Effect of Leaf and Stem Blight on Growth and Root Yield of Paeonia lactiflora Pallas (작약 지상부 고사가 뿌리의 생육과 수량에 미치는 영향)

  • The pathogenic fungi associated with blight of leaf and stem in peony were leaf spot (Alternaria sp.), powdery mildew (Erysiphe aquilegiae) and rust (Cronartium flaccidum). The infection of leaf spot and powdery mildew begins from late April to midMay and rust was infected in early June. Blight time of aerial part in peony started from late May and the ratio of blight on leaf and stem was more than 50% in late Aug. Yields of root by the incidence time of blight of leaf and stem were 69.1% in late June, 65.4% in late July and $87.6{\sim}92.7$% in August and September. The number of root of more than 10mm in root diameter blighted in late June and July was much lower than in August, but the paeoniflorin content in the former was much higher than the latter.

Development of Fungal Sapstain in Logs of Japanese Red Pine and Korean Pine (소나무와 잣나무 원목에서 변재변색 발생)

  • This study was conducted to investigate the influence of storage time on the sapstain development of Japanese red pine and Korean pine logs during storage in log yard, and their stain characteristics. Japanese red pine and Korean pine trees were harvested and cut into logs in mid January of 2001. These logs were transported to the two local sawmills where they were closely stacked in remote parts of log yard. The logs were then sampled destructively by cutting seven to nine 3-cm long discs along the length of each log at intervals of 3, 4, 5, 6, and 8 month after felling. The stain coverage and maximal radial penetration of stain were measured from the discs of the sampled logs after the isolation of causal staining fungi. The sapstain was primarily infested by the attack of bark beetles and the species of bark beetle was identified as Tomicus piniperda. The main fungal species isolated from stained wood was Ophiostoma species. Based on the present study, the logs could be stored in log yard until May without stain; but stain development was rapid after May and the severity of stain increased proportionally with storage time. Korean pine was more susceptible to fungal stain than Japanese red pine. During summer storage, decay started to develop in logs and the main species were identified as Tyromyces sp. and Schizophyllum commune. Information provided in this paper would be very useful to develop more effective control strategies for sapstain prevention in Japanese red pine and Korean pine logs.

Influence of substituted phenylcarbamoyl group on the fungicidal activites of a new 5,6-dihydro-2-trifluoromethyl-1,4-oxathiincarboxanilide derivatives (새로운 5,6-dihydro-2-trifluoromethyl-1,4-oxathiincarboxanilide 유도체의 항균활성에 미치는 치환-phenylcarbamoyl group의 영향)

  • New thirty derivatives of 5,6-dihydro-2-trifluoromethyl-1,4-oxathiin carboxanilide as substrate(S) were synthesized and their fungicidal activities in vivo against rice sheath blight(Rhizoctonia solani) and wheat leaf rust(Puccinia recondita) were examined. The structure activity relationships(SAR) between the activities($pI_{50}$) and a physicochemical parameters of substituents(X) at the phenylcarbamoyl group were analyzed using the adaptive regression analysis method. The 3-methoxy, 11, 3-isopropyloxy, 13 and 3-isopropyl substituent, 25 as X on the phenylcarbamoyl group exhibited the most highest fungicidal activity against the two fungi. The fungicidal potency of the (S) against Puccinia recondita was higher than Rhizoctonia solani. In case of Rhizoctonia solani, the molecular hydrophobicity(${\pi}>0$) and resonance effect(R<0) by meta-alkyl substitutents with electron donating were important factors in determining fungicidal activity. And the HOMO energy(HOMO>0), ABSQ, sum of absolute values of the atomic charges on each atom and specific polarizability(Sp.Pol<0) of (S) were significantly influential towards fungicidal activity against Puccinia recondita.. The interaction between (S) and receptor agonist from the based on SAR studies proceeds through charge-control reaction, and conditions to show higher activity has been also discussed.

Growth Inhibition of Sclerotium Cepivorum Causing Allium White Rot by Serratia plymuthica Producing Chitinase (Serratia plymuthica AL-1이 생산하는 chitinase에 의한 대파 흑색썩음균핵병균의 생육억제)

  • An allium rhizobacterium Serratia plymuthica AL-1 was previously selected as a biocontrol agent of allium white rot. The chitinase from S. plymuthica AL-1 produced in medium containing colloidal chitin was purified by ammonium sulfate precipitation (40~70%), affinity adsorption, column chromatography on DEAE-sephadex A-50 and sephadex C-200 gel filtration. The enzyme was purified 10.8-fold with a yield of 7.3% from the starting culture broth. The purified chtinase gave a single band on sodium dodecyl sulfate polyacrylamide gel electrophoresis, it's molecular weight was estimated to be 55 kDa. The optimum pH and temperature of the purified enzyme were pH 5.5 and $55^{\circ}C$, respectively and it is stable up to $50^{\circ}C$ and maintains around 90% of its activity for 60min. The enzyme were activated by $Ca^{2+}$, $Mn^{2+}$ and $Mg^{2+}$ and inhibited by $Cu^{2+}$, SDS, $\rho$-CMB, MIA, respectively. The purified chitinase showed broad spectrum of antifungal activities against plant pathogenic fungi Sclerotium cepivoruin, Alternana alternnta, Colletotrichum glceosporioidrs, Phoma sp., Sclerotinia sclerotiorum, Stemphylium solani, Fusarium oxysporium f. sp. niveum but rarely inhibited Phytophthora capsici and Pythium ultimum.. The purified chitinase from S. plymuthica AL-1 caused swelling, lysis, deceleration and degradation of the hyphal tips of S. sczerotiorum causing allium white rot. It suggest that S. prymuthica AL-1 chitinase play an important part in the bifunctional chitinase / lysozyme activity.

  • An indigenous antagonistic bacterium Bacillus sp. 7079 was isolated from a local soil sampled at Kyongju area in Korea . The strain has strong antagonistic ability which was originated from multifunctional mechanisms of chitinase and antibiotic and is a powerful antagonistic biocontrol agent against red-pepper rotting fungus Phytophthora capsici and Wilt fungus Fusarium oxysporum. The chitinase might degrade the cell wasll for Fusarium species. The selected Bacilus sp. 7079 was identified as a Bacillus amyloliquefaciens 7079. The maximal production of the chitinase from B, amyloliquefaciens 7079 were obtained in chitin-yeast extract medium containing 0.7%, $K_2$$HPO_4$, $0.2KH_2$$PO_4$, 0.1% ($NH_4$)$_2$$SO_4$, 0.05% sodium cirate, 0.01% $MgSO_4$$7H_2$O, 0.1% yeast extract and 0.1% colloidal chitin after cultivation of 3 days at pH 7.0 and $30^{\circ}C$. The best carbon and nitrogen sources for the production of the chitinase from B amyloliquefaciens 7079 were determined to be 0.1% colloi- dal chitin and 0.15% proteose peptone NO 3 respectively, The antagonistic activity of B amyloliquefaciens 7079 was confirmed using P. capsici by in vivo pot test with red-pepper plant.

Development of Microbial Inoculant Using By-product of Oriental Herbal Medicine (한약재박을 이용한 미생물제제의 개발)

  • The development of microbial inoculant was conducted using a by-product of oriental herbal medicine. The constituent of the by-product, which was high in organic matter, was 11.3% of crude protein, 5.1% of crude lipid, 49.7% of NDF (neutral detergent fiber), and 33.8% of ADF (acid detergent fiber). Microorganisms isolated from the by-product of oriental herbal medicine were 35 species. Among them, 6 bacterial species, 4 fungal species, 2 actnomycetes species, and 1 yeast species were effective in the utilization of the by-products. The 13 strains screened were tested for the plant growth-promoting effect in soybean seedling. BL-333 strain was found to increase the soybean yield by about 23% as compared with control. The strain BL-333 was identified as Paenibacillus marcerans. P. marcerans BL-333 showed high anti-fungal activities against virulent fungi, especially Fusarium sp. and Collectotrichum sp. Yields of plants which were inoculated with microbial inoculant prepared with P. marcerans BL-333 and by-product of oriental herbal medicine were found to be higher than control by $3{\sim}24%$. The yield was especially promoted in lettuce, radish, chinese cabbage and cucumber plants.

A Study on Lichen Diversity in the Placenta Chamber of Prince King Sejong's Sons, Seongju (성주 세종대왕자 태실에 분포하는 지의류상 연구)

  • A lichen is a complex microorganism comprising fungi and algae. Various lichens are commonly found in cultural heritage and are reportedly related to weathering. Lichens are commonly studied in Europe; however, few reports exist on lichens in Korean cultural heritage. The present study was conducted as part of the Korean National Arboretum's "A study on the Fungus and Lichen." We report the lichen diversity found at the Placenta Chambers of King Sejong's Sons, Seongju. A total of 65 lichens were collected from 19 placenta chambers and one memorial stone. We identified 40 species belonging to 10 genera, 17 families, and 23 orders. Morphological identification revealed that foliose lichens in the form of leaves(24, 36.9%) and crustose lichens in the form of crust(24, 36.9%) were distributed at a higher percentage than that of fruticose lichen in the form of branches(17, 26.2%). Xanthoparmelia coreana(foliose lichen) and Lepraria sp.(crustose lichen) were the most collected lichens and were distributed on 19 chambers. In addition, Clodonia, a fruticose lichen, had the highest species diversity with eight species. The highest species diversity of lichens was identified on the chamber of Prince Kyeyang(16 lichens), whereas the chamber of Prince Geumsung had the lowest species diversity(five lichens). This study will be used as primary data for the study of distribution of lichen diversity in Korean cultural heritage.

Prevention from microbial post-harvest injury of fruits and vegetables by using grapefruit seed extract, a natural antimicrobial agent (천연항균제처리에 의한 과채류의 선도유지 및 병해방지에 관한 연구 -저장중 병리적 장해 방지를 중심으로-)

  • In order to retain the freshness of fruits and vegetables and to reduce the rate of disease damage, grafruit seed extract (GPSE), natural microorganism control agent, was applied during the preservation process of fresh fruits and vegetables. GFSE showed an effective inhibitory action against plant putrefactive bacteria and fungi which were involved in the decay of fruits and vegetables. Minimal inhibitory concentrations for GFSE against the microbes were in the range of 50 to 2,000 ppm. Direct observation of microbial cells and spores using electron microscopy showed their function was destroyed by the treatment of the dilute solutions of GFSE. Fresh Welsh onions, onions and red peppers treated with GFSE and stored in polyethylene film (0.1 mm) retained better quality in color and texture than the non-treated control. GFSE was efficient in controlling the germination of potatoes. It was observed that GFSE would reduce disease damages and have bactericidal and fungicidal properties during the storage of such fruits and vegetables as zucchinis, cucumbers, tomatoes and mandarin oranges.

Impacts of Chemical Properties on Microbial Population from Upland Soils in Gyeongnam Province (경남지역 밭 토양 화학성분이 미생물 생태에 미치는 영향)

  • Soil management for environment-friendly agriculture depends on the effects of soil microbial activities and soil fertility. To improve soil health for the upland crops, this study evaluated a relationship between soil chemical properties and soil microbial diversities at 25 sites in upland soils in Gyeongnam Province. The average nutrients in the upland soils were 1.7 times for available phosphorous, 1.4 times for exchangeable potassium and 1.5 times for exchangeable calcium higher compared to recommend concentrations in the upland soils. We found a significant positive correlation between the soil organic matter and the soil microbial biomass C (p<0.01). Contents of organic matter and dehydrogenase in the inclined piedmont soils were significantly higher than those in the other topographical soils (p<0.05). In addition, concentrations of organic matter and microbial biomass C in the loam soils were significantly higher than in the silt loam soils (p<0.05). In principal component analyses of chemical properties and microbial populations in the upland soils, our findings suggested that available phosphorous should be considered as potential factor responsible for the clear upland soils differentiation. The soil organic matter was positive correlation with Bacillus sp. and fungi, whereas soil pH was also positive correlation with Pseudomonas sp. in upland soils.

Inhibition of in Vitro Growth of Three Soil-borne Turfgrass Diseases by Antagonistic Bacteria from Composted Liquid Manure (가축분뇨액비의 길항미생물에 의한 토양전염성 병원균의 생육억제 효과)

  • This study was conducted to test in vitro the antagonistic effect of composted liquid manure (CLM) against soil-borne turfgrass pathogenic fungi, Rhizoctonia solani AG-2-2 (IIIB) (brown patch), R. solani AG-2-2 (IV) (large patch), and Sclerotinia homoeocarpa (dollar spot) for environmentally friendly turfgrass management. CLMs were collected from 9 livestock excretion treatment facilities around the country including Gunwi (GW), Hapcheon (HC), Hoengseong (HS), Icheon (IC), Iksan (IS), Muan (MA), Nonsan (NS), and Yeoju (YJ). CLMs of IC, GW, and IS showed s ignificant (p < 0.05) mycelium growth inhibition that was 17.8%, 20.4%, and 48.0% against R. solani AG-2-2 (IIIB), R. solani AG-2-2 (IV), and S. homoeocarpa, respectively. A t otal of 110 bacterial isolates were obtained from the CLMs that showed antagonistic effects. Among them, 5, 4, and 10 microbe isolates showed promising antifungal activity against mycelium growth of R. solani AG-2-2 (IIIB), R. solani AG-2-2 (IV), and S. homoeocarpa, respectively. The bacterial isolates ICIIIB60, GWIV70, and ISSH20 effectively inhibited the mycelial growth of three soil-borne turfgrass pathogens. Selected bacterial isolates were identified as Alcaligenes sp., Bacillus licheniformis Ab2, and B. subtilis C7-3 through 16s rDNA gene sequence analysis. Among 5 fungicides, the most compatible fungicide with ICIIIB60, GWIV70, and ISSH20 was tebuconazol, toclofos-methyl and toclofos-methyl, respectively. These findings suggested that CLMs could be effectively used not only as organic liquid fertilizer sources but also as biological control agents for soil-borne turfgrass diseases such as brown patch, large patch, and dollar spot.
