Essential Oil Prepared from Cymbopogon citrates Exerted an Antimicrobial Activity Against Plant Pathogenic and Medical Microorganisms

  • Essential oils are mixtures of volatile, lipophilic compounds originating from plants. Some essential oils have useful biological activities including antimicrobial, spasmolytic, antiplasmodial, and insect-repelling activities. In this study, we tested the antimicrobial activity of essential oil prepared from the aromatic plant, Cymbopogon citrates, against three important plant pathogenic and medical microorganisms, Pectobacterium carotovorum, Colletotrichum gloeosporioides, and Aspergillus niger. It effectively inhibited the growth of the bacterium, Pectobacterium carotovorum, in a dose-dependent fashion, and 0.5% of the oil inhibited the growth of bacteria completely. Similarly, the essential oil inhibited the growth of plant pathogenic fungus, Colletotrichum gloeosporioides, and the addition of 1% of essential oil completely inhibited the growth of fungus even after 5 days of culture. Finally, it effectively inhibited the growth of the medically and industrially important fungal species, Aspergillus spp. These results suggest that the essential oil from Cymbopogon citrates may be an environmentally safe alternative to inhibit antimicrobial agents for various uses.

The Composition of Essential Oil from Nepeta cataria and Its Effect on Microorganism

  • We analyzed the total yields and composition of essential oils in leaf extracts of Nepeta cataria by Gas Chromatography Mass Spectrometry (GC-MS). Thirty-six compounds representing 97.0% of total oil were detected. The major constituents of essential oils in Nepeta cataria were nepetalactone (90.9%), unidentified compound (Retention time 17.35; 1.82%), 1,8-cineol (1.49%), ${\beta}-caryophyllene$ (1.12%), and ${\beta}-pinene$ (1.078%). The volatile compounds in leaf extracts of N. cataria concentrated to nepetalactone ($88.83{\sim}93.33%$) remarkably. In the essential oil of N. cataria cis,trans-nepetalactone ($30.2{\sim}37.8%$) and cis,cis-nepetalactone ($31.5{\sim}37.0%$) were found as the main constituents. The effects of essential oil of N. cataria on the growth of six microorganisms (Bacillus cereus, B. subtilis, B. amyloliquefaciens, Escherichia coli, Staphylococcus aureus subsp. aureus, and Pseudomonas aeruginosa) were investigated. The essential oil of N. cataria had strong inhibitory effect on the growth of three fungal species (Bacillus cereus, B. subtilis, and B. amyloliquefaciens). The essential oil from N. cataria was found to have a low antimicrobial activity against Staphylococcus aureus subsp. aureus, while no activity were found against Escherichia coli and Pseudomonas aeruginosa. Results indicate the significant antimicrobial effect, which may be depended on the yield of nepetalactone.

피부사상균 균사의 형태학적 변화를 통한 일본잎갈나무 정유의 항진균 활성 효과 구명 (Evaluation on Anti-Dermatophyte Effect of Larix (kaempferi) Essential Oil on the Morphological Changes of Eermatophyte Fungal Hyphae)

  • 본 연구는 피부사상균 Epidermophyton floccosum, Trichophyton mentagrophytes, Trichophyton rubrum에 대한 일본잎갈나무 정유의 항진균 활성을 평가하고, 항진균 유효성분을 구명하고자 하였다. 또한 일본잎갈나무 정유에 노출된 피부사상균의 균사를 전자현미경으로 관찰하여 일본잎갈나무 정유의 항진균 효과를 균사의 형태학적 변화를 통하여 밝히고자 하였다. 일본잎갈나무 정유의 주성분은 (-)-bornyl acetate였으며, MIC는 모든 균에 대해 125 ppm을 나타냈다. 한천희석법을 통한 항진균 활성 평가에서는 모든 균에 대해 500 ppm 이상에서 100%의 활성, 100 ppm의 낮은 농도에서도 50% 이상의 활성을 나타냈다. SEM과 TEM을 통해 균사를 관찰한 결과, 일본잎갈나무 정유에 노출된 E. floccosum은 정상세포와 다른 균사 형태가 관찰되었다. 팡이실 모양의 격막을 갖고 있는 곧고 매끈한 표면의 정상세포와는 달리, 처리구에서는 균사 표면에 주름이 있었으며 균사가 터지거나 접히고 부풀어 오른 것, 세포소기관과 세포벽이 붕괴된 것도 관찰되었다. 분획을 통해 일본잎갈나무 정유의 항진균 유효성분을 확인한 결과, (-)-${\tau}$-muurolol, (+)-terpinen-4-ol, ${\alpha}$-terpineol, ${\alpha}$-cadinol 등의 terpene alcohol 화합물이었다.

Fungal Biotransformation of Monoterpenes Found in Agro-Industrial Residues from Orange and Pulp Industries into Aroma Compounds: Screening Using Solid Phase Microextraction

  • The biotransformation of monoterpenic agro-industrial wastes (turpentine oil and essential orange oil) was studied. More than 40 fungal strains were isolated from Brazilian tropical fruits and eucalyptus trees and screened for biotransformation of the waste substrates. Solid phase microextraction was used to monitor the presence of volatile compounds in the headspaces of sporulated surface cultures. The selected strains were submitted to submerged liquid culture. The biotransformation of R-(+)-limonene and ${\alpha},\;{\beta}-$ pinenes from the oils resulted in ${\alpha}-terpineol$ and perillyl alcohol, and verbenol and verbenone, respectively, as the main products. The selected strains were also placed in contact with ${\alpha}-$ and ${\beta}-$ pinenes standards. It was confirmed that verbenol, verbenone, and ${\alpha}-terpineol$ were biotransformation products from the terpenes. A concentration of 90 mg/L of verbenone was achieved by Penicillium sp. 2360 after 3 days of biotransformation.

신경교(神經膠) 성상세포(星狀細胞)의 세포자감사(細胞自減死)에 있어서 박하오일의 효과(效果) (Effects of Peppermint Oil on Apoptosis of Astrocytes)

  • Recently, essential oils are used for aromatherapy. Most essential oils are said to be anti-bacterial; some may be anti-viral or anti-fungal. I investigated the effects of peppermint pure essential oil on the heat shock-induced apoptosis in human astrocyte cell line CCF-STTGI. In previous studies, heat shock has been reported to induce the apoptosis or programmed cell death through the activation of caspase-3. We studied the heat shock-induced apoptosis through flow cytometry, DNA electrophoresis, and giemsa staining. Interestingly, these events were inhibited by pretreatment of peppermint pure essential oils in CCF-STTGl cells. Peppermint oil also inhibited the heat shock-induced apoptosis in primary cultured rat astrocytes. In addition, this Peppermint essential oil inhibited the heat shock-induced activation of caspase-3. These results suggest that peppermint pure essential oils may modulate the apoptosis through the activation of the interleukin-I -converting enzyme-like protease.

Phytochemical Screening, Isolation, Characterization of Bioactive and Biological Activity of Bungkang, (Syzygium polyanthum) Root-bark Essential Oil

  • Bungkang (Syzygium polyanthum) is a medium to tall plant which produces medicinal root-bark, the plant is normally found along inland river bank and produces small white flowers and fruits. Essential oils are among the most interesting components of the plant extracts consisting mostly of monoterpenoid or sesquiterpenoids. They are used as therapeutic agents in ethno, conventional, and complementary alternative medicines. Investigation and evaluation of the essential oil of Syzygium polyanthum as well as the antibacterial, antioxidant and antifungal activity was ascertained. The experiment was performed. 100 chemical constituents were obtained and two pure compound was isolated as Eugenol (1) and Farnesol (2). Significant growth inhibition of Staphylococcus aureus, (ATCCⓒ25923) Klebsiellia pneumonia (ATCCⓒ19155), Salmonella typhi (ATCCⓒ14028) and Escherichia coli (ATCC©25922) and the fungal strains Aspergillus flavin, Aspergillus niger, Candida, tropicalis, and Fusarium oxysporium was observed from the essential oil at concentration of 500 ㎍/mL. Antioxidant potential was observed to be strong of 18.42 ㎍/mL when compared to the control of 15.23 ㎍/mL. The result indicated that the oil obtained from root-bark of Syzygium polyanthum can be considered as an agent for antioxidant, antibacterial and antifungal in pharmaceutical food and cosmetic industries trails.

난황유를 이용한 단감 흰가루병 방제 (Control of Powdery Mildew on Sweet Persimmom Using Cooking Oil and Yolk Mixture in the Orchard)

  • 2010년부터 2011년까지 2년 동안 경상남도농업기술원 단감연구소 포장에서 단감 흰가루병(P. kakicola)을 방제하기 위하여 난황유를 3회 지상부에 살포한 다음 5일 후 병해 방제효과를 검정하였다. 난황유 처리구에서 병반 면적율을 조사한 결과 13.4%였으며, 단감 흰가루병에 대한 방제효율은 80.5%이었다. 무처리구에서 68.6% 발생하였다. 난황유를 처리한 후 주사전자현미경으로 관찰한 흰가루병의 균사는 형태적으로 세포가 원형질막이 파괴되어 세포내용물이 소실되어 쭈그러졌다. 이것은 난황유가 흰가루병의 세포벽이나 세포막을 파괴하여 생긴 것으로 사료된다.

Systematic Analysis of the Anticancer Agent Taxol-Producing Capacity in Colletotrichum Species and Use of the Species for Taxol Production

  • Paclitaxel (taxol) has long been used as a potent anticancer agent for the treatment of many cancers. Ever since the fungal species Taxomyces andreanae was first shown to produce taxol in 1993, many endophytic fungal species have been recognized as taxol accumulators. In this study, we analyzed the taxol-producing capacity of different Colletotrichum spp. to determine the distribution of a taxol biosynthetic gene within this genus. Distribution of the taxadiene synthase (TS) gene, which cyclizes geranylgeranyl diphosphate to produce taxadiene, was analyzed in 12 Colletotrichum spp., of which 8 were found to contain the unique skeletal core structure of paclitaxel. However, distribution of the gene was not limited to closely related species. The production of taxol by Colletotrichum dematium, which causes pepper anthracnose, depended on the method in which the fungus was stored, with the highest production being in samples stored under mineral oil. Based on its distribution among Colletotrichum spp., the TS gene was either integrated into or deleted from the bacterial genome in a species-specific manner. In addition to their taxol-producing capacity, the simple genome structure and easy gene manipulation of these endophytic fungal species make them valuable resources for identifying genes in the taxol biosynthetic pathway.

The Major Postharvest Disease of Onion and Its Control with Thymol Fumigation During Low-Temperature Storage

  • Onion (Allium cepa L.) is one of the major vegetable crops in Korea that are damaged and lost by pathogenic fungal infection during storage due to a lack of proper storage conditions. The aim of this study was to determine an appropriate control measure using thymol to increase the shelf life of onions. To control fungal infections that occur during low-temperature storage, it is necessary to identify the predominant fungal pathogens that appear in low-temperature storage houses. Botrytis aclada was found to be the most predominant fungal pathogen during low-temperature storage. The antifungal activity of the plant essential oil thymol was tested and compared to that of the existing sulfur treatments. B. aclada growth was significantly inhibited up to 16 weeks with spray treatments using a thymol solution. To identify an appropriate method for treating onions in a low-temperature storage house, thymol was delivered by two fumigation treatment methods, either by heating it in the granule form or as a solution at low-temperature storage conditions (in vivo). We confirmed that the disease severity was reduced up to 96% by fumigating thymol solution compared to the untreated control. The efficacy of the fumigation of thymol solution was validated by testing onions in a low-temperature storage house in Muan, Jeollanam-do. Based on these results, the present study suggests that fumigation of the thymol solution as a natural preservative and fungicide can be used as an eco-friendly substitute for existing methods to control postharvest disease in long-term storage crops on a commercial scale.

Producing Alkaline Lipase by Fusarium oxysporum Using Unconventional Medium Components

  • This study reports the use of different inducing agents (olive, soybean, and used frying oils) and culture mediums [synthetic medium (SM), whey protein, and corn steep liqueur (SL)] to optimize the production of lipase by Fusarium oxysporum. A relationship among the inoculum size, presence of a fat source, fungal growth, and lipase production was evident during the fermentation. The best results were achieved when the inoculum was grown in SM or SL and the fermentation was developed in SM with frying oil as the inducing agent. The maximum activity (about 15 U/mL) was obtained after a 72 hr cultivation.
