Ganoderma lucidum의 생육에 미치는 홍삼박의 영향 (Effect of ginseng residue on the growth of Ganoderma lucidum)

  • 홍삼제품 제조 과정중의 부산물인 홍삼박의 활용 가능성을 조사하기 위하여 여러 가지 담자균의 배양에 홍삼박 추출물을 첨가하여 배양한 후 균체 증식량, 단백질 및 amino acid함량 등을 조사하였으며 그 결과를 보고하는 바이다. 수집 된 담자균 중 G. lucidum과 P. ostreatus가 액체배양에서 균체증식이 양호한 균주로 선발되었으며, 홍삼박 추출물을 0.2% 첨가하였을 때 균체증식량이 가장 많았으며 단백질 함량은 0.5% 첨가하였을 때 l-3%가 증가하였다. Amino acid함량은 인삼박 추출물을 0.2% 첨가하였을 때는 거의 변화가 없었으나 0.5% 첨가하였을 때에는 G. lucidum, P. ostreatus 모두 상당히 증가하였으며 그 증가폭 중 cysteine이 가장 컸다.

인삼백 첨가가 알콜발효용균의효소생성에 미치는 영향 (Effect of Red Ginseng Residue on Various Enzyme Production of Alcohol Fermentation Koji)

  • The effect of red ginseng residue on the several enzyme activities of the koji and alcohol fermentation were investigated. The koji showed maximum values of amylase and cellulase activity when it was prepared by 30% red ginseng residue and 70% wheat bran, and of protease activity when it was prepared by 40% red ginseng residue and 60% wheat bran-${\alpha}$ amylase activity of the koji during its fermentation was increased rapidly until 4 days and there after it was increased slowly, but ${\beta}$-amylase was rapidly increased after 3 days fermentation. During the preparation of the koji, the acidic, neutral protease and cellulase activities showed the maximum value after 3 days fermentation and the alkaline protease showed the maximum value within 4-6 days fermentation. On the otherhand, fermented broth, containing 6%(v/v) alcohol, could be obtained when the substrate was saccharified by the koji, based on 25% red ginseng residue and 75% wheat bran, prior to alcohol fermentation.

효모생육에 미치는 홍삼박의 영향 (Effect of Ginseng Residue Extract on Yeast Growth)

  • 인삼의 부산물중에서 박의 활용가능성을 조사하기 위하여 박의 구성성분, ext의 DPPH 환원력, 효모생육에 미치는 영향 및 효모균체내의 amino acid함량등에 미치는 영향등을 조사한 결과를 요약하면 다음과 같다. 박내에는 총당이 43~46%, 조단백이 14~15%, n-BuOH ext함량은 0.18% 였으며 alcohol박의 물ext는 alcohol ext에 비해 약 46%정도의 DPPH환원력이 남아있었으며 박ext를 첨가하고 효모를 배양시켰을 때 효모의 생육에 필요한 영양물질이나 생육을 촉진하는 물질이 박ext내에 존재하고 있다는 것을 알았으며 알콜박의 물추출물을 첨가하여 배양한 효모균체의 amino acid함량이 다른 ext를 첨가한 효모균체보다 많았다. 박에 cellulase를 0.1%, 20분간 처리로 박ext의 수율을 250%정도 증가시킬 수 있었다.

홍미삼 알콜 추출박을 이용한 산성다당체 다량 함유 홍삼 엑기스 제조 (Preparation of Red Ginseng Extract Rich in Acidic Polysaccharide from Red Tail Ginseng Marc Produced After Extraction with 70% Ethyl Alcohol)

  • In this study, we investigated the appropriate conditions to extract acidic polysaccharide and to prepare red ginseng extract being rich in acidic polysaccharide from red tail ginseng marc produced after manufacturing alcoholic extract from red tail ginseng. Amount of acidic polysaccharide in red tail ginseng marc was about 11%. The best condition for the extraction of acidic polysaccharide from the marc was using of 3~5 mg of $\alpha$-amylase/g residue/15 ml of distilled water, and the amount of acidic polysaccharide in water extract of the residue treated with $\alpha$-amylase was about 27%. So, it is possible to manufacture red ginseng extract being rich in acidic polysaccharide using water extract of red tail ginseng alcoholic residue as extraction solvent. From the above results, we suggest that red tail ginseng residue produced by manufacturing alcoholic extract of red tail ginseng has high potencies in the utilization of waste material.

홍삼박 볶음처리 추출액이 알콜해독에 미치는 효과 (Effect of Ginseng Extract Residue Roasted on Alcohol Detoxification)

  • Alcohol and acetaldehyde concentrations were measured in the blood and brain of rats which were treated with 20% alcohol (control group) or co-administered 20% alcohol with ginseng extract residue roasted (test group). There was no change in blood alcohol concentration between control and test group. However, the brain alcohol concentration was lowered in the test group which was treated for seven days. The concentration of aldehyde in the brain and blood was lowered in the test group. The activities of monoamine oxidase b in various regions of brain were recovered to normal group in the test groups. However, the Quantities of naloxone binding receptors were not changed by ginseng extract residue roasted.

  • Korean red ginseng and water extract residue of red ginseng roots were treated with dry heat and incorporated in PDA medium to examine the effect of the materials on induced tolerance against mercury chloride and mycelial growth of Fusarium oxysporum. Ginseng residue was not effective in the inducement of tolerance to mercury chloride regardless of dry heat treatment. However, the heat treatment of ginseng and ginseng residues stimulated the mycelial growth of the fungus. The materials responsible for the detoxification appeared to be water-soluble. The stimulation of the fungal mycelial growth on the media by the heat treatment was highest in the water extract of ginseng. Due to the heat treatment, the mycelial growth was also slightly increased in n-hexane and methanol extracts of ginseng, compared with the ginseng fractions without dry heat treatment.

안전사용기준에 따라 살포된 살균제 Tolclofos-methyl, Azoxystrobin 및 Difenoconazole의 인삼 중 잔류량 (Residues of Tolclofos-methyl, Azoxystrobin and Difenoconazole in Ginseng Sprayed by Safe Use Guideline)

  • The aim of this work is to assess the safety of fungicide tolclofos-methyl, difenoconazole and azoxystrobin in ginseng sprayed by safe use guideline. When tolclofos-methyl was sprayed on ginseng by safe use guideline, the residue amounts (MRL) of it in ginseng was 0.13 mg/kg which is below than 0.3 mg/kg, maximum residue limit established by Korea Food & Drug Administration (KFDA). The residue amounts of ginseng parts, head part was 0.37 mg/kg and main body part was 0.13 mg/kg. In case of difenoconazole, the residue amounts in ginseng was 0.81 mg/kg. which was exceed the MRL, 0.2 mg/kg. By the analyze results of ginseng part, the residues of head and main body part were 3.01 and 0.40 mg/kg, respectively. In experiment of vinyl mulching, the residue amount of difenoconazole in ginseng was 0.05 mg/kg. The residue amounts of azoxystrobin in ginseng sprayed by safe use guideline was 0.14 mg/kg. This residue was not exceed the MRL 0.5 mg/kg. The residue amounts by ginseng parts was 0.51 mg/kg for head part and 0.28 mg/kg for main body part. In case of vinyl mulching, the residue amount of azoxystrobin was 0.02 mg/kg.

제초제 Quinclorac이 인삼생육에 미치는 영향 (Effect of Herbicide Quinclorac on Ginseng Growth)

  • In this experiment, germination test of ginseng seed and growth test of two year old ginseng in soil incorporated with quinclorac (3,7-Dichloro-8-quinoline carboxylic acid) were conducted to evaluate phytotoxicity caused by quinclorac residue in soil. Ginseng seed germiilation was hardly affected, but growth of ginseng seedling was retarded by quinclorac. The phytotoxicity of quinclorac on ginseng seedling was expressed at the lowest level tested, 3.75 g a.i./10a (118 of recommended rate for rice cultivation). The representative phytotoxicity in ginseng were decreased root weight, shortened leaf width and leaf length, and weak lateral root Quinclorac residue decreased by 1/4 in pot soil after 2 months and below 2 ppm in the paddy soil applied at usual rate (30 g a.i./10a) after 1∼ 2 years. Below 2 ppd ginseng did not show any phytotoxicity symptom.

살충제 Cypermethrin 및 살균제 Diethofencarb의 살포방법에 따른 인삼 중 잔류 (Residues Amounts of Cypermethrin and Diethofencarb in Ginseng Sprayed by Safe Use Guideline)

  • This study is conducted in order to know the residue patterns of insecticide cypermethrin and fungicide diethofencarb in ginseng sprayed by various application methods. Two pesticides were sprayed separately on ginseng using traditional, soil and vinyl mulching applications. The vinyl mulching application was that head part of ginseng protected from pesticides. When cypermethrin was sprayed on ginseng by traditional application, its residue amount in ginseng was 0.25 mg/kg which exceeded 0.1 mg/kg, maximum residue limit(MRL) established by Korea Food & Drug Administration(KFDA). But in case of vinyl mulching and soil application, its residue amounts were 0.04 and 0.07 mg/kg, respectively. The residue amount of diethofencarb in ginseng was 3.01 mg/kg which exceeded the MRL, 0.3 mg/kg. Further, in case of vinyl mulching and soil application, its residue amounts were 1.71 and 9.39 mg/kg which exceeded the MRL 0.3 mg/kg. Although the residue amounts of both pesticides exceeded the MRLs for ginseng, we can explain that pesticides remained in head part of ginseng is probably the reason why higher pesticides residue levels were observed.

Production of Hydrolyzed Red Ginseng Residue and Its Application to Lactic Acid Bacteria Cultivation

  • Enzymatic treatment conditions for red ginseng residue (RGR) were investigated to apply RGR as a microbial medium. Polysaccharide hydrolyase and protease were screened to obtain high solid and carbohydrate yields, and a good degree of carbohydrate hydrolysis. The optimal dosage and reaction time for Viscozyme, the chosen polysaccharide hydrolyase, were found to be 1.0% (w/w) and 3 h, respectively. Of the tested proteases, Flavourzyme, whose optimal dosage was 0.5% (w/w), was selected. Co-treatment with the optimal dosages of Flavourzyme and Viscozyme increased solid yield, carbohydrate yield, and degree of carbohydrate hydrolysis by 76%, 65%, and 1,865%, respectively, over levels in non-treated RGR. The culture characteristics of Leuconostoc mesenteroides strain KACC 91459P grown in enzymatically hydrolyzed red ginseng residue (ERGR) and RGR suspensions were compared. After cultivation for 6 h, the viable cell counts of both cell suspensions rapidly increased to $1.3{\times}10^9$ colony-forming units (CFU)/g. Moreover, while the viable cell population drastically decreased to $2.4{\times}10^6\;CFU/g$ for cells grown in RGR medium, it was maintained in cells fermented in ERGR medium for 24 h.
