxic effect of ROS(P<0.005). 2. In contrast to control group, 7.6, 15.6, 31.25, 62.5 and $250{\mu}g/ml$ of total saponin increased ALP activity significantly. 3. Otherwise, 500 and $1000{\mu}g/ml$ of total saponin decreased ALP activity significantly(P<0.005). 4. As the time span increases, $31.25{\mu}g/ml$ of total saponin increased ALP activity. 5. Cycloheximide decreased saponin-indueced ALP actitity in ROS(P<0.005). These results suggest that Ginseng total saponin stimulates the ALP activity of rat osteoblastic cells.

  • Mountain ginseng is a perennial crop rarely found in the deep mountains of Korea. The medicinal effect of the mountain ginseng is well known as a panacea in traditional Chinese medicine for a long time. But scientific studies to elucidate the medicinal effect of the mountain ginseng have never been made on account of lack of sample. Recently an improved method of adventitious root culture system through the use of bioreactor has been developed in Panax ginseng that seems to be a reliable way of commercialization of root derived secondary metabolites. This experiment was conducted to evaluated chemotherapeutic effect against human cervical cancer cells by cisplatin (CDDP) and extract of tissue cultured mountain ginseng (ETCMG). CDDP and ETCMG-induced apoptotic cell death in human cervical cancer cell line, HeLa was confirmed by the analysis of cell growth, morphological changes, DNA fragmentation, flow cytometry showed that ETCMG is an inducer of apoptosis and synergizes with CDDP. These results suggest that ETCMG present evidence of anticancer effect and could have a possibly natural therapeutic potential in cervical cancer patients.

Antagonistic activity of Streptomyces apecies against Fusarium solani causing ginseng root rot (인삼뿌리 썩음 병균 Fusarium solane에 대한 Streptomyces species의 길한작용)

  • Antagonistic effects of Streptomyces species aganinst Fusarium solani causing ginseng root rot were investigated in terms of chitinase activity and growth inhibition in vitro. Among 131 isolates of streptomycetes obtained from ginseng cultivating soil, 9 isolates producing large clear zone around the colony on a chitin agar medium were selected for further study. All 9 isolates produced chitinase in a range from 0.10 to 0.38 U lysing cells of F. solani and inhibited germination of the conidia. In the ten-fold condentrated culture filtrate of S. alboniger ST59 and S. roseolilacinus ST129, the number of conidia of F. solane was reduced to about 20% of original count within 14 days. When S. alboniger ST59 and F. solani were grown simultaneously in the mineral saly medium, chitinase activity increased with incubation period, whereas mycelial volume of F. solani decreased. In a chitin added mineral salt medium, chitinase activity increased during the first four days and maintained steady level until the 8th day, and increased thereafter. S. alboniger ST59 lysed mycelia, conidia and even chlamydospores of F. solani. It is probable that the antagonistic activity of this streptomycete against F. solani is the lysis of fungal cell wall by streptomycete producing chitinase affected by antifungal substances.

Effects of Boron Application on the Outbreak of Granville wilt in Flue-cured Tobacco (붕소시용에 잎담배입고병발생에 미치는 영향)

  • This experiment was carried out to study the effect of boron application on the physiological response and the outbreak of Granville wilt flue-cured tobacco. NC2326 and Coker 86(flue-cured tobacco) were transplante dand investigated in all field culture plots for this experiment. Elongation of shoots were retarded, and stem heights were shortened in all boron applied plots and the more boron applied, the greater these tendency was. The ratio of root branching and root discoloration, and contraction by Granville wilt were docreased markedly in boron plots. Amount of harvested loaves (yield) was increased greatly in 2 and 4kg plots of borax and boric acid per 10a, and the increasing tendency was greater in NC2326 variety than Coker 86. Phenol compounds content in harvested leaves were higher in boron applied plots than non-applied, and was higher in Coker 86, Granville wilt resistant variety, than NC2326. The more boron applied, the higher nicotine content in the harvested leaves was.

  • Ion-pair reverse phase HPLC techniques were used to compare the contents of IAA and IAAsp in the embryogenic and non-embryogenic calli derived from ginseng (Panax ginseng C.A. Meyer) root tissues. The contents of IAA and IAAsp of the embryogenic callus were much higher (7 to 18 X respectively) than those of non-embryogenic callus. There is a distinct fluorescent peak of an unknown component in the HPLC profile of the extract for indolic compounds from non-embryo-genic callus. This distinct difference may be employed as a promising parameter to screen the culture pieces for obtaining the calli with high potential for embryoid formation.

  • Optimal regeneration conditions of ginseng transformants were studied. It has been known that Ferritin Light Heavy Chain (FLHC) gene remove the several heavy metal by combination, store and transport. To obtain the ginseng tolerant to heavy metal, binary vector was introduced in Agrobacterium by tri-parental mating and then Agrobacterium tumefaciens MP90/FLHC was selected on the AB media and MS media containing kanamycin. Explants were co-cultured with Agrobacterium tumefaciens MP90/FLHC, which contained NPT II as a selectable marker, tadpole ferritin heavy chain (FLHC) gene and human ferritin light chain gene and then a number of embryos were induced. The induced embryo transferred to shooting media consisting of MS medium supplemented with GA 10 mg/L. As a result of examination that induced the normal growth of transfomants, transformants showed the equivalent growth in both root and shoot on the media containing the 1/3 MS.
