Effect of Nitrite, Thiocyanate and Ascorbic acid on N-nitrosamine Formation in Salted and Dried Yellow Corvenia under Simulated Gastric Digestion (염건조기의 인공소화시 Nitrite, Thiocyanate 및 Ascorbic acid가 N-nitrosamine의 생성에 미치는 영향)

  • This experiment was conducted to investigate the effect of NA formation in the human body with salted and dried yellow corvenia(Gulbi, Pseudosciaena manchurica) steamed for 30 minutes and incubated in 10 ml simulated saliva and 40 ml gastric juice at 37。C for 2 hours. And the experiment studied the NA formation after nitrite, thiocyanate and ascorbic acid treatment under simulated gastric conditions. N-nitrosodimethylamine(NDMA) was not detected when nitrite was not added to the digestate mixture. And then increasing the nitrite concentration from 0.5 mM to 8 mM gave a slight increase in the NDMA formation at all level of Gulbi utilized in the experiment. In contrast to the change of the nitrite concentration, the increase of the amount of Gulbi didn't show any marked influence on NDMA formation. At all level of thiocyanate(1∼6.4 mM) tested, concentration of NDMA was still proportional to the nitrite concentration. In the catalytic ability of thiocyanate at another high level of nitrite(100 mM) no catalytic activity was observed up to the level of 8mM thiocyanate. The same amounts of salted fish(10 g) and ascorbic acid levels in each single digestate were used to prevent NDMA formation. No prevention effect was observed at nitrite concentration of 0.5 mM and 1 mM. Most importantly, the decrease of NDMA concentration, by the increase of ascorbic acid absolutely, was remarkably shown when nitrite concentration was high. On the contrary, according to the degree of ascorbic acid, the higher the amount of nitrite the higher the prevention of NDMA formation.

Effect of Ethanol Extract of Dried Chinese Yam (Dioscorea batatas) Flour Containing Dioscin on Gastrointestinal Function in Rat Model

  • In this study, a 40% ethanol extract of Chinese yam flour (Dioscoreae rhizoma), containing $177{\pm}58{\mu}g/mL$ of dioscin, was tested in order to evaluate its pharmacological effects on the gastrointestinal tracts of Sprague-Dawley rats. Via the ingestion of the Chinese yam extract, the secretion of gastric acid was suppressed in the rats, and gastrointestinal motility increased by as much as 10%. The fecal quantity of rats fed on the Chinese yam extract also increased, by more than 40% as compared with that of the controls. The Chinese yam extract was found not to affect the growth of normal intestinal bacteria. However, a great deal of lactose-fermenting bacteria was observed in the fecal samples of rats fed for 6 weeks on 2% Chinese yam extract. This finding would appear to suggest that Chinese yam extract not only induces an improvement in digestive capability, but also affects the conversion of some intestinal flora to helpful bacteria. Our serochemical analyses indicated that serum glucose, neutral lipid, and total cholesterol levels were reduced to some degree by long-term feeding on Chinese yam extract. This finding bolsters the notion that Chinese yam extract may prove helpful as a digestion-aiding agent for patients suffering from hyperglycemia or hyperlipidemia.

  • A Helicobacter pylori urease inhibitor from Rubus coreanus Miquel has been isolated and partially characterized for aiming to Prevent H. pylori growth and decrease harmful accumulation of ammonia in human gastric mucosa. We screened urease inhibitory activities in 519 extracts library prepared by solvent extraction from 173 kinds of edible plants, medicinal herbs, herbs and seaweeds using a colorimetric urease assay system. As results of primary and secondary screening, 70% acetone extract of Rubus coreanus Miquel was selected as potent candidate, showing about 24% inhibitory activity. The acetone extract was sequentially partitioned into RCE/RCWI and RCB/RCW2 layers with ethyl acetate and butanol. The major active component in RCW2, water layer from butanol fractionation was revealed to be peptidic or proteinous substance by inhibitory activity determination after pronase digestion and periodate oxidation. RCW2-IIIc a was isolated by sequential column chromatography on DEAE-Toyopearl 650C, Butrl-Toyopearl 650M and Sephadex LH-20. The isolated urease inhibitor RCW2-IIIc $\alpha$, was highly pure proteinous substance with molecular weight of 13kDa by high-performance gel permeation liquid chromatography. RCW2-IIIc$\alpha$ has about 5 times higher inhibitory activity than 70% acetone extract, showing high stability against heat treatment and peptic digestion.

  • Body fluid(津液) is a general term for normal mucus in human body, including saliva, gastric juice, intestinal juice and articular fluid in joints as well as tear, running nose, sweat, etc.. The formation of Body fluid goes through two phases. First phase is digestion of food at stomach, and then evaporation and classification of energy at Triple warmer(三焦). More technically speaking, Body fluid is divided into the Jin(津) & the Aek(液). Aek is a very mild and water-like fluid, runs deep into the internal organs. Jin is a thick and sticky liquid, running shallow under the skin or in the joints of limbs. Major roles of body fluid over the body are to moisturize the internal organs, flesh, skin, etc., to soften the joints, to fill the bone marrow, and to balance Yin and Yang. This article deals with the role of body fluid and how to differentiate them, the liquid metabolism in the human body, and the formation and change of sweat, urine, tear, spit, bone marrow, etc.. The imbalance of Yin and Yang and disturbance of Triple warmer's evaporation lead into lumbago, leg weakness and edema is also explained here.

Effects of Protein and Iron Concentrations on Iron Solubility in Black Tea Infusion (단백질 함량 및 철분 농도의 변화에 따라 홍차 추출물이 철분의 용해도에 미치는 영향)

  • Tannins in plant foods and beverages may produce antinutritional or toxic effects although some proteins with high affinity for tannins seem to function as defense mechanism to tannin toxicity. Our objectives were to investigate of tea tannins, iron and protein and to evaluate the role of proteins in tannin effects on iron solubility. Iron solubility in vitro was measured using tea with and without proteins. Mixtures of tea, protein in varying concentrations(either gelatin or bovine serum albumin), and iron(eithe 10 or 50ug/mL) were prepared. Controls contained water in place of tea. Iron bioavailability was assessed by measuring iron solubility in the simulated gastric condition with pepsin digestion. Bound iron was removed by centrifugation and soluble in tea alone. When iron concentratin was 10ug/mL, addition of small amounts of protein to tea dramatically reduced iron solubility, but solubility of iron increased in the tea mixturea as the concentration of protein was increased. The percnetage of iron that precipitated was much greater at 10ug Fe/mL than the values at 50ug Fe/mL suggesting that the iron binding sites on the tea-protein complex was saturated. These results suggest that interactions of iron with tea tannins are influenced by the concentratins of protein and iron.

Changes of Somatostatin-Immunoreactive Cells on the Stomach of Ovariectomized Rats

  • The changes on the regional distributions and frequencies of somatostatin-immunoreactive (IR) cells in the fundus and pylorus of the stomach of osteoporotic Sprague-Dawley rats induced by ovariectomy were studied by immunohistochemical methods. The experimental animals were divided into two groups, one for non-ovariectomized group (Sham) and the other for ovariectomized group (OVX). Samples were collected from the fundus and pylorus regions at the 10 th week after ovariectomy or sham-operation. Somatostatin-IR cells were observed in both regions of the stomach regardless of ovariectomy. Most of these IR cells in the mucosa of the fundus or pylorus were generally spherical or spindle in shape (open type cell) while cells found in the gastric gland regions were round in shape (close type cell). Significantly lower number (P<0.01) of somatostatin-IR cells were detected in OVX as compared with Sham in the fundus and pylorus. In the present study, the density of somatostatin in the stomach was markedly decreased. Therefore, these changes in density of somatostatin-IR cells detected in this study may support the speculation that the development of gastrointestinal symptoms in osteoporosis such as impairments of calcium and some lipids, frequently encountered in patients with postmenopausal osteoporosis because the changes in gastrointestinal endocrine density would reflect the change in the capacity of producing these hormones and regulating gut motility and digestion.

Effect of Stevia rebaudiana on the Bioactive Compounds from Agarwood Leaf (Aquilaria spp.) by Lactic Fermentation and Spray Drying

  • Agarwood (Aquilaria spp) has high economic value. However, essential oil production from agarwood is a time-consuming process. Additionally, agarwood leaves have not been utilized even though they contain various bioactive ingredients. In this study, agarwood leaves were fermented using Lactobacillus plantarum ATCC 8014 with or without Stevia (4, 8, and 12%; v/v). The fermented fluid was mixed with maltodextrin (15%; w/v) and subjected to spray drying (inlet temperature, 120℃; outlet temperature, 65-70℃). The contents of polyphenols, polysaccharides, saponins, and flavonoids and the viability of L. plantarum were determined. Fermentation enhanced the levels of bioactive compounds. The contents of polyphenol (69.19 ± 4.05 mg GAE/g of sample), polysaccharide (20.75 ± 0.98 mg GE/g of sample), saponin (305.23 ± 4.21 mg OAE/g of sample), and flavonoid (7.86 ± 0.72 mg QE/g of sample), and the viability of L. plantarum (8.72 ± 0.17 log CFU/ml) were markedly upregulated in the samples containing Stevia (12%; v/v). This indicated that the supplementation of Stevia during fermentation decreases the fermentation time (9 h), upregulates bioactive compound production in agarwood leaves, enhances microencapsulation during spray drying, and increases the viability of L. plantarum under simulated gastric digestion conditions.

  • Oriental mineral medicines are single or mixture of more than one mineral species or rock/fossil which are used to treat disease. Mineral medicines remove harmful or useless substances to decrease toxicity and secondary effects, and cause the manufacture of medical compounds with increased efficacy. The extraction test is an accepted in vitro system to predict the bioaccessibility of major and minor elements from mineral medicine. It incorporates gastrointerstinal tract parameters representative of a human body that including stomach and small intestinal pH which are the same as digestion condition. The bioaccessibility of a mineral medicine is the fraction that is soluble in the gastrointestinal environment and is available for absorption. Reaction path modeling in the human body can predict digestion with gastric fluid as well as absorption in the small intestine, existence in body fluids and reaction progress of the exhaust process according to pH conditions in body. Also reaction path modeling can predict bioavailability, which is equal to existence rate in the body and the form and amount of a medicine in the body after intake. The study results from predicating the existence form mineral medicines in the body, and proving the effective ingredient using bioaccessibitily and human risk assessment, suggest these that should be necessary data for new medicine development.

Swelling and Drug Release Characteristics of PVP Hydrogel Polymerized by $\gamma$-Irradiation Method (김마선 조사법으로 합성한 PVP하이드로겔의 팽윤과 약물방출특성)

  • The short and variabke transit of drug throught GI tracj and the inter-and intra-subject variations of the transit restrict the sustained drug absorption after oral adminstration. These restrictions may be solved by retaining the dosage forms in the stomach. Then the dosage form will act as a platform which releases the drug slowly and makes the GI absorption occur for a long time. In this study, as the platforms, PVP hydrogels were synthesized by chemical and y-irradiation method in the cylindrical test tube. The chemical method means the synthesis of the hydrogel by heating the mixed solution of N-vinyl-2-pyrrolidone [monomer], acrylated albumin [crosslinking agent], 2, 2'-agobis(2-methylpropionitrile) [initiator] and proxyphylline [drug] at $65^{\circ}C$ for 5 hr. The $\gamma$-irradiation method means the synthesis of the hydrogel by irradiation with $^{60}$ Co $\gamma$-ray of the mixed solution of the monomer, acrylated albumin, and flurbiprofen [drug] at room temperature with total 0.2 Mrad for 3 hr. Our intention is to design the hydrogel tablet (diameter : 1.20 cm, thickness : 0.60 cm) which swells in the gastric fluid after oral administration to such a size that passing through the pylorus could be inhibited during the period of drug release. After releasing drug, the hydrogel should be degraded by the enzymeatic digestion in the stomach, or by hydrolysis and eventually solubilized. Thus, in votro tests were performed to examine the factors that affect swelling and drug release from the PVP hydrogels. Experimental results show that the hydrogels swell to a size larger than the diameter of the pylorus(l.3$\pm$0.7 cm) and the hydrogel prepared by the chemical method is digested by pepsin. But the hydrogel prepared by the $\gamma$-irradiation method was not digested by the pepsin and just collapsed with time. Thus, the swelling of the hydrogel synthesized by $\gamma$-irradiation was independent albumin acrylation time and pepsin concentration. But drug content and radiation dose affected the swelling and drug release kinetics of the hydrogel. Drug release from the hydrigels was prolonged up to about 24 hr. Therefore, it was concluded that by adjusting these factors, the albumin-crosslinked PVP hydrogel synthesized by $\gamma$-irradiation method is expected to be retained in the stomach for up to 60hr and be a potential platform of drugs for long-term GI absorption.
