Development of mouse fetus brains can be defined morphologically and functionally by three developmental stages, embryo day (ED) 16, postnatal stage one week and eight weeks. These defined stages of brain development may be closely associated with differential gene expression rates due to limited cellular resources such as energy, space, and free water. Complex patterns of expressed genes and proteins during brain development suggests the changes in relative concentrations of proteins rather than the increase in numbers of new gene products. This study was designed to evaluate early protein expression pattern in mouse fetus brain. The mouse brain proteome of fetus at ED 15.5, and 19.5 was obtained using 2-dimensional gel electrophoresis (DE). Analysis of the 2-DE gels in pH 3-10 range revealed the presence of 15 differentially expressed spots, of which 11 spots were identified to be known proteins following MALDI-TOF analysis; 3 spots were up-regulated and 8 spots were down-regulated in the mouse fetus brain at ED 15.5. UP-regulated proteins were identified as MCG18238, isoform M2 of pyruvate kinase isozymes M1/M2, isoform 2 of heterogeneous nuclear ribonucleoprotein K, heterogeneous nuclear ribonucleoprotein H2, creatine kinase B-type, 40S ribosomal protein SA and hemoglobin subunit beta-H1. Down-regulated proteins were putative uncharacterized protein, lactoylglutathione lyase and secreted acidic cysteine rich glycoprotein. Our results revealed composite profiles of mouse fetus brain proteins related to mouse fetus development by 2-DE analysis implying possible roles of these proteins in neural differentiation.
A critical element in tissue engineering approaches is a control of the mechanical properties of polymer scaffolds to regulate cell phenotype, which may lead to clinically successful tissue regeneration. In this study, we hypothesized that gel stiffness could be a key factor to manipulate adhesion and proliferation of different types of cells. RGD-modified alginate gels with various mechanical properties were prepared and used as a substrate for MC3T3-E1 and H9C2 cells. Adhesion and growth rate of MC3T3-E1 cells in vitro were increased in parallel with an increase of gel stiffness. In contrast, those of H9C2 cells were decreased. This approach to control the mechanical properties of polymer scaffolds depending on the cell types may find useful applications in the tissue engineering.
Cyst formation is a major characteristic of ADPKD and is caused by the abnormal proliferation of epithelial cells. Renal cyst formation disrupts renal function and induces diverse complications. The mechanism of cyst formation is unclear. mIMCD-3 cells were established to develop simple epithelial cell cysts in 3-D culture. We confirmed previously that Mxi1 plays a role in cyst formation in Mxi1-deficient mice. Cysts in Mxi1 transfectanted cells were showed by collagen or mebiol gels in 3-D cell culture system. Causative genes of ADPKD were measured by q RT-PCR. Herein, Mxi1 transfectants rarely formed a simple epithelial cyst and induced cell death. Overexpression of Mxi1 resulted in a decrease in the PKD1, PKD2 and c-myc mRNA relating to the pathway of cyst formation. These data indicate that Mxi1 influences cyst formation of mIMCD-3 cells in 3-D culture and that Mxi1 may control the mechanism of renal cyst formation.
The inheritance of isoenzymes in young coat tissues of trembling aspen (Populus tremuloides Michx.) was studied by electrophoretically analysing five parental clones and their full-sib progenies obtained by crossing one female clone to four male clones. The distal 2cm section of vigorous young roots of 70 seedlings per family were subjected to horizontal starch gel electrophoresis. The resulting gels were tested for activity of 7 enzyme systems. Evidences for the inheritance of isoenzymes observed indicated that the isoenzyme variants of every isoenzyme zone were under control of codominant alleles at a single locus. Chi-square test of joint segregation data of the two loci, 6-PGD-2 and PGI-2, indicated that the pair of loci was not linked.
This paper studies the effect of the compressive strength for combined alkali-activated slag mortars. The effect of activators such as alkali type and dosage factor on the strength was investigated. The alkalis combinations made using five caustic alkalis (sodium hydroxide (NaOH, A series), calcium hydroxide ($Ca(OH)_2$, B series), magnesium hydroxide ($Mg(OH)_2$, C series), aluminum hydroxide ($Al(OH)_3$, D series), and potassium hydroxide (KOH, E series)) with sodium carbonate ($Na_2CO_3$) were evaluated. The mixtures were combined in different dosage at 1M, 2M, and 3M. The study results showed that the compressive strength of combined alkali-activated slag mortars tended to increase with increasing sodium carbonate. The strength of combined alkali-activated slag mortars was better than that of control cases (without sodium carbonate). The result from scanning electron microscopy (SEM) analysis confirmed that there were reaction products of calcium silicate hydrate (C-S-H) and alumina-silicate gels from combined alkali-activated slag specimens.
The purpose of the study were to characterize the proteins secreted by elongating caprine conceptus, to identify a group of low molecular weight proteins as retinol-binding protein (RBP), to identify RBP cell-specific localization in conceptus tissue, and to demonstrate that the conceptuses secreted continuously RBP during the time period maternal recognition of pregnancy. Caprine conceptuses were removed from the uterus between days 16 and 22 of pregnancy, the time period maternal recognition of pregnancy. Isolated conceptuses were cultured in a modified minimum essential medium in the presence of radiolabeled amino acids. Proteins synthesized and secreted into medium were analyzed by fluorography of two-dimensional polyacrylamide gel electrophoresis and fluorography. At least five proteins showed consistently a grouping of spots with characteristic location on two-dimensional gels. A major low molecular weight protein consisted of two major isoforms (pI 5.3-6.0) of similar molecular mass (21 kDa) was identified as RBP by using antiserum against RBP. Presence of RBP in conceptus culture medium and uterine flushings between days 16 and 22 of pregnancy were determined by immunoprecipitation and Western blotting using anti-RBP serum. In immunocytochemical study, strong immunostaining for RBP was localized in trophectoderm and endoderm of conceptus. These results clearly demonstrated that the caprine conceptus was active in protein synthesis as early as day 16 of pregnancy. Secretion of RBP by caprine conceptuses (days 16-22) coincident with the rapid transformation of the conceptus from a spherical blastocyst to a filamentous structure. Production of RBP by the elongating conceptuses may be indicative of an important role for conceptus RBP in the transport, availability and metabolism of retinol during maternal recognition of pregnancy.
Precursor gels with the composition of $xZrO_2{\cdot}(100-x)SiO_2$ systems (x=10, 20 and 30 mol%) were prepared by the sol-gel method. Kinetic parameters, such as activation energy, Avrami's exponent, n, and dimensionality crystal growth value, m, have been simultaneously calculated from the DTA data using Kissinger and Matusita equations. The crystallite size dependence on tetragonal to monoclinic transformation of $ZrO_2$ was investigated using XRD, in relation to the fracture toughness. The crystallization of tetragonal $ZrO_2$ occurred through 3-dimensional diffusion controlled growth(n=m=2) and the activation energy for crystallization was calculated using Kissinger and Matusita equations, as about $310{\sim}325{\pm}10kJ/mol$. The growth of $t-ZrO_2$, in proportion to the cube of radius, increased with increasing heating temperature and heat-treatment time. It was suggested that the diffusion of Zr4+ions by Ostwald ripening was rate-limiting process for the growth of $t-ZrO_2$ crystallite size. The fracture toughness of $xZrO_2{\cdot}(100-x)SiO_2$ systems glass ceramics increased with increasing crystallite size of $t-ZrO_2$. The fracture toughness of $30ZrO_2{\cdot}70SiO_2$ system glass ceramics heated at $1,100^{\circ}C$ for 5 h was $4.84Mpam^{1/2}$ at a critical crystaliite size of 40 nm.

Conventional geotechnical engineering soil binders such as ordinary cement or lime have environmental issues in terms of sustainable development. Thus, environmentally friendly materials have attracted considerable interest in modern geotechnical engineering. Microbial biopolymers are being actively developed in order to improve geotechnical engineering properties such as aggregate stability, strength, and hydraulic conductivity of various soil types. This study evaluates the geotechnical engineering shear behavior of sand treated with xanthan gum biopolymer through laboratory direct shear testing. Xanthan gum-sand mixtures with various xanthan gum content (percent to the mass of sand) and gel phases (initial, dried, and re-submerged) were considered. Xanthan gum content of 1.0% sufficiently improves the inter-particle cohesion of cohesionless sands 3.8 times and more (up to 14 times for dried state) than in the untreated (natural) condition, regardless of the xanthan gum gel condition. In general, the strength of xanthan gum-treated sand shows dependency with the rheology and phase of xanthan gum gels in inter-granular pores, which decreases in order as dried (biofilm state), initial (uniform hydrogel), and re-submerged (swollen hydrogel after drying) states. As xanthan gum hydrogels are pseudo-plastic, both inter-particle friction angle and cohesion of xanthan gum-treated sand decrease with water adsorbed swelling at large strain levels. However, for 2% xanthan gum-treated sands, the re-submerged state shows a higher strength than the initial state due to the gradual and non-uniform swelling behavior of highly concentrated biofilms.

The non-Newtonian flow curves of sodium bis-(2-ethylhexyl)sulfosuccinate-water lamellar liquid crystals were obtained in various concentrations and temperatures by using a cone-plate rheometer. By applying non-Newtonian flow equation to the flow curves for AOT-water lamellar liquid crystal samples, the rheological parameters were obtained. Particular attention is given to the hysteresis loop detected when the liquid crystal samples are shear under increasing-decreasing shear stress modes which result in thixotropic and dilatant behavior. Sodium bis-(2-ethylhexyl)sulfosuccinate-water lamellar liquid crystals behave as weak gels when they are subjected to shear flow, but when the applied stress surpasses the yield stress, they exhibit non-linear viscoelasticity. Upon decreasing shear rate, the dispersion still preserves much of its structure and consequently its shear stress remains higher than the values measured in the increasing shear rate mode.
Porcine beta-defensin-1 (PBD-1) gene plays an important role in the innate immunity of pigs. The peptide encoded by this gene is an antimicrobial peptide that has direct activity against a wide range of microbes. This peptide is involved in the co-creation of an antimicrobial barrier in the oral cavity of pigs. The objective of the present study was to detect polymorphisms, if any, in exon-1 and exon-2 regions of PBD-1 gene in Large White Yorkshire (LWY) and native Ankamali pigs of Kerala, India. Blood samples were collected from 100 pigs and genomic DNA was isolated using phenol chloroform method. The quantity of DNA was assessed in a spectrophotometer and quality by gel electrophoresis. Exon-1 and exon-2 regions of PBD-1 gene were amplified by polymerase chain reaction (PCR) and the products were subjected to single strand conformation polymorphism (SSCP) analysis. Subsequent silver staining of the polyacrylamide gels revealed three unique SSCP banding patterns in each of the two exons. The presence of single nucleotide polymorphisms (SNPs) was confirmed by nucleotide sequencing of the PCR products. A novel SNP was found in the 5'-UTR region of exon-1 and a SNP was detected in the mature peptide coding region of exon-2. In exon-1, the pooled population frequencies of GG, GT, and TT genotypes were 0.67, 0.30, and 0.03, respectively. GG genotype was predominant in both the breeds whereas TT genotype was not detected in LWY breed. Similarly, in exon-2, the pooled population frequencies of AA, AG, and GG genotypes were 0.50, 0.27, and 0.23, respectively. AA genotype was predominant in LWY pigs whereas GG genotype was predominant in native pigs. These results suggest that there exists a considerable genetic variation at PBD-1 locus and further association studies may help in development of a PCR based genotyping test to select pigs with better immunity.
