도라지 수집종의 형태적 특성과 SSR마커에 의한 유연관계 분석 (Morphological Characteristics and Genetic Diversity Analysis of Platycodon grandiflorum (Jacq.) A. DC Determined Using SSR Markers)

  • Background : Plant breeding requires the collection of genetically diverse genetic resources. Studies on the characteristics of Platycodon grandiflorum resources have not been carried out so far. The present study was carried out to discriminate P. grandiflorum based on morphological characteristics and genetic diversity using simple sequence repeat (SSR) markers. Methods and Results :We collected 11 P. grandiflorum cultivars: Maries II, Hakone double white, Hakone double blue, Fuji white, Fuji pink, Fuji blue, Astra white, Astra pink, Astra blue, Astra semi-double blue and Jangbaek. Analyses of the morphological characteristics of the collection were conducted for aerial parts (flower, stem and leaf) and underground parts (root). Next, the genetic diversity of all P. grandiflorum resources was analyzed using SSR markers employing the DNA fragment analysis method. We determined that the 11 P. grandiflorum cultivars analyzed could be classified by plant length, leaf number and root characteristic. Based on the genetic diversity analysis, these cultivars were classified into four distinct groups. Conclusions : These findings could be used for further research on cultivar development using molecular breeding techniques and for conservation of the genetic diversity of P. grandiflorum. Moreover, the markers could be used for genetic mapping of the plant and marker-assisted selection for crop breeding.

  • This study was conducted to estimate the genetic improvements by selection criteria using the genetic parameters and breeding values for population of abalone Haliotis discus hannai. Genetic parameters and breeding values were estimated using all measurement data of growth traits (shell length, shell width and total weight) at 18 and 30 months old after artificial fertilization for 3,029 individuals produced in April 2014. Growth traits all exhibited moderate heritability (0.253-0.354). So it is considered that family selection will be more advantageous than individual selection. It was found that a higher genetic improvement could be expected when selecting the top 10% based on the breeding values of total weight rather than other traits. In particular, a higher genetic improvement could be expected when selecting the top 10% at 30 months old than 18 months old after artificial fertilization. This seems to be because the selection differential and heritability were higher at the 30 months old. Therefore, by estimating genetic parameters and breeding values of a population for production of the next generations by stage of growth, if they are used properly in selection and mating according to the improvement direction, it is considered that more breeding effects can be expected.

Investigation of Genetic Diversity between Wild-caught and Hatchery-reared Rock Bream (Oplegnathus fasciatus) Using Microsatellite DNA Analysis

  • Marine fisheries are important natural resources and must be maintained, especially fish species that are important sources of food. Despite the increase in stocking programs to maintain fisheries with artificially raised fish, the genetic impact stocking has on the wild fry population has not been addressed. Genetic variation in rock bream, Oplegnathus fasciatus, within and between wild-caught parents and the $F_1$ generation produced by them in 1 day was assayed using nine highly variable micro satellite markers. The nine micro satellite loci used in this study displayed diverse polymorphisms, and in total, 98 different alleles were observed over all loci. Differences in genetic variability of the $F_1$ offspring compared to their wild-caught parents (brood stock) were observed in terms of allele frequency, gene diversity, and heterozygosity. Although the $F_1$ generation of rock bream was missing 16% of the micro satellite alleles, no significant reduction was found in mean heterozygosity of the $F_1$ population compared to the brood stock. Eight of nine loci showed significant Hardy-Weinberg equilibrium (HWE) deviations in the $F_1$ population, while the brood stock deviated from HWE at three micro satellite loci (KOF85, KOF360 and KOF374). These deviations showed mostly a deficit of heterozygotes. Our results provide evidence for genetic differences in the $F_1$ hatchery offspring compared to their wild-caught parents and reinforce the need for a series of consecutive egg collections to avoid the loss of genetic variability. This also further underscores the importance of monitoring genetic variability of hatchery populations for the conservation of natural rock bream resources.

Effect of Glycine on L-Ornithine Production by a Citrulline Auxotroph of Brevibacterium ketoglutamicum and Stoichiometric Analysis

  • The effects of glycine on cell growth and L-omithine production were investigated in shake-flask and jar fermentor cultures of a citrulline auxotrophic mutant, Brevibacterium ketoglutamicum BK 1046. In the shake-flask culture, the optimal concentration of glycine for L-ornithine production was found to be 20 g/l. In the jar fermentor culture with the glycine at an initial concentration of 20 g/l, L-ornithine production increased by 28%, compared to that of the culture with no glycine added. 37 g/l of L-ornithine was produced when additional feeding of glycine (5 g/l) was made. This was a significant improvement in L-ornithine production compared to that (ca. 24 g/l) of the corresponding batch culture conducted without glycine. According to the stoichiometric analysis with the batch fermentation results, the experimental and theoretical L-ornithine yields based on the glucose consumption were 0.24 and 0.59, respectively. This indicates that the performance of L-ornithine fermentation can further be improved by the supplementation of glycine and the development of a mutant strain possessing a higher growth yield.

  • To examine whether the signal sequence of Bacillus endo-1, 4-glucanase can act functionally in a yeast, a lower eucaryote, two recombinant plasmids were constructed and introduced into Saccharomyces cerevisiae: recombinant plasmid pGCMC10 containing the complete signal sequence of Bacillus endoglucanase, and pGCMC11 without the signal sequence. Secretion of endoglucanase into culture medium was obtained with the yeast transformant containing plasmid pGCMC10. The secreted endoglucanase was glycosylated and was apparently processed to be about 36 kilodaltons (KDa) and 43KDa proteins. The glycosylated endoglucanase from yeast transformant was more thermostable than the nonglycosylated endoglucanase from Escherichia coli transformant.

유전적 다양성이 고려되지 않은 어미 관리에 의한 양식 넙치(Paralichthys olivaceus)의 유전적 다양성의 변화 (Genetic Variability of Farmed Olive Flounder (Paralichthys olivaceus) Populations Managed with no Consideration of Genetic Diversity)

  • 우리나라의 주요 양식 대상종인 넙치에 있어 유전적 조성에 대한 고려없이 구성된 양식 넙치 어미 집단에서 세대의 경과에 따른 집단의 유전적 다양성의 변화를 알아보기 위하여 총 8개의 microsatellite DNA markers를 이용하여 분석하였다. 동일한 수정란 생산업체에서 2003년에 생산된 양식 넙치와, 동일한 집단내에서 생산된 넙치로 어미 그룹이 완전히 교체된 2006년 및 2007년에 생산된 넙치 집단을 비교한 결과, 대립유전자 수와 이형접합기대치의 비교에서 2003년산 집단의 경우 9.75개와 0.796에서 2006년 7.78개와 0.785로 유전적 다양성이 다소 감소되는 경향이었다. 또한 대립유전자의 빈도에 있어서도 몇몇 대립유전자의 경우 소실되거나 빈도에 변화를 보이는 등 왜곡되어 나타났다. 그러나 여러 가지 다양성 지표들에서 수치상 감소를 나타내고 있는 이러한 결과들은 비록 유의적인 수준에서 유전적 다양성의 축소라고는 판단되지 않지만 양식 넙치 집단이 HWE에 어긋나 있어 세대가 진행될수록 집단의 유전적 다양성은 감소될 것으로 예상되었다.

Study of Genetic Diversity among Simmental Cross Cattle in West Sumatra Based on Microsatellite Markers

  • A study was conducted to assess the genetic diversity among Simmental Cross cattle in West Sumatra using microsatellite DNA markers. A total of 176 individual cattle blood samples was used for obtaining DNA samples. Twelve primers of microsatellite loci as recommended by FAO were used to identify the genetic diversity of the Simmental Cross cattle population. Multiplex DNA fragment analysis method was used for allele identification. All the microsatellite loci in this study were highly polymorphic and all of the identified alleles were able to classify the cattle population into several groups based on their genetic distance. The heterozygosity values of microsatellite loci in this study ranged from 0.556 to 0.782. The polymorphism information content (PIC) value of the 12 observed loci is high (PIC>0.5). The highest PIC value in the Simmental cattle population was 0.893 (locus TGLA53), while the lowest value was 0.529 (locus BM1818). Based on the genetic distance value, the subpopulation of the Simmental Cross-Agam and the Simmental Cross-Limapuluh Kota was exceptionally close to the Simmental Purebred thus indicating that a grading-up process has taken place with the Simmental Purebred. In view of the advantages possessed by the Simmental Cross cattle and the evaluation of the genetic diversity results, a number of subpopulations in this study can be considered as the initial (base) population for the Simmental Cross cattle breeding programs in West Sumatra, Indonesia.

It sets direction of the New-Hanok Type models development based on Expert advice and the literature, and was reviewed a primal reason system of Korea as an extraction base of genetic factors. Result: Then Through the framework of the vertical axis (the form), the horizontal axis (space), It extracted the genetic factors of the Korea Traditional Architecture, classified the genetic factors extracted as the structure(layout, construction, space), features, traditional beauty, investigated the content of the form representation and spatial meaning, and were characterized. Based on the result, It were comprehensive the genetic factors extracted as plan Elements for inheriting of the traditions.

Assessment of Genetic Diversity and Population Structure on Kenyan Sunflower (Helianthus annus L.) Breeding Lines by SSR Markers

  • In crop breeding program, information about genetic dissimilarity on breeding resources is very important to corroborate genealogical relationships and to predict the most heterozygotic hybrid combinations and inbred breeding. This study aimed to evaluate the genetic variation in Kenyan sunflower breeding lines based on simple sequence repeat (SSR). A total of 83 alleles were detected at 32 SSR loci. The allele number per locus ranged from 2 to 7 with an average of 2.7 alleles per locus detected from the 24 sunflower accessions and the average value of polymorphic information contents (PIC) were 0.384. A cluster analysis based on the genetic similarity coefficients was conducted and the 24 sunflower breeding resources were classified into three groups. The principal coordinates (PCoA) revealed 34% and 13.38% respectively, and 47.38% of total variation. It was found that the genetic diversity within the Kenyan sunflower breeding resources was narrower than that in other sunflower germplasm resources, suggesting the importance and feasibility of introducing elite genotypes from different origins for selection of breeding lines with broader genetic base in Kenyan sunflower breeding program.

  • Purpose: The purpose of this review article is to introduce how the Korean Society of Genetic Nursing (KSGN) has evolved and tried to translate genomic knowledge to nursing practice, and then to suggest the future role of genetic nurses in Korea. Methods: A literature review was performed and the current status of genetic counselling in Korea was explored. Then the educational and clinical experiences of the authors were incorporated. Finally, the main activities of Korean nursing for genetics were identified. Results: Two types of genetic counsellor certification have been issued in Korea: one is issued by the Korean Society of Genetic Medicine, another by the Korean Society of Breast Cancer since June 2011. A few Korean nursing researchers have continuously performed research related to genetic nursing and undertook several research projects funded by the government since 2003. In February 2011, KSGN was established and is now trying to establish further international networks. Conclusion: Nursing genetic experts should be trained to integrate all specialties for genetic counselling, so they can provide holistic genetic services including ethical, legal, and social issues (ELSI).
