Wheat wild relatives that have never been domesticated contained useful genetic resources such as the resistance to abiotic and biotic stresses. Leymus racemosus is one of the wild species. It can grow in a harsh environment like seaside and distribute by healthy rhizomes. Also, it has a useful genetic resource such as salt tolerance and different diseases resistance. Wheat (Triticum aestivum L. cv. Chinese Spring; CS) was crossed with L. racemosus. Wheat-L. racemosus disomic addition lines were produced. The purpose of this study is to identify protein expression in each disomic addition line compared to CS. We performed two-dimensional electrophoresis. Two-dimensional gels stained with coomassie brilliant blue (CBB), a total of 1566 differentially expressed proteins were identified by Progenesis Same Spots software from the cultivars. However, a total of 90 protein spots were identified to be either present or absent or showing significantly differential expression when the difference threshold was set to more than 1.5 fold. However, out of the 90 differentially protein spots, a total of 74 spots were sorted for mass spectrometry analysis. The identified proteins may provide important clues for better understanding the molecular changes in the chromosomes carrying Leymus racemosus.
Antibodies were raised against fusion proteins of the N-terminus and a region containing the GDNQ (Gly-Asp-Asn-Gln) polymerase motif of the L (polymerase) protein of sonchus yellow net virus (SYNV). Immunoblot analyses using these antibodies revealed the presence of the L protein in purified SYNV preparations and in nuclear extracts from infected tobacco. The serological analyses and detection in a polyacrylamide gels suggested that the L protein is present in at least a 20 fold lower abundance than the G, N, M1 and M2 proteins, and has size corresponding to a molecular weight of over 200 kDa as predicted from nucleotide sequence data. Electron microscopy with gold-labelled antibodies was used to localize the N, M2, and G proteins of SYNV in thin sections of infected tissue. When sections of SYNV-infected tissue were treated with antisera against total SYNV proteins and N protein, gold label could be detected in both the viroplasms and in virus particles. With the anti-M2 protein antiserum, the gold label was strongly localized in the viroplasms but only limited labelling of the virus particle sonly. Limited labelling of the L protein was observed in the viroplasms and the virus particles, presumably because of the low abundance of L protein in the tissues.
Wound healing and antibacterial effects of solcoseryl-micronomicin combination gel on an open wound were studied in mice. A simple model was designed for assessing the effects. Using the model, we compared the efficacy of a combined topical gel of solcoseryl and micronomicin with those gels of solcoseryl or micronomicin alone. From the results of our experiment, the wound healing effect of open wounds by treatment with the combination gel was significantly enhanced when compared with those by treatment with solcoseryl gel or micronomicin gel alone. And the antibacterial effect of the combination gel was higher than those of solcoseryl gel or micronomicin gel alone.
LAS (lithium aluminosilicate) sol was synthesized using the hydrolysis-condensation reaction of TEOS, chelated Al(OBus)3 and LiNO3 with H2O in alcohol (ethanol+2-propanol) medium. Lowering Li content by a factor of 1/2 significantly enhanced densification and retarded the crystallization of LAS gel by ~30$0^{\circ}C$. Dense LAS specimen with essentially pore-free microstructure was obtained by sintering the sol-gel derived gel at 80$0^{\circ}C$ for 4 h and annealing at 120$0^{\circ}C$ for 2 h. Similary, a mixed colloidal processing was attempted as a convenient, alternative route for the fabrication of dense LAS sintered body. The $\beta$-spodumene seeding (~0.8 ${\mu}{\textrm}{m}$) in the sol-gel derived LAS modified the sequence of phase transformations and lowered the temperature of crystallization by ~12$0^{\circ}C$. Combining the epitaxial seeding with the sol-gel process, we could lower the crystallization temperature to the sintering temperature range (~80$0^{\circ}C$) and, demonstrate a possibility of making the viscous sintering/crystallization as a continuous as a continuous unit process.
In order to improve the performance of green emitting phosphors for plasma display panel, the $Zn_2SiO_4:Mn,Al$ phosphors were synthesized using sol-gel technique and studied using SEM and VUV photoluminescence spectrometer. pH values of the starting solutions (pH = 0.5$\~$2.34) were controled by HCl as the catalysis of hydrolysis and wet gels were dried at $80^{\circ}C$ and $120^{\circ}C$, respectively. We investigated the effects of pH and drying temperatures during sol-gel processes. The results indicated that the phosphor prepared at pH = 1 showed the maximum emission intensity in both drying conditions and the effect of pH of the starting solution on morphology were increased with particle size as HCl and phosphor dried at high temperature showed more spherical and smaller particles than at low.

Undergoing large volumetric changes upon incremental environmental stimulation, hydrogels are interesting materials which hold immense potentials for utilization in a wide array of applications in diverse industries. Owing to the large magnitudes of deformation it undergoes, swelling induced instability is a commonly observed sight in all types of gels. In this work, we investigate the instability of photo-thermal sensitive hydrogels, produced by impregnating light absorbing nano-particles into the polymer network of a temperature sensitive hydrogel, such as PNIPAM. Earlier works have shown that by using lights of different intensities, these hydrogels follow different swelling trends. We investigate the possibility of utilizing this fact for remote switching applications. The analysis is built on a thermodynamic framework of inhomogeneous large deformation of hydrogels and implemented via commercial finite element software, ABAQUS. Various examples of swelling induced instabilities, and its corresponding dependence on light intensity, will be investigated. We show that the instabilities that arise have their morphologies dependent on the light intensity.
The effects of salt concentration and heating conditions on the thermal gelation properties of surimi produced from mechanically deboned chicken meat (MDCM) were investigated. Chicken surimi was manufactured by washing (MDCM: 0.5% NaCl=1:4), standing, straining and centrifuging. The fat, water-soluble protein and heme pigment in the MDCM were removed by increasing washing cycles. The compressive force of the chicken surimi increased as the concentration of salt was increased from 0% to 5%. Total gel strength of the surimi measured by texture profile analysis showed a maximum in the range 3-5% NaCl. Microstructural analysis showed that the unfolding network structure of the surimi gel began to appear at NaCl concentrations>2%. The optimum heating condition for gelation was $90^{\circ}C$ for 40 min as this resulted in maximum values for measures of gel strength including compressive force, hardness, fracturability, adhesiveness, springiness, gumminess, chewiness and resilience. Chicken surimi gel formed by cooking at a heating rate of $1^{\circ}C/min$ to $90^{\circ}C$ showed better a texture than gels produced at $1.85^{\circ}C/min$. Our result show that a lower rate of heating improves chicken surimi gelation.
In vitro analyses of type I signal peptidase activities require protein precursors as substrates. Usually, these pre-proteins are expressed in vitro and cleavage of the signal sequence is followed by SDS polyacrylamide gel electrophoresis coupled with autoradiography. Radioactive amino acids have to be incorporated in the expressed protein, since the amount of the in vitro expressed protein is usually very low and processing of the signal peptide cannot be followed by SDS polyacrylamide gel electrophoresis alone. Here we describe a rapid and simple method to express large amounts of a protein precursor in E. coli. We have analyzed the effect of ionophors as well as of azide on the accumulation of expressed protein precursors. Azide blocks the function of SecA and the ionophors dissipate the electrochemical gradient across the cytoplasmic membrane of E. coli. Addition of azide ions resulted in the formation of inclusion bodies, highly enriched with pre-apo-plastocyanine. Plastocyanine is a soluble copper protein, which can be found in the periplasmic space of cyanobacteria as well as in the thylakoid lumen of cyanobacteria and chloroplasts, and the pre-protein contains a cleavable signal sequence at its N-terminus. After purification of cyanobacterial pre-apo-plastocyanine, its signal sequence can be cleaved off by the E. coli signal peptidase, and protein processing was followed on Coomassie stained SDS polyacrylamide gels. We are optimistic that the presented method can be further developed and applied.
Purpose: Both silica gel and activated carbon black particles were adopted for use as PCM absorbed porous media applicable as construction materials. To investigate usable methods for absorbing PCM into the media, they were soaked into PCM and also tested for enhancement of PCM absorption into them. Method: To test PCM absorption into some porous media such as both ${\varphi}1{\sim}2mm$ and $10{\mu}m$ silica gels, and $50{\mu}m$ activated carbon black, $43^{\circ}C$ PCM was used as a laten heat material. The method, soaking into PCM was applied to this study, and the media were moderately rotated by centrifuge to have the extra PCM flow out. DSC analysis was conducted to investigate the melting and solidifying of the PCM absorbed into the porous media. Result: It was found that PCM was absorbed into the porous media by over 85 wt% of all particles. In addition, it was noted that the ultrasonic vibrator was accelerating the PCM absorption into the particles to three times higher speed than simple soaking. Centrifuge was adopted to remove extra PCM sticking on the particle surfaces and extra PCM was moderately removed from the surfaces of the particles. DSC analysis indicated that the latent heat of the absorbed PCM particles was 160 J/g, and the melting temperature was approximately $40^{\circ}C{\sim}50^{\circ}C$.

Immobilization technique by PVA encapsulation is an effective alternative for wastewater treatment. However little information is available about the effect of PVA characteristics on physical properties as an encapsulation media. This study aims at investigating the effect of the preparation methods of PVA and filler addition to media on the solubility of PVA. The solubility decreased as the freezing temperature decreased and the vacuum-drying was applied. Addition of the PAC (Powered Activated Carbon) and organoclay decreased the solubility of the PVA gels. Organoclay was more effective for lowering the solubility about 25% than the PAC. Nitrification with the PVA-coating media was less sufficient than with the polyurethane media due to the mass transfer restriction for oxygen and nutrients.
