Antioxidative and Anti-inflammatory Effects of Aurantii Fructus Immaturus Pharmacopuncture (지각 약침액의 항산화 및 항염증 효과에 관한 연구)

  • Objectives : Ulcerative colitis is a chronic relapsing inflammatory disease in the gastrointestinal tract. We investigated whether Aurantii fructus immaturus (AFI) pharmacopuncture has antioxidative and anti-inflammatory effects. Methods : in vitro experiments, 1,1-diphenyl-2-picryl hydrazyl (DPPH) free radical scavenging activity, superoxide dismutase (SOD) activity, prevention on $H_2O_2$-induced cell death in RAW264.7 cell line, DNA fragmentation, and cyclooxygenase-2 mRNA expression induced by lipopolysaccharide (LPS), were analyzed to investigate antioxidative and anti-inflammatory effect of AFI pharmacopuncture. in vivo experiment, a murine model of dextran sulfate sodium (DSS)-induced colitis was used to examine the effect of AFI pharmacopuncture on CV12 at different doses of 5 ${\mu}l$, 0.5 ${\mu}l$, 0.05 ${\mu}l$ for 10 days. Body weight, colon length and macroscopic features were investigated. Results : AFI pharmacopuncture showed DPPH free radical scavenging and SOD active effects in a dose-dependent manner. AFI pharmacopuncture showed a protective effect against $H_2O_2$-induced cell injury and also attenuated LPS-induced COX-2 mRNA expression. In a DSS- induced colitis murine model, however, AFI pharmacopuncture at CV12 had no anti-inflammatory effects. Conclusions : The present results suggest that AFI pharmacopuncture extract may have anti- inflammatory and antioxidative effects in vivo test, but further research on the underlying mechanism is required.

Screening and Extraction Condition of Anti-skin Aging Elastase Inhibitor from Medicinal Plants (각종 약용 식물로부터 피부노화 억제 관련 elastase 저해물질의 탐색 및 추출조건)

  • To develop a new anti-skin aging cosmetics or functional foods by using elastase inhibitor, a potent elastase inhibitor was screened from various extracts of medicinal plants and its optimal extraction condition was investigated. Methanol extracts of Rubi fructus showed the highest elastase inhibitory activity of 85%. The elastase inhibitor of Rubi fructus was maximally extracted when it was treated with 80% methanol at $50^{\circ}C$ for 12hr and its elastase inhibitory activity $(IC_{50})$ was 0.52 mg.

Water Extracted Evodiae Fructus Possesses Immunomodulatory Activities on Cyclophosphamide Induced Immunesuppression (오수유 열수추출액이 Cyclophosphamide 유도 면역억제에 미치는 효과)

  • Evodiae Fructus(EF) has been used as Traditional medicine for the treatment of headache, abdominal pain, hemorrhage, and menorrhae in many Asian countries. The present study was conducted to investigate the immunomodulatory effect on cyclophosphamide(CY)-induced immunesuppression of water extracted EF(EFE). In the mouse spleen cell proliferation assay, EFE enhanced mitogenic activity and restored the CY-induced cell suppression. In the nitric oxide(NO) assay, EFE inhibited NO production and iNOS protein levels in LPS-stimulated RAW 264.7 cells. In the GC-MS analysis, many ingredients of EFE were detected by solvents. These results indicated that EFE can modulate immune response through immune cell proliferation, the regulation of NO production and the inhibition of CY-induced immunotoxicity.

Antiallergic and Anti-inflammatory Effects of the Viticis Fructus (만형자의 항알레르기 및 항염증효과)

  • This study was carried out to the determine the effects of the water extract of Viticis Fructus (Verbenaceae, WEVF) on experimental allergic reactions and inflammation. WEVF was anally administered to mice for high and fast absorption. WEVF inhibited compound 48/80-induced systemic anaphylaxis and immunoglobulin E (IgE)-mediated local allergic reaction. Histamine releasing from mast cells was reduced by WEVF, which was mediated by modulation of intracellular calcium. In addition, WEVF decreased the gene expression of pro-inflammatory cytokines in phorbol 12-myristate 13-acetate plus calcium ionophore A23187 (PMACI)-stimulated HMC-1 cells. These findings demonstrate that the WEVF possesses antiallergic and anti-inflammatory activities, which may be mediated by reducing the release of mediators such as histamine from mast cells and weakening the inflammatory action of these mediators.

The Ameliorating Effect of Hoveniae Semen Seu Fructus Extract against Ethanol-induced Psychomotor Alterations in Rats (흰쥐에서 지구자 추출물의 에탄올 유도 숙취 행동 개선 효능)

  • Ethanol consumption causes psychomotor alterations. Hovenia Semen seu Fructus (HS), widely distributed in Korea, China, and Japan, has been reported to have beneficial effects on acute alcohol-induced liver injury. The present study sought to assess the effects of HS extract on ethanol-induced psychomotor alterations in rats. Sprague-Dawley rats were orally (p.o.) given ethanol (4 g/kg) (ethanol group) to induce psychomotor alterations. A separate group (HS-treated groups), were treated with different dosages of HS (50, 100, and 200 mg/kg, p.o.), 30 minutes before ethanol treatment. The control group received only the vehicle (saline). Ethanol-induced psychomotor alterations were evaluated in the open-field, rota-rod, hanging wire, and cold swimming test. In addition, blood ethanol and acetaldehyde concentrations were also measured. Behavioral evaluations and blood analysis were carried out 0.5, 1, 2, 4, and 8 hours after ethanol administration. Pre-treatment of HS ameliorated ethanol-induced alterations in the open-field, rota-rod, and cold swimming test, significantly evident in 2 and 4 hours after ethanol treatment. These improvements coincided with decrease in blood ethanol and acetaldehyde concentration. Based on these results, the present study suggests that HS may have ameliorating effects against ethanol-induced psychomotor alterations.

A Study on The Natural Dyeing of Crataegi fructus Extracts (산사 추출물을 이용한 천연염색 연구)

  • In this study, the dyeing properties and functionalities of Crataegi fructus extract were investigated for the purpose of application to new natural dye resources. The effects of dyeing conditions(concentration of dye, dye bath temperature, dyeing duration and dye bath pH) and mordanting on dye uptake and color changes were also examined. The study also estimated the colorfastness, antibacterial properties, and ultraviolet protection properties of the dyed and mordanted silk fabrics. The dye uptake increased in tandem with the dye concentration and dye bath temperature. The highest K/S values were obtained at the following conditions: a dye concentration of 100%(v/v), a dyeing period of 60 minutes, a dye bath temperature of $90^{\circ}C$, and a dyeing pH of 3. The colorfastness of the material when dry cleaned or rubbed ranged from good to excellent and fastness to light was rated at grade 2-3. The change in the color grade when exposed to washing was not good; however, the stain of washing fastness was good at a range of 4-5. In regards to the functional property aspects, the dyed and mordanted fabrics exhibited excellent results with a 99.9% reduction rate, and excellent ultraviolet protection factors.

The Ameliorative Effect of Rubi Fructus on DSS-induced Colitis in Mice

  • Ulcerative colitis (UC) is an inflammatory bowel disease and a chronic gastrointestinal disorder. Rubi Fructus (RF), the fruit of Rubus coreanus Miquel, is known to exert several pharmacological effects including anti-oxidative, anti-obesity and anti-inflammatory properties. However, the improving effect and mechanism of RF on intestinal inflammation is not been fully understood. The purpose of this study was to investigate the regulatory effect of RF on dextran sulfate sodium (DSS)-induced colitis in mice. We evaluated the effects of RF on DSS-induced clinical signs by analyzing weight loss and colon length. The inhibitory effects of RF on inflammatory mediators such as prostaglandin E2 (PGE2), cyclooxygenase (COX)-2, as well as the activation of nuclear factor-κB (NF-κB), were determined in colitis tissue. Our data indicated that mice treated with DSS showed clinical symptoms of colitis, including weight loss, colon length decrease and diarrhea. However, we observed that RF treatment significantly improved these clinical symptoms of weight loss, colon length decrease and diarrhea induced by DSS. RF inhibited the enhanced levels of COX-2 and PGE2 caused by DSS. We also showed that the anti-inflammatory mechanism of RF by suppressing the activation of NF-kB in DSS-treated colon tissues. Collectively, the findings of this study indicate the prospect of developing new drugs from RF for UC treatment.

Ameliorative Potential of Rengyolone Against CCI-induced Neuropathic Pain in Rats

  • The sciatic nerve is the largest nerve among the peripheral nerves, and the damage to the sciatic nerve is caused by mechanical and physical pressure. This is an important disease that consumes a lot of time and money in the treatment process. Among them, research on relieving nerve pain caused by damage to the peripheral sciatic nerve has been made efforts to prevent and treat this disease through various methods such as drugs, natural products, electrical stimulation, exercise therapy, and massage. Existing treatments are not very effective in neurological pain, and countermeasures are needed. Forsythia Fructus, used in this study, has been used as a therapeutic agent for infectious diseases and a pain reliever for cancer from the past, and in past studies, it has been known to properly control the inflammatory response. In this study, rengyolone, a physiologically active substance of Forsythiae Fructus, was administered to rats that caused chronic left nerve pain to verify the pain relief effect. As a result of the experiment, it was found that mechanical pain and cold stimulation pain were significantly reduced in the rengyolone-treated group compared to the non-administered group. In addition, it was found that nerve growth factor (NGF) mRNA expression was significantly reduced and Cyclin-dependent kinase 2 (Cdc2) expression was increased in the rengyolone administration group. This increase in NGF expression is thought to be related to rengyolone's anti-inflammatory regulatory mechanism. It is expected that the reduced NGF was directly involved in pain relief.

The Study about Relationship between Treatment of Ulcerative Colitis and Intestinal Bacteria According to Characteristic of Herbal Medicine (한약재 특성에 따른 궤양성 대장염 치료과 장내세균 상관관계 연구)

  • Many modern people are exposed to chronic inflammatory diseases, such as inflammatory bowel disease (IBD), atopic dermatitis and immune disorder. Among those chronic diseases, the incidence ratio of IBD has been increased. IBD, including Crohn's disease and ulcerative colitis (UC), is known to cause abnormal inflammation in intestinal tissue. UC is accompanied by abdominal pain, bloody stool and diarrhea. Many therapeutic agents, such as sulfasalazine, corticosteroids, immunosuppressive agents, have been used for treating UC. However, those agents have side-effects and temporary effects on UC. The aim of this study was to investigate the effect of herbal medicine on UC and relationship between UC and intestinal bacteria according characteristic of herbal medicine. To determine the effect of herbal medicine on UC, various herbal medicine were chosen within oriental medicine category such as cheongyeol and onyeol medicine. In this study, we found carthami fructus, included in cheongyeol medicine, had stronger effect than onyeol medicine. Also, we determined influence of carthami fructus against lactic acid bacteria. Catthami fructus and lingon berry extracts affected the composition of mice intestinal bacteria in mice fecal. The symptoms of UC could be regulate by using herbal medicine, according to characteristic of herbal medicine. Also, herbal medicine might be change body condition to healthy by controlling intestinal bacteria composition. Herbal medicine characteristic could be a therapeutic agent by revealing relationship between intestinal bacteria and UC.

Antitumor Activity of Corni Fructus Ethanol Extract in Sarcoma-180 Cancer Cells (산수유 에탄올 추출물의 Sarcoma-180 세포에 대한 항암 효과)

  • To develop Corni Fructus as a cancer preventive food material, the in vitro cytotoxicities and in vivo antitumor activities of various concentrations of 80% Corni Fructus ethanol extract (CFEE) were investigated using sarcoma-180 cancer cell. Viability was decreased and cell death rate was increased in both dose- and time-dependent manners in cells treated with CFEE at 10, 100, 300, and $500\;{\mu}g/mL$ concentrations for 24, 48, and 72 hr. Proliferation was also inhibited more than 60% in cells treated with CFEE at the $100\;{\mu}g/mL$ concentration for 48 hr. In addition, the morphology of cells treated with CFEE at the 100 and $500\;{\mu}g/mL$ concentrations was distorted with shrunken cell masses and lower cell numbers compared to the control cells. In the cells treated with CFEE, the formation of apoptotic bodies and nuclear condensation were observed in dose dependent manners. CFEE also increased DNA fragmentation values at the 100 and $500\;{\mu}g/mL$ concentrations. The apoptosis induced by CFEE was connected to the proteolytic activation of caspase-3. When CFEE was administered at 100 and 300 mg/kg, ip, for 7 consecutive days in mice inoculated with sarcoma-180 cancer cell, the life span of the mice was found to be longer than that of the control mice that did not receive the extract. These results suggest that Corni Fructus may be used as a potential cancer preventive food material.
