The objectives of present study is to investigate genetic damage of radiation in mammalian male germ cell and. to establish available screening method for determining genetic hazard by radiation. Several methods were employed to measure the genetic damage of radiation as follows: Sperm head counts, frequency occurrence of sperm with abnormal head shape, fertility, activity of LDH-X, and the induction of unscheduled DNA synthesis (U.D.S.) in male mouse were performed with the passing of time after irradiation by making use of the sequence of event that occurs during spermatogenesis. Sperm head counts and activity of LDH-X in testes were gradually reduced by increased radiation dose and with the passing of the time after irradiation. Frequency occurrence of sperm with abnormal head shape, sterile period, and the induction of unscheduled DNA synthesis were increased by increased radiation dose. It is suggested that since germ cell is a direct reflection of genetic complement, the use of male germ cell is rapid and convenient method for measuring genetic damage by radiation.
The purpose of this paper is to find an optimal weaving connection method of sweater threads while weaving intarsia sweater by the genetic algorithm. The objective function was devised to minimize labor cost and lessen the amount of thread usage. In order to create the parental population group in the genetic algorithm, we developed five thread connection methods. Besides, elite chromosome screening methods for the offspring group was selected both to the whole chromosome thread elite and to a color-coded elite thread chromosome. Commonly used diamond pattern in Intarsia sweater manufacturing was applied to the experiments. The experimental results showed that thread system saved the labor and material costs than woven method under the existing software. When weaving Intarsia sweater in the field, we can apply the developed genetic algorithm to improve productivity of weaving connection method.

We investigated antioxidant capacity in leaves of 14 coniferous trees under field conditions. We focused on understanding the species characteristics on antioxidant systems and screening the coniferous tree species with the best antioxidant systems using their characteristics. The antioxidant capacity of 14 coniferous trees was divided into three groups. First group was Thuja orientalis and Chamaecyparis obtusa and those species had the highest content of ${\beta}$-carotene and xanthophyll. Second group, C. obtusa and Juniperus chinensis, used antioxidant enzymes to mitigate stress. C. obtusa represented high activity at superoxide dismutase (SOD), glutathione reductase (GR), and peroxidase (POD), and J. chinensis exhibited high activity at SOD, POD, catalase (CAT). Third group employed antioxidant such as ascorbic acid and ${\alpha}$-tocopherol. The antioxidant content of T. orientalis was the highest while that of Pinus parviflora and C. obtusa were the lowest. Few species belonged in three groups simultaneously, and most species belonged in at least one or two groups. In summary, we proposed that C. obtusa and T. orientalis had the highest antioxidant capacity while P. parviflora and P. desiflora for. multicalus had the lowest antioxidant capacity.
Purpose: The purpose of this micro-ethnography is to examine whether science and societal changes impact family communication patterns among a convenience sample of 16 Korean women. Methods: The authors observed family communication in the context of a new breast cancer genetic screening and diagnostic testing program to detect BRCA gene mutations in Korean women at highest risk. Results: Analysis of in-depth interviews and field notes taken during participant observation illustrated that communication patterns in families vary according to a woman's position in the family. If a grandmother tests positive for a gene mutation, her daughters make decisions on her behalf; they open and maintain the communication channel among family members. If a housewife is diagnosed with cancer and a genetic mutation, she immediately consults her husband and her sisters. The husband creates an open communication channel between his wife, his parents and his siblings. As a result, a woman's cancer is a concern for the whole family not merely a woman's secret or crisis. Conclusion: Cultural differences are important to consider when designing new genetic service programs in different countries.
Cardiomyopathy is a major cause of death worldwide. Based on pathohistological abnormalities and clinical manifestation, cardiomyopathies are categorized into several groups: hypertrophic, dilated, restricted, arrhythmogenic right ventricular, and unclassified. Dilated cardiomyopathy, which is characterized by dilation of the left ventricle and systolic dysfunction, is the most severe and prevalent form of cardiomyopathy and usually requires heart transplantation. Its etiology remains unclear. Recent genetic studies of single gene mutations have provided significant insights into the complex processes of cardiac dysfunction. To date, over 40 genes have been demonstrated to contribute to dilated cardiomyopathy. With advances in genetic screening techniques, novel genes associated with this disease are continuously being identified. The respective gene products can be classified into several functional groups such as sarcomere proteins, structural proteins, ion channels, and nuclear envelope proteins. Nuclear envelope proteins are emerging as potential molecular targets in dilated cardiomyopathy. Because they are not directly associated with contractile force generation and transmission, the molecular pathways through which these proteins cause cardiac muscle disorder remain unclear. However, nuclear envelope proteins are involved in many essential cellular processes. Therefore, integrating apparently distinct cellular processes is of great interest in elucidating the etiology of dilated cardiomyopathy. In this mini review, we summarize the genetic factors associated with dilated cardiomyopathy and discuss their cellular functions.

The purpose of this study is to report a successful twin pregnancy and delivery in a female patient with X-linked dominant incontinentia pigmenti (IP) who underwent assisted reproductive technology followed by preimplantation genetic screening (PGS). A 29-year-old female with IP had a previous history of recurrent spontaneous abortion. A molecular analysis revealed the patient had a de novo mutation, 1308_1309insCCCCTTG(p.Ala438ProfsTer26), in the inhibitor of the kappa B kinase gamma gene located in the Xq28 region. IVF/ICSI and PGS was performed, in which male embryos were sexed using array-based comparative genomic hybridization (aCGH). After IVF/ICSI and PGS using aCGH on seven embryos, two euploid male blastocysts were transferred with a 50% probability of a viable male pregnancy. The dizygotic twin pregnancy was confirmed and the amniocentesis results of each twin were normal with regard to the mutation found in the mother. The patient delivered healthy twin babies during the 37th week of gestation. This case shows the beneficial role of PGS in achieving a successful pregnancy through euploid male embryo gender selection in a woman with X-linked dominant IP with a history of multiple male miscarriages.

Screening for drought and salinity tolerance was undertaken for selected Philippine upland rice landraces during germinative and seedling stages to identify varieties which can potentially be grown in marginally dry and saline soils. While increasing PEG and NaCl concentrations caused obvious signs of injury to all rice genotypes, considerable varietal differences were noted in the nature of responses providing evidence that these genotypes possess broad intraspecific genetic variations for drought and salt tolerance. Inconsistent responses of these varieties during both growth stages highlight complexities involved in stress responses and underscore the futility of utilizing a single stage in the rice plant's life cycle for physiological screening. Notwithstanding these perplexing responses, G_Katiil and Ml-Pilit Tapul were observed to thrive relatively well despite increased salt and drought stress during early growth stages and may therefore possess genes needed in crop improvement efforts for drought and salinity tolerance. While these results do not reflect the entire spectrum of adaptive expression to drought and salinity stress during the life cycle of the upland rice plant, they nonetheless provide an easy, reliable and reproducible method for preliminary identification of drought and salt tolerant rice varieties.
Small molecule tyrosine kinase inhibitors targeting HER 2 receptors have emerged as an important therapeutic approach in inhibition of downstream proliferation and survival signals for the treatment of breast cancers. Recent drug discovery efforts have demonstrated that naturally occurring polyphenolic compounds like delphinidin have potential to inhibit proliferation and promote apoptosis of breast cancer cells by targeting HER2 receptors. While delphinidin may thus reduce tumour size, it is associated with serious side effects like dysphonia. Owing to the narrow therapeutic window of delphinidin, the present study aimed to identify high affinity compounds targeting HER2 with safer pharmacological profiles than delphinidin through virtual screening approaches. Delphinidin served as the query parent for identification of structurally similar compounds by Tanimoto-based similarity searching with a threshold of 95% against the PubChem database. The compounds retrieved were further subjected to Lipinski and Verber's filters to obtain drug like agents, then further filtered by diversity based screens with a cut off of 0.6. The compound with Pubchem ID: 91596862 was identified to have higher affinity than its parent. In addition it also proved to be non-toxic with a better ADMET profile and higher kinase activity. The compound identified in the study can be put to further in vitro drug testing to complement the present study.
Background: Recently, the pork industry of Thailand faced an epidemic of highly virulent strains of porcine reproductive and respiratory syndrome virus (PRRSV), which spread throughout Southeast Asia, including the Lao People's Democratic Republic and Cambodia. Hence, the rapid and on-site screening of infected pigs on a farm is essential. Objectives: To develop the new aptamer as a biosensor for detection PRRSV which are rapid and on-site screening of infected pig. Methods: New aptamers against PRSSV were identified using the combined techniques of capillary electrophoresis, colorimetric assay by gold nanoparticles, and quartz crystal microbalance (QCM). Results: Thirty-six candidate aptamers of the PRRSV were identified from the systematic evolution of ligands by exponential enrichment (SELEX) by capillary electrophoresis. Only 8 out of 36 aptamers could bind to the PRSSV, as shown in a colorimetric assay. Of the 8 aptamers tested, only the 1F aptamer could bind specifically to the PRSSV when presented with the classical swine fever virus and a pseudo rabies virus. The QCM was used to confirm the specificity and sensitivity of the 1F aptamer with a detection limit of 1.87 × 1010 particles. Conclusions: SELEX screening of the aptamer equipped with capillary electrophoresis potentially revealed promising candidates for detecting the PRRSV. The 1F aptamer exhibited the highest specificity and selectivity against the PRRSV. These findings suggest that 1F is a promising aptamer for further developing a novel PRRSV rapid detection kit.
