This study was conducted to investigate the genetic analysis of esterase isozymes in maize with tillers. The materials used for the study were stele tissue for five day old seedlings of IK inbred line with tiller and A-type inbred line with no tiller, their Fl and F,. The methods employed for the study were same as previous report by Lee and Choe. A total of thirteen isoesterase enzyme bands were identified and five zones were distinguised according to both migration distance and genetic segregation patterns. The E$\sub$0.3/, E$\sub$0.4/ and E$\sub$0.5/ loci appeared from orgin to 0.5cm migration distance were controlled by the two alleles in (IK/A-type)F$_2$ and the E$\sub$0.3/+E$\sub$0.4/ of variants was controlled by codominant alleles. The E$\sub$1.0/, E$\sub$1.2/, E$\sub$1.5/ and E$\sub$1.8/ loci appeared from 1.0cm to 1.8cm were also controlled by the two alleles. However, the null band was functioned alleles. The E$\sub$2.8/, E$\sub$3.0/ and E$\sub$3.5/ loci appeared from 2.8cm to 3.5cm migration distance were very active and near location. A total of individuals with two paried bands of these loci were more than those of three paired bands(x$^2$=0.327$\^$**/). The activity of bands appeared over 3.8cm were very low and these were controlled by the two alleles. In above results, genetic segregations of stele tissue of maize with tillers were suggested to be controlled by Mendelian genetic laws.
Background: Combining risk factors for prostate cancer into a predictive tool may improve the detection of prostate cancer while decreasing the number of benign biopsies. We compare one such tool, age multiplied by prostate volume divided by total serum PSA (PSA-AV) with PSA density and detection of primary malignant circulating prostate cells (CPCs) in a Chilean prostate cancer screening program. The objectives were not only to determine the predictive values of each, but to determine the number of clinically significant cancers that would have been detected or missed. Materials and Methods: A prospective study was conducted of all men undergoing 12 core ultrasound guided prostate biopsy for suspicion of cancer attending the Hospital DIPRECA and Hospital de Carabineros de Chile. Total serum PSA was registered, prostate volumecalculated at the moment of biopsy, and an 8ml blood simple taken immediately before the biopsy procedure. Mononuclear cells were obtained from the blood simple using differential gel centrifugation and CPCs identified using immunocytchemistry with anti-PSA and anti-P504S. Biopsy results were classed as positive or negative for cancer and if positive the Gleason score, number of positive cores and percent infiltration recorded. Results: A total of 664 men participated, of whom 234 (35.2%) had cancer detected. They were older, had higher mean PSA, PSA density and lower PSA-AV. Detection of CPCs had high predictive score, sensitivity, sensibility and positive and negative predictive values, PSA-AV was not significantly different from PSA density in this population. The use of CPC detection avoided more biopsies and missed fewer significant cancers. Conclusions: In this screening population the use of CPC detection predicted the presence of clinically significant prostate cancer better than the other parameters. The high negative predictive value would allow men CPC negative to avoid biopsy but remain in follow up. The formula PSA-AV did not add to the predictive performance using PSA density.
Introduction: In the mass spectrometry-based proteomics, biological samples are analyzed to identify proteins by mass spectrometer and database search. Database search is the process to select the best matches to the experimental mass spectra among the amino acid sequence database and we identify the protein as the matched sequence. The match score is defined to find the matches from the database and declare the highest scored hit as the most probable protein. According to the score definition, search result varies. In this study, the difference among search results of different search engines or different databases was investigated, in order to suggest a better way to identify more proteins with higher reliability. Materials and Methods: The protein extract of human mesenchymal stem cell was separated by several bands by one-dimensional electrophorysis. One-dimensional gel was excised one by one, digested by trypsin and analyzed by a mass spectrometer, FT LTQ. The tandem mass (MS/MS) spectra of peptide ions were applied to the database search of X!Tandem, Mascot and Sequest search engines with IPI human database and SwissProt database. The search result was filtered by several threshold probability values of the Trans-Proteomic Pipeline (TPP) of the Institute for Systems Biology. The analysis of the output which was generated from TPP was performed. Results and Discussion: For each MS/MS spectrum, the peptide sequences which were identified from different conditions such as search engines, threshold probability, and sequence database were compared. The main difference of peptide identification at high threshold probability was caused by not the difference of sequence database but the difference of the score. As the threshold probability decreases, the missed peptides appeared. Conversely, in the extremely high threshold level, we missed many true assignments. Conclusion and Prospects: The different identification result of the search engines was mainly caused by the different scoring algorithms. Usually in proteomics high-scored peptides are selected and low-scored peptides are discarded. Many of them are true negatives. By integrating the search results from different parameter and different search engines, the protein identification process can be improved.
The fibrinolytic activities of soluble proteins extracted from young leaves of Camellia japonica L. were studied. Fibrinolvity activity of extract from partitions of C. japonica L. showed 1.6-2.0 times higher than plasmin used as positive control. The fibrinolytic enzyme was confirmed directly from young leaves of C. japonica L. by a fibrin Plate and fibrin zymography. The protein was composed of a single polypeptide and its apparent molecular weight was found to be 45 kDa, as judged by SDS-polyacrylamide gel electrophoresis. The optimum pH and temperature for the fibrinolytic activity were pH 5.5 and $30^{\circ}C$, respectively. Also, the fibrinolytic activity was clearly inhibited by PMSF and TLCK, suggesting that it is a member of the trypsin-like serine protease. All these results suggest the protease is a fibrinolytic enzyme belong to a family of trypsin-like serine protease.
The objectives of this research were to (1) identify the membrane fouling potential due to different fractions of NOM (2) correlate the physicochemical properties of NOM and membranes with the adsorption of humic substances on membrane (4) find out the effect of membrane physical and chemical washing according to membrane material. The static adsorption test and adsorption test showed that hydrophobic organics adsorbed much more quickly than hydrophilic organics. In case of the effect of membrane properties on the adsorption of organic fractions, the adsorption rate ratio(a) of hydrophobic membrane (0.016, 0.077) was greater than that of hydrophilic membrane (0.010, 0.033) regardless of the kind of organic fractions. This suggests that the UF membrane fouling were occurred mainly by internal pore size decreasing due to adsorption of organic into pore surface for hydrophobic membrane, and by sieving of organics and forming a gel layer on the membrane surface for hydrophilic membrane. In conclusion, the decrease in the pore volume, which was caused by the organic adsorption into the internal pore, was greater with the hydrophobic membrane than with the hydrophilic membrane. In case of the effect of membrane properties on permeate flux, the rate of flux decline for the hydrophobic membrane was significantly greater than that for the hydrophilic membrane.

The purposes of this study are to confirm ways to reduce bacteria of minimally processed agricultural products, using antibacterial foods that are easily available in home and to improve bacterial hygienic condition of them. We chose garlic-allicin, ginger-gingerol, green tea-catechin, cinnamon-cinnamic aldehyde, wasabi-allyl isothiocyanate as antibacterial foods and their unique antibiotic materials. We confirmed the better washing effect when these antibiotic extracts were used, compared to washing effect by only distilled water. Their antibiosis was proved by statistical processing. PFGE (Pulsed Field Gel Electrophoresis) of Bacillus cereus shows continuous contamination probability of minimally processed vegetables by same product suppliers and the necessity of systematic measures against bacterial contamination.

The recycling rate of recovered paper in Korea is the highest in the world, 92%, but remanufacturing yield is low due to the extremely poor quality of the paper. The poor quality, in turn, influences to the reject amount in deinking process. To increase the yield of old newspaper recycling process, hydrophobic degree of inorganic pigments of deinking stock must be reduced. To determine the hydrophobicity, Pitch Potential Deposit Tester (PDT) was newly designed and applied with respect to the SB latex property of various quality used in Korea; its hydrophobic degree according to Tg, gel content, charge and particle size of latex and optimum designing condition of SB latex. And below are the conclusions: 1. The reason of excessive reject from old newspaper deinking process for total amount of printed ink is loss of inorganic pigments. When lipase, a biochemical catalyst, was applied with the purpose of preventing inorganic pigments loss about more than 70% of total reject weight and promoting hydration of pulp for deinking, deinking process yield of pre flotation secondary stage increased remarkably without any changes of deinking efficiency. 2. Lipase improved deinking stock by cutting ester linkage on surface of hydrophobic materials to promote its hydration. From this, it reached the conclusion that hydration degree of stock exercises significant effect on flotation deinking process yield. 3. Inorganic alkali promotes hydration of deinking stock. But there have been needs for more fundamental measures other than inorganic alkali of promoting hydration for yield improvement. For this, this study intended to find out reasons of chemical properties change on surface of hydrophobic material by change of pH. 4. Pitch Deposit Test (PDT) was performed for understanding principle of why surface of coating flake from OMG is hydrophobic and why it becomes hydrophilic when pH of stock is alkaline. As a result of this test, it is determined that swelling property by change of pH of latex film, which were used as coating adhesive is the reason for hydrophobic change. 5. Hydrophilicity of coating flake increased with hydrophilic pigments. And as more of SB Latex adhesive was used and higher of calcium hardness of stock became, its hydrophilicity decreased. SB Latex adhesive film is reformed by mechanical friction. For having hydrophilicity under neutral pH, strong bruising action such as kneading is required. 6. Because swelling of adhesive film decreases as Tg of SB latex gets lower and mean diameter gets smaller, it shows hydrophobicity under neutral pH. This lowers hydrophilicity of coating flake, which leads to easy elimination with flotation reject on DIP process. Therefore, for improving future flotation yield, it is necessary to develop to use eco-friendly clean SB latex by raising Tg and increasing mean diameter for recycling, and as a result, to reduce excessive loss of coating flake as a reject from deinking process.
This study was carried out to extract the natural antioxidants from Bovine bile and to investigate their effects on various antioxidant activities. It also characterized the patterns of antioxidants by GC/FID and GC/MS. The antioxidative activities and chemical structure of the antioxidant were elucidated by examining the effects of biological activity and the analysis of GC/MS. The antioxidant materials extracted from bovine bile were isolated and purified by silica gel column chromatography and TLC. It was confirmed that there were effects of antioxidants such as Xanthine Oxidase(XO) and Glutathione-S-Transferase(GSH-T) on antioxidative activities. When they were compared with BHT, bile extracts showed the relative effects of 51.2% on the antioxidant activity, the inhibition effects of 48.3% on XO activity, and the synergism effects of 85.7% on the GSH-T activity. According to the results of investigation at neuron cell of mouse, the rate of cell activity in the treatment of 6mM glutathione was 96%, While it in the treatment of 140mg of bile extract was 78%. Based on the TLC analysis of EtOAc extracts from the Bovine bile, the antioxidant activity appeared at $R_f$ value, 0.72. These results suggested that the antioxidant may be coprostan 3-ol.

Purpose: The purpose of this study was to verify that the expressions of angiogenin, transforming growth factor-beta(TGF-${\beta}$), vascular endothelial growth factor(VEGF), human apurinic/apyrimidinic endonuclease(APEX) and tumor necrosis factor-alpha(TNF-${\alpha}$) were associated with the tumorigenesis of the oral squamous cell carcinoma(OSCC). Materials and Methods: Fifty-one samples of OSCC and fifteen normal oral mucosae were obtained to analyze the expression levels of above five factors. mRNA expressions were quantified by the quantitative competitive PCR(QC-PCR) method. After 2% agarose gel electrophoresis stained with ethidium bromide, the concentration of mRNA was calculated by a digital image analysis system. The expression levels of angiogenin, TGF-${\beta}$, VEGF, APEX and TNF-${\alpha}$ were compared by unpaired Student's t-tests between cancer and normal tissues. We analyzed statistically to find the cut-off values that would be useful as diagnostic markers, and the linear regression analysis between every two factors of these five factors by SAS system. Results: All of these five factors (angiogenin: P<0.0037, TGF-${\beta}$: P<0.0001, VEGF: P<0.0102, APEX: P<0.0023, TNF-${\alpha}$: P<0.0074) were significantly correlated with OSCC. In the analysis to find the cut-off values for the diagnosis, we could not find any value that had a reasonable sensitivity and specificity. In the linear regression analysis, there were correlations between angiogenin and TNF-${\alpha}$, TGF-${\beta}$ and VEGF, TGF-${\beta}$ and APEX, TGF-${\beta}$ and TNF-${\alpha}$, VEGF and APEX, VEGF and TNF-${\alpha}$, APEX and TNF-${\alpha}$. Conclusion: Our results suggest that not only angiogenin, TGF-${\beta}$, VEGF, APEX and TNF-${\alpha}$ are significantly associated with the tumorigenesis, but also the close relationship between these factors might enhance the tumorigenesis of OSCC. We can not find clinical availability for diagnosis.

PURPOSE: Previous studies have documented the lack of ultrasound's non-thermal effects on nerve conduction using frequencies of 1 MHz and 870 kHz. The purpose of this study was to determine the biophysical effects of continuous ultrasound on median local forearm temperature and motor nerve conduction velocities using frequencies of 3.0 MHz. SUBJECTS: Twelve healthy subjects (6 males, 6 females, age $22.30{\pm}2.41$ yrs, weight $61.33{\pm}10.16$ kg, height $167.58{\pm}8.04$ cm) without a history of neurological or musculoskeletal injury to their dominant arm volunteered for this study. METHODS AND MATERIALS: Each subject received a total of five treatments, one each at .0, 0.5, 1.0, 1.5, 2.0 W/$cm^2$ of 3 MHz continuous ultrasound on the anterior surface of the middle area of dominant forearm for 10 minutes. Dependent measures for forearm local temperature and median motor nerve conduction velocity (MNCV) were taken pretreatment and immediately post-treatment. One-way ANOVA were used for each dependent measure. RESULTS: The posttreatment forearm local temperature were differed significantly (p<0.001) between intensities of ultrasound. The posttreatment forearm local temperature of the ultrasound treated with 1.0 w/$cm^2$, 1.5 w/$cm^2$ and 2.0 w/$cm^2$ were significantly higher than 0.5 w/$cm^2$ and 0.0 w/$cm^2$ of ultrasound (p<0.05). The posttreatment median MNCV were differed significantly from the respective pretreatment velocities (p<0.001). The MNCV of the ultrasound treated with 0.0 w/$cm^2$ and 0.5 w/$cm^2$ were significantly (p<0.05) slower than that observed pretreatment, while the three ultrasound intensities produced significantly increased posttreatment MNCV: 1.0 w/$cm^2$ and 1.5 w/$cm^2$ and 2.0 W/$cm^2$. The posttreatment MNCV at 2.0 w/$cm^2$ and 1.5 w/$cm^2$ was significantly faster than that at 0 w/$cm^2$, 0.5 w/$cm^2$ and 1.0 w/$cm^2$ (p<0.05), the MNCV at 1.0 w/$cm^2$ was significantly faster than that associated with 0 w/$cm^2$ and 0.5 w/$cm^2$ of ultrasound (p<0.05). CONCLUSIONS: The decreased median motor forearm local temperature and MNCV of the ultrasound treated with 0.0 w/$cm^2$ and 0.5 w/$cm^2$ were attributed to the cooling effect by ultrasound transmission gel. Local forearm temperature and nerve conduction velocity were directly related to the intensity of ultrasound. Alterations in MNCV from ultrasound on healthy nerves appeared to be related to temperature changes induced by thermal effects of ultrasound.
