Quality of Irradiated Plain Yogurt during Storage at Different Temperatures

  • To develop a safer yogurt for immuno-compromised or allergy patients and to extend shelf-life, a plain yogurt was irradiated with doses of 0, 1, 3, 5, and 10 kGy using a gamma ray and the chemical and microbiological quality and allergenicity change were investigated. There was no difference in the content of protein, total solid, and amino acids of the plain yogurt by irradiation treatment and different storage temperatures (4, 20, and $35^{\circ}C$). The lactic acid bacterial counts of irradiated plain yogurt had approximately 3-decimal reduction at 3 kGy, and no viable cell at 10 kGy regardless of storage time and temperature. The binding ability of rabbit antiserum to milk proteins in irradiated plain yogurt showed that 10 kGy of irradiation produced significantly higher binding ability than other treatments. Sensory evaluation indicated that only appearance of the plain yogurt irradiated at 3 kGy or higher had a lower value than the non-irradiated control when stored at $20^{\circ}C$. Results suggest that irradiation of plain yogurt does not significantly affect the chemical and sensory quality of plain yogurt, but can extend the shelf-life, possibly reduce allergenicity, and provide a safer product.

  • The purpose of the present experiment was to investigate the micronuclei (MN) frequency in cytokinesis-blocked (CB) cells after various doses of gamma-rays in two species (fish and fowl) and so to contribute to the clarification of the question whether these species are suitable as a target organism in the test system. The frequencies of binucleated cells, and gamma-ray-induced MN in CB cells at several doses were measured in three donors of two species. No binucleated cell was noted in erythrocyte. The peaks of binucleated lymphocyte formation were found at a concentration of 2% phytohaemagglutinin (PHA) and $3{\mu}g/m{\ell}$ cytochalasin B (Cyt-B) in fish at 144 hours after incubation and 2% PHA and $6{\mu}g/m{\ell}$ Cyt-B in fowl at 72 hours after incubation. But the micronucleus counts failed to show any evidence of radiation damage. Measurements performed after irradiation showed a dose-related decrease in the formation of binucleated cells in each of the donors studied. Results indicated that the assays were not suitable for this due to blastization inhibition (binucleation failure) after irradiation. We concluded that the use of CB cell from fish and fowl for detecting the results of mdiation exposure was highly questionable.

Detection of DNA Instability Induced from Tissue Culture and Irradiation in Oryza sativa L. by RAPD Analysis

  • As a consequence of tissue culture of rice, RAPD analysis was peformed to determine whether extended culture periods as undifferentiated calli affected the subsequent genetic constancy, and whether any resulting DNA rearrangements could be detected between sibling plants produced from the same callus. Somaclonal variation was induced at the initial stage of tissue culture and it increased with the length of culture maintenance. Of the 192 total bands, the number of polymorphic bands was 22 (11.5%), 33 (17.2%), and 49 (25.5%) in the callus of 1,3, and 6 months culture, respectively. A significantly higher level of genotypic polymorphisms between regenerants from two different somaclones was also detected, although all the regenerants were derived from a single genotype. In comparison of DNA polymorphisms between regenerants from non-irradiated and from irradiated calli, a scope of variation spectrum by gamma ray irradiation was larger than that by tissue culture. Consideration must be given to this genomic variation where attempts are to be made to use desirable somaclonal variants for plant breeding purpose and in genetic engineering program.

  • It was determined whether photostimulated luminescence (PSL) is applicable to the detection of post-irradiation of foods by measuring PSL photon counts for unirradiated and irradiated pepper powder, dried herbs, fresh shrimp, potato, soybean, dried fig, chestnut, dried squid, and dried cod. The samples were irradiated with $^{60}Co\;{\gamma}-ray$ source and PSL photon counts were measured for them. The photon counts of unirradiated samples were lower than 2,000 for potatoes and less than 1,000 for the others. The photon counts of the irradiated samples except dried figs (0.5 kGy), chestnuts (0.2 kGy), and dried squid (0.5 kGy) were higher than 1,000, and the photon counts increased with dose. Thus, PSL might be applied to the primary detection of irradiated foods.

  • A calculation is presented of air-scattered gamma rays using the modified single-scattering approximation. The air-scattered tissue dose rates are calculated for a general purpose taking into account (a) the buildup and exponential attenuation, (b) the energy spectrum at the position of question and (c) the geometrical scattering volume in three dimensions. These calculations have been further modified to render them applicable to a typical field irradiation facility which is surrounded by a shield wall and in which the source is fitted with a beam collimating device. The results of the calculation include the energy spectra, angular distribution and tissue does rates at source-receiver separation distances of from 35m to 300m. The comparison shows that the present method developed may be generally adequate for the gamma-ray air-scattering problems in field irradiation facilities if energy and angular distribution at the shield are unimportant.

Hydrogen Bonding Effect on γ-Ray Irradiated Poly(vinyl alcohol) Hydrogels in Different Drying Conditions

  • Three-dimensional network hydrogels were prepared by ${\gamma}$-irradiation of aqueous solutions of poly(vinyl alcohol) (PVA) and glycerol (Gly). Oven-drying was used to measure the gel fraction (G), hydration (H) or swelling behavior (S) of the prepared hydrogels. This study made a hypothesis that hydrogen bonds due to addition of glycerol and change of dry states such as freeze-drying (FD), room-drying (RD) and oven-drying (OD) acts on the G, H, and S. Interesting results on the hydrogen bonding effect in the prepared hydrogels are monitored at different drying conditions. The FD samples have a higher G values with increase in glycerol content as compared with the OD and RD samples. The formation of strong hydrogen bonding network among Gly molecules and hydrogel matrix was considered as the main driving force, resulting in the changes in the G, H, and S of the hydrogels under different drying conditions.

  • Taraxacum platycarpum (TP) has been used for years without restriction on usual dose for its non-toxic nature and nonexistence of the side effects. To develop antimicrobial hydrogel, poly (vinylalcohol) (PVA) hydrogels containing the aqueous extracted TP as an antimicrobial agent were prepared by using gamma-rays irradiation. The antimicrobial activities of the TP hydrogels were tested against Staphylococcus aureus and Staphylococcus epidermidis by disc diffusion method. The inhibition zones (IZ) of the TP extracts and TP hydrogels against S. aureus were 16 mm and 20 mm and against S. epidermidis was 10 mm and 13 mm, respectively. In conclusion, the TP hydrogel that has an excellent antimicrobial activity was proved to be a valuable material for functional skin care.

Impacts of Irradiation Sources on Quality Attributes of Low-salt Sausage during Refrigerated Storage

  • This study was performed to investigate the impacts of irradiation sources on quality attributes of low-salt sausage during refrigerated storage. Control sausage was prepared with 1.5% sodium chloride (NaCl), whereas low-salt sausage was formulated with 0.75% NaCl (a 50% reduction; L-control). Sausage samples were vacuum-packaged, and low-sausages were irradiated with gamma-ray, electron-beam and X-ray at 5 kGy, respectively. The samples were stored at $4^{\circ}C$ for 28 d to determine changes in quality attributes. The pH of low-salt sausages was unaffected by irradiation at 5 kGy (p>0.05). Higher redness values were found at irradiated low-salt sausages compared to control (p<0.05). The hardness, gumminess and chewiness of control sausage were higher than those of low-salt sausages (p<0.05). However, there were no significant differences in the textural parameters between low-salt sausage treatments. The overall sensory acceptability score of irradiated/low-salt sausages were lower than L-control due to decreased scores for cooked meat flavor but increased radiolytic off-flavor (p<0.05). The initial 2-thiobarbituric acid-reactive substances (TBARS) values of irradiated/low-salt sausages were higher than control and L-control (p<0.05). However, the TBARS values of irradiated treatments were significantly lower than control at the end of storage. Irradiation could effectively inhibit the microorganism growth (total aerobic bacteria, coliforms, Enterobacteriaceae, and Pseudomonas spp.) in low-salt sausages (p<0.05). Therefore, our findings show that irradiation could be to improve microbial safety of low-salt sausages, and suggest that further studies should be necessary to reducing radiolytic off-flavor of irradiated/low-salt sausages.

Variations in Sweetpotato Regenerates from Gamma-ray Irradiated Embryogenic Callus

  • Radiation induced and somaclonal variations were investigated in the regenerates from gamma irradiated and controlled embryogenic callus (EC) of sweetpotato cvs., Yulmi and White Star by morphological, RAPD and AFLP analysis. Most (approx. 90%) of the EC produced somatic embryos developed into plantlets after being transferred to the auxin-free medium. The frequency of morphological variants derived from the irradiated callus ranged from 3 to 7.8% compared to 0.1-1.1% of that derived from the non-irradiated. Morphological variants were selected from the regenerates and analyzed by RAPD and AFLP procedures. RAPD polymorphisms of Yulmi and White Star regenerates from irradiated calli were 8.8% and 6.1%, respectively. However, the polymerphisms among regenerates from the non-irradiation treatment in these two cultivars were non-detectable and 3%, respectively. AFLP polymorphisms of Yulmi and White Star regenerates from irradiated calli were 29.9% and 28.6%, respectively. while the frequencies for those form non-irradiated calli were 8.5% and 5.6%, respectively. Reduction of Estrogenic Activity by Gamma-ray Treatment (감마선 처리에 의한 에스트로겐 활성 저감 연구)

  • In this study, degradation of estrone (E1) and $17{\alpha}$-ethynylestradiol (EE2) by gamma-irradiation and subsequent reduction of estrogenic activity as a function of absorbed dose were conducted using the yeast two-hybrid assay. Relative potency of E1 and EE2 compared to estrogenic activity of $17{\beta}$-estradiol (E2) was found to be 0.0144 and 0.1605, respectively. More than 90% of E1 and EE2 (both $5.0{\times}10^{-6}M$) was removed at an absorbed dose of 5 kGy, but more than 40% of estrogenic activity still remained. The addition of $TiO_2$ catalyst appeared to improve the removal efficiency of E1 and decrease estrogenic activity while there was no significant effect for EE2. Additionally, the calculated estrogenic activity of E1 and EE2 based on a regression model was well correlated with the observed activity.
