A Study on the Quality Characteristics of Jeung-Pyun by the Addition of Chitosan-oligosaccharide (키토산 올리고당 첨가량에 따른 증편의 품질에 관한 연구)

  • The purpose of this study was to investigate the quality characteristics of Jeung-Pyhn by the addition of chitosan-oligosaccharide (COS, Chitoligo-L). Jeung-Pyun is a traditional Korean fermented rice cake leavened by yeast. To develop a functional Jeung-Pyun, COS was added to the rice at the levels of 0, 2, 4 and 6% of the 2-hour-soaked rice. Physicochemical, sensory, rheological and inner structure characteristics of Jeung-Pyun were examined. The pH values of COS Jeung-Pyun batters were decreased with fermentation time. In sensory evaluation, the uniformity of the grain and overall quality of Jeung-Pyun added with 2% COS had the highest score among the samples. In He rheological properties measured with rheometer, the hardness of Jeung-Pyun with COS was higher than control. Cohesiveness of Jeung-Pyun added with 4% COS and gumminess, brittleness and springiness of Jeung-Pyun added with 2% COS were the highest among the samples. The inner structure observed by scanning electron microscope showed that Jeung-Pyun added with 2% COS had the largest and the most uniform air cells. In conclusion, Jeung-Pyun with 2% COS had positive effects on the qualify of Jeung-Pyun.

  • The functional properties of novel sugar derivative sweeteners, fructo-oligosaccharide, maltitol, sorbitol and high maltose syrup(HMS) were examined for their humactant effect, lactic acid bacterial growth, Streptococcus mutants growth and relative sweetness compared to sucrose. Sorbitol exhibited remarkably high water activity reducing capacity, whereas fructo-oligosaccharide and maltitol showed the same level as sucrose. Maltitol showed distinct anti-bacterial(bacteriocidic) effect against Stc. mutants and most of lactic acid bacteria tested except for L. plantarum. The molar basis relative sweetness of sugar derivatives in comparison with 1%(w/w) level of sucrose were 0.69 for Neosugar(fructo-oligosaccharide), 0.21 for sorbitol, 0.50 for maltitol and 0.27 for HMS.

Effects of Dietary Oligosaccharide on the Blood Glucose and Serum Lipid Composition in Streptozotocin-Induced Diabetic Rats (Streptozotocin 유발 당뇨쥐의 혈당 및 혈중 지질조성에 미치는 Oligosaccharide의 영향)

  • This study was conducted to examine the effects of dietary oligosaccharide on the blood glucose and serum lipid composition in streptozotocin (STZ)-induced diabetic rats. Sprague-Dawley male rats weighing 150$\pm$10g were randomly assigned to one normal and four STZ-induced diabetic groups. Diabetic groups were classified to basal diet(DM group) 10% xylooligosaccharide diet(DM-XO group) 10% isomaltooligosaccharide(DM-IMO group) and 10% fructooligosaccharide (DM-FO). Diabetes was experimentally induced by intravenous injection of 50 mg/kg of body weight of STZ in citrate buffer (pH 4.3) after feeding of experimental diets for 4 weeks. The rats were fed with experimental diet for further 4 weeks in diabetic state. The oligosaccharide diets were not effected on the body weight food intakes and food efficiency ratio. The oligosaccharide diets were also not effected on the body weight food intakes and food efficiency ratio. The oligosaccharide diets were also not effected on the weights of liver kidney and small intestine but the weight of cecum was significantly increased on the groups of xylooligosaccharide and isomaltooligosaccharide diet. The levels of oral glucose tolerance test was more effectively improved by DM-XO group. The levels of blood glucose were markedly lower in oligosaccharide supplemented groups than that of DM group. The levels of blood glucose were markedly lower in oligosaccharide supplemented groups than that of DM group. Activities of two intestinal enzymes such as lactase and sucrase in DM-XO and DM-FO groups were lower than that of DM group while activity of maltase was lower only in DM-XO in DM-FO groups than that of DM-group respectively. The levels of serum triglyceride in DM-XO group were lower than that of DM-group respectively. The levels of serum triglyceride in DM-XO groups were lower than that of DM group however was no significant differences among the oligosaccharide groups. These results suggest that dietary oligosaccharide may act as functional food to be capable of improving carbohydrate and lipid metabolism in diabetic rats.

  • Various recombinant deletion mutants were constructed from cycloinulo - oligosaccharide fructanotransferase(CFTase) gene of Xanthomonas oryzae #5 . The mutants were expressed in Escherichia coli DH5${\alpha}$. We were able to obtain three recombinant proteins were purified, and examine their CFTase and hydrolyzing activity. N-terminal deletion mutant had both CFTase activity and hydrolyzing activity. however, in C-terminal and N,C-terminal deletion mutant disappeared CFTase activity, but hydrolyzing activity remained. From there results, it seems that the C-terminal region(amino acid $1173{\sim}1333$) is important for cyclization.

Quality Characteristics of the Hasuo (Polygoni multiflori Radix) Muffin Prepared with Different Types of Sweeteners (하수오 머핀 제조시 당 종류가 품질에 미치는 영향)

  • The purpose of this study was to develop a functional hasuo (Polygoni Multiflori Radix) muffin prepared via the replacement of sucrose (SM) with oligosaccharide (fructooligosaccharide, isomaltooligosaccharide, galactooligosaccharide). The fructooligosaccharide-added hasuo muffin showed the highest weight while the volume and specific volume were lower in the oligosaccharide-added group than in the sucrose-added sample. The pH of the sucrose-added sample was higher than that of the oligosaccharide-added sample. The lowest lightness value was found in the sucrose-added sample, but the highest redness and yellowness values were found in the sucrose-added sample. The springiness and cohesiveness were highest in the fructooligosaccharide-added sample. The DPPH-radical-scavenging activity of the hasuo muffin including sucrose was 38.67, while that including the oligosaccharide group ranged from 42.87 to 65.95. In the sensory evaluation of the hasuo muffin, the fructooligosaccharide-added sample obtained the highest score in after-swallow, flavor, taste, and texture. The hasuo muffin with fructooligosaccharide and isomaltooligosaccharide obtained high scores in overall acceptability. These results suggest that hasuo muffin with fructooligosaccharide and isomaltooligosaccharide have the potential to become commercially successful muffins.

Effect of Citrus Pectin Oligosaccharide Prepared by Irradiation on High Cholesterol Diet B6.KOR-ApoE Mice

  • Effect of citrus pectin oligosaccharides produced by irradiation was studied on the ability to improve lipid metabolism and hypercholesterolemia in mice fed high cholesterol diets. A total of 35 mice were divided into 5 groups and fed the following diets for 6 weeks: normal diet (C), 0.5% cholesterol (CH), 0.5% cholesterol+5% non-irradiated pectin (P), 0.5% cholesterol+5% irradiated pectin at 20 kGy (PIR), and 0.5% cholesterol+5% irradiated at 20 kGy and dialyzed (PIR-F). CH group had significantly higher serum triglycerides, total cholesterol, and low density lipoprotein (LDL)-cholesterol contents than pectin oligosaccharide-treated groups (p<0.05). Triglycerides and total cholesterol contents was the lowest in C and PIR-F and followed by PIR and P group, and CH group had significantly higher LDL-cholesterol. Serum high density lipoprotein (HDL)-cholesterol content in C group was not different from that in CH and P groups, but lower than that of PIR and PIR-F groups. These results suggest that pectin oligosaccharides produced by irradiation can reduce the levels of serum triglyceride, total cholesterol, and LDL-cholesterol in the blood of mice fed high-cholesterol diets and therefore, irradiation can be used as a tool to produce functional oligosaccharides from citrus pectin.

  • BACKDROUND/OBJECTIVE: Constipation is a condition that can result from intestinal deformation. Because humans have an upright posture, the effects of gravity can cause this shape deformation. Oligosaccharides are common prebiotics and their effects on bowel health are well known. However, studies of the physiological functionality of a product that contains both lactulose and galactooligosaccharides are insufficient. We investigated the constipation reduction effect of a dual-type oligosaccharide, Dual-Oligo, in loperamide-treated rats. MATERIALS/METHODS: Dual-Oligo consists of galactooligosaccharides (15.80%) and lactulose (51.67%). Animals were randomly divided into four groups, the normal group (normal), control group (control), low concentration of Dual-Oligo (LDO) group, and high concentration of Dual-Oligo (HDO) group. After 7 days of oral administration, fecal pellet amount, fecal weight, watercontent of fecal were measured. Blood chemistry, short-chain fatty acid (SCFA), gastrointestinal transit ratio and length and intestinal mucosa were analyzed. RESULTS: Dual-Oligo increased the fecal weight, and water content of feces in rats with loperamide-induced constipation. Gastrointestinal transit ratio and length and area of intestinal mucosa significantly increased after treatment with Dual-Oligoin loperamide-induced rats. A high concentration of Dual-Oligo tended to produce more acetic acid than that observed for the control group, and Dual-Oligo affected the production of total SCFA. Bifidobacteria concentration of cecal contents in the high-concentration oligosaccharide (HDO) and low-concentration oligosaccharide (LDO) groups was similar to the result of the normal group. CONCLUSIONS: These results showed that Dual-Oligo is a functional material that is derived from a natural food product and is effective in ameliorating constipation.

  • Milk oligosaccharides are the complex mixture of six monosaccharides namely, D-glucose, D-galactose, N-acetyl-glucosamine, N-acetyl-galactosamine, L-fucose, and N-acetyl-neuraminic acid. The mixture is categorized as neutral and acidic classes. Previously, 25 oligosaccharides in bovine milk and 115 oligosaccharides in human milk have been characterized. Because human intestine lacks the enzyme to hydrolyze the oligosaccharide structures, these substances can reach the colon without degradation and are known to have many health beneficial functions. It has been shown that this fraction of carbohydrate can increase the bifidobacterial population in the intestine and colon, resulting in a significant reduction of pathogenic bacteria. The role of milk oligosaccharides as a barrier against pathogens binding to the cell surface has recently been demonstrated. Milk oligosaccharides have the potential to produce immuno-modulation effects. It is also well known that oligosaccharides in milk have a significant influence on intestinal mineral absorption and in the formation of the brain and central nervous system. Due to its structural resemblance, bovine milk is considered to be the most potential source of oligosaccharides to produce the same effect of oligosaccharides present in human milk. This review describes the characteristics and potential health benefits of milk oligosaccharides as well as the prospects of oligosaccharides in bovine milk for use in functional foods.

A Study on the Dextran Formation and Inner Structure of Jeung-Pyun during the Fermentation by the Added Chitosan-Oligosaccharide (키토산 올리고당 첨가 증편 발효 중 Dextran 형성과 증편의 내부구조에 관한 연구)

  • This study was carried out in order to investigate the dextran formation and the internal structure of the chitosan-oligisaccharide(COS) Jeung-Pyun during the fermentation. To develop a functional Jeung-Pyun, COS was added to the rice at the level of 0, 2, 4, and 6% to the 2-hour-soaked rice. The pH, dextran and reducing sugar contents of COS batter, specific volume and the internal structure of COS were analyzed as a function of fermentation time. The specific volume of COS peaked at the 3 hour fermentation. The pH values of COS batters reduced with the fermentation time. The dextran content of the control batters peaked at the 7 hour fermentation. In contrast, the dextran contents of COS batters reduced up to 7th hour of fermentation, and then increased slowly until 20th hour of the fermentation process. From the air pore size and distribution of COS observed by SEM, the COS fermented fur 3 hour was judged as the best. It was concluded that COS prevented the fermation of dextran at the early stage of fermentation process.

Antimicrobial Activity of Yacon K-23 and Manufacture of Functional Yacon Jam (야콘 K-23의 항균성 및 기능성 야콘잼의 제조)

  • Yacon (Polymnia sonchifolia Poepping & Endlicher), which contains fructo-oligosaccharide, is low calorie functional food. Yacon is efficacious against cholesterol, obesity, and diabetes. Yacon K-23 has antibacterial activity against Gram-positive (Listeria monocytogenes KCTC 3710 and Staphylococcus aureus KCTC 1927) and negative (Salmonella typhimurium KCTC 2514, Escherichia coli KCCM 11591, Proteus vulgaris KCTS 2512, and Pseudomonas aeruginosa KCCM 11803), Yacons A (water extract) and B (crushed juice) showed clear zone of 10mm, while Cassiae Semen, Acanthopanacis Cortex, Angelica gigas showed 7-10mm. Yacon jam showed good sensory attributes, suggesting it could be utilized as food ingredient. Hardness, adhesiveness, and strength of strawberry jam with or without aloe vera were higher than those of yacon jam with or without aloe vera.
