Activity Determination, Kinetic Analyses and Isoenzyme Identification of Gamma Glutamyltransferase in Human Neutrophils

  • Gamma-glutamyltransferase (GGT, EC 2.3.2.2) which hydrolyzes glutathione (GSH), is required for the maintenance of normal intracellular GSH concentration. GGT is a membrane enzyme present in leukocytes and platelets. Its activity has also been observed in human neutrophils. In this study, GGT was purified from Triton X-100 solubilized neutrophils and its kinetic parameters were determined. For kinetic analyses of transpeptidation reaction, $\gamma$-glutamyl p-nitroanilide was used as the substrate and glycylglycine as the acceptor. Apparent $K_m$ values were determined as 1.8 mM for $\gamma$-glutamyl p-nitroanilide and 16.9 mM for glycylglycine. The optimum pH of GGT activity was 8.2 and the optimum temperature was $37^{\circ}C$. It had thermal stability with 58% relative activity at $56^{\circ}C$ for 30 min incubation. L-serine, in the presence of borate, was detected as the competetive inhibitor. Bromcresol green inhibited neutrophil GGT activity as a noncompetetive inhibitor. The neutrophils seem to contain only the isoenzyme that is present in platelets. We characterized the kinetic properties and compared the type of the isoenzyme of neutrophil GGT with platelet GGT via polyacrylamide gel electrophoresis (PAGE) under a standart set of conditions.

  • ${\gamma}$-Glutamyltransferase (GGT: E.C. 2.3.2.2.) is a glycoprotein enzyme which is involved in glutathione metabolism and amino acid transport through the plasma membrane. It is distributed widely in several organs including liver and kidney. Several isozymes of GGT have been reported and some of the isozymes may be associated with hepatocarcinogenesis. We have produced six monoclnal antibodies (mAbs) against GGT purified from the liver of 2-acetamidofluorene (AAF) treated rats. All of the six mAbs were obtained by immunizing mice with liver GGT Six hybridomas which produced anti-GGT Abs were extensively subcloned and injected into the peritoneal cavity of BALB/c mice to obtain large quantities of Abs. These mAbs were purified from ascites by ammonium sulfate precipitation and protein A sepharose CL-4B column chromatography. Using these mAbs we preformed enzyme-linked immunosorbent assay (ELISA), radioimmunoassay (RIA), immunohistochemistry (IHC), and autoradiography (ARG) to study the distribution of GGT isozyme in tissue. The results indicate that GGT-mAb 1 is specific for the AAF treated liver GGT, GGT-mAb 5 for the normal liver GGT, and GGT-mAb 6 for the normal kindey GGT. These mAbs may be used to evaluate the distribution of GGT isozymes in different tissues.

연령과 성별에 따른 정상 혈청 Gamma-glutamyltransferase와 관상동맥질환 위험인자와의 관계 (Relations between Normal Serum Gamma-glutamyltransferase and Risk Factors of Coronary Heart Diseases according to Age and Gender)

  • 혈중 gamma-glutamyltransferase (GGT)는 지금까지 간기능장애와 대표적인 알코올 섭취 관련 지표로서 폭넓게 사용되어 왔다. 현재 다양한 질환과의 관련성이 밝혀지면서 그 관심이 커지고 있다. 본 연구에서는 고도의 음주군에 해당하는 대상자를 제외하고 GGT 수치가 정상 범위에 속하는 대상자들에서 GGT 수치에 따른 관상동맥 질환 위험 요인과의 관련성을 살펴보고, 연령과 성별에 따른 특성을 비교 검토 하였다. 분석 자료는 2011 국민건강영양조사 데이터를 이용하였다(N=3,619). GGT(IU/L) 수준에 따라 사분위수(quartile) 네 군으로 나누었을 때 GGT기준은 남성의 경우 순서대로 10~20, 21~27, 28~38, 39~71 IU/L 이었고, 여성의 경우 각각 6~12, 13~16, 17~22, 23~42 IU/L 였다. 대부분의 변수에서 평균치는 GGT 4분위수로 갈수록 수치가 높게 나타났으나, 남성에서 연령과 저밀도콜레스테롤은 GGT 2분위수에서 수치가 가장 높았다. FRS와 10년 관상동맥 질환 위험도는 남성에서 GGT 2분위수에서 유의하게 높았고, 여성의 경우 GGT 4분위수에서 유의하게 높았다. 여성의 경우 GGT 수준이 높아질수록 연령이 증가하였으나, 남성의 경우 GGT 2분위수에서 가장 높았다. 연령 70대에서 남성의 경우 1,2분위수 분포가 가장 많고, 여성의 경우 3,4분위수 분포가 많아 그 차이가 뚜렷하게 나타났다. GGT 수치가 정상 범위에 속하더라도 GGT 수치가 증가함에 따라 여러 위험인자와 관련성이 있음을 증명하였다. 그러나 GGT 수치 증가와 함께 관상동맥질환 관련 수치와 10년 위험도 관련 예측치 증가에 대한 부분은 연령과 성별에 따라 서로 다른 양상을 보였다.

농촌지역 고혈압 전단계 성인의 혈청 Gamma-Glutamyltransferase와 고혈압 발생률간의 관련성 (Association Between Gamma-Glutamyltransferase and Hypertension Incidence in Rural Prehypertensive Adults)

  • Objectives: Prehypertension is associated with a higher risk of developing hypertension compared with normotension. Yet, factors predicting the development of hypertension among prehypertensive people are ill-understood. This prospective cohort study was performed to examine if serum gamma-glutamyltrasferase (GGT) within a normal range can predict the future risk of hypertension among prehypertensive adults. Methods: Study subjects were 293 prehypertensive persons >30-years-of-age who participated in a community-based health survey in 2003 and who were followed up in 2008. Sex-specific quartiles of baseline serum GGT were used to examine association with 5-year hypertension incidence. Results: Baseline serum GGT within normal range predicted the risk of developing hypertension for 5 years only in prehypertensive women. Adjusted relative risks were 1.0, 3.7, 3.6, and 6.0 according to quartiles of baseline serum GGT (P for trend <0.01). This pattern was similarly observed in non-drinkers. However, serum GGT was not associated with incident hypertension in men. Different from serum GGT, baseline serum alanine aminotransferase, another well-known liver enzyme, did not predict the risk of incident hypertension in both genders. Conclusions: Even though baseline serum GGT within normal range strongly predicted the future risk of hypertension, it was observed only in women, Although underlying mechanisms of this association are currently unclear, serum GGT can be used to select a high risk group of hypertension in prehypertensive women.

일부 남성근로자에서 ${\gamma}-GTP$와 관련된 요인에 대한 단면연구 (A Cross-Sectional Study on ${\gamma}-GTP$ and its Related Factors in Male Workers)

  • Objective : To investigate gamma-Glutamyltransferase (${\gamma}-GTP$) and its related factors in male industrial workers. Method : Five hundred and tony male workers without heart disease, diabetes mellitus, renal disease, hepatitis, and other liver diseases were surveyed in October 1998. Blood samples were collected to test for ${\gamma}-GTP$, total-cholesterol and fasting blood glucose. A self-administered questionnaire survey on life style was also done. Results : The total geometric mean value of ${\gamma}-GTP$ was 30.6 U/L. According to a univariate analysis: age, BMI(body mass index, $kg/m^2$), alcohol consumption, current smoking, stress, diastolic blood pressure, and blood total cholesterol were significantly associated with ${\gamma}-GTP$(p<0.05). From a multiple regression analysis: BMI, alcohol consumption, current smoking, diastolic blood pressure and total-cholesterol were significantly related to ${\gamma}-GTP$(p<0.05). Coffee consumption was negatively related to ${\gamma}-GTP$, but not significantly. Conclusion : We recommend that a change in health behavior (i.e. reducing alcohol intake, controlling BMI and not smoking) is necessary to decrease ${\gamma}-GTP$ in male workers.

Model Based on Alkaline Phosphatase and Gamma-Glutamyltransferase for Gallbladder Cancer Prognosis

  • Purpose: To evaluate the prognostic value of alkaline phosphatase (ALP) and gamma-glutamyltransferase (GGT) in gallbladder cancer (GBC). Materials and Methods: Serum ALP and GGT levels and clinicopathological parameters were retrospectively evaluated in 199 GBC patients. Receiver operating characteristic (ROC) curve analysis was performed to determine the cut-off values of ALP and GGT. Then, associations with overall survival were assessed by multivariate analysis. Based on the significant factors, a prognostic score model was established. Results: By ROC curve analysis, $ALP{\geq}210U/L$ and $GGT{\geq}43U/L$ were considered elevated. Overall survival for patients with elevated ALP and GGT was significantly worse than for patients within the normal range. Multivariate analysis showed that the elevated ALP, GGT and tumor stage were independent prognostic factors. Giving each positive factor a score of 1, we established a preoperative prognostic score model. Varied outcomes would be significantly distinguished by the different score groups. By further ROC curve analysis, the simple score showed great superiority compared with the widely used TNM staging, each of the ALP or GGT alone, or traditional tumor markers such as CEA, AFP, CA125 and CA199. Conclusions: Elevated ALP and GGT levels were risk predictors in GBC patients. Our prognostic model provides infomration on varied outcomes of patients from different score groups.

농촌 지역 남성들에서 혈청 Gamma-glutamyltransferase 활성도와 혈압 및 혈청 지질의 관련성 (Blood Pressure and Serum Lipids in Relation to Serum Gamma-glutamyltransferase Activity in Men of Rural Area)

  • 농촌 지역 남성 주민들을 대상으로 혈청 GGT 활성도와 혈압 및 혈청 지질의 관련성을 검토해 보기 위하여 경상남도 내 농촌 지역인 울주군 두동면과 창녕군 장마면에 거주하는 40세 이상의 남성으로서 1997년과 2000년에 실시한 주민 건강 검진에 참여하였던 사람들 중 379명의 자료를 분석하였다. 혈청 GGT 활성도의 수준을 중앙값을 기준으로 이분하여 두군에서의 수축기및 이완기 혈압, 혈청총 콜레스테롤, HDL-콜레스테롤의 평균치를 비교하였고 중성지방은 중앙값을 비교하였으며 통계학적 유의성은 t-검정과Wilcoxon rank sum 검정을 이용하여 검토하였다. 교란변수로 작용할 가능성이 있는 연령, 신체비만지수와 커피음용에의한영향은공분산분석으로보정하였다. 수축기 혈압과 이완기 혈압은 비음주자에서 GGT 활성도 수준이 높은 경우에 더 높은 값을 보였고 통계학적 유의수준에 접근하였다(P=0.055와 P=0.074). 음주자에서도 역시GGT 활성도가 높은 경우에 더 높은 값을 보였으나 통계학적 유의성은 없었다. 혈청 총 콜레스테롤은 비음주자에서 GGT 활성도가 높은 경우에 더 높은 값을 보였으며 통계학적 유의수준에 접근하였다(P=0.052). 음주자에서는 GGT 활성도 수준에 따라 비슷한 값을 보였다. 중성지방은 비음주자와 음주자 모두에서 GGT 활성도가 높은 경우에 더 높은 값을 보였다(P=0.035와 P=0.002). 이상의 소견으로 보아 혈청 GGT 활성도의 증가는 비음주자에서 혈청 중성지방, 총 콜레스테롤 및 혈압의 상승과 관련성이 있음을 암시한다.

The Anti-hepatotoxic Effect of Ginseng in Rats: Meta-analysis

  • The aim of this meta-analysis was to systematically investigate the anti-hepatotoxic effect of ginseng in rats induced toxicity which damage to liver. Primary researches were gained on the ScienceDirect database, the DBpia, and the KISS, and the data about the effect factors in plasma and in enzyme were listed as many as possible. The effect factors were alanine transaminase (ALT), aspartate transaminase (AST), liver aminopyrine N-demethylase (AD), liver aniline hydroxylase (AH), liver 3,4-Methylenedioxyamphetamine (liver MDA), cytochrome P450 (P450), serum alkaline phosphatase (ALP), serum lactate dehydrogenase (LDH), cytochrome b5 (Cyto b5), glutathione reductase (GR), Liver glutathione S-transferase (GST), liver glutamyltransferase (GT), Liver (${\gamma}-GCS$), serum liver 3,4-Methylenedioxyamphetamine (serum MDA), serum sorbitol dehydrogenase (SDH), serum total protein (TP), serum ${\gamma}-glutamyltransferase$ (${\gamma}-GT$). To investigate the effect of ginseng, the mean difference (MD) between the group of rats induced by toxicity (RH) and the group of rats induced by toxicity with ginseng (RHG) were combined, and the significance of MDs were tested. The combined MDs were checked the biases caused by heterogeneity among studies and the publication biases, and adjusted by using random effect model and trim and fill method, respectively. The effect about ALT, AST, ALP, LDH, SDH, TP and ${\gamma}-GT$ in plasma factors were significant, and about AD, liver MDA, P450, Cyto b5, GR, GST, GT and ${\gamma}-GCS$ in enzyme factors were significant. The treatment with ginseng supplementation was significantly effected on plasma and enzyme factors of damaged-rats.

The treatment with ginseng significantly affected the plasma and enzyme levels in rats induced with toxicity.
