Genetic Relationships and Phylogeny of the Asplenium antiquum Makino (Aspleniaceae) and its relative species based on RAPD Analysis

  • This study characterized the genetic variations of 13 populations of Asplenium antiquum and its relative species using randomly amplified polymorphic DNA (RAPD) markers. A total 88 scorable RAPD bands were generated by the 12 random oligo primers and were analyzed by Nei and Li's genetic distance. High genetic variability was detected between A. antiquum and A. nidus, with the range from 0.568 to 0.682. And slightly low genetic variations showed within the populations of same species. Seven populations of A. antiquum showed slight differences (0.000-0.216), and five populations of A. nidus showed similar low genetic variations (0.114 to 0.171). Two individuals from Sup-seom Island which are growing in might be the regenerated one from abroad. A. antiquum were clustered as two groups (Group I, Group II) by UPGMA phenogram. And five populations of A. nidus were clustered as two groups correlated with geographical distribution. The RAPD data was very useful to define the genetic variations and to discuss the phylogenetic relationships among A. antiquum and the related species..

Genetic diversity and phylogenetic analysis of genus Paeonia based on nuclear ribosomal DNA ITS sequence

  • The genus Paeonia belongs to the family Paeoniaceae having significant medicinal and ornamental importance. The present investigation was undertaken with an aim to understand phylogenetic relationships of three Paeonia species (P. lactiflora, P. obovata, and P. suffruticosa) that are widely distributed in China, Korea, and Japan, using nuclear ribosomal DNA (nrDNA) internal transcribed spacer (ITS) sequence and to compare the phylogeny results with investigations reported earlier using existed sequences of the same species. The size variation obtained among sequenced nrDNA ITS region was narrow and ranged from 722 to 726 bp. The highest interspecific genetic distance (GD) was found between P. lactiflora and P. suffruticosa or P. obovata. The phylogram obtained using our nrDNA ITS sequences showed non-congruence with previous hypothesis of the phylogeny between section Paeonia and section Moutan of genus Paeonia. This result was supported by the phylogenetic relations showed in the phylogram constructed with existed sequences in NCBI. The present study suggested that P. obovata belonging to section Paeonia was phylogenetically closer to P. suffruticosa representing section Moutan of genus Paeonia than P. lactiflora belonging to section Paeonia. The main reason of the paraphyly of section Paeonia is thought to be nucleotide additivity directly caused by origin hybridization. This study provides more sequence sources of genus Paeonia, and will help for further studies in intraspecies population, and their phylogentic analysis and molecular evolution.

Intraspecific Molecular Phylogeny, Genetic Variation and Phylogeography of Reticulitermes speratus (Isoptera:Rhinotermitidae)

  • Population structure was investigated in Reticulitermes speratus populations in the Korean Peninsula and the Japanese Archipelago. All trees derived from analyses of the combined sequence dataset of two mitochondrial genes, COII and COIII, showed that R. speratus populations cluster into two major clades comprising the Korean/southern Japanese populations and the northern Japanese populations. Analysis of population genetic structure showed strong genetic partitioning between populations of the two clades. To understand historical migration routes and current distributions, the phylogeographic history of R. speratus was inferred from intra-/interspecific phylogeny and divergence times estimated between the clades of the phylogenetic tree. The estimated migration route and divergence time of ancestral R. speratus are congruent with recent paleogeographic hypotheses involving land-bridge connections between the Asian continent and the Japanese Archipelago. We suggest that ancestral R. speratus separated into northern and southern Japanese populations after its migration into the Japanese main islands from East China during the early Pleistocene via the East China Sea basin, which may have been exposed during that period. The Korean populations seem to have diverged recently from southern Japanese populations; this may explain the current distribution of R. speratus in the Japanese Arachipelago, and account for why it is restricted to northern areas of the Tokara Strait.

한국산 털보톡토기상과 (톡토기목, 곤충강)의 계통과 종분화 (Phylogeny and Speciation of Entomobryoidea (Collembola, Insecta) from Korea)

  • 톡토기목 가운데 털보톡토기상과에 속하는 3개 과의 종들간의 유연관계를 조사하기 위하여 이들 과에 속하는 3종 및 인접 상과에 속하는 3과 4종에 대해 유전자분석을 실시하 였다. 이를 위해서 전기영동을 실시하고 대립유전자빈도를 구한 후 유전적차이치(D)를 이용 하여 UPGMA 방법으로 dendrogram을 작성하였다. 그 결과 분절아목 중 털보톡토기상과에 속하는 마디톡토기과, 털보톡토기과 및 가시톡토기과의 종들이 함께 유집되었고, 인접상과인 보라톡토기과, 어리톡토기과 및 혹무늬톡토기과의 종들이 함께 유집되었다. 한편 가시톡토기 과의 종은 털보톡토기과의 종과 그리고 혹무늬톡토기과의 종은 보라톡토기과의 종과 가깝게 유집이 되기는 하나, 이들 종간의 유전적 차이값이 매우 높아 일부 학자의 견해대로 한 과 에 속하는 두 아과로 놓기보다는 별개의 과로 두는 것이 타당하다고 판단되었다. 아울러 굴 톡토기는 어리톡토기과와 유집되기는 하나 유전적차이가 크므로 독립된 과로서의 성격이 인 정된다. 이 결과는 형태형질 및 18S rDNA 염기서열을 분석한 결과와도 대체로 일치하였다.

Phylogenetic analysis of Neottia japonica (Orchidaceae) based on ITS and matK regions

  • To elucidate the molecular phylogeny of Neottia japonica, which is a terrestrial orchid distributed in East Asia, the internal transcribed spacer (ITS) of nuclear DNA and the matK of chloroplast DNA were used. A total 22 species of 69 accessions for ITS and 21 species of 114 accessions for matK phylogeny were analyzed with the maximum parsimony and Bayesian methods. In addition, we sought to establish a correlation between the distribution, morphology of the auricles and genetic association of N. japonica with phylogenetic data. The phylogenetic results suggest that N. japonica is monophyletic and a sister to N. suzukii in terms of the ITS phylogeny, while it is paraphyletic with N. suzukii in terms of the matK phylogeny. N. japonica and N. suzukii show similar morphologies of the lip and column, they both flower in April, and they are both distributed sympatrically in Taiwan. Therefore, it appears to be clear that N. japonica and N. suzukii are close taxa within Neottia, although there is incongruence between the nrDNA and cpDNA phylogenies of N. japonica. The incongruence between the two datasets may have various causes, meaning that further studies are needed to confirm the evolutionary process of N. japonica. The phylogenetic status of N. kiusiana, which was not included in previous studies, was as a sister to N. nidus-avis. Meanwhile, the ITS and matK phylogenies are unsuitable for identifying genetic associations with the characteristic of auricles. The phylogenetic topologies of Korean, Taiwanese and mainland Chinese individuals suggest that the populations of N. japonica in Korea originated from China's mainland and island areas. The characterization of regional gene differences could provide useful preliminary data for future studies.

Intraspecific Phylogeny of the Korean Water Deer, Hydropotes inermis argyropus (Artiodactyla, Cervidae)

  • The water deer, Hydropotes inermis (Cervidae), is native to China and Korea and has two subspecies of the Chinese water deer (Hydropotes inermis inermis) and Korean water deer (Hydropotes inermis argyropus). To date, only the Korean water deer has been reported in South Korea. In this study, however, an intraspecific phylogeny and haplotype analysis based on mitochondrial cytochrome oxidase I indicated that both Korean and Chinese water deer are found in South Korea. The populations of the two Korean genetic lineages did not show distinct geographic distributions. Further morphological studies on the Korean water deer will be required to confirm its taxonomic status.

Genetic Variation of Rice Populations Estimated Using nrDNA ITS Region Sequence

  • The rice belonging to Oryza sativa is not only has significant economic importance, for it is the major source of nutrition for about 3 billion all around the world. But also plays a vital role as a model organism, because it has a number of advantages to be a model plant, such as efficient transformation system and small genome size. Many methods and techniques have been conducted to attempt to distinguish different Oryza sativa species, such as amplified fragment length polymorphism (AFLP), random amplified polymorphic DNA (RAPD), simple sequence repeat (SSR) and so on. However, studies using sequence analysis of internal transcribed spacer (ITS), a region of ribosomal RNA has not been reported until now. This study was undertaken with an aim to understand the phylogenetic relationships among sixteen isolates of Oryza sativa collected from abroad and fifteen isolates collected from Korea, using ribosomal RNA (rRNA) internal transcribed spacer (ITS) sequences to compare the phylogeny relationships among different Oryza sativa species. The size variation obtained among sequenced nuclear ribosomal DNA (nrDNA) ITS region ranged from 515bp to 1000bp. The highest interspecific genetic distance (GD) was found between Sfejare 45 (FR12) and Anapuruna (FR15). Taebong isolate showed the least dissimilarity of the ITS region sequence with other thirty isolates. This consequence will help us further understanding molecular diversification in intra-species population and their phylogenetic analysis.

Studies on Biological Diversity of Firefly in Japan

  • Taxonomic and phylogenetic studies of firefly in Japan have been reviewed. Fourty-six lampyrid species and one rhagophthalmid are distributed in the Japanese Islands including the Ryukyus. Recently, molecular phylogenetic approaches have been employed in the systematic study of firefly using mitochondrial and luciferase genes. Based on the molecular phylogenetic trees, evolutionary process of flashing patterns related strictly to mating behavior was estimated. Furthermore, genetic diversity studies revealed geographic differentiation patterns within species, and conservation measures of firefly were proposed to protect genetic resources endemic to the localities.
