Protective Effects of Angelica keiskei Extracts Against D-Galactosamine (GalN)-induced Hepatotoxicity in Rats

  • Although the vegetable Angelica keiskei (AK) has widely been utilized for the purpose of general health improvement among Korean population, its functionalities are not very well defined. In this study, we examined the effects of methanol extract of AK in rats on the biochemical changes induced by two hepatotoxins, D-galactosamine (GalN) and carbon tetrachloride ($CCl_4$). AK was orally administered once daily for 7 days to male rats at 200 and 500 mg/kg, before hepatotoxins. Effects of AK were assessed 24 hr later. AK pretreatments at 200 and 500 mg/kg significantly blunted GalN-induced elevation in liver lipid peroxidation, plasma aspartate-transaminase (AST) and alanine-transaminase (ALT) activities. AK also prevented, after 500 mg/kg but not after 200 mg/kg, the GalN-induced elevation in triglyceride, total cholesterol and LDL-cholesterol levels. Differently from against GalN-induced toxicity, AK did further elevate the $CCl_4$-induced rise in AST, ALT and lipid peroxidation. These results suggest that AK, when pre-administered prior to GalN, exerted protective effects against GalN-induced hepatotoxicity, in contrast however, AK exacerbated that induced by $CCl_4$. To explore possible mechanism for the toxicity-potentiating effects of AK on $CCl_4$, the activity of hepatic drug metabolism after AK treatment was assessed. It was observed that AK increased the activity of aniline hydroxaylase, a cytochrome P450 isoenzyme responsible for metabolic activation of $CCl_4$. This finding suggests that hepatoprotective effects of AK are not equally expected depending on hepatotoxins employed.

  • Application of steam distillation on the analysis of pesticide residue in soil, water and crops was studied using the nitrogen distillation apparatus. Most pesticides which were extracted by organic solvents could be analyzed by the steam distillation method. For instance, distillations of PCNB, ${\gamma}-BHC$, ${\alpha}-or$ ${\beta}-endosulfan$, IBP, diazinon, fenthion, fenitrothion, alachlor, butachlor, pretilachlor, metolachlor, pendimethalin, benthiocarb and molinate were possible, but not simazine, atrazine and nitrofen. The optimum volume of distillate for a sufficient extraction of pesticide varied according to kind of pesticide. In general, the volume needed was little for carbamate, but much more for organochlorine. When the definite amount of distillate was obtained and then the condenser was washed by acetone, the optimum volume of distillate was less. Using the steam distillation method, the amount of organic solvent needed for one extraction of pesticide from soil, water and vegetable was less than the conventional solvent extraction method, and the analytical procedure became simpler. The process of concentration and clean up was mostly unnecessary, although a ghost peak was apparent in the gas chromatogram according to the kind of materials distilled.

Effect of Light Quality during Priming and Germination Temperature on Pepper Seed Germinability (파종 전 Priming과 광질 처리, 발아온도에 따른 고추종자의 발아율)

  • Pepper (Capsicum annum L.) used as a spice or fruit vegetable has been transplanted for cultivation to accompany the production of its seedlings in general. The experiment was done to measure the effect of its cultivars (Daemyng; Wanggochu), priming (chemicals; concentration; period), light quality (red; far-red; blue; dark) during priming, and germination temperature (25 or 15$^{\circ}C$ constant; 25/15$^{\circ}C$ alternating) on the rate of germination done under incandescent lamps until 9 days after sowing. Seed germination was better when primed with Ca(NO$_3$)$_2$ than with KNO$_3$. Priming for 6 to 12 days using the former chemical enhanced the final germination rate and shortened the elapsed days to 50% germination, $T_{50}$, compared to no-priming. Under $25^{\circ}C$ constant germination temperature, primed seeds at darkness or far-red light showed the highest rate until 5 days after sowing or the lowest one since 7 days after sowing, respectively. The germination response at 25/15$^{\circ}C$ alternating temperature did not followed the lowest rate of 15$^{\circ}C$ constant germination temperature but inclined to that of $25^{\circ}C$ constant recorded the greatest one. Under 3 germination temperature red light treated during priming elevated the rate since 7 days after sowing compared to the other light treatments, and the germination rate of each cultivar was affected by no-priming or priming, light quality during priming. which imply their interaction to the seed germination.

Dietary Factors and Serum and Urinary Electrolytes Affecting Blood Pressure in Adolescents (사춘기 혈압에 영향을 미치는 식이 인자 및 혈액과 뇨중 무기 이온 농도)

  • Correlations of adolescents' blood pressure with dietary factors and blood or urinary electrolytes were investigated in this study. Through the screening for blood pressures of 960 middle school students aged 13-16 years, 30 students with the mean value of SBP and DBP above 96th percentile(high blood pressure group) and the other 30 with the mean blood pressure between 48th and 52th percentiles(normal blood pressure group) were selected as the subjects. SBP/DBP of the high and talc normal blood pressure groups were $141.8{\pm}9.0$ / $83.6{\pm}5.1$ mmHg and $116.4{\pm}3.5$ / $69.8{\pm}3.7 mmHg$, respectively. The average values of age, weight, height, BMI, degree of relative crude physical activity, and family income were not different between two groups. Among nutrients for which intakes were determined by 24 hour-recall and expressed as percent RDA, intakes of total and animal calcium were lower in the high blood pressure group compared to the normal, and were negatively correlated with both SBP and DBP even when the effects of related general and other dietary factors were excluded. Whereas energy, total, animal and vegetable protein, total and animal lipid, and riboflavin were higher in intake in the high blood pressure group than in fille normal, and they were in positive correlations with SBP and/or DBP. Vitamin A and ascorbic acid intakes were also negatively correlated with DBP, though ascorbic acid intake was not different between two groups. High blood pressure group preferred higher concentration of thin rice gruel than normal group and talc preferred salt concentrations was ill positive correlation with SBP. Serum and urinary levels of Ca, Mg, Ca/Mg ratio, Na, K and Na/K ratio did not show any differences between two groups. However serum Ca/Mg retio was in negative and se겨m Mg was ill positive correlations with SBP, and urinary Na excretion was negatively correlated with SBP. Calcium intake showed negative correlation with serum Ha and Na/K ratio. Our data indicated that dietary intake of Ca, energy, protein, lipid, vitamin A and C as well as salt preference were associated with blood pressure in adolescents. It might be assumed that Ca lowered blood pressure by increasing serum Ca/Mg ratio and decreasing serum Ca/K ratio though urinary excretion of Na.

The Association between Changes in Food and Nutrient Intakes and Changes in Bone Metabolic Indicators in Postmenopausal Women with Osteopenia after a 12-week Intervention of Nutrition Education and Aerobic Exercise (폐경 후 골감소증 여성에 대한 12주간의 영양교육과 운동 중재 전.후 식품 및 영양소 섭취량 변화와 골밀도 지표 변화와의 관계)

  • Few studies investigated the effects of nutrition education and exercises in women with osteopenia. This study examined the relationship between changes in dietary intakes and changes in indicators related to bone health in postmenopausal women with osteopenia (-2.5 ${\leq}$ T-score ${\leq}$ 1) after a 12-week intervention. Thirty-one postmenopausal women aged > 50 years residing in Seoul were recruited and participated in nutritional education regarding bone health and general nutrition practices and aerobic exercises (three times a week; 60 min per session). Twenty-five subjects completed the study and were eligible for the analysis. Bone mineral density (BMD) at femoral neck was measured by dual energy x-ray absorptiometry. Serum calcium, osteocalcin, and intact parathyroid hormone (PTH) were also measured. Dietary intake was estimated by using a one-day 24 recall by a clinical dietitian. After 12 weeks, meat consumption increased (P = 0.028) but vegetable intake decreased (P = 0.005). Intakes of animal protein (P = 0.024), vitamin B1 (P = 0.012) and vitamin $B_2$ (P = 0.047) increased, and sodium intake decreased (P = 0.033). Intact PTH (P = 0.002) decreased and osteocalcin (P = 0.000) increased, however, BMD decreased (P = 0.000). Changes in mushroom consumption were positively correlated with femoral neck BMD (r = 0.673, P = 0.003). Changes in animal iron intake were negatively correlated with intact PTH (r = -0.488, P = 0.013) but were positively correlated with osteocalcin (r = 0.541, P = 0.005). These results suggested that the association between animal iron intake and biochemical markers of bone turnover may play an important role in bone metabolism. Further studies are needed to shed light on complicated mechanisms of diet, hormonal levels of bone metabolism, and bone density.

The Current Quality Control and State of Scorched Particles in Infant formula in Korea (한국 조제분유의 초분 관리 실태 및 현황)

  • Contents of scorched particles in infant formula were studied to provide a scientific evidence for standardization of Food Regulation in Korea. The specification of scorched particles in infant formula in Korea was satisfied with CODEX, USDA and so on. But food regulations for scorched particles in baby food are not mentioned in Korea. Nowadays social interests of scorched particles in infant formula are being increased in the respect of safety for baby food. The composition and contents of scorched particles were analyzed with Scanning Electron Microscope-Energy Dispersive X-ray Spectrometer(SEM-EDS) and Inductively Coupled Plasma Mass Spectrometer(ICP-MS). The results indicate that the scorched particles consist Ca, Na, Fe, Mg and trace elements derived from the ingredient of infant formula. Infant formula are composed of milk, skimmed milk, whey, mixed vegetable oils, minerals and vitamins. These results also show that the contents of scorched particles of infant formula in Korea are little or similar level to those reported in other countries. In general, heavy metals derived from scorched particle are originally used as a source of minerals in infant formula. And it has been thought that they doesn't effect on Provisional Tolerable Weekly Intake set by FAO/WHO for contents and composition of scorched particles.

Comparison and Analysis on Characteristics for recycling of Multifarious Food Waste (음식물쓰레기의 자원화를 위한 배출업종별 성상 및 특성의 비교분석)

  • This study was executed for utilizing it as basic data in appropriate recycling way and design by examining and analyzing various characteristics of food waste which is being discharged from various restaurant and apartment house. In general, there were differences in restaurants classification and Japanese restaurant showed big differences as compared to other restaurants. Vegetable had the highest composition and its contents had big difference according to seasons. In alien substances, oyster shells were the highest and restaurants showed high rate of alien substances than apartments, therefore it was required to divide them in advance. Salinity was the highest in Japanese restaurants and most restaurants had rate of up to 1%, so it was not appropriate for recycling. However, it was considered that if there is cleaning operation in the whole process, there might not be a problem. Feed Ingredient had 28% of fiber, 25% of protein, and 11% of fats. We analyzed noxious germs and heavy metal too. However microorganisms of etiological cause were not detected and each harmful material showed less values of control concentration. In the result of analysis of each characteristic, it is required to consider collection rate from restaurants and apartments and seasonal cause into design regarding recycling. And apartments are compatible to compost than feed and food waste from Chinese, flour restaurant and collective feeding facilities are compatible for feed, as it is evaluated.

Risk Factors for Cervical Cancer in Rural Areas of Wuhan China: a Matched Case-control Study

  • Cervical cancer is a serious public health problem in developing countries. We investigated possible risk factors for cervical cancer in rural areas of Wuhan China using a matched case-control study with 33 women diagnosed with cervical cancer and 132 healthy women selected from the same area as matched controls. A questionnaire, which included questions about general demography conditions, environmental and genetic factors, the first sexual intercourse, first marriage age, age at first pregnancy, pregnancy first child's age, female personal health history, social psychological factors, dietary habits, smoking and alcohol status and other living habits was presented to all participants. At the same time, HPV infection of every participant was examined in laboratory testing. Results showed HPV infection (P<0.000, OR=23.4) and pregnancy first child's age (P<0.000, OR=13.1) to be risk factors for cervical cancer. Menopause (P=0.003, OR=0.073) was a protective factor against cervical cancer. However, there was no indication of associations of environmental (drinking water, insecticide, disinfectant) genetic (cancer family history), or life-style factors (smoking status, alcohol status, physical training, sleep quality), including dietary habits (intake of fruit and vegetable, meat, fried food, bean products and pickled food) or social psychological factors with cervical cancer. The results suggest that the risk of cervical cancer in Chinese rural women may be associated with HPV infection, menopause and the pregnancy first child's age.

Wahsing Effect of Micor-Bubbles and Changes in Quality of Lettuce (Lacutuca sativa L.) during Storage (마이크로버블에 의한 상추의 세척효과 및 저장 중 품질변화)

  • We assessed quality changes in and washing effects (time and method) on lettuce (Lactuca sativa L.) treated with micro-bubbles. Samples were treated with micro-bubbling for 1, 3, or 5 min, and the 5-min treatment yielded the best results in terms of reduced total microorganism counts, sensory aspects, and degree of washing. Total microorganism counts were 4.30 log colony-forming units (CFU)/g in unwashed lettuce(CT), 4.10 log CFU/g in hand-washed lettuce (HW), 3.98 log CFU/g in conventional, bubble-washed lettuce (BW) and 3.25 log CFU/g in micro-bubble-washed lettuce (MW). In comparison, total counts of samples examined after 10 days of storage were 7.00 log CFU/g for CT, 6.19 log CFU/g for HW, 6.02 log CFU/g for BW, and 5.89 log CFU/g for MW. The lowest counts were seen after micro-bubble treatment. BW and MW samples showed significantly higher counts than did CT and HW samples. In general, BW and MW samples did not vary significantly in count numbers. MW showed a 2.3-fold lower residual pesticide level compared with CT, and also had the lowest level of impurities. HW and BW samples were not well washed.

Comparison of Packaging Methods to Prolong the Freshness and Quality of Korean Head Cabbage (Brassica rapa). (알배추 포장 방법에 따른 품질 및 선도 비교)

  • The effect of packaging and storage methods in enhancing the shelf life and improving the postharvest quality of the Korean head cabbage (Brassica rapa) used for wrapping vegetables was studied at 10℃. The wrapping cabbage was packed using four types of packaging and storage materials: (A) Perforated PP film; (B) Non-perforated PP film; (C) PVC film for wrapping; and (D) non-packaged as control. The quality parameters, such as fresh appearance, weight loss, hue angle, moisture content, hardness, and SSC of wrapping cabbage were investigated. The weight loss of wrapping cabbage showed a significant difference between the one packaged with film and the non-packaged as control. The general appearance of Korean cabbage stored at 10℃ was not significantly affected by the packaging treatments. However, Korean head cabbage packaged with perforated film tend to show a better external appearance compared with those exposed to the other packaging during three weeks of storage at 10℃. The inside appearance, hue angle, moisture content, hardness, and SSC, gradually decreased during the storage period. No remarkable change in the measured items were observed in Korean cabbage packaging methods. In this experiment, the Korean cabbage packaged inside a PP film with holes, and stored at 10℃ temperature had the most desirable outcome of extending the head cabbage's shelf life and appearance quality. Results suggest that perforated packaging treatment combined with low storage temperature could be an effective method in prolonging the shelf life of Korean cabbage for wrapping vegetable.
