Role of certain nutritional supplements and biological regulators in the epilepsy

  • Certain dietary contents, biological supplements might influence the occurrence or treatment of epilepsy. Some studies have found that the supplementation with individual nutrients reduced seizure frequency or improved other aspects of health in patients with epilepsy. Potentially beneficial dietary interventions include treating blood glucose dysregulations. Identifying and avoiding allergenic foods, and avoiding suspected triggering agents such as alcohol, aspartame, and monosodium glutamate. The Atkins diet (very low in carbohydrates) is a less restrictive type diet that may be effective in some cases. Nutrients that may reduce seizure frequency include vitamin B6, magnesium, vitamin E, manganese, taurine, dimethylglycine, and omega-3 fatty acids. Administration of thiamine may improve cognitive function in patients with epilepsy. Supplementation with folic acid, vitamin B6, biotin, vitamin D, and L-carnitine may be needed to prevent or treat deficiencies resulting from the use of anticonvulsant drugs. Vitamin K1 has been recommended near the end of pregnancy for women taking anticonvulsants. Melatonin may reduce seizure frequency in some cases, and progesterone may be useful for women with cyclic exacerbations of seizures. In most cases, nutritional therapy is not a substitute for anticonvulsant medications. However, in selected cases, depending on the effectiveness of the interventions, dosage reductions or discontinuation of medications may be possible. However, nutrient supplementation may be necessary to prevent or reverse the effects of certain deficiencies that frequently result from the use of antiepileptic drugs.

An analysis of the Gongjindan's ingredients and its efficacy on anti-oxidation (공진단(拱辰丹)의 성분 분석 및 갱산화(坑酸化) 작용에 미치는 영향)

  • Objectives : For the congenital feeble, the Gongjindan is useful medicine. The experiment is to estimate the value of the Gongjindan as therapeutic agent preventing against aging with an analysis of the ingredients and the bio-activating effects by enzymologic methods. Methods : General ingredients' of the Gongjindan's extract were analyzed first and the quantitative analysis of a reducing sugar, a soluble protein, an amino acid and minerals was made. The Gongjindan, which is extracted, concentrated, and freeze drying with water, ethanol and chloroform, have got applied for the experiment. The effects on electronic donating ability, SOD-like activity, nitric oxide inhibition, xanthine oxidase inhibition, whitening effect have been investigated in the physiological activity measurement of function experiment. Results : The contained amino acid, in order of high amount, is Arginine, Alanine, Glutamic acid, Proline and the contained free amino acid is Glutamic acid, Leucine, Lysine, Phenylalanine. The derivative of free amino acid is ${\gamma}-Aminoisobutyric$ acid, Phosphoserine, Taurine. And the Gongjindan is containing 13 species of minerals in order of Ca>K>Na>Mg>Fe>AI>Mn. Then, to assure of the Gongjindan's capability of anti-oxidation, these following subjects-polyphenol, electronic donating ability, SOD-like activity, nitric oxide inhibition, xanthine oxidase inhibition, tyrosinase inhibition- are analyzed and show high activity especially the most in ethanol extracts. Conclusion : With this analysis of ingredients, the Gongjindan is containing many materials functioning as anti-oxidation, anti-aging, anti-fatigue, neurotransmitter and immune agent. Moreover, In every water, ethanol, chloroform extracts, the Gongjindan's capability of anti-oxidation is confirmed so that we can apply to patients' treatment clinically.

Nutritional and Digestive Properties of Israeli Carp Cyprinus carpio as Affected by Size, Part and Commercial Value (향어(Cyprinus carpio)의 크기별, 부위별 및 상품성별 영양 및 소화 특성 비교)

  • This study was conducted to compare sensory and nutritional and digestive properties of the Israeli carp Cyprinus carpio muscle (ICM) by size [normal large (NLF) and small (NSF) fish], part [ventral (VM) and dorsal (DM) muscle], and commercial value [NLF, recessive (RF) or deformed (DF) fish]. In NLF, the major amino acids were lysine, aspartic acid, and glutamic acid, whereas aspartic acid and glutamic acid were predominant in NSF, DM, VM, DF, and RF. Taurine content increased with fish size, and was higher in DM than in VM, and higher in NLF than in DF but lower than that in RF. Regardless of ICM size, part, and commercial value, potassium and iron were expected in the health function, but calcium and zinc were not expected. The major fatty acids present in ICM were 16:0, 18:1n-9, and 18:2n-6, regardless of size, part, and commercial value. The digestion rate of ICM by digestive organ was lower in NLF than in NSF, and lower in VM with a high lipid content than in DM, and lower in DF than in RF.

Effect of Hepatoprotective Agents and Bile Acids on TNF-${\alpha}$ Production in Macrophage Cell Lines (간 보호제 및 담즙산류들이 마크로파지 세포주에서 TNF-${\alpha}$ 분비에 미치는 효과)

  • The effect of hepatoprotective agents and bile acids on tumor necrosis factor-alpha, (TNF-${\alpha}$) production in murine and human macrophage cell line (RAW264.7 and U937) was inve stigated. The hepatoprotective agents including silymarin and its major component, silybin, significantly inhibited TNF-alpha production in a concentration dependent manner ($IC_50$ of silybin=67.7${\mu}g$/ml (140.3${\mu}g$M)). In differentiated U937 cells, especially, silybin showed more effective inbitory activity ($IC_50$=35.1${\mu}g$g/ml (72.7${\mu}g$M)). These results suggest that silymarin and silybin may inhibit TNF-alpha production in the process of hepatic diseases in human. However, biphenyldimethyl dicarboxylate (DDB) was not effective. In the case of bile acids, chenodeoxycholic acid (CDCA) showed a concentration dependent inhibitory effect on TNF-alpha production ($IC_50$ of CDCA= 71.5${\mu}g$g/ml (182.1${\mu}g$M)). In contrast, glycine or taurine conjugated form (G-CDCA or T-CDCA) restored to the control level or significantly increased TNF-${\alpha}$ production. And also ursodeoxycholic acid (UDCA) and its conjugated forms (G-UDCA and T-UDCA) showed a variety of patterns on TNF-${\alpha}$ production by changes of functional groups and concentration. These results also indicate that bile acids may regulate TNF-${\alpha}$ production in normal hepatic function or disease conditions.

The Effect of Aquapuncture with Hepatonics on the Recovery in Artificially Induced Hepatic Damaged Dogs (간기능강화제의 수침이 간손상 유발견의 회복에 미치는 영향)

  • The present experiment was performed in order to know the treatment effect of aquapuncture with hepatonics on recovery in artificially induced hepatic damaged dogs by carbon tetrachloride. The animals were divided into a control and two experimental groups (aquapuncture with taurine into Gan-su acupoint: Aa-I group and aquapuncture with taurine into blank acupoint in the thigh: Aa-II group). The changes of serum enzyme activities (ALT, AST and ALP), serum total protein contents, protein fractions and pathohistological findings of the liver were examined after application of aquapunctiuf treauent The results obtained through this experiment were summarized as follows : The serum ALT activities tendeded to decrease in experimental group compared with those of control group. Significances were detected at 5th (p<0.05) and 7th (p<0.05) day in Aa-I group and 7th day (p<0.05) in Aa-II group, respectively. Low value was fecund in Aa-I group compared with Aa-II groups but significance was not observed between two experimental groups. The serum AST activities in experimental group showed decreasing tendency compared with those of control group. Significances were observed at 2nd (p<0.05) and 5th (p<0.05) day in Aa-I group and 2nd (p<0.05) day in Aa-II groups respectively. Aa-I group showed lower values than those of Aa-II groups however, no significance was detected between experimental groups. The serum ALP activities of experimental group showed a slight decrease compared with those of control group, however, significance was not detected among all groups. The serum total protein content in experimental group showed tendency of increase compared with control group. Significance was found at 2nd day (p<0.05) in Aa-I group, but there was no significance in Aa-II group. Further significant increase of total protein content was seen at 1st day (p<0.05) in Aa-I group compared with Aa-II group. The change of serum albumin content in experimental group showed tendency of increase compared with control. Significant increases were detected at 1st (p<0.01) and 2nd (p<0.01) day in Aa-I group, respectively. Aa-II group showed increase compared with control groups but significance was not observed. Further significant increase was at 1st day (p <0.05) in Aa-I group compared with Aa-II group. The change of ${\beta} $-globulin in Aa-I group was slightly decreased compared with control group. Aa-II group was similar to controls but significance was not observed among all groups. The change of P-globulin content in Aa-I group showed tendency of increase compared with control and Aa-II group showed the tendency of decrease compared with control. The change of ${\gamma}$-globulin content experimental group showed tendency of increase compared with control, however, significance was not detected among groups. The change of A/G ratio in Aa-I group showed tendency of increase compared with control group and Aa-II group was similar to controls but no significance was found among groups. As for pathohistological observations the grade of hepatocellular vacuolized degeneration and necrosis in Aa-I group was milder than those of control and Aa-II groups and the change of Aa-II group was similar to that of control. Considering above finding collectively, it was thought that aquapuncture of Gan-su acupoint with hepatonics was more effective than aquapuncture of blank acupoint for the recovery of hepatic damage.

Changes of Physicochemical Components and Antioxidant Activity of Garlic During its Processing (흑마늘 가공 중 이화학적 성분 및 항산화 활성의 변화)

  • This study was performed to confirm of biological function of black garlic, it heated for 11 days at $40{\sim}90^{\circ}C$ (first step: heate for 2 days at $90^{\circ}C$, second step: heated for 4 days at $80^{\circ}C$, third step: heated for 4 days at $60^{\circ}C$ and fourth step: heated for 1 day at $40^{\circ}C$). Samples were analyzed physico-chemical characteristics and antioxidant activity. Hunter L, a and b values were decreased during processing, and then inner part Hunter values were highly decreased at the second step. The moisture contents were decreased to 58.48${\pm}$0.41 g/100 g at fourth step. pH was also acidified to pH 4.22${\pm}$0.02, but O.D. value at 420 nm was increased during processing of black garlic. At fourth step, total phenolics and flavonoids contents were increased about 1.9 and 2.6 folds than first step sample. Also, total pyruvate and thiosulfinate contents were increased about 1.6 and 5.8 folds as change of total phenolic and flavonoid contents, respectively. Fructose contents were the highest level among free sugars and its contents increased to 2,454.45${\pm}$4.20 mg/100 g. Contents of sucrose and maltose were decreased during processing of black garlic. The contents of total minerals were the highest at fourth step (1,009.20${\pm}$6.91 mg/100 g) during its processing. Contents of glutamic acid, proline and aspartic acid were detected higher than other composition amino acids. Taurine and ethanolamine were not detected in the first step sample, but they were detected 0.88${\pm}0.60{\sim}1.06{\pm}$0.04 and 0.28${\pm}0.4{\sim}0.5{\pm}$0.09 mg/100 g in next processing step, respectively. DPPH radical scavenging ability of water and ethanol extracts from black garlic was increased during its processing. Abilities of DPPH radical scavenging were the highest in fourth step sample, its abilities were 67.40${\pm}$0.21% in 1,000 ${\mu}g/ml$ of water extracts. Reducing power was also significantly higher in water extract than ethanol extract on the whole.

Compositional Change of Hepatic Bile Acid by Multiple Administration of DWP305, a Combined Preparation Containing Ursodeoxycholic Acid and Silymarin, in Rats (흰쥐에서 Ursodeoxycholic Acid 및 Silymarin을 함유한 의약조서울(DWP305)의 연용투여에 의한 간내 담즙산 조성변화)

  • DWP305, a preparation containing combination of ursodeoxycholic acid(UDCA), silymarin and vitamins ($B_1\;and\;B_2$), is a drug currently being developed for hep atic disorders. In order to evaluate the changes in hepatic function by multiple oral administration(2 and 4 weeks) of DWP305 in rats, several biochemical parameters in blood, bile acid composition, and the accumulation of UDCA and lithocholic acid(LCA),a toxic metabolite formed by enterobacteria, were examined using HPLC. In blood biochemical findings, DWP305 did not affect the normal level and there was no difference in total bile acid composition for UDCA, cholic acid(CA), deoxycholic acid(DCA), chenodeoxycholic acid(CDCA) and LCA compared to the UDCA administered group, although total ratio of UDCA and CA was different from normal group. In case of ratio of taurine and glycine conjugated forms, DWP305(186mg/kg as a UDCA) administered group was also similar to normal group and UDCA administered group, while high dosing of DWP305 was not different in the ratio of UDCA administered group(930mg/kg) but normal group. And the ratio of LCA was in order of UDCA(930mg/kg), DWP305(930mg/kg as a UDCA), UDCA(186mg/kg) and DWP305(186mg/kg as a UDCA) administered group, which was less than 4%. The free form of UDCA as well as most of bile acids was not detected at all in rat liver, indicating that there's no accumulation. These results suggest that multiple dosing of DWP305 in rats may not affect hepatic biotransformation and metabolism of bile acids.
