The purpose of this study was to introduce the differential standard of the types of Angelicae Pubescentis Radix. We established the classificatory standard according to the external and internal morphology and the physicochemistrical pattern for the four types of Aucklandiae Radix. The results follow: 1. At the external shape, Angelica pubescens f. biserrata has rising lenticel and dark brown surface, Its section is light gray and its xylem is yellow gray. Aralia continentalis has big stem mark in upper portion, its section has no dense tissue with yellow gray. Heracleum hemsleyanum has dark brown surface and little lenticel, its cortical layer is white yellow and xylem is isabella and powdered. 2. At the internal shape, Angelica pubescens f. biserrata has phloem with half of the root and rare vessel. Aralia continentalis has phloem with two-thirds of the root and it has broad cutting and putting vessel that formed 1-2 row. Heracleum hemsleyanum also has phloem with half of the root and wide scattered latex tube and many large vessel. 3. At the TLC pattern, Heracleum hemsleyanum has remarkable dark spot at $R_{f}$ 0.23 on the sulphuric acid color pattern test, but others have faint. 4. At the HPLC pattern, all samples have generally patterns. But Angelica pubescens f. biserrata shows diminutive continentalic acid content and the peak at Rt 20.278min comes out on Heracleum hemsleyanum, but do not come out on Aralia continentalis and Angelica pubescens f. biserrata. Heracleum hemsleyanum has remarkable peak at Rt 20.278min, but shows no peak at Rt 29.023min unlike Aralia continentalis or Angelica pubescens f. biserrata. Also Aralia continentalis and Angelica pubescens f. biserrata show one remarkable peak at Rt 29.023, but Heracleum hemsleyanum do not show. Consequently, Aralia continentalis and Angelica pubescens f. biserrata are comparable whit continentalic acid content and Heracleum hemsleyanum is comparable with the peak at Rt 20.273 and Rt 29.023. So it is thought that content of continentalic acid and the peaks at Rt 20.278 and Rt 29.023 can apply to differentiate a species from other. It is considered the results of this study will be furnished the basis to succeeding studies and it is needed to extensive comparative study for the same genus-degree of relatedness.
To examine the natural regeneration in the natural mixed-broadlived forest, flowering, fruiting, seed-fall, and seed viabilities of Acer ukurunduense Trautv. & C.A. Mey. (AU) were investigated in Mt. Jungwang, Gangwon-do, from 2009 to 2013. The flower of AU consisited many male and bisexual flowers on the raceme (unusual panicle) and the arrangement of two sex morphs are differed for each inflorescence. Flowering dates are differed between sex morph in the same inflorescence. Stamens are stop growing and disappeared after pollinated pistil begin to grow in bisexual flowers, and male flowers have vestial pistil. The flowers of AU might be pollinated by Apis mellifera, Thyris fenestrella seoulensis, Cerambycidae sp., Andrenidae sp. and Ctenophora sp., and had some mechanism to prevent feom self pollination. The number of flower buds per inflorescence is 189 on June 8, and that of young samaras per inflorescence is 41.2 on June 21. At last ripened samaras per inflorescence is reduced 33.4 on September 5. Mean annual seedfall of AU was 6,720 ea/ha (ranged 670~17,930). Rates of sound seeds are highest 43.2%, and those of damaged or decayed seeds are 41.8%,. Those of undeveloped and empty seeds are 10.2% and 4.8%, respectively. Successful regeneration of AU might be in masting year and on the gap sites with proper conditions to germinate and grow. To understand the natural regeneration of the species, Genus Acer, further study on the fruiting habit, pre- and post-dispersal seed viability, and annual variation on these factor should be needed.

Bacteria play an important role in biogeochemical cycles in marine environments and their functional attributes in ecosystems depend primarily on species composition. In this study, seasonal variation of bacterial diversity was investigated by pyrosequencing of 16S rDNA in surface sediment and overlying seawater collected in the south East Sea, planned for the site of $CO_2$ sequestration by the carbon capture and storage (CCS) project. Gammaproteobacteria was dominant in the sediment in most seasons, whereas Alphaproteobacteria was a most dominant group in the overlying water. Thus, the bacterial diversity greatly differ between sediment and seawater samples. On the genus level, bacterial diversity between two habitats was also different. However, the number of genera found over 5% were less than 10 in both habitats and the bacterial community was composed of a number of diverse minor or rare genera. Elevation of $CO_2$ concentration during a $CO_2$ storage process, could result in change of bacterial diversity. Thus, this study will be very useful to access the effect of $CO_2$ on bacterial diversity and to predict functional change of the ecosystem during the process of CCS project.

Bacterial endophytes may be important factors in plant growth and ecologically relevant functions in rice. Using pyrosequencing technology, we analyzed the composition of endophytic bacterial communities that colonized the roots of rice cultivated in long-term fertilized (APK) and non-fertilized (NF) paddy soils. A total of 1,900 reads were obtained from 2 samples. All sequences were classified into 177 OTUs (APK sample) or 72 OTUs (NF sample) at a 97% similarity cut-off. Twenty-two OTUs were shared between the 2 samples, and these were also the most dominant OTUs in both samples. Proteobacteria was the most dominant phylum with 90.2%, followed by Actinobacteria (7.1%) and Bacteroidetes (1.1%). Furthermore, Pseudomonas was the most abundant genus in both samples. We observed clear differences in the structure of the endophytic bacterial community structure between the 2 samples. Notably, the distributions of Alphaproteobacteria and Gammaproteobacteria were markedly different. The diversity index of the APK sample was higher than that of the NF sample. These findings showed that the endophytic bacterial community of rice roots was affected by the presence of fertilizers in the rice field soil.

The gut microbiome influences the health and well-being of dogs. However, little is known about the impact of breed on the fecal microbiome composition in dogs. Therefore, we aimed to investigate the differences in the fecal microbiome in three breeds of dog fed and housed under the same conditions, namely eight Maltese (8.0 ± 0.1 years), eight Miniature Schnauzer (8.0 ± 0.0 years), and nine Poodle dogs (8.0 ± 0.0 years). Fresh fecal samples were collected from the dogs and used to extract metagenomic DNA. The composition of the fecal microbiome was evaluated by 16S rRNA gene amplicon sequencing on the MiSeq platform. A total of 840,501 sequences were obtained from the 25 fecal samples and classified as Firmicutes (32.3-97.3% of the total sequences), Bacteroidetes (0.1-62.6%), Actinobacteria (0.2-14.7%), Fusobacteria (0.0-5.7%), and Proteobacteria (0.0-5.1%). The relative abundance of Firmicutes was significantly lower in the Maltese dog breed than that in the other two breeds, while that of Fusobacteria was significantly higher in the Maltese than in the Miniature Schnauzer breed. At the genus level, the relative abundance of Streptococcus, Fusobacterium, Turicibacter, Succinivibrio, and Anaerobiospirillum differed significantly among the three dog breeds. These genera had no correlation with age, diet, sex, body weight, vaccination history, or parasite protection history. Within a breed, some of these genera had a correlation with at least one blood chemistry value. This study indicates that the composition of the fecal microbiome in dogs is affected by breed.

N-acyl-homoserine lactone (AHL)-mediated quorum sensing (QS) plays a major role in development of biofilms, which contribute to rise in infections and biofouling in water-related industries. Interference in QS, called quorum quenching (QQ), has recieved a lot of attention in recent years. Rhodococcus spp. are known to have prominent quorum quenching activity and in previous reports it was suggested that this genus possesses multiple QQ enzymes, but only one gene, qsdA, which encodes an AHL-lactonase belonging to phosphotriesterase family, has been identified. Therefore, we conducted a whole genome sequencing and analysis of Rhodococcus sp. BH4 isolated from a wastewater treatment plant. The sequencing revealed another gene encoding a QQ enzyme (named jydB) that exhibited a high AHL degrading activity. This QQ enzyme had a 46% amino acid sequence similarity with the AHL-lactonase (AidH) of Ochrobactrum sp. T63. HPLC analysis and AHL restoration experiments by acidification revealed that the jydB gene encodes an AHL-lactonase which shares the known characteristics of the α/β hydrolase family. Purified recombinant JydB demonstrated a high hydrolytic activity against various AHLs. Kinetic analysis of JydB revealed a high catalytic efficiency (kcat/KM) against C4-HSL and 3-oxo-C6 HSL, ranging from 1.88 x 106 to 1.45 x 106 M-1 s-1, with distinctly low KM values (0.16-0.24 mM). This study affirms that the AHL degrading activity and biofilm inhibition ability of Rhodococcus sp. BH4 may be due to the presence of multiple quorum quenching enzymes, including two types of AHL-lactonases, in addition to AHL-acylase and oxidoreductase, for which the genes have yet to be described.
Due to the recent economic development, the diet style has become more and more westernized in Korea, which increased the concern of our well-beings. Our well-beings are also associated with the gut microbiota which vary depending on the dietary intake. In this study, we compared gut microbiome shifted by two diets: high-fat diets (HFD) and low-fiber diet (LFD) based on 16S rRNA gene sequences using MiSeq. Compared to the control diet, LFD and HFD treatments significantly decreased species richness, while there was no difference in species evenness. Both diet treatments significantly increased the relative abundance of the Proteobacteria (p<0.05), especially the genus Sutterella. Bacteroidetes was significantly decreased in HFD groups, where the family S24-7 was decreased most. On the other hand, significant difference between HFD and LFD was seen among Firmicutes, where the abundance of family Lachnospiraceae was lower in LFD groups (p<0.05). PICRUSt-based metabolic difference analyses showed LFD treatment significantly decreased metabolisms of amino acid, carbohydrate and methane (p<0.01). In contrast, HFD significantly increased amino acid metabolism (p<0.05). Glycan biosynthesis and metabolism were significantly increased in both treatment groups (p<0.01). Our results suggest that long-term unbalanced dietary intakes induce gut dysbiosis, leading to metabolic and colonic disorders.
Nontuberculous mycobacteria (NTM) are a major cause of opportunistic infections in immunocompromised patients, making the reliable and rapid identification of NTM to the species level very important for the treatment of such patients. Therefore, this study evaluated the usefulness of the novel target genes tuf and tmRNA for the identification of NTM to the species level, using a PCRrestriction fragment length polymorphism analysis (PRA). A total of 44 reference strains and 17 clinical isolates of the genus Mycobacterium were used. The 741 bp or 744 bp tuf genes were amplified, restricted with two restriction enzymes (HaeIII/MboI), and sequenced. The tuf gene-PRA patterns were compared with those for the tmRNA (AvaII), hsp65 (HaeIII/HphI), rpoB (MspI/HaeIII), and 16S rRNA (HaeIII) genes. For the reference strains, the tuf gene-PRA yielded 43 HaeIII patterns, of which 35 (81.4%) showed unique patterns on the species level, whereas the tmRNA, hsp65, rpoB, and 16S rRNA-PRAs only showed 10 (23.3%), 32 (74.4%), 19 (44.2%), and 3 (7%) unique patterns after single digestion, respectively. The tuf gene-PRA produced a clear distinction between closely related NTM species, such as M. abscessus (557-84-58) and M. chelonae (477-84-80-58), and M. kansasii (141-136-80-63-58-54-51) and M. gastri (141-136-117-80-58-51). No difference was observed between the tuf-PRA patterns for the reference strains and clinical isolates. Thus, a diagnostic algorithm using a tuf gene-targeting PRA is a promising tool with more advantages than the previously used hsp65, rpoB, and 16S rRNA genes for the identification of NTM to the species level.
Cotton plants were sampled and ranked according to their resistance to Verticillium wilt. In total, 642 endophytic fungi isolates representing 27 genera were recovered from Gossypium hirsutum root, stem, and leaf tissues, but were not uniformly distributed. More endophytic fungi appeared in the leaf (391) compared with the root (140) and stem (111) sections. However, no significant difference in the abundance of isolated endophytes was found among resistant cotton varieties. Alternaria exhibited the highest colonization frequency (7.9%), followed by Acremonium (6.6%) and Penicillium (4.8%). Unlike tolerant varieties, resistant and susceptible ones had similar endophytic fungal population compositions. In three Verticillium-wilt-resistant cotton varieties, fungal endophytes from the genus Alternaria were most frequently isolated, followed by Gibberella and Penicillium. The maximum concentration of dominant endophytic fungi was observed in leaf tissues (0.1797). The evenness of stem tissue endophytic communities (0.702) was comparatively more uniform than the other two tissues. Eighty endophytic fungi selected from 27 genera were evaluated for their inhibition activity against highly virulent Verticillium dahliae isolate Vd080 in vitro. Thirty-nine isolates exhibited fungistasis against the pathogen at varying degrees. Seven species, having high growth inhibition rates (${\geq}75%$), exhibited strong antifungal activity against V. dahliae. The antifungal activity of both volatile and nonvolatile metabolites was also investigated. The nonvolatile substances produced by CEF-818 (Penicillium simplicissimum), CEF-325 (Fusarium solani), CEF-714 (Leptosphaeria sp.), and CEF-642 (Talaromyces flavus) completely inhibited V. dahliae growth. These findings deepen our understanding of cotton-endophyte interactions and provide a platform for screening G. hirsutum endophytes with biocontrol potential.

Seven isolates showing strong proteolytic activity, KYC 1028, 1100, 1134, 1139, 1151, 1159, and 1182, were collected. Out of them, the broth of KYC 1134 and KYC 1139 showed the high proteolytic activity measured by azocazein. To determine 16S rDNA sequences for identification, 16S rDNA of seven isolates were amplified and compared with the 16S rDNA sequences of other myxobacteria at NCBI. It is evident from the phylo-genetic tree that the isolates belong to the genus Myxococcus. Sharing high percentage similarity values with myxobacteria, the 16S rDNA sequences were involved in two species, Myxococcus macrospores and M. Fulvus. Biochemical characteristics of KYC 1134 broth, which showed the highest proteolytic activity, showed increased activity 8 times to seven days after culture, and protein production were increased gradually and stopped at five days. The broth had optimal temperature at $60^{\circ}C$ for proteolytic activity, and stability of pH was ranged from pH 5 to 10, at $50^{\circ}C$ and 60, respectively. To classify proteases being in the broth, ten inhibitors were determined and only bestatin showed 27% inhibition effect. The inhibition result demonstrates that the broth contains kinds of amino peptidases and other exopeptidases.
