제주도 자생 참억새의 종자발아에 관한 연구 (Studies on Seed Germination of Miscanthus sinensis Native to Jeju Island)

  • The study was conducted to find out the harvesting time, storage duration, cold treatment and sterilization on seed germination of Miscanthus sinensis. The optimum harvesting time of the Miscanthus seed in native to Jeju island was December. And the seed germination passible 3 years after harvesting. When the seeds had cold treatment, seed germination nate was 74% rather than 63% of non-treatment. For seed sterilization treatment, soaking in solution of Benomyl 7% during from 3 hours to 12 hours germinated 9% over. But the non-treatment on 24 hours treatment reduced the seed germination of Miscanthus sinensis.

  • Germination and vegetative growth of Enteromorpha multiramosa Bliding from Pyoson, Cheju Island were investigated in laboratory under various combinations of temperature (5-$25^{\circ}C$) and salinity (8-48$^{\circ}C$). Percent level of germination was relatively high at all combinations of the two factors. The highest value among the combinations was revealed at 15$^{\circ}C$ and 32$\textperthousand$. Dry weight also was fairly high at all levels of combination with maximum value at 2$0^{\circ}C$ and 32$\textperthousand$. Analysis of variance for germination and growth was completed respectively and polynomial prediction models were constructed. F ratio revealed that all factors had a significant effect (p<0.001) on percentage of germination and dry weight, and their interactions also were significant (p<0.001), although the F ratio of interactions was far less than that for either the separate effect of temperature or salinity. Response surface of polynomial equation represented that temperature influenced less than salinity on germination, while it effected remarkably on vegetative growth, so the Enteromorpha multiramosa was kept to visible macrothalli from winter to spring in Cheju Island.

  • The study was conducted to determine the optimal basal media and the concentration of plant growth regulator for germination of seeds and growth of plantlet from Dendrobium monile. The results obtained were summarized as follows. Germination was similar in light and dark condition, but the growth of plantlet after germination was better under dark than under light condition in several media. Germination was best in Hyponex and Kyoto solution medium among the 9 media tested. The number of roots/shoot was most in the Hyponex medium containing 0,1ppm NAA and 1.Oppm BA.

  • Various presowing seed treatments have been used to enhance the rates of germination and afterward seedling emergence. Seeds treated by them occasionally have not shown the same rate in indoor and field. The presowing treatments considering germination mechanism and factors affecting germination should be totally included in indoor experiments so that the results drawn could be reproduced in the field. Seed drying after the treatments and field conditions after sowing especially able to show Phytochromemediated responses related to light treatment must be determined prior to force the treatments to seeds. After set up these prerequisites to promote the rates of germination and emergence, many seed treatments proposed so far must be fixed how to be done; alone, sequential and simultaneous.

  • Influence of temperature, relative humidity, spore washing and spore drying on conidial germination of Alternaria porri(Ell.) Cif. was studied. Maximum conidial germination occurred at 100% relative humidity prevailing for 6 hours or more at $25^{\circ}C$. Conidial germination decreased with increase in number of spore washings. Drying of conidia for more than half an hour caused significant decrease in germination. In all the experiments, conidial germinatio increased with increase in incubation period.

The Effect of Salinity (NaCl) on the Germination and Seedling of Sugar Beet (Beta vulgaris L.) and Cabbage (Brassica oleracea L.)

  • This study was carried out to investigate seed germination and seedling of cabbage and sugar beet in four treatments of salinity including 0 (control), 0.5, 1.0 and 1.5% NaCl. The results showed that different treatments of salinity had considerable effects on the germination and root and shoot length of cabbage and sugar beet. Percent of germination in both species showed significant decrease with increasing salinity up to 1.5% NaCl. This decrease was more evident in cabbage when compared to sugar beet. The required time for germination increased with high levels of salinity. The seedling growth of both species were inhibited by all salinity levels. Particularly at 1.0 and 1.5% NaCl, no measurable length was observed in cabbage and sugar beet. At 0.5% NaCl root growth of both plant species was more affected as compared to shoot growth by salinity.

  • Antifungal activity of 2-hydroxy 4,4'6'trimethoxy chalcone individually was tested against spore germination of ten fungi of different genera. Efficacy of the chemical was also tested against conidial germination and other growth parameters of Erysiphe pisi on excised pea leaves. 2-Hydroxy 4,4'6'trimethoxy chalcone inhibited spore germination at all the concentrations. Maximum inhibition was observed at 2000 ppm where more than 78 per cent inhibition of spore germination was observed in Ustilago cynodontis, Alternaria brassicicola, A. solani and Aspergillus flavus. It also reduced conidial germination of E. pisi significantly, when applied as pre-inoculation treatment.

  • It is generally known that electron beam has sterilization effects and can activate plant germination and growth. Compared to electron beam, electrical pulse has not been frequently studied with respect to the biological application. In this study, we have analyzed the effects of high voltage pulse on seed germination and growth using various plant species. We have used the high voltage generator for examining seed's responses to the high voltage pulse. The operating voltage and currents of the generator are about 300 kV and 30 kA, respectively. Pulse width is 60 ns. High voltage pulse has slightly activated germination and growth of radish during early stage. Various levels of germination and growth are observed in different plant species after treated with high voltage pulse.

  • This study was carried out to investigate the effect of sucrose, boric acid, and calcium nitrate in optimum pH and relative humidity on pollen germination and pollen tube growth of boxthorn (Lycium chinense M.). Medium containing 100mg/L of $H_3BO_3$ showed maximum pollen germination and pollen tube elongation, 64.8% and $920{\mu}m$. Additionally supplied calcium nitrate to the medium lowered pollen germination rate and pollen tube elongation. Here we report the optimum conditions for the pollen germination and pollen tube development was observed at 10% sucrose, 1.0% agar, pH 6.0, and 100% relative humidity.

  • Germination of Italian ryegrass and sorghum were measured at a range of temperatures and with PEG treatment at several different levels of concentration and treatment period to evaluate the effects of osmoconditioning with polyethylene glycol(PEG-8000). The results obtained are summarized as follows: 1. Cumulative germination percentage of intact Italian ryegrass seeds did not show much difference among temperatures, while that of sorghum seeds decreased markedly at low temperature. 2. Italian ryegrass seeds osmoconditioned with 20% PEG solution for two days at 10^{\circ}C.$ accelerated germination at 10^{\circ}C.$, with injurious effect of osmoconditioning at higher temperature for many days. 3. Osmoconditioning with 20% PEG solution for two or four days at 10^{\circ}C.$ increased germination of sorghum seeds, while the seeds treated at higher temperatures for long period suppressed germination.
