포도당 억제현상은 유전자 조작 및 induder에 의해 감소될 수 있다. UA8G와 GALl TATA box 사 이에서의 유전자 삭제는 포도당 억제현상을 줄이고 갈락토스가 존재하지 않는 조건에서 지속적인 유전자 발현을 도모했다. 상대적 inducer의 양(갈락토스/포 도당 농도의 비)은 유전자 발현 및 포도당 억제현상 에 영향을 주었다. 포도당 억제현상은 상대적 inducer의 양이 증가함에 따라 2-5배 정도 감소하였다. 또한 유전자 발현은 플라즈미드의 수에 좌우된다. 배지에 갈락토스만 았을 경우 유전자 발현은 플라즈 마드의 수가 증가함에 따라 증가하였다. 반면에 배지에 포도당과 갈락토스가 함께 있는 경우 (2% G Glu+2% Gal), 플라즈미드의 수는 유전자 발현에 별다른 영향을 주지 못했다. 그러나 높은 상대적 md ducer 양이 배지에 있는 경우 (0.4% Glu+0.8% Gal), 플라즈미드의 수가 증가함에 따라 유전자 발 현이 증가하였다. 즉, 포도당 억제현상을 줄임으로써 유전자 발현효율을 높이고자 할 때 갈락토스의 농도 를 증가시키는 경우보다는 포도당의 농도를 낮춤으 로써 상대적 inducer의 양음 높여 유전자 발현을 유 도하는 방법이 보다 효율적인 것으로 나타났다.
효모에서 대랑의 물질생산계를 구축하기 위하여 먼저 여러 종류의 베터의 이용이 가능한 다양한 영양요구성 marker를 지니며 형질전환율이 향상된 효모숙주를 선별 개량하였다. 벡터의 제작에 사용되는 프로모터로는, 효모의 여러 유전자 중에서 그 활성이 매우 높은 해당계의 효소 GAP-DH의 구조 유전자 GAP를 이용하기로 하여, 효모염색체 DNA중에서 GAP 프로모터를 분리하여 이용하기 쉽게 변형하였다. 분리된 GAT promoter의 기능을 검토하기 위하여, reporter로 APase의 구조유전자 PHO5'를 이용하여 세포내의 copy수가 상이한 발현 벡터를 제작하여 GAP 프로모터에 의한 APase의 활성 및 전사산물을 측정한 결과, 정상적인 전사가 이루어 졌으며, 효소활성도 높게 나타났으며, 벡터의 copy수에 의한 효소활성의 차이도 감지되었다.
미꾸라지(Misgrunus mizolepis)의 DNA로부터 클론된 핵기질 부착부위(MAR)를 포함하는 발현벡터인 pUC19N6-luc 벡터를 구성하였다. 이를 물고기 CHSE-214 세포주에 electroporation으로 transfection 시킨 후 유전자의 발현율, 벡터의 copy 수 및 염색체내 삽입 양상을 luciferase 활성도 분석, PCR 및 Southern blotting를 통해 분석하였다. 대조군 발현벡터에 luciferase 유전자는 전형적인 transient 발현양상을 나타내는데 비해, 미꾸라지 MAR가 포함된 pUC19N6-luc 벡터의 luciferase 유전자의 발현은 transfection 후 5일째부터 급격히 증가하는 양상을 보였다. Transfection된 CHSE-214 세포내에서 pUC19N6-luc 벡터는 대조군 벡터에 비해 높은 copy 수를 유지하였으나, 염색체내 삽입은 거의 비슷한 시간에 일어났다. 결론적으로 transfection 후 시간경과에 따른 pUC19N6-luc 벡터내의 luciferase 유전자의 발현 증가에 미치는 MAR의 효과는 벡터 copy수 증가 때문이 아니라, 염색체내 삽입후 형성되는 전사활성구조의 형성에 기인하는 것으로 판단된다.
Copy number variations (CNVs), important genetic factors for study of human diseases, may have as large of an effect on phenotype as do single nucleotide polymorphisms. Indeed, it is widely accepted that CNVs are associated with differential disease susceptibility. However, the relationships between CNVs and gene expression have not been characterized in the horse. In this study, we investigated the effects of copy number deletion in the blood and muscle transcriptomes of Thoroughbred racing horses. We identified a total of 1,246 CNVs of deletion polymorphisms using DNA re-sequencing data from 18 Thoroughbred racing horses. To discover the tendencies between CNV status and gene expression levels, we extracted CNVs of four Thoroughbred racing horses of which RNA sequencing was available. We found that 252 pairs of CNVs and genes were associated in the four horse samples. We did not observe a clear and consistent relationship between the deletion status of CNVs and gene expression levels before and after exercise in blood and muscle. However, we found some pairs of CNVs and associated genes that indicated relationships with gene expression levels: a positive relationship with genes responsible for membrane structure or cytoskeleton and a negative relationship with genes involved in disease. This study will lead to conceptual advances in understanding the relationship between CNVs and global gene expression in the horse.

Several transgenic marine medaka Oryzias dancena strains harboring a green fluorescent protein (GFP) reporter construct regulated by an endogenous ${\beta}$-actin promoter were established and their expression characteristics in relation to transgene copy numbers were examined in 21 transgene genotypes. Most of the transgenic strains displayed transgene insertion patterns typical of microinjection-mediated introduction of foreign DNA into fish embryos, characterized by the random integration of multiple transgene copies (ranging from 1 - 282 copies per cell), often accompanied by the formation of concatemer(s), as assessed by genomic Southern blot hybridization analysis and qPCR. Transgenic strains showed ubiquitous and continued temporal and spatial expression patterns of the transgenic GFP during most of their life cycle, from the embryonic stage to adulthood, enabling assessment of the expression pattern of the endogenous ${\beta}$-actin gene. However, a comparative evaluation of transgene copy numbers and expression levels showed that copy number-dependent expression, the stability of the ubiquitous distribution and expression efficiency per transgene copy varied among the transgenic strains. Fluorescence expression levels were positively correlated with absolute transgene copy numbers, whereas the expression efficiency per transgene copy was inversely related to the number of transgene integrant copies. Data from this study will guide the selection of potentially desirable transgenic strains with ubiquitous expression of a fluorescent transgene, not only in this marine medaka species but also in other related model fish species.
The SecY is a central component of the protein export machinery that mediate the translocation of secretory proteins across the plasma membrane, and has been known to be rate-limiting factor of secretion in Escherichia coli. In order to study the extracellular protein secretion in Gram-positive microorganism, we have, constructed strains harboring more than one copy of the gene for SecY. Firstly, the gene, for B. subtilis SecY and its promoter region was subcloned into pDH32 and the chimeric vector was inserted into amyE locus by homologous recombination. Secondly, low copy number vector, pCED6, was also used for subcloning the secY gene and for constructing a strain which harbors several copies of secY. The KH1 cell which harbor two copies of secY on the chromosome excreted more extracellular proteins than the wild type PB2. Moreover, the KH2 cells which harbor several copies of secY in pCED6 vector excreted more extracellular proteins than the KH1 cells. Here, we found that the capacity of protein secretion is partly controlled by the number of secY and it is suggested that SecY has also an important role in protein secretion in B. subtilis, a gram positive microorganism, as like in E. coli. This will promote the use of B. subtilis as a host for the expression of useful foreign gene and excretion of precious proteins.
TAR(Transformation-Associated Recombination) cloning 법은 복잡한 고등생물의 게놈으로부터 유전자나 특정 염색체 부위를 선별적 분리를 가능하게 한다. 이 방법은 목적으로 하는 염색체 부위의 주변에 존재하는 비교적 짧은 게놈 염기서열에 대한 정보를 필요로 하며, spheroplast 형질전환 과정에서 게놈 DNA와 5'-와 3'-표적배열을 지닌 TAR vector 사이에서 일어나는 상동성 재조합과정에 의해 이루어진다. 본 연구에서는 single-copy 유전자의 클론닝에 필요한 specific hook의 최소 크기를 조사하였고, 서로 다른 TAR vector(radial과 unique vector)의 유용성을 조사하기 위해 동일한 single-copy 유전자의 클론닝을 통해 비교하였다. 그 결과, hHPRT 유전자에 대한 TAR cloning의 빈도는 hook의 길이가 750 bp∼63 bp의 범위에서 동일하게 나타났다. radial hook을 사용한 경우보다 unique hook을 사용하였을 경우 형질전환체의 수는 약 20배정도의 감소를 보였으나 목적으로 하는 재조합체의 분리 빈도는 두 배 이상 증가하였다. 그러므로 본 연구에서 two-unique TAR vector는 선별력이 높으므로 일반적 TAR cloning에 사용할 수 있으며, radial TAR vector의 경우는 병리학적 표본과 같이 제한된 게놈 DNA를 사용하는 경우에 더 적합하다고 볼 수 있다. 또한, single-copy 유전자의 분리에 필요로 하는 specific hoot의 최소 길이는 약 60 bp로도 가능하다는 것을 확인하였다.
전분 분해능력을 갖는 알콜생산용 효모를 만들기 위해 Saccharomyces cerevisiae에 glucoamylase 유전자인 STA를 도입하였다. 도입된 형질의 발현증대를 위해 STA 유전자의 promoter 부위를 alcohol dehydrogenase isoenzyme I 유전자의 promoter 부위와 치환 시켜준 재조합 플라스미드를 재조하였으며 안정성을 증진시키기 위해 centrometer 부위를 치환시킨 결과 glucoamylase의 발현이 증가하였으며, STA 유전자와 centromere를 갖고 있는 재조합 플라스미드는 여러세대가 거듭되어도 비교적 안정하게 유지되었으나 낮은 copy 수로 인해 형질전환체의 효소 역가와 형질전환 빈도는 낮아졌다. STA 유전자가 도입되어 형질전환된 다배체 산업용 효모는 액화 과정만을 거친 주정생산 배지(액화액)에서 원래의 알콜 생산용 효모에 비해 훨씬 많은 양의 알콜을 생산해 내었다. 그러나 centromere를 보유하는 플라스미드에의한 산업용 효모의 형질전환에는 실패하였다.

Copy number variations(CNVs) are known as one of the most important factors in susceptibility to genetic disorders because they affect expression levels of genes. In previous studies, pyrosequencing, mini-sequencing real-time polymerase chain reaction(PCR), invader assays and other techniques have been used to detect CNVs. However, the higher the copy number in a genome, the more difficult it is to resolve the copies, so a more accurate method for measuring CNVs and assigning genotype is needed. PCR followed by a quantitative oligonucleotide ligation assay(qOLA) was developed for quantifying CNVs. The aim of this study was to compare the two methods for detecting and quantifying the CNVs of duplicated gene: the published pyrosequencing assay(pyro_CNV) and the newly developed qOLA_CNV. The accuracy and precision of the assay were evaluated for porcine KIT, which was selected as a model locus. Overall, the root mean squares(RMSs) of bias and standard deviation of qOLA_CNV were 2.09 and 0.45, respectively. These values are less than half of those of pyro CNV.
