Basic Studies on the Breeding of Fiber Flax (Linum usitatissimum L.) in Korea (섬유용 아마(Linum usitatissimum L.)의 육종에 관한 기초적 연구)

  • The earliness to flowering was completely dominant over the lateness, and the short plant height and stem length were partially dominant over the tall. The heavy stem weight, however, was recessive to the light. Heritability values for the flowering period, plant height, dry stem weight and fiber ratio were high, while those of days to initial flowering, stem length and fiber weight were low. Flowering period, plant height, stem weight, dry stem weight and fiber ratio were closely related to fiber weight or fiber yield. The selection index estimated jointly the plant height ($X_1$), dry stem weight($X_2$) and fiber weight($X_3$); that is 0.0020$X_1$-0.0047$X_2$-0.0181$X_3$, was the most efficient one for the selection practices. The plant height was the most reliable character for the increased genetic advances and the relative selection efficiences. Effects of locations and genotype-environment interactions were highly significant in most of the characters investigated.

Immunocompetence Status of White Plumage Naked Neck versus Normally Feathered Broilers in Tropical Climate

  • The study was undertaken to evaluate the effect of naked neck gene on mortality, cell mediated and humoral immune response in white plumage broiler population. The mortality of homozygous naked neck (Na/Na) broilers (11.71%) was comparatively lower than that of heterozygous naked neck (Na/na) (12.28%) and normally feathered (na/na) (13.59%) broilers. The humoral immune response was measured against (1% v/v) sheep red blood cells (SRBC) for total haemagglutinin (HA) antibody, 2-mercaptoethanol resistance (MER) or (IgG) antibody and 2-mercaptoethanol sensitive (MES) or (IgM) antibody titre on 7 days post-immunization. The titre was expressed as log2 of the highest dilution which shows complete haemagglutination. Total HA titers of Na/Na and Na/na (11.05$\pm$0.53 and 11.09$\pm$0.38) were comparatively higher than that of na/na (10.26$\pm$0.42). The MES antibody titre of Na/Na (8.50$\pm$0.53) and Na/na (7.63$\pm$0.45) broilers were significantly higher as compared to na/na (6.11$\pm$0.32) broilers. The MER titre of na/na genetic group (4.15$\pm$0.42) was significantly higher than Na/Na (2.55$\pm$0.37) and comparatively higher than Na/na (3.45$\pm$0.38) broilers. In vivo cell response to phytohaemagglutinin-P (PHA-P), measured as Foot Index (FI) in mm expressed significantly higher response in Na/na (0.473$\pm$0.05) and Na/Na (0.413$\pm$0.04) broilers as compared to na/na (0.304$\pm$0.03) broilers. The result of present study suggested that white plumage naked neck broilers had better immune response as compared to normally feathered broilers.

Prevalence of obesity, hypertension, and hyperlipidemia in Gunpo children of low economic status (군포시 저소득층 소아의 비만도, 혈압 및 지질검사)

  • Purpose : This study aimed to assess the prevalence of obesity, hypertension, and hyperlipidemia in children from low-income families in Gunpo and to evaluate whether economic status affects the prevalence of obesity. Methods : Between October 2007 and March 2008, 341 children (167 girls and 174 boys; age, 6 to 13 years) were enrolled in this study. All these children came from families who earned minimum wages and who were supported by government. We measured height, weight, and blood pressure and performed laboratory examinations, including total cholesterol, high-density lipoprotein cholesterol, and triglycerides. Body mass index (BMI) was calculated using weight and height. We compared the prevalence of obesity, hypertension, and hyperlipidemia in children from low-income families with the prevalence of these maladies found in other studies. Results : Prevalence of obesity ($BMI{\geq}95^{th}$) was 7.1%. Prevalence of hypertension and hyperlipidemia was 8.2% and 16.7%, respectively. In the obese group, prevalence of hypertension and hyperlipidemia was 25.0% and 45.8%, respectively. Obesity and hyperlipidemia were slightly more frequent in our study than those found in other reports, and the prevalence of hypertension in the obese group was very high compared with statistics from the Korean National Health and Nutrition Examination Survey (25% vs. 19.5%) and those from other reports. Conclusion : Low-income status was associated with an increased incidence of obesity, hyperlipidemia, and hypertension. Hypertension showed an especially strong association with economic status, which seemed to correlate with genetic, environmental, and dietary effects.

Investigating Organ Toxicity Profile of Tenofovir and Tenofovir Nanoparticle on the Liver and Kidney: Experimental Animal Study

  • Tenofovir nanoparticles are novel therapeutic intervention in human immunodeficiency virus (HIV) infection reaching the virus in their sanctuary sites. However, there has been no systemic toxicity testing of this formulation despite global concerns on the safety of nano drugs. Therefore, this study was designed to investigate the toxicity of Tenofovir nanoparticle (NTDF) on the liver and kidney using an animal model. Fifteen adult male Sprague-Dawley (SD) rats maintained at the animal house of the biomedical resources unit of the University of KwaZulu-Natal were weighed and divided into three groups. Control animals (A) were administered with normal saline (NS). The therapeutic doses of Tenofovir (TDF) and nanoparticles of Tenofovir (NTDF) were administered to group B and C and observed for signs of stress for four weeks after which animals were weighed and sacrificed. Liver and kidney were removed and fixed in formal saline, processed and stained using H/E, PAS and MT stains for light microscopy. Serum was obtained for renal function test (RFT) and liver function test (LFT). Cellular measurements and capturing were done using ImageJ and Leica software 2.0. Data were analysed using graph pad 6, p values < 0.05 were significant. We observed no signs of behavioural toxicity and no mortality during this study, however, in the kidneys, we reported mild morphological perturbations widening of Bowman's space, and vacuolations in glomerulus and tubules of TDF and NTDF animals. Also, there was a significant elevation of glycogen deposition in NTDF and TDF animals when compared with control. In the liver, there were mild histological changes with widening of sinusoidal spaces, vacuolations in hepatocytes and elevation of glycogen deposition in TDF and NTDF administered animals. In addition to this, there were no significant differences in stereological measurements and cell count, LFT, RFT, weight changes and organo-somatic index between treatment groups and control. In conclusion, NTDF and TDF in therapeutic doses can lead to mild hepatic and renal histological damage. Further studies are needed to understand the precise genetic mechanism.

Variation of Leaflet Traits and Their Association with Agronomic Traits of Soybean Germplasm (콩 유전자원의 소엽형질 변이와 농업형질과의 관계)

  • To determine variations in leaflet length (LL), leaflet width (LW), leaflet size (LS), and leaflet shape index (LSI), and their association with eight agronomic traits, characterization data of 884 soybean accessions which were grown in the autumn of 1992 in Taiwan were analyzed. LL ranged from 4.3 to 14.7 cm, and LW ranged from 2.8 to 9.7 cm. Also, LS (LL $\times$ LW) ranged from 12.1 to 124.6 $\textrm{cm}^2$. The absolute variation of LL, LW, and LS was not large because of limitation in vegetative growth by short day length. None was classified as a large leaflet based on the International Board for Plant Genetic Resources (IBPGR) descriptors. LSI (LL /LW) ranged from 1.21 to 3.06, and three accessions were classified as narrow leaflet. There were differences in ranges and means of LL, LW, LS, and LSI between and within temperate and tropical accessions. LL, LW, LS, and LSI had highly significant positive correlations with seven agronomic traits and highly significant negative correlation with 100-seed weight except LW for all accessions. There was variation in the closeness of association among leaflet traits, and between and within temperate and tropical accessions. Generally, LL, LW, and LS were more closely associated with days to flowering, plant height at $R_1$ and $R_8$, number of pods per plant; LSI was more closely associated with 100-seed weight than other traits.

Assessment on Ecological Characteristics of Vegetation in the Trail of Adjacent to Molundae (몰운대 탐방로 주변 식생의 생태적 특성 평가)

  • The objective of this study is to analyze the flora and forest vegetation of trails with high visitor density at Molundae. Nine quadrats of $20{\times}20m$ were selected for the survey. The survey was conducted from April to October 2010. The obtained results are summarized as follows. Plot1, plot2, plot3, plot4, plot6, and plot7 were located at slopes of $5{\sim}20^{\circ}$, 17~40m above sea level, and were formed with the colony of Japanese black pine(Pinus thunbergii Parl) and Japanese black pine(Pinus thunbergii Parl)-white oak(Quercus aliena Blume). Tree layer had the height of 8~20m, and the coverage of 50~70%, while subtree layer had the height of 3-8m, and the coverage 30~80%. On the other hand, shrub layer had the height of 2~4m, and the coverage of 10~30%, and herb had the height of 0.2~1m and coverage 5~20%. Plot5, plot8, and plot9 were located at the summit areas of 57~78m above sea level, and $0^{\circ}$ slope. Japanese black pine(Pinus thunbergii Parl) formed a community there. Tree layer was 8~20m high, and covered 60~70%, of the area, and subtree layer was 6~8m high, and coverage 30~40%. Shrub layer had the height of 2~6m, and the coverage of 30%, while herb layer had the height 0.2~2m, and the coverage 20-80%. Succession does not occur in the surveyed areas which have high visitor density. Artificially planted sawtooth oak(Quercus acutissima) trees were found to disturb succession and formation of multi-layer vegetation, resulting in the ecologically unstable forest. Therefore, the researcher suggested the strategy of managing the vegetation in the conclusion. This study has the limit in that the plots selected for the survey reflected only part of various trails in the Molundae area. It is necessary to suggest the vegetation management plans by selecting more diverse trail areas in consideration of the visitor density and behaviors, and analyzing the changes in vegetation quantitatively in order to manage the vegetation in urban areas damaged by visitors more effectively.

Genotypes of Lysophospholipase I (LYPLA1) Gene Associate with Carcass Traits of the Jeju Crossbred Cattle (Hanwoo × Jeju Black cattle) and Hanwoo Populations (제주도산 한우와 제주흑한우 집단의 도체형질에 대한 Lysophospholipase I (LYPLA1) 유전자형의 효과)

  • Genotypes of the nucleotide substitution g.23655332G>C of SNP marker rs385360448 at Lysophospholipase I (LYPLA1) gene intron 7, were tested for their effect on the carcass traits of Hanwoo and Jeju Crossbred cattle ($F_1$ progeny produced between Hanwoo ${\times}$ Jeju Black cattle) populations on Jeju Island. In the Hanwoo steer population, the meats containing LYPLA1 rs385360448 G/- genotypes showed significantly higher marbling scores and greater texture indices, compared to those of rs385360448 C/C homozygous animals (p<0.05). However, the LYPLA1 genotypes were not associated with the levels of carcass weight, backfat thickness, eye muscle area (EMA), meat color, and fat color (p>0.05). On the other hand, in the JCC steer population, the LYPLA1 G/- harboring meats showed significantly greater EMA levels, compared to those of C/C homozygotes (p<0.05). The results of the present study indicate that the LYPLA1 genotypes could alter the levels of intramuscular fat deposition, texture index, and eye muscle area via phospholipid metabolism in the Longissimus dorsi muscle of the cattle. These findings suggested that LYPLA1 genotypes may effect molecular genetic markers in the improvement of carcass traits of Hanwoo and Jeju Black industrial cattle populations on Jeju Island.

Genetic signature of strong recent positive selection at interleukin-32 gene in goat

  • Objective: Identification of the candidate genes that play key roles in phenotypic variations can provide new information about evolution and positive selection. Interleukin (IL)-32 is involved in many biological processes, however, its role for the immune response against various diseases in mammals is poorly understood. Therefore, the current investigation was performed for the better understanding of the molecular evolution and the positive selection of single nucleotide polymorphisms in IL-32 gene. Methods: By using fixation index ($F_{ST}$) based method, IL-32 (9375) gene was found to be outlier and under significant positive selection with the provisional combined allocation of mean heterozygosity and $F_{ST}$. Using nucleotide sequences of 11 mammalian species from National Center for Biotechnology Information database, the evolutionary selection of IL-32 gene was determined using Maximum likelihood model method, through four models (M1a, M2a, M7, and M8) in Codeml program of phylogenetic analysis by maximum liklihood. Results: IL-32 is detected under positive selection using the $F_{ST}$ simulations method. The phylogenetic tree revealed that goat IL-32 was in close resemblance with sheep IL-32. The coding nucleotide sequences were compared among 11 species and it was found that the goat IL-32 gene shared identity with sheep (96.54%), bison (91.97%), camel (58.39%), cat (56.59%), buffalo (56.50%), human (56.13%), dog (50.97%), horse (54.04%), and rabbit (53.41%) respectively. Conclusion: This study provides evidence for IL-32 gene as under significant positive selection in goat.

An Array-Type RGB Sensor for Precision Measurement of pH

  • As pH is a widely used index in chemical, medical, and environmental applications, research on pH sensors has been active in recent years. This study obtained RGB values by measuring the reflected light from a liquid sample to detect fine changes in pH, and performed mathematical modeling to investigate the relationship between the detected optical signal and pH value. Also, the trends in pH changes were easily identified by analyzing RGB values and displaying them in the color coordinate for easy visualization of data. This method implemented a user-friendly system that can measure and analyze in real time. This system can be used in many fields such as genetic engineering, environmental engineering, and clinical engineering, because it not only can measure pH but also replaces a colorimeter or turbidimeter.

Diversity and Phylogenetic Analysis of Culturable Marine Bacteria Isolated from Rhizosphere Soils of Suaeda japonica Makino in Suncheon Bay (순천만 칠면초의 근권으로부터 분리된 해양세균의 다양성 및 계통학적 분석)

  • Bacterial diversity was studied in the rhizosphere of Suaeda japonica Makino, which is native to Suncheon Bay in South Korea. Soil samples from several sites were diluted serially, and pure isolation was performed by subculture using marine agar and tryptic soy agar media. Genomic DNA was extracted from 29 pure, isolated bacterial strains, after which their 16S rDNA sequences were amplified and analyzed. Phylogenetic analysis was performed to confirm their genetic relationship. The 29 bacterial strains were classified into five groups: phylum Firmicutes (44.8%), Gamma proteobacteria group (27.6%), Alpha proteobacteria group (10.3%), phylum Bacteriodetes (10.3%), and phylum Actinobacteria (6.8%). The most widely distributed genera were Bacillus (phylum Firmicutes), and Marinobacterium, Halomonas, and Vibrio (Gamma proteobacteria group). To confirm the bacterial diversity in rhizospheres of S. japonica, the diversity index was used at the genus level. The results show that bacterial diversity differed at each of the sampling sites. These 29 bacterial strains are thought to play a major role in material cycling at Suncheon Bay, in overcoming the sea/mud flat-specific environmental stress. Furthermore, some strains are assumed to be involved in a positive interaction with the halophyte S. japonica, as rhizospheric flora, with induction of growth promotion and plant defense mechanism.
