우리 나라의 주요 고추 재배지와 미국, 대만, 호주, 아르헨티나에서 수집된 44 개 고추 더뎅이병균(Xanthomonas campestris pv. vesicatoria)균주간의 유전적변이를 genomic DNA의 restriction fragment length polymorphism(RFLP)에 의해 분석하였다. Genomic DNA RFLP profiles을 cluster 분석하여 얻은 dendrogram에서 지리적 기원이 다른 44개 균주들은 11개 RFLP 그룹으로 분류되었다. 외국 균주들은 genomic DNA의 RFLP 분석에 의해 모두 각각 다른 RFLP 그룹으로 분류되었다. 외국 균주들 중에서 미국 균주는 우리 나라 일부 균주들과 밀접한 유전적 관련성을 가지고 함께 cluster를 이루었는데, 이것은 이 균주들이 동일한 고추 더뎅이병균의 조상 균주 집단에서 유래했으리라는 것을 시사해 준다. 우리 나라 균주들은 6개의 RFLP 그룹으로 분류되었다. 대부분의 우리 나라 균주들은 가까운 cluster를 이루며 미국 균주를 제외한 외국 균주들과 뚜렷하게 구분되었다. 그러나 우리 나라 균주들 중에서 마산에서 수집된 Ms93-1은 다른 우리 나라 균주들과 뚜렷하게 구분되었다. 유전적으로 격리된 균주의 출현은 우리 나라에서 지리적 기원이 다른 고추 더뎅이병균 균주 사이에 이미 발생한 다양한 유전적 분화의 결과라고 추론된다.
Nuclear DNA was isolated from the sperm cells representing genetic characteristics and genomic polymorphisms of rainbow trout by polymerase chain reaction(PCR) amplification of DNA using arbitrary primers. Genomic DNA fingerprints were generated from rainbow trout sperm DNA by polymerase chain reaction amplification using 20 arbitrary decamers as primers. Out of these primers, 4 generated 17 highly reproducible RAPD markers, producing almost six polymorphic bands per primers. Four of 6 primers tested generated amplified fragments which were polymorphic between different individuals. Polymorphic DNA fragments were reproducibly amplified from independent DNA preparations made from individuals. Rainbow trout was distinctly observed 3 specific DNA markers (2. 3, 2.0 and 1.3kb) in bandsharing. Individual fragments generated using the same arbitrary primer, demonstrated that a single primer detected at least three independent genomic polymorphisms in rainbow trout sperm DNA. The RAPD polymorphism generated by this primer may be used as a genetic marker for individual identification The RAPD-PCR technique has been shown to reveal informative polymorphism in many species of fish. The present results demonstrate that RAPD markers are abundant, reproducible and provide a basis for future gene mapping and MAS in these important aquaculture species using RAPD polymorphic markers. It is concluded that RAPD polymorphisms are useful as genetic markers for fish breed differentiation.
A silkworm Bombyx mori genomic DNA library was constructed from polyphagous J111 strain and unpolyphagous $C_3$ strain to develop the genomic study by DNA makers. Genomic DNAs of two strains were digested with restriction enzyme EcoRI and ligated into pUC18. The ligated plasmids were transferred into E. coli host strain DH5$\alpha$. When the genomic DNAs were hybridized with insert DNAs from transformant, could be categorized from hybridization patterns to three groups as high repetitive sequence, moderately repetitive sequence, and low-copy number sequences. A total of 219 clones containing single or low-copy number sequence inserts were examined for any polymorphisms between two strains of J111 and $C_3$. Forty six clones showed RFLPs and 10 of these clones were used as a probe of analysis of $F_2$ population derived from crossing between J111 and $C_3$ strain. The genetic inheritance tested with each clones will be important tools to construct the genetic map of the silkworm, Bombyx mori.
Polychlorinated biphenyls (PCBs) are persistent pollutants in aquatic environments, often causing the decline or disappearance of wild populations. In this study, we used a random amplified polymorphic DNA (RAPD) assay to evaluate the effects on the genomic DNA of olive flounder embryo and larval stages of exposure to Aroclor 1254 at concentrations of 1, 5, 10, 20, and $40{\mu}g/L$. We compared RAPD fingerprints of exposed and non-exposed samples. Polymorphisms were revealed as the presence and/or absence of DNA fragments between the two samples. A dose-dependent increase in the number of polymorphic bands was observed with Aroclor 1254 treatment. Also, RAPD profiles of animals exposed to Aroclor 1254 exhibited an increase in the frequency values (FV) compared to the control. A phenogram constructed using neighbor-joining method indicated that genomic template stability in developing embryo and larval stages was significantly affected at ${\geq}5{\mu}g/L$. This study suggested that DNA polymorphisms detected by RAPD analysis could be used as an investigative tool for environmental toxicology and as a useful biomarker in early life stages for the detection of potential genotoxicants.
The polymorphisms of several genes including Leptin (LEP), beta-lactoglobulin (LGB) and Prolactin receptor (PRLR) have been shown to affect milk composition traits in dairy cattle. But, the effects of these polymorphisms on the milk traits of Philippine water buffalo are still unclear. In the Philippines, buffalo are the major milk producers most of which are the Philippine carabao (PC), the American Murrah Buffalo (AMB) and Bulgarian Murrah Buffalo (BMB). The LEP, LGB and PRLR genes are considered to be associated with milk production traits. The objective of the present study was to identify the single nucleotide polymorphisms (SNPs) in the LEP, LGB and PRLR genes of PC, AMB and BMB and to investigate the effect of the SNPs on milk production traits in these buffalo. Genetic polymorphisms were screened by DNA sequencing and 12 SNPs were detected in BMB; 5 SNPs were in LEP exon3 region (G14227A, G14343A, T14502C, C14526T, G14603A); 5 SNPs were in LGB exon 2 region (G1861C, A1900G, G1901T, T1948C, G1949A); 2 SNPs were in PRLR exon 6 (T59047C, T59109C). Also, 12 polymorphism sites between cattle and buffalo were identified. Our analysis of the association between SNPs and milk production traits should be useful in future studies of buffalo breeding to improve lactation performance.
The objective of this study is to develop the gene based sequence tagged site (STS) markers in wheat. The euchromatin enriched genomic library was constructed and the STS primer sets were designed using gene based DNA sequence. The euchromatin enriched genomic (EEG) DNA library in wheat was constructed using the $Mcr$A and $Mcr$BC system in $DH5{\alpha}$ cell. The 2,166 EEG colonies have been constructed by methylated DNA exclusion. Among the colonies, 606 colonies with the size between 400 and 1200 bp of PCR products were selected for sequencing. In order to develop the gene based STS primers, blast analysis comparing between wheat genetic information and rice genome sequence was employed. The 227 STS primers mainly matched on $Triticum$$aestivum$ (hexaploid), $Triticum$$turgidum$ (tetraploid), $Aegilops$ (diploid), and other plants. The polymorphisms were detected in PCR products after digestion with restriction enzymes. The eight STS markers that showed 32 polymorphisms in twelve wheat genotypes were developed using 227 STS primers. The STS primers analysis will be useful for generation of informative molecular markers in wheat. Development of gene based STS marker is to identify the genetic function through cloning of target gene and find the new allele of target trait.

A key aspect of genomic research in the “post-genome era”is to associate sequence variations with heritable phenotypes. The most common variations in the human genome are single nucleotide polymorphisms (SNPs) that occur approximately once in every 500 to 1,000 bases. Although analyzing the phenotypic outcome of these SNPs is crucial to facilitate large-scale association studies of genetic diseases, detection of SNPs from an extended number of human DNA samples is often difficult, labor-intensive and time-consuming. Recent development in SNP detection methods using DNA microarrays and mass spectrophotometry has allowed automated high throughput analyses, but such equipments are not accessible to many scientists. In this study, we demonstrate that a simple PCR-based method using primers with a mismatched base at the 3'-end provides a fast and easy tool to identify known SNPs from human genomic DNA in a regular molecular biology laboratory. Results from this PCR amplification of specific alleles (PASA) analysis efficiently and accurately typed the Q576R polymorphism of human IL4 receptor from the genomic DNAs of 29 Koreans, including 9 samples whose genotype could not be discerned by the conventiona1 PCR-SSCP (single strand conformation polymorphism) method. Given the increasing attention to disease-associated polymorphisms in genomic research, this alternative technique will be very useful to identify SNPs in large-scale population studies.
This study was carried out to analyze the genetic polymorphisms of genomic DNA of blood and meat for conservation of the genetic resources and genetic improvement of Korean Native goat. The genetic identification between Korean Native goat and imported goat was examined using RAPD(random amplified polymorphisms DNAs) analysis with 30 Korean Native goat, 10 hybrid, 10 imported goat. 10 Korean native goat meat and 10 imported goat meat. The results obtained from this study were summarized as follows: 1. Genomic DNA from Korean native goat, hybrid and imported goat could be obtained above about 23kb size using 0.5% agarose gel electrophoresis and the ratio of optical density at 260nm to that at 280nm was between 1.7 and 2.0 using UV spectrophtometer instrument. 2. In the results of the gene identification between Korean Native goat and hybrid, and imported goat using RAPD methods with random primer of 110 kinds, only Korean native goat showed a specific band at about 369bp using a random primer OPO-19 (5'-CAA ACG TCG G-3'), but imported goat and hybrid not showed. 3. Although sea slaters Ligia have a significant role in rocky shore habitats, their taxonomic entities have not been clearly understood. In this study, we investigated whether genetic variation inferred from a nuclear genetic marker, namely amplified fragment length polymorphism (AFLP), would conform to that of a mitochondrial DNA marker. Using both the mitochondrial DNA marker and the AFLP marker amplified by the six selective primer sets, we analyzed 95 Ligia individuals from eight locations from East Asia. The direct sequencing of mitochondrial 16S rRNA gene revealed three distinct genetic lineages, with 9.8-11.7 Kimura 2-parameter genetic distance. However, the results of AFLP genotyping analysis with 691 loci did not support those of mitochondrial DNA, and revealed an unexpectedly high proportion of shared polymorphisms among lineages. The inconsistency between the two different genetic markers may be explained by difference in DNA evolutionary history, for example inheritance patterns, effective population size, and mutation rate. The other factor is a possible genomic island of speciation, in that most of the genomic parts are shared among lineages, and only a few genomic regions have diverged.
In view of the importance of muscle-specific creatine kinase (CKMM) gene as a genetic factor for athletic performance, we investigate the relationship between elite athletic performance and two restriction fragment length polymorphisms (Ncol and Taql RFLPs) in the CKMM gene. Genomic DNA was extracted from white blood cells of 98 unrelated male Korean elite athletes and 04 sedentary controls, respectively. Two genetic polymorphisms in the CKMM gene were detected by the polymerase chain reaction and the digestion with restriction endonucleases, Ncol and Taql, respectively. There were no significant associations between two genetic polymorphisms in the CKMM gene and elite athletic performance or clinical parameters in our subjects. Therefore, these findings suggest that two genetic polymorphisms in the CKMM gene may not be useful as genetic markers to predict the athletic performance in male Koreans.
