Plant Growth-Promoting Activity of Endophytic Fungi Isolated from the Roots of Native Plants in Dokdo Islands (독도 자생식물의 뿌리로부터 분리된 내생균의 식물생장촉진 활성)

  • Endophytic fungal strains were isolated from the roots of six species plants in the Dokdo islands. Native plant samples, such as Artemisia japonica, Chenopodium album and Solanum nigrum were isolated from Dongdo, and those such as Cyrtomium falcatum, Dianthus longicalyx and Tetragonia tetragonoides were isolated from Seodo. In total, thirty two fungal strains were isolated from these native plants. To identify the fungal strains, polymerase chain reaction (PCR) amplification of internal transcribed spacer (ITS: containing ITS1, 5.8s and ITS2 region) regions was done with universal primers ITS1 and ITS4. Endophytic fungi of four species were isolated from A. japonica, eight species from C. album, three species from S. nigrum, three species from C. falcatum, three species from D. longicalyx and eleven species from T. tetragonoides. Culture filtrates (CF) of isolated endophytic fungi were used to treatwaito-c rice seedlings to test plant growth-promoting activity. As a result of bioassay, Ca-5-2-2 strain isolated from C. album expressed highest plant growth-promotion activity. Of all the endophytic fungi isolated, Penicillium sp., Fusarium sp. and Aspergillus sp. were the most abundantly distributed fungal strains in the six plants used in this study.

Nematode-Trapping Fungi Showed Different Predacity among Nematode Species (선충 종류별 4종 포식성곰팡이의 포식력 차이)

  • Nematode-trapping fungi develop trap and consume nematodes are an important part of the subsoil ecosystem and they share a special predator-prey relationship. Four nematode-trapping species, there with adhesive network, Arthrobotrys oligospora, A. sinensis, A. thaumasia and one with constricting ring, Drechslerella brochopaga were collected from soils in Korea and tested their predacity against 12 different nematode species. They were three feeding groups, plant-parasitic (Meloidogyne incognita and Pratylenchus penetrans), fungivorous (Aphelenchus avenae), bacteriovorous (Betlerius sp. and Diplogasteritus sp. in diplogasterid, Panagrolaimus labiatus, P. multidentatus in panagrolaimid, Mesorhabditis irregularis, Pelodera strongyloides and Rhabditis sp., in rhabditid, and Acrobeloides sp. in cephalobid). Results showed that nematode-trapping fungi successfully captured most of nematodes in Petri dish in the group of plant-parasitic nematodes and rhabditids, moderately and variably in other nematodes in 15 days. But it didn't captured A. avenae and Acrobeloides sp. both belongs to c-p group 2. Numbers of Acrobeloides sp. and A. avenae even increased during the test period. The results of this study indicated that nematode-trapping fungi may have specificity among nematode species.

  • The yields and biological efficiency depending on the capacity of culture bottles in sawdust cultivation was very high in using 850㎖ bottle. In reduction ratio of sawdust, lignin and holocellulose, mixed fungi among the three cultures showed the most reduction ratio.

Frequency and Wood Decaying Ability of Butt-rot Fungig Isolated form Larch (Larix leptolepis) (낙엽송근주심재부후병균(根株心材腐朽病菌)의 분포빈도(分布貧道)와 목재열화능력(木材劣化能力))

  • Butt-rot fungi were isolated from decayed portion in butt rotted larch trees[Larix leptolepis(Sieb. et Zucc.) Gord.]. From a total of 216 affected trees in 38 forests of five provinces, Sparassis crispa(Wulf. ex Fr.) Fr. was isolated with the highest frequency, i.e.. 28% of the total trees. Laetiporus sulphureus(Fr.) Bond. et Sing, with 19%, and Phaeolus schweinitzii(Fr.) Pat. with 8%. However, none of these fungi was isolated from 45% of the trees. Wood decaying abilities and interaction of four basidiomycetous and three non-basidiomyo:etous fungi isolated from butt rotted trees were investigated. The weight loss of larch wood blocks, when exposed to the culture of a fungus for six months, was 19.6% for P. schweinitzii, 10% for S. crispa, 8.2% for L. sulphurous, 5.1% for Coriolus hirsutus, and less than 4.4% for three non-basidiomycetous fungi. In incubation with combinations of the fungi, the wood decay activity of S. crispa, L. sulphurous and C. hirsutus was increased by the non-basidiomycetous fungi of Geotrichum candidacy, G. sp. and Verticillium sp., but that of P. schweinitzii was not.

  • Culture conditions affecting mycelial growth and sporulation of P. theae, SP2, SP3 and P. longiseta which causing leaf blight on oriental persimmon leaf blight have been investigated. The optimum temperature for the mycelial growth and sporulation on potato dextrose agar was $25{\sim}30^{\circ}C$ in all the fungi, but was inhibited and finally arrested at 10 and $30^{\circ}C$. The optimum pH for mycelial growth and sporulation were ranged at pH 4.5~5.0 and 5.0~6.0 in all the fungi. Lenonian agar, potato sucrose agar and oatmeal medium were good culture media for the mycelial growth and sporulation of all the fungi. The effective sources of nitrogen and carbon for the mycelial growth were tryptone, glycine, starch, dextrose, galactose and lactose. Glutamic acid, peptone and tryptone were good nitrogen sources for sporulation of the fungi. Sucrose, starch and galactose were also good carbon sources for sporulation. Generally, vitamins had no effect on mycelial growth and sporulation. The pH of the potato dextrose broth inoculated with SP2, SP3 and P. theae and P. longiseta was changed from 7.0 to 4.5~4.7 and 5.0~5.4 after incubating for 10 days, respectively. But, the initial pH of the medium adjusted to 5.0 was lowered to 4.5~4.7 after incubating for 10 days.

Antifungal Activity of Securinine against Some Plant Pathogenic Fungi

  • The alkaloid securinine was assessed against spore germination of some plant pathogenic and saprophytic fungi (Alternaria alternata, Alternaria brassicae, Alternaria brassicicola, Curvularia lunata, Curvularia maculans, Curvularia pallenscens, Colletotrichum musae, Colletotrichum sp., Erysiphe pisi, Helminthosporium echinoclova, Helminthosporium spiciferum, Heterosporium sp.). Spore germinations of all the tested fungi were inhibited. Alternaria brassicicola, C. lunata, C. pallenscens and H. spiciferum were highly sensitive as complete inhibition of spore germination was observed at very low concentrations (200 ppm).

  • This survey was conducted to identify the fungal pathogens on the imported seeds during two months from September to October in 1985. Twenty-four species of fungi were detected from six kinds of the imported seeds and the following eight species of fungi have not been reported before in Korea.

  • Monacroporium ullum sp. nov. the captures nematodes on adhesive spherical knobs was isolated from soil around Codonopsis lanceolata at Ulleung island, Korea. The spindle-shaped conidia sized $17-44{\mu}m\;(26.5{\mu}m)\;long,\;7-10{\mu}m\;(8.1{\mu}m)$ wide, containing from 1 to 4 cross-walls but most often divided by 2 septa (47%). Resting bodies sized $57{\times}30{\mu}m$. Arthrobotrys amerospora has almost spherical non-septate conidia with a small truncate protuberance at the base and sized $20-27{\mu}m\;(23.3){\mu}m$ long and $11-17{\mu}m\;(14.1){\mu}m$ wide. Conidiophores are somewhat longer $362.8{\mu}m\;(311-418{\mu}m)$ than its original description ($75-250{\mu}m$).

  • The tetrahydropahnatine alkaloid was assayed against spore germination of some saprophytic and pathogenic fungi e.g., Alternaria solani, A. brassicicola, A. brassicae, A. alternata, Erysiphe pisi, Curvularia lunata, C. pallescens, C. maculans, Curvularia species, Colletotrichum species, C. musae, Helminthosporium echinoclova, H. pennisetti, H. spiciferum, and Heterosporium sp. It inhibite spore germination of all the fungi tested. Colletotrichum spp. Curvularia lunata, Helminthosporium spiciferum and Heterosporium sp. were most sensitive as complete inhibition of spore germination was observed at very low concentration(200 ppm).

  • The endophytic fungi were isolated from red pine trees (Pinus densiflora) that were distributed in three sites of Chungbuk, Korea. Twenty fungal isolates were isolated from 16 trees and divided into 8 groups by morphological characters. The fungal isolates were identified using the sequences of ITS region of rDNA; Lophodermium complex, Sydowia polyspora, Hymenula sp., Sistotrema brinkmannii, Septoria pini-thunbergii, Earliella sp. Lophodermium spp. were the most frequently found fungal species the across sites and firstly detected from Pinus species in Korea by molecular work.
