비육돈에 있어 황기, 인삼, 양파 혼합물의 급여가 성장 밑 도체 특성에 미치는 영향 (Effects of Dietary Astragalus membranaceus, Ginseng and Onion Complex on Growth Performance and Carcass Characteristics in Finishing Pigs)

  • 본 연구의 목적은 생약제 및 양파 혼합물의 급여가 비육돈의 성장 및 도체 특성에 미치는 영향을 평가하기 위하여 실시하였다. 3원 교잡종(Durox$\times$Yorkshire$\times$Landrace) 비육돈 72두을 공시하였으며 시험개시시의 체중은 체중은 75.66$\pm$1.86 kg 이었다. 처리구로는 옥수수-대두박 위주의 식이에 NRC(12)의 영양소 요구량에 따라 처리한 구(Control: 기초식이), 대조구 식이내 생약제 및 양파 혼합물을 0.25% 첨가한 구(AGO 0.25: 기초식이+0.25% 생약제 및 양파 혼합물), 대조구 식이내 생약제 및 양파 혼합물을 0.50% 첨가한 구(AGO 0.50: 기초식이 + 0.50% 생약제 및 양파 혼합물)로 3개 처리를 하여 처리당 4반복, 반복당 6마리씩 완전임의 배치하였다. 본 사양시험에 사용한 생약제 및 양파 혼합물은 인삼, 황기 그리고 양파 분말을 함유한 것을 사용하였다. 전체 시험기간 동안, 생약제 및 양파 혼합물의 첨가수준이 증가함에 따라 식이효율 및 등지방 두께가 증가하는 경향을 나타내었으나 유의적인 차이는 보이지 않았다(p>0.05). 도체등급에 있어서는 생약제 및 양파 혼합물의 첨가수준이 증가함에 따라 A등급 출현율이 높은 것으로 평가되었다(linear effect, p<0.01). 대조구 및 AGO 0.50 처리구와 비교하여 AGO 0.25 처리구가 혈청내 총 콜레스테롤 및 LDL+VLDL 콜레스테롤 농도가 낮은 것으로 평가되었으나 유의적인 차이는 보이지 않았다(p>0.05). 도축한 돈육의 등심부의 육색에 있어서 L-$^{*}$ , a-$^{*}$ , b-$^{*}$ 값에 있어서도 유의적인 차이를 보이지 않았다(p>0.05). 생약제 및 양파 혼합물 급여에 따른 증체량 개선 효과로 인해 증체당 식이비는 대조구가 830원, AGO 0.50 처리구가 813원으로 대조구와 비교하여 2.05%의 식이비 절감 효과를 나타내었다. 결론적으로 비육돈 식이내 생약제 및 양파 혼합물의 첨가가 성장 촉진을 갖는 것으로 사료되나 도체특성에는 영향을 미치지 않는 것으로 판단된다.

내백 수삼 전분의 특성 (The Physicochemical Properties of Starch from Inside White Fresh Ginseng)

  • Among many kinds of constituents of ginseng root, starch was the most noticeable component related to inside white in fresh ginseng. The ash of inside white section was higher in content and greenish gray in color. The water binding capacity and ash content of the starch from inside white fresh ginseng were higher than those of starch from normal fresh ginseng, but amylase content was not significantly different. Ash of inside white fresh ginseng starch contained higher in Ca, Mn and Fe content. Complexes of small granular starch and amino acids were rich in swelling power, and solubility by temperature change were similar between inside white fresh ginseng starch and normal one, but the former was more easily swelled than the latter.

화장품소재로서의 인삼복합물의 항염증 및 미백효능 검증 (Anti-inflammatory and whitening effects of ginseng complex as a cosmetic material)

  • 본 연구는 화장품 소재로서 인삼복합물의 가능성을 확인하기 위한 것이다. 이를 위해 우리는 인삼복합물 에탄올 추출물을 사용하여 항염증, 미백 효과에 대한 생물학적 활성 평가를 수행하였다. 시료는 인삼(A), 상황(B), 리기다소나무(C)를 각 70% 에탄올로 추출한 후 (A)1 : (B)1 : (C)0.5의 비율로 혼합한 시료를 사용하였다. 대식세포(RAW 264.7 cells)내에서 시료의 항염증 효과를 평가하기 위해 MTT assay를 이용한 샘플의 독성평가와 nitric oxide 생성 저해 활성 및 염증관련 단백질 및 유전자의 발현량을 확인하였다. 시료의 미백 효과를 평가하기 위해서 멜라노마(B16F10 cell)를 이용해 MTT assay를 통한 샘플의 독성평가, cellular tyrosinase 저해율 측정 및 멜라닌 생합성량을 측정하였다. LPS로 유도된 RAW 264.7 세포내에서 시료의 nitric oxide 저해 활성 결과는 $25{\mu}g/mL$ 농도에서 71.2%의 우수한 효능을 나타내었으며 western blot 실험결과 iNOS, COX-2 단백질의 발현은 농도 의존적으로 감소하는 것을 확인하였다. Tyrosinase 활성 억제 실험에서 인삼복합물의 농도 $50{\mu}g/mL$에서 36.8%의 감소를 나타냈으며 멜라닌 생합성 억제 평가 결과에서 농도 $50{\mu}g/mL$에서 47.8%의 감소를 나타냈다. 상기 실험 결과로부터 인삼복합물의 우수한 항염증, 미백 효능을 확인하였으며 향후 안전한 천연 화장품 원료로 사용될 수 있음을 확인하였다.

알시안블루 색소를 이용한 인삼다당체의 함량 분석 (Analysis of Panax ginseng Polysaccharide by Alcian Blue Dye)

  • Polysaccharide contents in Panax ginseng roots were evaluated by a spectrophotometry, utilizing the complex formation of ginseng polysaccharide with alcian blue dye in 50 mM ammonium biphos-phate, pH 4.2. The polysaccharide content in red ginseng was about three times higher than that in fresh ginseng when both were extracted with water, and increased about two times when red ginseng was extracted with an alkaline solution. The determination of polysaccharide in various parts of ginseng revealed that main roots contained the component more than fine roots. Fresh ginseng sections stained by the dye showed polysaccharide mainly was found in cortex and combium but not in epidermis.

Taxonomy of fungal complex causing red-skin root of Panax ginseng in China

  • Background: Red-skin root of Asian ginseng (Panax ginseng) significantly reduces the quality and limits the production of ginseng in China. The disease has long been thought to be a noninfectious physiological disease, except one report that proved it was an infectious disease. However, the causal agents have not been successfully determined. In the present study, we were to reveal the pathogens that cause red-skin disease. Methods: Ginseng roots with red-skin root symptoms were collected from commercial fields in Northeast China. Fungi were isolated from the lesion and identified based on morphological characters along with multilocus sequence analyses on internal transcription spacer, β-tubulin (tub2), histone H3 (his3), and translation elongation factor 1α (tef-1α). Pathogens were confirmed by inoculating the isolates in ginseng roots. Results: A total of 230 isolates were obtained from 209 disease samples. These isolates were classified into 12 species, including Dactylonectria sp., D. hordeicola, Fusarium acuminatum, F. avenaceum, F. solani, F. torulosum, Ilyonectria mors-panacis, I. robusta, Rhexocercosporidium panacis, and three novel species I. changbaiensis, I. communis, and I. qitaiheensis. Among them, I. communis, I. robusta, and F. solani had the highest isolation frequencies, being 36.1%, 20.9%, and 23.9%, respectively. All these species isolated were pathogenic to ginseng roots and caused red-skin root disease under appropriate condition. Conclusion: Fungal complex is the causal agent of red-skin root in P. ginseng.

Safety of red ginseng and herb extract complex (RHC) in menopausal women: A randomized, double-blind, placebo-controlled trial

  • Background: Various treatments are used to relieve menopausal symptoms for women. However, herbal substances are frequently used as complementary and alternative therapies as other treatments can increase ovarian and breast cancer risk. While the herbal substances' therapeutic effect is essential, the safety of their use is considered more important. This study aims to confirm the safety of red ginseng and herb extract complex (RHC), which are used to relieve menopausal symptoms. Methods: This randomized, double-blind, placebo-controlled clinical study recruited and divided 120 women experiencing menopausal symptoms into the RHC and placebo groups (60 women per group). Subjects were administered with 2 g RHC or placebo daily for 12 wk. Adverse reactions, female hormonal changes, and uterine thickness were observed and recorded on wk 0, 6, and 12. Hematologic and blood chemistry tests were also conducted. Results: The reactions of the subjects who received RHC or placebo at least once were analyzed. A total of six adverse reactions occurred in the RHC group, while nine occurred in the placebo group; common reactions observed in both groups were genital, subcutaneous tissue, and vascular disorders. However, there was no statistically significant difference between the administration groups (p = 0.5695), and no severe adverse reactions occurred in both groups.

Comparative analysis of the transcriptomes and primary metabolite profiles of adventitious roots of five Panax ginseng cultivars

  • Background: Various Panax ginseng cultivars exhibit a range of diversity for morphological and physiological traits. However, there are few studies on diversity of metabolic profiles and genetic background to understand the complex metabolic pathway in ginseng. Methods: To understand the complex metabolic pathway and related genes in ginseng, we tried to conduct integrated analysis of primary metabolite profiles and related gene expression using five ginseng cultivars showing different morphology. We investigated primary metabolite profiles via gas chromatography-mass spectrometry (GC-MS) and analyzed transcriptomes by Illumina sequencing using adventitious roots grown under the same conditions to elucidate the differences in metabolism underlying such genetic diversity. Results: GC-MS analysis revealed that primary metabolite profiling allowed us to classify the five cultivars into three independent groups and the grouping was also explained by eight major primary metabolites as biomarkers. We selected three cultivars (Chunpoong, Cheongsun, and Sunhyang) to represent each group and analyzed their transcriptomes. We inspected 100 unigenes involved in seven primary metabolite biosynthesis pathways and found that 21 unigenes encoding 15 enzymes were differentially expressed among the three cultivars. Integrated analysis of transcriptomes and metabolomes revealed that the ginseng cultivars differ in primary metabolites as well as in the putative genes involved in the complex process of primary metabolic pathways. Conclusion: Our data derived from this integrated analysis provide insights into the underlying complexity of genes and metabolites that co-regulate flux through these pathways in ginseng.

  • The developmental processes of the protein body are studied on endosperm cells of Panax ginseng during seed maturation periods. The spherosome, mitochondria, rough endoplasmic reticulum, and ribosome are observed and then are gradually increased in early endosperm cells. Protein body developed from vesicles produced by the rough endoplasmic reticulum and was formed at the enlarged ends of rough endoplasmic reticulum. Also, vacuole-like protein body was observed in associated with rough endoplasmic reticulum. Golgi complex is observed in associated with vacuole and its vesicles containing proteinaceous granules moved and accumulated to the vacuole. Proteinaceous granules are deposited in the spherical or oval shaped vacuole and gradually, vacuole is surrounded by the multi-membranous structure. Rough endoplasmic reticulum, ribosome, Golgi complex, and vacuole are observed in associated with protein body formation.

작약에서 분리한 Cylindrocarpon destructans의 인삼에 대한 병원성 검정 및 분류학적 고찰 (Virulence Assays and Genetic Reclassification to Assess the Pathogenicity of Cylindrocarpon destructans Isolated from Peony in Ginseng)

  • 작약에서 분리된 Cylindrocarpon destructans KACC44663 균주의 인삼에 대한 병원성 여부를 밝혀내고자 병원성 검정을 실시하였고 새로운 분류체계에 따른 균의 재분류를 위해 계통학적 유연관계를 분석하였다. 작약으로부터 분리된 KACC44663 균주가 인삼에 병을 일으키는 것을 확인하였고, 새로운 분류체계에 의해 I. radicicola species complex 내 위치하는 Ilyonectria robusta로 동정하였다. 이는 국내에서 타 기주로부터 분리된 병원균이 인삼에 뿌리썩음병을 일으키는 것을 증명한 첫 사례이다.
