Antithrombotic Activity of Sunghyangjunggisan

  • As apart of our continuing search for antistroke agents from the herbal medicinal resources, we examined in vitro, ex vivo and in vivo the possibility of Sunghyangjunggisan and its ingradients as a novel antithrombotic agent. In vitro ADP- and collagen-induced rat platelet aggregations were potently inhibited by Arisaematis Rhizoma, Cinnamomi Cortex and Zingiberis Rhizoma in a dose-dependent manner, but not by Sunghyangjunggisan. However, Sunghyangjunggisan significantly inhibited ex vivo rat platelet aggregation. Arisaematis Rhizoma, Atractylodis Rhizoma Alba, and Pinelliae Rhizoma also significantly inhibited ex vivo rat platelet aggregation. Sunghyangjunggisan, Alpiniae Fructus and Zingiberis Rhizoma showed significant protection from death due to pulmonary thrombosis in mice. Therefore, Sunghyangjunggisan can express the antithrombotic action, when it is orally administered.

Purgative Activities of Whangryunhaedoktang and Chunghyuldan

  • The purgative activities of Whangryunhaedoktang (WT), Chunghyuldan (CD) and Rhei Rhizoma were measured to choose herbal formulae available for stroke patients suffering from constipation. When the laxative activity of Rhei Rhizoma and CD (the same dose as Rhei Rhizoma) compared, Rhei Rhizoma was more potent than CD. WT contained Gardeniae Fructus, which has been used as a laxative, as a ingredient. Nevertheless, WT did not show the purgative activity. WT and CD did not stimulate the transportation of small intestine. However, CD weakly stimulated the transportation of large intestine than Rhei Rhizoma. Accordingly, we believe that CD can be used as a useful purgative for stroke patients with constipation.

  • 후박의 주성분인 magnolol과 honokiol을 분리, 치주병인균 (혐기성그람음성균: Porphyromonas gingivalis 381, W50, A741-27, Prevotella intemedia 25611, 9336, G8-9K-3, 통기성그람음성균: Actinobacillus actinomycetemcomitans)에 대한 항균력을 조사하고 교원질 분해 활성, 섬유아세포재생능, 치은상피세포재생능, 치주인대세포재생능, cytokine생산 억제력시험을 하여 치주조직재생능을 관찰하였다. 후박과 대추의 안전성을 확인하기 위하여 일반임상증상, 체중측정, 피부반응을 관찰하였다. 아울러 이들 약물을 주성분으로 한 제제의 품질평가를 위하여 활성성분의 분리법 및 주성분인 magnolol과 honokiol을 지표로하여 HPLC 분석조건을 검토하였다.

Effects of betaine on the glutamate-induced neurotoxicity in primary cultured chicken brain cells

  • The neuroprotective effect of betaine, one of the , components of Lycii Fructus, on glutamate-induced neurotoxicity in primary cultured chicken brain cells were examined. Betaine was found to attenuate glutamate-induced neurotoxicity at the concentration of 5-10 mM in both morphological and chemical aspects. The pretreament of chicken brain cells with 5-10 mM betaine for 2 hr at the 12th day of culture before the 40 min-exposure to 500${\mu}$M glutamate significantly increased the survival rate of nerve cells in chicken brain. Betaine could also raise the decreased LDH-level due to the neurotoxicity induced with 100${\mu}$M glutamate in chicken braill cells. LDH value was decreased to 63% of control level in chicken brain cells at the time of 48 hr after the exposure to glutamate. However, the pretreament of chicken brain cells with 5 mM betaine for 2 hr before the exposure to glutamate could prevent the decrease of LDH-level in brain cells showing 90% of control level. Nevertheless, tile remarkable neuroprotective effect of betaine on the glutamate-inducer in neurotoxicity in cultured chicken brain cells could not be observe when betaine was simultaneously administered with glutamate.

Antitumor constituents from some Korean medicinal plants

  • For the screening of bioactive natural products, the benzene or methanol extracts from 93 medicinal plants of Korea were prepared, and tested for the cytotoxicity against L1210 cells and for the antitumor action (Bae et al., 1992 and 1996). Of 93 extracts tested, 6 samples showed a cytotoxicity in both benzene and methanol extract, 39 samples in benzene and 13 samples in methanol extract. The benzene extract of the root of Scutellaria indica L., Sophora fIavescens Solander ex Aiton, Carpesium abrotanoides L., Gymnaster koraiensis (Nakai) Kitamura, Pyrola japonica Klenze, and Forsythiae Fructus showed a potent cytotoxic activity. This observation led to isolate active cytotoxic components, some of which demonstrated some antitumor action. In addition, the structure-activity relationship was discussed.

  • The total 31 prescriptions of Gok Dal(穀疸), recorded in all the medical books, were investigated. The results are as follows 1. The most used prescription of Gok Dal(穀疸) was INJINHOTANG(茵蔯蒿湯). The next ones are GOKDALHUAN(穀疸丸), SOONJUNGTANG(小溫中湯). 2. The total 79 kinds of medicines were prescribed for Gok Dal(穀疸). Fructus Gardeniae(梔子) was the most popular medicine and Herba Artemisiae Scopariae(茵蔯), Radix et Rhizoma(大黃), Pericar pium Citri NobiLis Viride(靑皮), Rhizoma Atractyloclis(蒼朮) follows. 3. Chung Youl(淸熱) medicines was the most popularly medicines, Sam Sub Li Su(渗濕利水) medicines. Pa Juk Chu(破積聚) medicine So Do(消導) medicine follows. 4. Compared to other jaundices, Gok Dal(穀疸) has been cured by the case of Pa Juk Chu(破積聚) medicines, So Do(消導) medicines.

A Study on Conductant drugs in a prescription (방제구성(方劑構成)에 있어서 사약(使藥)으로 이용되는 약물(藥物)에 관한 연구(硏究))

  • I studied conductant drug(使藥) in a prescription. As results of these studies, conductant drug(使藥) with meaning of medicinal guide(引經) were Achyranthis Bidentatae Radix(牛膝), Platycodi Radix(桔梗), Cinnabaris(朱砂), Sal Maritimus(食鹽), alcoholic drink(酒,) Cimicifugae Rhizoma(升麻) and Bupleuri Radix(柴胡). Conductant drugs with meaning of harmonizing medicines in a prescription were almost Glycyrrhizae Radix(甘草). And sometimes Mel(蜂蜜) was used with meaning of harmonizing medicines in pill. And Jujubae Fructus was used with meaning of harmonizing medicines in purgation medicines.

  • Angelicae gigantis radix is the root of the perennial plant, which belongs to the family Umbelliferae. However, this herbal drug is represented quite different chemical components according to its different genus name, though other herbal drugs (i.e. Leonuri Herba, Xanthii Fructus and so on) show similar constituents on the same name. The root of Angelica gigas containing the coumarin compounds is commonly used in Korea, while Angelica sinensis and Angelica acutiloba including phthalide compounds are used in China and Japan, respectively as Angelicae gigantis radix. In this paper, a nearinfrared spectroscopic method was developed to determine genus name of Angelica spp., especially A. gigas and A. sinensis which are commonly misused in herbal markets. X-ray fluorescence spectrometry and electronic nose have been also applied as nondestructive methods to discriminate A. gigas from A. sinensis according to their specific properties.

Study on the antimicrobial activities of herbal extracts against Helicobacter pylori (Helicobacter pylori에 대한 항균활성을 가진 천연물의 검색과 항균력 비교 연구)

  • Infection with Helicobacter (H.) pylori is strongly associated with duodenal and gastric ulcers. Substantial epidemiological data has revealed that high rates of H. pylori infection might be related to high rates of gastric cancer. In this study, a medicinal herbal extracts were examined and screened for anti-H. pylori activity. Seventy percent ethanol was used for herbal extraction. For anti-H. pylori activity study, the inhibitory zone tests with 74 herbal compounds were conducted. As the results, thirteen compounds including Cinnamomi Cortex, Magnoliae Cortex and Meliae Fructus were revealed strong anti-H. pylori activities.

Antimicrobial Effects of Naringenin alone and in Combination with Related Flavonoids (나린게닌의 항균력에 미치는 관련 후라보노이드의 병용효과)

  • As part of our search for less toxic antimicrobial substances from natural resources, naringin was isolated from feels of Citri fructus and then hydrolyzed to naringenin. The antimicrobial activity of naringenin was examined by measuring the minimal inhibitory concentration(MIC) against fourteen species of bacteria. The antimicrobial activity of narngenin in combination with rutin or hesperetin was evaluated by checkerboard method. Among fourteen species tested, the antimicrobial activity of naringenin was the most prominant against Staphylococcus aureus and Shigella boydii showing MIC of $100\;{\mu}g/ml$ for both species. Combinations of naringenin with rutin or hesperetin showed synergism against several species of bacteria, but no antagonism was observed.
