Objectives : Obesity is an important cause of diabetes, and lipotoxicity causes insulin resistance. In this study, we investigated the effects of Salvia miltiorrhiza on high fat diet-induced obese type 2 diabetic mouse models. Methods : Diabetes was induced in ICR male mouse (23~25 g) with Surwit's high fat, high sucrose diet. Mice were divided into 4 groups (n=10) of normal, control, Salvia miltiorrhiza, and metformin. After 8 weeks, body weight, OGTT, fructosamine, lipid profile, serum level of adiponectin and leptin, epididymal fat pad, liver weight and epididymal adipocyte size were measured. Results : Salvia miltiorrhiza significantly reduced oral glucose tolerance levels, fructosamine serum level, epididymal fat weight, and epididymal adipocyte size. Salvia miltiorrhiza also increased HDL-cholesterol, adiponectin and leptin serum levels. Conclusions : These results show that Salvia miltiorrhiza improves insulin resistance. Therefore we suggest that Salvia miltiorrhiza would be an effective treatment for obese type 2 diabetic patients.
This study investigated the hypoglycemic effects of mulberry leaf (M), green tea (G), and a mixture of mulberry leaf and green tea (MG) in rats with streptozotocin-induced diabetes. Male Sprague-Dawley rats were divided into the non-diabetic (N) and diabetic groups. The experimental animals were divided into four groups and fed the following for four weeks: a normal diet (N), the STZ+normal diet (STZ), the STZ+5% mulberry leaf (STZ-M), STZ+5% green tea (STZ-G), and the STZ+5% mixture of mulberry leaf and green tea (STZ-MG). The whole blood glucose level of the STZ-M, STZ-G and STZ-MG groups significantly decreased compared to the STZ group two weeks later. The serum glucose and fructosamine, and urine glucose levels in the STZ-M, STZ-G and STZ-MG groups were lower than in the STZ group. The levels of serum insulin were higher in the STZ-M, STZ-G and STZ-MG groups than in the STZ group. However, serum glucose, fructosamine and insulin levels, and urine glucose levels were not significantly different among the STZ-M, STZ-G and STZ-MG groups. These results indicate that mulberry leaf, green tea, and their mixture help prevent or attenuate the progression of diabetes in rats with STZ-induced diabetes.
Objectives: This study was designed to investigate the anti-obesity, anti-diabetic and anti-inflammatory effects of Platycodi radix on obese type 2 diabetes mouse model. Methods: Obese type 2 diabetes mouse model was induced by Surwit's high fat, high sucrose diet for 8 weeks. Models were divided into 4 groups of normal diet (ND, n=10), high fat and high sucrose diet (HFD, n=10), high fat and high sucrose diet with Platycodi radix (PR, n=10), and high fat and high sucrose diet with Metformin (Met, n=10). Body weights were measured every week. After 7 weeks fasting, blood sugar and oral glucose tolerance tests were conducted. After 8 weeks blood samples were taken from mouse hearts and analyzed biochemically. Lipid profile, fructosamine, leptin and weight of epididymal fat pad and liver were measured. Adipose tissue macrophage percentage was analyzed by fluorescence-activated cell sorting (FACS). Results: Compared with the HFD group, body weight, glucose level, fructosamine, weight of epididymal fat pad and adipose tissue macrophage percentage decreased in the PR group. Conclusions: These results suggest that Platycodi Radix has anti-obesity, anti-diabetic, and anti-inflammatory effects on obese type 2 diabetes mouse model.
Objectives: This study was undertaken to evaluate anti-glycemic and anti-lipid effects of Supungsunkihwan-gagambang, which is composed of Cornus officinalis, Dioscorea Batatas Decaisne, Auantii fructus and Platicodon grandiflorum. Methods: Diabetes was induced in ICR male mice ($24{\pm}1g$) with Surwit's high fat and high sucrose diet. Mice were divided into 3 groups (n=10) of normal, control and Supungsunkihwan-gagambang. The Supungsunkihwan-gagambang group was given 5% herbal medicine in their diet. The animals were fed on each experimental diet for 8 weeks. Body weight was assessed every week. At the 7th week, the fasting blood sugar (FBS) and oral glucose tolerance test (OGTT) were conducted in all experimental groups. After 8 weeks, fructosamine, lipid profile, epididymal fat weight, liver weight and white adipose tissue (WAT) size were measured. Results: Supungsunkihwan-gagambang significantly reduced FBS, OGTT and fructosamine. It also increased high density lipoprotein (HDL) cholesterol and significantly reduced triglyceride/HDL cholesterol ratio, total cholesterol/HDL cholesterol ratio and WAT size. Conclusions: These results show that Supungsunkihwan-gagambang improves anti-glycemic and anti-lipid effect in high fat diet-induced obese mice. Therefore we suggest that Supungsunkihwan-gagambang could be an effective treatment for patients with type 2 diabetes.
We have studied the effects of buckwheat diet on serum glucose and lipid metabolism in 9 NIDDM volunteers during 2 weeks. The subjects were given dietary counseling in their own homes at 2-3 day intervals throughout experimental periods and the dietary intake were determined by interview and record methods. The intake of calorie, carbohydrate, protein and fat during the buckwheat diet period were not significantly different compared with control diet and body weight was maintained within 1-2kg. The mean total glycohemoglobin, fructosamine, total cholesterol and LDL-cholesterol levels at the end of buckwheat diet were significantly lower than the end of control diet (P<0.05). Fasting serum glucose, insulin and HDL-chloesterol levels were slightly decreased. The mean triglyceride level was increased but it was not significant. These results indicate that buckwheat diet is an effective therapeutic regimen for the control of metabolic derangements in diabetes mellitus.
Two spaniel dogs were presented with a history of lethargy, weakness, refractory hypoglycemia, and episodic seizure. A pancreatic mass on the distal end of the left limb was identified through screening tests including dualphase CT angiography in each patient. Canine insulinoma was suspected with low blood glucose, low fructosamine, and normal to high serum insulin levels. Exploratory laparotomies were performed, and partial pancreatectomy using an ultrasonic-activated scalpel was achieved without enzyme leakage or fistula formation after surgery. The lesions were definitively diagnosed as insulinoma via histopathologic examination. One dog has been doing well until POD 870 with medication; in the other dog, diffuse hepatic nodules noted during surgery were confirmed as a liver metastasis. This dog died at POD 45. This case report addresses the utility of the ultrasonic scalpel in partial pancreatectomy for the treatment of canine insulinoma and differences in prognosis according to disease stage.

The effects of pan-fired (PM) and fermented (FM) Cudrania tricupidata tea leaves on $\alpha$-glucosidase inhibitory activity, oral glucose tolerance, blood glucose levels and serum lipids profiles in streptozotocin (STZ)-induced hyperglycemic rats were investigated. The $\alpha$-glucosidase inhibitory activity of FM ethanol extracts (20 mg/mL) was higher (92.5%) than that of raw dried leaves (RM) (69.1%) and PM (54.6%). In addition, the results of a glucose tolerance test revealed that the glucose levels of hyperglycemic rats that were fed PM and FM ethanol extracts and then orally administered glucose began to decrease after 60 minutes, but recovered after 120 minutes. However, the blood glucose levels in the hyperglycemic control group did not begin to decrease for 360 minutes. Additionally, the results of animal experiments that were conducted over five weeks to compare the dietary effects of PM and FM following hyperglycemic induction to the effects on the hyperglycemic control group (DM) were as follows: The body weight gain and FER of the treated rats were $12.9{\sim}16.9%$ higher than those of the DM group, whereas the amounts of feed and water intake by the treated rats were $6.8{\sim}10.1%$ lower. Additionally, the levels of blood glucose and serum fructosamine decreased by $27.3{\sim}39.8%$ and $6.7{\sim}20.0%$, respectively, in the treated rats. Moreover, the serum triglyceride, total cholesterol and LDL-cholesterol concentrations in the treated rats were $24.9{\sim}27.1%$, $15.9{\sim}17.4%$ and $33.8{\sim}38.4%$ lower, respectively. Finally, the HDL-cholesterol contents were $20.5{\sim}24.8%$ higher in the treated rats than in the control group. The above results suggest that PM and FM exerts an anti-hyperglycemic effect that occurs due to the inhibition of $\alpha$-glucosidase activity as well as via prevention and/or inhibition of changes in the serum lipid profile. In addition, the results of this study revealed that the synthetic anti-hyperglycemic effect of FM was greater than that of PM. However, further detailed studies are needed to confirm these results.
This study analyzed the distribution of the blood glucose level according to the fasting status. Moreover, a relationship was analyzed between fasting blood glucose level and glycemic control indicators. A total of 707 outpatients, who visited Dankook University Hospital, were included and classified into either the fasting group and the non-fasting group. The mean blood glucose level of each group was calculated and analyzed by sex, age, and clinic. In addition, blood glucose, HbA1c, fructosamine, and 1,5-AG were measured in 153 fasting health check-up patients, and the correlation between the blood glucose level and glycemic control indicators was evaluated. Blood glucose averages between the two groups (non-fasting 111.9 vs. fasting 103.6 mg/dL) were different (p<0.05); and the mean difference was lower in women (4.8 mg/dL) than in men (12.2 mg/dL). A significant difference of the median glucose values among the age groups was only observed in the non-fasting group (Kruskal-Wallis test, p<0.01), and not in the fasting group. A 1,5-Anhydroglucitol was estimated to be significantly correlated with the fast blood glucose level in the range of the criteria of impaired fasting glucose (IFG). We presented an assessment of the distribution of blood glucose level in accordance with the fasting status among outpatients, and estimated that 1,5-anhydroglucitol was well correlated with the fasting blood glucose than fructosamine and HbA1c, through the analysis of results of health screening subjects. It is suggested that the use of glycemic indicators that reflect short-term blood glucose control can be used together with the blood glucose measurement in the screening of diabetes mellitus.
Purpose: Diabetic complications are a major concern to manage progression of diabetes. Production of advanced glycation end products (AGEs) due to high blood glucose is one of the mechanisms leading to diabetic complications. Multiple pharmacologic AGE inhibitory agents are currently under development, but clinical applications are still limited due to safety issues. Thus, it is necessary to identify a safe anti-glycation agent. It is known that burdock roots have antioxidant, anti-inflammatory, and anti-cancer activities. The objective of the present study was to investigate the inhibitory role of burdock roots on the formation of high glucose-induced glycation of bovine serum albumin (BSA). Methods: In this study, glycation of BSA by glucose, galactose, or fructose at $37^{\circ}C$ for 3 weeks was assessed based on levels of ${\alpha}$-dicarbonyl compounds (early-stage glycation products), fructosamine (intermediate products of glycation), and fluorescent AGEs (late-stage glycation products). In order to compare the inhibitory actions of burdock root extract in AGE formation, aminoguanidine (AG), a pharmacological AGE inhibitor, was used as a positive control. Results: BSA glycation by glucose, fructose, and galatose was dose- and time-dependently produced. Burdock root extract at a concentration of 4 mg/mL almost completely inhibited glucose-induced BSA glycation. The results demonstrate that burdock root extract inhibited AGE formation with an $IC_{50}$ value of 1.534 mg/mL, and inhibitory activity was found to be more effective than the standard anti-glycation agent aminoguanidine. This study identified a novel function of burdock root as a potential anti-glycation agent. Conclusion: Our findings suggest that burdock root could be beneficial for preventing diabetic complications.
A Spitz (5-years old, castrated male) and a Maltese (9-years old, castrated male) were presented with weight loss, polyuria/polydipsia (PU/PD) and depression. Diabetic ketosis was diagnosed based on clinical signs, physical examinations and screening tests (CBC, serum chemistry and urinalysis). The dogs were treated with NPH initially. However, NPH was inadequate to control blood glucose level due to the short duration of the action (< 5 hours). Because of the poor glycemic control with NPH, the dogs showed diabetic complications including weight loss and cataract. After change to glargine, the duration of insulin action was extended up to 11 hours. As a result, there was significant improvement in clinical signs and serum fructosamine concentrations. This study suggests that glargine is useful as a long-acting insulin in dogs which have poor glycemic control due to the short duration of NPH.
