Influence of Panax Ginseng on the Responce of Btressful Stimuli in the Experimental Animal exposed to Various Stress (인삼주정추출액이 스트레스에 폭로된 동물의 생체반응에 미치는 영향)

  • Three hundred gram of Korean ginseng root was extracted with 95% ethanol on a boiling water bath for about 300 hr. Evaporation of alcohol yieled 50.2g of dark brown residue which was used by dissolving 4 mg of the residue in 1 ml of physiological saline. The ginseng group and the saline group received each day 0.5 ml per 100 g body weight of ginseng extract and physiological saline, respectively. Both the ginseng and saline group with stress were exposed to positive radial acceleration (1∼29g), cold (5$^{\circ}C$, 0$^{\circ}C$ &-10$^{\circ}C$) and heat (35$^{\circ}C$) environment, and surgical stress. After termination of the last stress, the tolerance, body weight, visceral organ weight, basal metabolism rate, rectal temperature, the number of erythrocyte and leucocyte, hemoglobin level, hematocrit ratio, total serum protein content and it's fraction and the content of adrenal ascorbic acid in the experimental animal exposed to stress were measured and at the corresponding periods, the same measurements were also carried out with the ginseng and the saline groups without stress exposure (serving as control). Results obtained were as follows. 1. Administration of ginseng does depressed the decrease of the tolerance, body weight, visceral organ weight, basal metabolism rate, the number of erythrocyte, hemoglobin value, hematocrit ratio and the A/G ratio in the mice and rats exposed to various stress. 2. The change of the rectal temperature, eosinophile counts, total serum protein content and the content of adrenal ascorbic acid of ginseng group that exposured to various stress facilitates the reaction to, and accelerates the recovery from the stress. 3. Even after hypophysectomy which served the link between the central and the peripheral portion of the stress mechanism, the adrenal ascorbic acid content of ginseng group decreased significantly more than that of the saline group 30 min. after administration of ACTH, while the value approached the normal level significantly closer in the ginseng group than in the saline group 1 and 2 hr after ACTH administration. Judging from the above results, it is concluded that administration of ginseng extract tolerated the experimental animals under the environment of stressfu1 stmuli, although the ginseng has no significant influence upon the stress mechanism in the absence of stressful stimuli. The site of action of the ginseng appears to be in the peripheral portion of the stress mechanism.

  • this study is conducted to evalute the effect of ginseng fraction components(ginseng extract siolution; GES ginseng protein ; GP ginseng saponin ; GSA ginseng residue ; GR) upon hy-perlipidemia and fatty liver induced by high fat administration. In doing so the serum liver and epididymal adpoid tissue have been examined for lipid component level cortisol and insulin level. The change of liver tissue has been observed by light and electron microscope. In the cortisol level all experimental groups were lower compared to control group. The liver of rats observed histochemically. Control group appeared to be fatty liver but GP and GSA group looks normal electron-microscopically. GES and GP group showed a slight improvement compa-red with control group.

  • This study was carried out to evaluate the residual characteristics of azoxystrobin in fresh ginseng and calculate its processing factors in processed products, such as dried ginseng, red ginseng and their extracts. Azoxystrobin was sprayed annually onto four-year-old ginseng according to its pre-harvest interval (PHI) for two years. Harvested ginsengs were processed according to the commercially well-qualified conventional methods provided by the Korea Ginseng Corporation. Limits of detection (LODs) of azoxystrobin in fresh ginseng and its processed products were 0.001 and 0.002 mg/kg, respectively. Also limits of quantitation (LOQs) in fresh ginseng and its processed products were 0.003 and 0.007 mg/kg, respectively. Recoveries of the analytical methods in fresh ginseng and its processed products ranged from 69.3 to 114.8%. Highest residue amounts in fresh ginseng and its processed products were 0.025 and 0.118 mg/kg, respectively. Processing factors of the processed products ranged from 1.85 to 3.17 in four-year-old ginseng and from 2.48 to 5.84 five-year-old ginseng.

  • A peroxidase[E.C.1.11.1.7] is very important enzymes,e.g., as preventive antioxidants. The function is connected with growth and specialization of plant. It makes from the peroxidase and other product to save itself When a plant have been under stress of environment. A class III peroxidase cDNA was isolated from the flower bud of Panax ginseng C.A. Meyer and named PgPrx3. The PgPrx3 is an ORF(open reading frame) of 1,065 bp and a amino acid of 355 residue. Used BioEdit software to compare the PgPrx3 amino acid sequence with other plants which have already known a result of Identity was Spinacia oleracea(70%), vigna angularis(71%), Nicotiana tabacum(69%) and Linum usitatissimym(65%). The peroxidase of Vigna angularis has high homology relationship with ginseng. for that reason, the PgPx3 is a member of class III peroxidase.

  • This study is conducted to evalute the effect of ginseng fraction component (ginseng extract solution, GES; ginseng protein, GP; ginseng saponin, GSA; ginseng residue, GR) upon hyperlipidemia and fatty liver induced by high fat administration. In doing so, the serum, liver and epididymal adpoid tissue have been examined for lipid components level and lipoprotein fraction. Feces bile acid and neutral sterol excretion have been also measured. 1'he results obtained from this study are as follows. 1. Serum, liver, epididymal lipid components of GP and GSA group were significantly lower than the controlgroup. 2. During the feeding experiment, VLDL and LDL increase while HDL decrease in all group. However the degree of VLDL and LDL increase and HDL decrease were signficantly small in GP and GSA group compared with control group. 3. In the excretion of bile acid and neutral sterol, all experiment group showed increased excretion in the comparison of control group.

Residual Characteristics and Behavior of Azoxystrobin in Ginseng by Cultivation Conditions (인삼 중 azoxystrobin의 재배방법별 잔류특성 및 행적)

  • To determine residual characteristics of azoxystrobin in ginseng under different cultivation conditions such as use of straw mat on cultivation soil and filling gap between ginseng stem and soil surface and also to elucidate its approximate behavior after spraying, 20% azoxystrobin suspension concentrate solution was sprayed 4 times onto 5-year-old ginseng with 10 days interval at a application rate of about 200 L/10 a and then residues in samples were analyzed. The residue level was lower in case of use of straw mat and filling the gap with soil than in case of no use of straw mat and no filling the gap, representing that use of straw mat and filling the gap with soil were contributed to decrease of pesticide residues in ginseng. A large portion of the test pesticide distributed onto ginseng leaf with a higher specific surface area. The amounts of azoxystrobin residues decreased in ginseng leaf, while increased on soil surface, as close to harvest. About 0.1% of azoxystrobin sprayed was distributed in ginseng root and 12.7-20.4% (mean 16.6%) of azoxystrobin could be decreased for dietary intake by removing of rhizome from ginseng root before intake.

Growth promotion effect of red ginseng dietary fiber to probiotics and transcriptome analysis of Lactiplantibacillus plantarum

  • Background: Red ginseng marc, the residue of red ginseng left after water extraction, is rich in dietary fiber. Dietary fiber derived from fruits or vegetables can promote the proliferation of probiotics, and it is a key technology in the food industry to increase the productivity of probiotics by adding growth-enhancing substances such as dietary fiber. In this study, the effect of red ginseng dietary fiber (RGDF) on the growth of probiotic bacterial strains was investigated at the phenotypic and genetic levels. Methods: We performed transcriptome profiling of Lactiplantibacillus plantarum IDCC3501 in two phases of culture (logarithmic (L)-phase and stationary (S)-phase) in two culture conditions (with or without RGDF) using RNA-seq. Differentially expressed genes (DEGs) were identified and classified according to Gene Ontology terms. Results: The growth of L.plantarum IDCC3501 was enhanced in medium supplemented with RGDF up to 2%. As a result of DEG analysis, 29 genes were upregulated and 30 were downregulated in the RGDF-treated group in the L-phase. In the S-phase, 57 genes were upregulated and 126 were downregulated in the RGDF-treated group. Among the upregulated genes, 5 were upregulated only in the L-phase, 10 were upregulated only in the S-phase, and 3 were upregulated in both the L- and S-phases. Conclusions: Transcriptome analysis could be a valuable tool for elucidating the molecular mechanisms by which RGDF promotes the proliferation of L.plantarum IDCC3501. This growth-promoting effect of RGDF is important, since RGDF could be used as a prebiotic source without additional chemical or enzymatic processing.

Monitoring of Pesticide Residues and Risk Assessment for Medicinal Plants (국내 유통 약용식물 중 잔류농약 모니터링 및 위해성 평가)

  • This study was conducted to monitor residual pesticides in ginseng and balloon flower roots and to assess their risk to human health. All of 112 samples consisted of ginseng and balloon roots were purchased from traditional domestic markets and supermarkets in nine provinces of Korea in 2012. Multi-residue analysis of 122 pesticides was conducted and the analysis was performed by gas chromatography-electron capture detector, gas chromatography- nitrogen/phosphorus detector, and high-performance liquid chromatography. Seven pesticides were detected in 12 root samples and the detection rate was 10.7%. The detected twelve root samples were 10 ginseng root samples and 2 balloon root samples. Pesticides detected in root samples were procymidone, kresoxim-methyl, endosulfan, cypermethrin, tralomethrin, tetraconazole and chlorfluazuron. Among them, two pesticides as tetraconazole in a balloon flower root and cypermethrin in a ginseng root exceeded the recommended maximum residue limit set by Korea Food and Drug Administration. Five pesticides detected from 10 root samples were identified as unregistered pesticides in Korea. In order to do risk assessment with Korean medicinal plant consumption, estimated daily intake of residual pesticides were determined and compared to acceptable daily intake, referring to %ADI values. The range of %ADI values was from 0.006% to 0.333%. Taken together, it demonstrates the pesticides found in the two root samples were below the safety margin, indicating no effect on human health.

  • Ginseng (Panax ginseng) is frequently used in Asian countries as a traditional medicine. The major components of ginseng are ginsenosides. Among these, ginsenoside compound K has been reported to prevent the formation of malignancy and metastasis of cancer by blocking the formation of tumor and suppressing the invasion of cancer cells. In this study, ginsenoside $Rb_1$ was converted into compound K, via secreted ${\beta}$-glucosidase enzyme from the Leuconostoc lactis DC201 isolated, which was extracted from Kimchi. The strain DC201 was suspended and cultured in MRS broth at $37^{\circ}C$. Subsequently, the residue from the cultured broth supernatant was precipitated with EtOH and then dissolved in 20 mM sodium phosphate buffer (pH 6.0) to obtain an enzyme liquid. Meanwhile, the crude enzyme solution was mixed with ginsenoside $Rb_1$ at a ratio of 1:4 (v/v).The reaction was carried out at $30^{\circ}C$ and 190 rpm for 72 hours, and then analyzed by TLC and HPLC. The result showed that ginsenoside Rb1 was transformed into compound K after 72 hours post reaction.
