Evaluations of Poly{vinyl alcohol)/Alginate Hydrogels Cross-linked by r-ray Irradiation Technique

  • In this work, we prepared hydrogels for wound dressing from a mixture of poly(vinyl alcohol) (PVA) and alginate using the $\^$60/Co ${\gamma}$-ray irradiation technique. We examined the physical properties of these hydrogels, including gelation, water absorptivity, and gel strength, to evaluate the applicability of these hydrogels for wound dressings. The biocompatibility of these hydrogels was also evaluated in vitro, in cultures of mouse fibroblasts, and in vivo, by subcutaneous implantation studies in rats. The gel content and strength increased upon increasing the radiation dose and upon decreasing the concentration of alginate. The degree of swelling was inversely proportional to the gel content and strength. The degree of cytotoxicity of the ${\gamma}$-ray-treated hydrogels was ca. 60% compared to the (-) control (serum) after 1 day of incubation. When the incubations were prolonged up to 2 days, the toxicity of all the samples decreased remarkably and reached that of the control. Subcutaneous implantation studies in rats indicated that foreign body reactions occurring around the implanted hydrogels were moderate and became minimal upon increasing the implantation time.

  • Postharvest diseases cause losses in a wide variety of crops around the world. Irradiation, a useful nonchemical approach, has been used as an alternative treatment for fungicide to control plant fungal pathogens. For a preliminary study, ionizing radiations (gamma, X-ray, or e-beam irradiation) were evaluated for their antifungal activity against Botrytis cinerea, Penicillium expansum, and Rhizopus stolonifer through mycelial growth, spore germination, and morphological analysis under various conditions. Different fungi exhibited different radiosensitivity. The inhibition of fungal growth showed in a dose-dependent manner. Three fungal pathogens have greater sensitivity to the e-beam treatment compared to gamma or X-ray irradiations. The inactivation of individual fungal-viability to different irradiations can be considered between 3-4 kGy for B. cinerea and 1-2 kGy for P. expansum and R. stolonifer based on the radiosensitive and radio-resistant species, respectively. These preliminary data will provide critical information to control postharvest diseases through radiation.

A proposed new configuration of a shuffle-dwell gamma irradiator

  • A gamma irradiator is a well-developed installation for gamma radiation sterilization. A "shuffle-dwell" mode is preferable for high dose applications. A novel configuration of a shuffle-dwell gamma irradiator is proposed to increase energy utilization and throughput, which would result in higher profitability. While the minimum distance between any irradiation position and each source pencil, the minimum distance between the neighboring irradiation positions and the size of source pencils are kept the same as the current configuration, the irradiation positions and source pencils are rearranged based on the fact that radiation is emitted in an isotropic fashion. The computational results suggest that the proposed configuration requires an 8.7% smaller area and exposes the product to 11.8% more gamma radiation in a 10.7% shorter irradiation time. In other words, the proposed configuration needs a smaller area and shorter irradiation time to have a better performance compared to the current shuffle-dwell gamma irradiator. Note that the claim is based primarily on an analytical calculation. Experimental and manufacturing among other practical considerations will be taken into account in the future work to exhaustively evaluate the performance of the proposed configuration and to compare it with that of the traditional configuration.

Adaptive Response Induced by Low Dose Ionizing Raditation in Human Cervical Carcinoma Cells

  • Adaptive response induced by low dese .gamma.-ray irradiation in human cervical carcinoma cells was examined. Cells were exposured to low dose of .gamma.-ray irradiation in human cervical carcinoma cells was examined. Cells were exposured to low dose of .gamma.-ray (1-cGy) followed by high doses of r-ray irradiation (0,1,2,3,5,7 and 9Gy for chlnogenic assay or 1.5Gy for micronucleus assay) with various time intervals. Survival fractions of cells in both low dose-irradiated and unirrated groups were analyzed by clonogenic assay. Surviva fractions of low dose-irradiated in cell survival was maximum when low and high dose irradiation time interval was 4 hr. Frequencies of micronuclei which is an indicative of chromosome aberration were also enutained from survival fractions analyzed by clonogenic assay, maximum when low and high dose irradiation time interval was 4hr. Frequencies of micronuclei which is an indicative of chromosome aberration were also enumerated in both low dose-irradiated and unirradiated groups. In consiststent with the result obtained from survival fractions analyzed by clonogenic assay, maximum reduction in frquencies of micronuclei was observed when low dose radiation was given 4 hr prior to high response to subsequent high dose .gamma.-ray irradiation in human cervical carcinomal cells. Our data suggest that one of the possible mechanisms of adaptive response induced by low dose rediation is the increase in repair of DNA double strand breaks in low dose radiation-adapted cells.

  • PVA nanofibers containing silver nanoparticles were prepared by two methods. The first method was electrospinning of irradiated solution. The prepared $PVA/AgNO_3$ solution was irradiated by gamma-rays. And then the irradiated solution was electrospun. The second method was irradiation of electrospun nanofibers. Nanofibers prepared by electrospinning of unirradiated $PVA/AgNO_3$ solution. The morphology of the nanofibers was observed with a SEM, TEM. When the irradiated $PVA/AgNO_3$ solution were electrospun, the average size of the Ag nanoparticles was increased, but their number was decreased.

  • The effect of ${\gamma}$-ray irradiation on the color of nucleus and cultured pearls was investigated. After ($^{60}Co$) ${\gamma}$-ray irradiation on the pearl nucleus, its color was altered from brownish to blackish brown or gray depending on irradiation dose. It was clearly found that the all samples are composed of aragonite. ESR spectra were based on the measurement of the paramagnetic species $CO_2$-radicals by the interaction with the ${\gamma}$-ray irradiation in $Mn^{2+}$ containing $CaCO_3$. In this study, we investigated optimal detection method of ${\gamma}$-ray irradiation to establish destructive or non-destructive test for pearl nucleus.

  • Recently, most countries in the world have pursued a denuclearization. So it has been of interest to increase to Nuclear weapon in such as North Korea's continued nuclear test. Pulsed gamma rays produced in the nuclear explosion and the space environment can give the big damage to the electronic device in a very short period of time. To confirm the extent of damage of these electronic devices, pulsed gamma irradiation facility that can occur in nuclear weapon or space environment are required. In this paper, we implemented the pulsed gamma-ray detection module and analyzed output of the irradiation test. We have experimented using an electron beam accelerator research facilities in Pohang Accelerator similar conditions to equip and Nuclear weapon. As a result, we confirmed that the pulsed gamma rays emitted by the gamma radiation and electron beam conversion device. The results of this paper will contribute to improve the reliability and accuracy of studies for utilizing pulsed gamma rays.

The Effect of Red Ginseng Extract on Superoxide Dismutase Activity in the Kidney of Gamma-ray Irradiated Mice (홍삼 투여가 방사선에 조사된 생쥐 신장의 Superoxide Dismutase에 미치는 영향)

  • This study was prepared to observe the change of enzyme activities in kidney treated with red ginseng extract in the gamma ray irradiated mice. Determine the activity of SOD, peroxidase, catalase in the kidney a period of 1 day, 2 day, 3 day, 4 day, 5 day after a saline injection or injection of red ginseng extract or gamma ray irradiatied group into four classify. The activity SOD and catalase showed a tendency to increase and recovery at the early state but pay no regard. Wherease, the activity of peroxide restored and increased pay regard. A physiological saline injection group after gamma ray irradiation showed a tendency to diminish after remakable increase of activity of SOD, peroxidase and catalase than control group. Injection group of red ginseng extract after gamma ray irradiation observed rapid recovery on activity of SOD, peroxidase, catalase than a saline injection group. Experimental result suggested that injection of red ginseng extract after irradiation have the recovery effect on the changed of activity of SOD, peroxidase and catalase against radiation injury.

  • Gamma ray irradiation mutagenesis was employed to get variants of Pleurotus eryngii with functionally enhanced and improved characteristics. Protoplasts released from P. eryngii were treated with gamma ray radiation under 0.25-1.25 KGy. Protoplast sample that showed fatality rate of 80% at the 0.25 KGy was spreaded on YPMGA (yeast, peptone, malt-extract, glucose, agar) and 500 mycelial colonies were randomly selected from the medium. Of them, 100 mutant strains with mycelial morphology and growth rate that differ to control strain were observed on PDA. The cellulase and laccase activity of 67 gamma ray-irradiated P. eryngii isolates with morphological variation were investigated. Among these, 5 isolates were higher cellulase. In addition, the genetic variation of the mutant strains was analyzed by PCR fingerprinting.

Effects of Gamma Irradiation on Nutrient Composition, Anti-nutritional Factors, In vitro Digestibility and Ruminal Degradation of Whole Cotton Seed

  • Whole cotton seed (WCS) has become one of the major feed ingredients in TMR for dairy cattle in Korea, and WCS for feed use is mostly imported from abroad. Since this genetically modified oil seed is usually fed to the animal in raw state, its germination ability, if last long, often causes concerns about ecological disturbances. In the process of looking for effective conditions to remove germination ability of WCS this study had the objectives to evaluate the nutritional effects of gamma irradiation at doses of 8, 10 and 12 kGy on changes in nutrient contents, anti-nutritional factors, in vitro digestibility and ruminal degradability. No significant differences were found in proximate analysis of nutrients between raw WCS and gamma irradiated one. Glycine and threonine contents significantly increased when the WCS was exposed to gamma ray as compared to untreated WCS (p<0.05). As for fatty acid composition, no significant differences were observed with the irradiation treatment. Free gossypol in WCS was decreased (p<0.05) by gamma irradiation treatment. Of the 3 different levels of gamma irradiation, a dose of 12 kGy was found to be the most effective in reducing free gossypol concentration. Results obtained from in situ experiment indicated that gamma irradiation at a dose of 10 kGy significantly (p<0.05) lowered rumen degradability of both dry matter and crude protein as compared with raw WCS. However, there were no significant differences in rapidly degradable and potentially degradable fractions of crude protein due to 10 kGy gamma irradiation. Overall, this study show that gamma irradiation at a dose of 10 kGy is the optimum condition for removing germination ability of WCS, and could improve nutritive value for the ruminant with respect to the decrease in both ruminal protein degradability and gossypol content of WCS.
