Studies on a New Alimentotherapy for Diabetic Patients (당뇨환자를 위한 새로운 식이요법제에 관한 연구)

  • Mulberry, Morifolium leaves and fruits, Lycii fructus, and Panax ginseng have been known to maintain normal blood sugar levels in folk medicine. Here we investigated the effects of coated rice (Sodangmi$^{TM}$) with these herb extracts to diabetes mellitus patients. Alloxan-induced diabetic mice (80 mg/kg bw, i.v.) separated into 4 groups. 2 groups received the mixed extracts of these plants (Sodans Extract$^{TM}$) orally everyday for 4 weeks. Positive control received Alloxan except Sodans Extract$^{TM}$. Negative control received neither Alloxan nor Sodang Extract$^{TM}$. Blood sugar levels, volume of water consumed, body weights of each group were measured and compared. Blood sugar levels in the Alloxan and Sodang Extract$^{TM}$-received groups got close to normal levels on 4th week after starting the experiments. Volume of water consumed also showed similar pattern. Blood sugar levels in positive control remained high through the experimental periods. Body weights in positive control remained low, but those in the extracts-received groups recovered to normal levels. The clinical trials for both of healthy volunteers and diabetes mellitus patients also showed Sodangmi$^{TM}$ could control blood sugar level. The blood sugar level decreased since 3rd week after starting to receive Sodangmi$^{TM}$, and the difference between the blood sugar level before and after the meal was decreased significantly by receiving Sodangmi$^{TM}$. From these results, we suggest that the functional rice with herb extracts could be used as a new alimentotherapy for diabetes mellitus patients.llitus patients.

Interleukin-5 Inhibition Assay of the Oriental Materia Medica Treatment by Sandwich ELISA on Mouse Splenocytes (ELISA를 이용한 IL-5 분비조절 한약물 Screening)

  • Background and Objective : Allergy is defined as an altered reactivity to an antigen, which can result in pathologic reactions upon subsequent exposure to that particular antigen. This study was to evaluate the effect of oriental materia medica on IL-5 secretion in the mouse spleen cell. Methods : We used the splenocytes of mouse 8 weeks after its birth, and then cultivated those into the 2 experimental groups and a control group for 48 hours. The culture media of the experimental groups were made of $1{\mu}g/ml$, $10{\mu}g/ml$ oriental materia medica, representative. And the culture media of control group was given no oriental materia medica. Then, we assayed the quantity of cytokine-expression by the sandwich ELISA. The quantities of cytokine-expression of the experimental groups were compared with that of the control group, which was standardized. These methods were used for all of the oriental materia medica treated. Results : Some oriental materia medica inhibit the secretion of IL-5 in both $1{\mu}g/ml$ and $10{\mu}g/ml$ culture media. These were Acori Rhizoma, Luffae Fasciculus Vascularis, Amomi Rotundi Fructus, Schisandrae Fructus, Biotae Semen, Clematis armandii, Dioscoreae Sativa Rhizoma, Coicis Semen, Sophorae Flos, Oroxyli Semen, Aurantii Semen, Pini Nodi Lignum. Conclusion : This study indicates that some oriental materia medica inhibit the secretion of IL-5 and are beneficial for allergic disease.

Anti-inflammatory Effect of LFR on LPS-stimulated THP-1 Cells

  • Background and Objective: Luffae Fructus Retinervus (LFR) is used for investigating symptoms of inflammation. We have evaluated the anti-inflammatory effect of LFR by analyzing the expression of pro-inflammatory cytokines. Materials and Methods : We differentiated THP-l cells into macrophage-like cells by treatment with PMA. Inflammation was induced by treatment with LPS and PMA. We determined the safe concentration of LFR by using the MTS and MTT assays and using PD 98059 as a negative control for comparison of the anti-inflammatory effect of LFR. Results : The MTS and MTT analysis showed that the cell survival rate was >80% within the LFR concentration range of 10-100 ng/ml and began to decrease to >80% at 1 ${\mu}g/ml$. By RT-PCR analysis, the gene expression of TNF-${\alpha}$, IL-8, TGF-${\beta}$, IL-6, IL-${\beta}$1, and IL-10 levels were down-regulated when monocyte-derived macrophages were treated with concentrations of LFR between 10 ng/mL and 100 ng/mL. Conclusion : We conclude that LFR exerts an anti-inflammatory effect by inhibiting the expression of pro-inflammatory activity. The results suggest a promising way to treat general inflammatory diseases.

Effects of Guaruhaebaekbanha-tang Extract on Beating Rate and LDH Activity in Cultured Rat Myocardial Cells (과루해백반하탕 추출물이 배양 심근세포의 박동수와 LDH 활성도에 미치는 영향)

  • To certify the protective effect of herbal medicine against oxygen free radical-induced myocardiotoxicity, cytotoxicity was measured using MTT, LDH activity and Beating rate assay in the presence of Guaruhaebaekbanha-tang(GHBT) extracts or single constituents of this prescription. Myocardial toxicity was evaluated in neonatal rat myocardiocytes in cultures. In the present study, xanthine oxidase/hypoxanthine(XO/HX) resulted in a decrease in cell viability, increases in LDH activity in culture medium and decreases in beating rate in cultured myocardial cells. In the effect of GHBT extract, it showed the prevention from the XO/HX-induced cardiotoxicity by the increases of cell viability and beating rate as well as the decrease of LDH activity. In the protective effect of Fructus Trichosanthis(FT), Bulbus Allii Macrostemi(BAM) and Rhizoma Pinelliae(RP), all the extracts were significantly effective in the protection of XO/HX-induced cardiotoxocity in cultured myocardial cells by the increase of beating rate as well as th decrease of LDH activity. From these results, they show that XO/HX is cardiotoxic in cultured myocardial cells derived from neonatal rat, and it suggests that GHBT, FT, SAM, RP extracts are positively effective in the blocking in XO/HX-induced cardiotoxicity.

Experimental Study of CheonghunHwadam-tang on the Cerebral Blood Flow in Rats (청훈화담탕이 국소뇌혈류량에 미치는 실험적 연구)

  • CheonghunHwadam-tang(CHT) have been used in oriental medicine for many centuries as a therapeutic agent of vertigo by wind, fire and phlegm. CheonghunHwadam-tangGamypang(CHTG) was CHT adding Aurantii Fructus(AF), Gastrodae Rhizoma(GR). The effects of CHT on the cerebral blood flow and cardiacvascular system is not known. The purpose of this Study was to investigate effects of CHT and CHTG on the regional cerebral blood flow(rCBF), mean arterial blood pressure(BP), cardiac muscle contractile force(CMF), heart rate(HR). The changes of cerebral blood flow and movement of cardiacvascular system(BP, CMF, HR) was determinated by Laser-Doppler Rowmetry(LDF). The results were as follows; 1. CHT extract increased rCBF, but decreased BP, HR in a dose-dependent manner. 2. AF extract accelerated rCBF and movement of cardiacvascular system in a dose-dependent manner. 3. GR extract increased significantly rCBF(10.0mg/kg, p<0.05) as well as accelerated BP and rCBF in a dose-dependent manner. 4. CHTG extract increased significantly rCBF(10.0mg/kg, p<0.01) in a dose-dependent, but was not changed movement of cardiacvascular system. This results suggest that CHTG is significantly treated more than CHT in vertigo by wind, fire and phlegm.

Hepatoprotective Activities of Gomisin A and Gomisin N (Gomisin A 및 Gomisin N의 간독성 보호작용)

  • The aim of this study was to investigate the protective activity of gomisin A and gomisin N, bioactive lignan components isolated from Schizandae Fructus, on hepatocyte injury induced by carbon tetrachloride($CCl_4$, 10 mM), t-butyl hydroperoxide(TBH, 0.5 mM), and D-galactosamine(GalN, 30 mM). Primary cultures of rat hepatocyte(18 h culture) were treated with $CCl_4$, TBH or GalN and various concentrations(0.1, 1, 10, $100{\mu}M$) of gomisin A or gomisin N. $CCl_4$ significantly increased the levels of lactate dehydrogenase(LDH), alanine aminotransferase(ALT), and aspartate aminotransferase(AST). These increases were inhibited by gomisin N. TBH significantly increased the level of AST; an increase that was inhibited by gomisin N. GalN markedly increased the levels of LDH and ALT, and these increases was significantly inhibited by both gomisin A and gomisin N. These results suggest that gomisin A and gomisin N have the hepatoprotective activity.

Partial Characterization of Bacillus thuringiensis var. kurstaki Temperature-sensitive Mutants (Bacillus thringiensis var. kurstaki 감온성 돌연변이주의 일부특성)

  • Partial characterization of B. thuringiensis var. kurstaki 3ab temperature-sensitive mutants was carried out through biochemical analyses, utilization tests of carbohydrate sources, antobiotic resistant test, hemolytic reaction test, growth measurement of Fructus gardenia sxtrant medium and toxicity test against mice. Six ts mutants, ts-U154, ts-U601, ts-U602, ts-U603, tsU-604, and ts-U788 could not produce urease, ts-U603 lost its motility, ts-U154 could not use salicin and cellobiose and ts-U603 not ribose. All ts mutants except ts-U154 and wild type strain were resistant to cephalothin, ampicillin, and penicillin. but ts-U154 was sensitive to the three. Four mutants, ts-U21, ts-U74, ts0U131 and ts-U154 did not form pigment colonies on the F. gardenia medium. All the mutants and wild type strain showed hemolysis reaction on the blood agar. The B. thuringiensis and mutants were not toxic to mice.

Inhibitory Effect of Coumarins on Nitric Oxide Production in LPS-Activated Murine Macrophages (쿠마린에 의한 RAW 264.7 세포주의 Nitric Oxide 생성 저해활성)

  • During the screening for inhibitors on nitric oxide (NO) production in LPS-activated murine macrophage, RAW 264.7 cells, two coumarins were isolated from chloroform extract of Ponciri Fructus. They were identified as imperatorin (10), phellopterin (11) on the basis of spectroscopic methods. The $IC_{50}$ values for NO formatiom were about $5.4\;{\um}M$ and $35.0\;{\um}M$, respectively, and then eleven coumarins were tested for the inhibitory effects on NO production in activated macrophages. All the test coumarins inhibited NO production in concentration-dependent manner and furanocoumarins (6-11) showed much more potent inhibitory effect than simple coumarins. Among the compounds examined xanthotoxin (8) was the most potent inhibitors of NO production $(IC_{50}=1.4\;{\mu}M)$. Analysis of the structure-activity relationship among these coumarins led to the conclusion that the substitution of C-5 position in furanocoumarins reduces greatly their inhibitory potency although the substitution of C-8 does not almost affect it.

Effects of the Extract of Samjasan on the Nitric Oxide Synthase Activity and the Level of Lipid Peroxide in Penis of Rats (삼자산(三子散)이 흰쥐 음경조직의 Nitric Oxide Synthase 활성 및 과산화지질 함량에 미치는 영향)

  • Objectives : The following are the results of the experimental studies of Samjasan (SJS) extract on the nitric oxide synthase (NOS) activity and the level of lipid peroxide in the penises of rats. Methods : Cnidii Fructus, Cuscutae Semen, Schizandrae Fructus constitute SJS. 150 g of crushed crude drug was extracted with methyl alcohol, under reflux, for 24 hours, three times; the total extractive was evaporated under reduced pressure to give 28.6 g. Results : In vitro, the SJS extract didn't effect the activity of NOS. However, the SJS extract decreased the activities, the ratio of type conversion of xanthine oxidase, the levels of lipid peroxide, In vitro, after administration of the SJS extract to rats, the activities and ratio of type conversion of xanthine oxidase decreased, but the activity of NOS and the content of nitrite increased. Also, the levels of the superoxide anion radical and lipid peroxide decreased in the penises of rats. But, after administration of the SJS extract to rats, the levels of glutathione did not increase. The effects of the SJS extract did better as the dosage and the length of treatment increased. Conclusions : These results suggest that the SJS extract decreases the activities of free radical generating enzymes which form lipid peroxide and increases the NOS activity in the penises of rats. Therefore, the SJS extract is capable of improving of sexual ability in rats.

Cytoprotective action of Rubi Fructus by modulation of Reactive Oxygen Species, peroxynitrite and $Ca^{2+}$ (복분자(覆盆子)의 세포내 ROS, $ONOO^-$ 생성 및 $Ca^{2+}$ 증가 억제에 의한 혈관내피세포 보호작용)

  • Objectives : Poststroke depression is a frequent and specific entity that impaires the rehabilliation and functional recovery of patients with hemiplegia. The author evaluated the effect of Banhahubak-tang(Banxiahoupotang) in patients with poststroke depression. Methods : 38 patients suffering from poststroke depression(determined by Diagnostic and Statistical Manual of Mental Disorders, revised. 3rd edition. and Beck Depression Inventory[BDI] cutoff $point{\geqq}10$) in Kyunghee Oriental hospital were randomized into two groups; treatment group(n=19) and control group(n=19). The treatment group was prescribed with Banhahubak-tang(Banxiahoupotang) three times a day fur a week. Control troop was prescribed with other herbal medicines used for stroke Patients three times a day for a week. Patients were evaluated by use of BDI scale, Modified Barthel Index, Depression of Ki score, Yin syndrome score, and Yang syndrome score. Among 38 patients, 24 patients got BDI scores above 21, which is the cut-off score for depression in Korean. The same procedures and assessments described above were applied. Results : Treatment group did not significantly improve compared with control group. Results yielded only slight significance (P=0.086). Especially. patients with poststroke depression as yin syndrome improved more significantly on BDI than those classified as yang syndrome. When BDI cutoff point for depression was defined as being ${\geq}\;21$, treatment group did not significantly improve compared with control group(P=0.114). However, patients with poststroke depression classified as yin syndrome were also significantly improved on BDI than those classified as yang syndrome. Conclusions : This study suggests that Banhahubak-tang(Banxiahoupotang) is significantly effective in patients with poststroke depression classified as yin syndrome.
