The ${\gamma}$-secretase complex represents an evolutionarily conserved family of transmembrane aspartyl proteases that cleave numerous type-I membrane proteins, including the ${\beta}$-amyloid precursor protein (APP) and the receptor Notch. All known rare mutations in APP and the ${\gamma}$-secretase catalytic component, presenilin, which lead to increased amyloid ${\beta}$-peptide production, are responsible for early-onset familial Alzheimer's disease. ${\beta}$-amyloid protein precursor-like (APPL) is the Drosophila ortholog of human APP. Here, we created Notch- and APPL-based Drosophila reporter systems for in vivo monitoring of ${\gamma}$-secretase activity. Ectopic expression of the Notch- and APPL-based chimeric reporters in wings results in vein truncation phenotypes. Reporter-mediated vein truncation phenotypes are enhanced by the Notch gain-of-function allele and suppressed by RNAi-mediated knockdown of presenilin. Furthermore, we find that apoptosis partly contributes to the vein truncation phenotypes of the APPL-based reporter, but not to the vein truncation phenotypes of the Notch-based reporter. Taken together, these results suggest that both in vivo reporter systems provide a powerful genetic tool to identify genes that modulate ${\gamma}$-secretase activity and/or APPL metabolism.
Gel dosimeters have unique advantages in comparison with other dosimeters. Until now, these gels have been used in different radiotherapy techniques as a reliable dosimetric tool. Because dose distribution measurement is an important factor for appropriate treatment planning in different radiotherapy techniques, in this study, we evaluated the ability of the N-isopropylacrylamide (NIPAM) polymer gel to record the dose distribution resulting from the mixed neutron-gamma field of boron neutron capture therapy (BNCT). In this regard, a head phantom containing NIPAM gel was irradiated using the Tehran Research Reactor BNCT beam line, and then by a magnetic resonance scanner. Eventually, the $R_2$ maps were obtained in different slices of the phantom by analyzing T2-weighted images. The results show that NIPAM gel has a suitable potential for recording three-dimensional dose distribution in mixed neutron-gamma field dosimetry.
Attention Deficit Hyperactivity Disorder (ADHD) is one of the most common neurological disorders in children. The diagnosis of ADHD in children is based on the interviews and observation reports of parents or teachers who have stayed with them. Since this approach cannot avoid long observation time and the bias of observers, another approach based on Electroencephalography(EEG) is emerging. The goal of this study is to develop an assistive tool for diagnosing ADHD by EEG classification. This study explores the frequency bands of EEG and extracts the implied features in them by using the proposed CNN. The CNN architecture has three Convolution-MaxPooling blocks and two fully connected layers. As a result of the experiment, the 30-60 Hz gamma band showed dominant characteristics in identifying EEG, and when other frequency bands were added to the gamma band, the EEG classification performance was improved. They also show that the proposed CNN is effective in detecting ADHD in children.

Purpose: We performed three-dimensional (3D) dose reconstruction-based pretreatment verification to evaluate gamma analysis acceptance criteria in volumetric modulated arc therapy (VMAT) for prostate cancer. Materials and Methods: Pretreatment verification for 28 VMAT plans for prostate cancer was performed using the COMPASS system with a dolphin detector. The 3D reconstructed dose distribution of the treatment planning system calculation (TC) was compared with that of COMPASS independent calculation (CC) and COMPASS reconstruction from the dolphin detector measurement (CR). Gamma results (gamma failure rate and average gamma value [GFR and γAvg]) and dose-volume histogram (DVH) deviations, 98%, 2% and mean dose-volume difference (DD98%, DD2% and DDmean), were evaluated. Gamma analyses were performed with two acceptance criteria, 2%/2 mm and 3%/3 mm. Results: The GFR in 2%/2 mm criteria were less than 8%, and those in 3%/3 mm criteria were less than 1% for all structures in comparisons between TC, CC, and CR. In the comparison between TC and CR, GFR and γAvg in 2%/2 mm criteria were significantly higher than those in 3%/3 mm criteria. The DVH deviations were within 2%, except for DDmean (%) for rectum and bladder. Conclusions: The 3%/3 mm criteria were not strict enough to identify any discrepancies between planned and measured doses, and DVH deviations were less than 2% in most parameters. Therefore, gamma criteria of 2%/2 mm and DVH related parameters could be a useful tool for pretreatment verification for VMAT in prostate cancer.
Phytochemical analysis on rice bran and its pitch was performed by the tool of TLC, UV- and IR-spetroscopy, so that it was found that they contained ${\gamma}-oryzanol$ and free sterol. GC-MS analysis of free sterol revealed that it was composed of ${\beta}-sitosterol$, campesterol and stigmasterol. Successive phytochemical analysis of ${\gamma}-oryzanol$ revealed that it was composed of ferulic acid ester of triterpene and sterol, respectively. Triterpene moieties of ${\gamma}-oryzanol$ were identified as follows: cycloartanol, cycloartenol, 24-methylenecycloartanol and unknown triterpene; And sterol moieties were found to be identical with free sterols. In addition, characteristic absorption band in UV spectrum (220-340 nm) was exclusively due to ${\gamma}-oryzanol$. Thus, it was suggested that rice brans of nearly all species of Oryza sativa can be quantitatively analyzed by UV absorption spectrometry, even when water soluble pigments was contained in the rice bran.
Positron Emission Tomography(PET) was introduced as a research tool in the 1970s and it took about 20 years before PET became an useful clinical imaging modality. In the USA, insurance coverage for PET procedures in the 1990s was the turning point, I believe, for this progress. Initially PET was used in neurology but recently more than 80% of PET procedures are in oncological applications. I firmly believe, in the 21st century, one can not manage cancer patients properly without PET and PET is very important medical imaging modality in basic and clinical sciences. PET is grouped into 2 categories : conventional(c) and gamma camera $based_{(CB)}$ PET. $_{CB}PET$ is more readily available utilizing dual-head gamma cameras and commercially available FDG to many medical centers at low cost to patients. In fact there are more $_{CB}PET$ in operation than cPET in the USA. $_{CB}PET$ is inferior to cPET in its performance but clinical studies in oncology is feasible without expensive infrastructures such as staffing, rooms and equipments. At Ajou university Hospital, CBPET was installed in late 1997 for the first time in Korea as well as in Asia and the system has been used successfully and effectively in oncological applications. Ours was the fourth PET operation in Korea and I believe this may have been instrumental for other institutions got interested in clinical PET. The fellowing is a brief description of our clinical experience of FDG CBPET in oncology.
In the nuclear medicine imaging, quality control (QC) process using quadrant bar phantom is fundamental aspect of evaluating the spatial resolution. In addition, QC process of gamma camera is performed by daily or weekly. Recently, Monte Carlo simulation using the Geant4 application for tomographic emission (GATE) is widely applied in the pre-clinical nuclear medicine field for modeling gamma cameras with pixelated cadmium telluride (CdTe) semiconductor detector. In this study, we modeled a pixelated CdTe semiconductor detector and quadrant bar phantom (0.5, 1.0, 1.5, and 2.0 mm bar thicknesses) using the GATE tool. Similarity analysis based on correlation coefficients and peak signal-to-noise ratios was performed to compare image qualities for various source to collimator distances (0, 2, 4, 6, and 8 cm) and collimator lengths (0.2, 0.4, 0.6, 0.8, and 1.0 cm). To this end, we selected reference images based on collimator length and source to collimator distance settings. The results demonstrate that as the collimator length increases and the source to collimator distance decreases, the similarity to reference images improves. Therefore, our simulation results represent valuable information for the modeling of CdTe-based semiconductor gamma imaging systems and QC phantoms in the field of nuclear medicine.
We propose a three-year Key Science Program (KSP) consisting of VLBI monitoring observations and single dish (SD) rapid response observations (RRO). The VLBI monitoring observations are comprised of ten 24-hr observations per year (every month) of about 30 gamma-ray brigt active galactic nuclei (AGNs) with Korea VLBI Network (KVN) at 22, 43, 86, and 129 GHz. The SD RROs may consist of twelve 7-hr observations per source (every week for 3 months after triggering) of gamma-ray flaring sources with two KVN SD telescopes at 22, 43, and 86 GHz in dual polarization. We expect one or two sources per year for the SD RROs. Gamma-ray flares of AGNs are known to be occured in innermost regions of relativistic jets which radiate in whole ranges of electromagnetic spectra due to synchrotron radiation, syschrotron self absorption, inverse-compton scttering, doppler boosting etc. Possible explanations of the gamma-ray flares in AGNs are a) shocks-in-jets propagating within jet flow and b) bending of the whole jets. For both cases, we should expect changes in polarization, luminosity, particle distribution, and structures of jets at mas-scale. The multifrequency simultaneous VLBI/SD observations with KVN are the best tool for detecting such changes correlated with gamma-ray flares. This KSP proposal aims to answer the fundamental questions about the basic nature of the flares of AGNs.
The effect of austenitizing temperature on the degree of carbides re-solutionizing, mean graine size, hardness and the volume fraction of retaind austenite ($V_{\gamma}$) etc., has been studied by means of metallography, X-ray diffractometry and hardness measurement in STD 11 tool steel. As austenitizing temperature increases, the amount of alloying elements which is re-dissolved into matrix increases, resulting in increase of $V_{\gamma}$, due to the chemical stabilization of austenite. The Vickers hardness value decreases with increasing austenitizing temperature, which is attributed to grain size as well the volume fractions of $V_{\gamma}$ and carbides. Theoretical diffraction intensity of (200) ${\alpha}^{\prime}$, (211) ${\alpha}^{\prime}$ (200) ${\gamma}$ and (220) ${\gamma}$ peaks obtained by $CuK_{\alpha}$ chracteristics X-ray (${\lambda}=0.15429nm$) was calculated, and quantitative analysis of $V_{\gamma}$ could be carried out by X-ray diffraction method. The resultant value is well coincided with the value obtained by image analysis method. When the quenched specimen is tempered above $200{\sim}400^{\circ}C$ for 30 min, the transition carbides i.e., MC and $M_2C$ in the size of about 20 nm begin to precipitate at $300^{\circ}C$.
In this study, we evaluated image quality for new Gadolinium Aluminum Gallium Garnet (GAGG) scintillation material based on the Geant4 Application for Tomographic Emission (GATE) simulation tool. The gamma camera detectors with GAGG and NaI scintillation were designed. In particular, we modeled modified body phantom by National Electrical Manufacturers Association International Electrotechnical Commission to evaluate the simulated images. To analysis the image performance, the contrast to noise ratio (CNR) and coefficient of variation (COV) were used by drawn the region of interests, respectively. Based on the CNR and COV results, the CNR value for GAGG material is higher approximately 17 % than NaI material. In addition, the COV value for GAGG material is lower approximately 17 % than NaI material. In conclusion, we confirmed the performnace of GAGG based gamma camera is useful to improve the image quality for the nuclear medicine instrumentation.
