Dry bridges have been widely applied in the Qinghai-Tibet Railway (QTR) to minimize the thermal disturbance of engineering to the permafrost. However, because the Qinghai-Tibet Plateau is an area with a high potential occurrence of earthquakes, seismic action can easily destroy the dry bridges. Therefore, a three-dimensional numerical model, with consideration of the soil-pile interactions, is established to investigate the thermal characteristics and their impact on the seismic response of the dry bridge in permafrost region along the QTR. The numerical results indicate that there exist significant differences in the lateral displacement, shear force, and bending moment of the piles in different thermal conditions under seismic action. When the active layer become from unfrozen to frozen state, the maximum displacement of the bridge pile reduces, and the locations of the zero and peak values of the shear force and bending moment also change. It is found that although the higher stiffness of frozen soil confines the lateral displacement of the pile, compared with unfrozen soil, it has an adverse effect on the earthquake energy dissipation capacity.
한대지역은 계절 변화에 따라 동결융해가 반복되는 상부의 활동층과 하부의 영하상태로 항시 유지되는 영구동토층으로 구성되어 있다. 한대지역에서 여름철에는 동결되어 있는 지반을 융해시키기 때문에 지반 강도저하 및 침하가 발생한다. 이러한 강도저하 및 침하는 상부 구조물의 구조적 안정성에 문제를 야기시켜 활동층을 포함하고 있는 한대지역에서는 지반의 온도를 항시 영하상태로 유지할 수 있는 동결지반 안정화 공법이 필요하다. 열사이펀이란 구조체 내부에 충전된 냉매의 자가적인 열순환에 의해 지반의 온도를 항시 영하상태로 유지할 수 있는 지반 안정화 공법 이다. 본 연구에서는 열사이펀의 지반 동결성능을 분석하기 위해 R-134a 냉매를 이용하여 충전율에 따른 열사이펀의 지반 온도제어를 실내실험과 수치해석을 통해 분석하고 열사이펀의 열전도율을 산정하였다.

There were three considerations in deciding optimal runner size in this study: minimum pressure at the gate that makes resin fully filled in the cavity, minimum runner size that compensates shrinkage of resin in the cavity, and frozen layer thickness formed in the runner during injection. Through the computer simulations the optimal runner size that satisfies those three considerations has been decided. Although the computational results among the softwares were slightly different, it was enough to predict the optimal runner size. The previous runner diameter was 8 mm and predicted optimal size was 5 mm. This was verified by injection molding experiment. Thus, the way of CAE application in deciding optimal runner size adapted in this study would be appropriated.

It was studied the phenomena of transient freezing of an inclined water-saturated enclosure. One side of the test section was cooled and the other sides were insulated. The effects of the initial temperature, the inclination angle on the temperature field and the shape of the ice-water interface were observed. In the beginning of freezing, with increasing the inclination angle the freezing rate was increased and in the stable density layer centered $4^{\circ}C$, the freezing was fast as the convective fluid flow became small. When the initial temperature was above the $4^{\circ}C$, the frozen thickness in the upper part of inclined surface was thinner than that in the lower part, but with time the frozen thickness of upper part was thicker than that of lower part, below the $4^{\circ}C$, the frozen thickness in the upper part was thicker than that of lower part from the begining, and above the $8^{\circ}C$ in the beginning upper part was thinner with concave, but with time thicker the upper part, vanishing concave.

Permafrost is frozen soil below $0^{\circ}C$ for two or more years. Surface of permafrost is called as active layer that seasonally thaws during the summer. Although the thawing of permafrost may deepen the active layer and consequently increase the microbial activity, the microbial community structure in this habitat has not yet been well described. In this study, we presented bacterial and archaeal diversity in the active layer soil from Resolute, Canada using pyrosequencing analysis. The soil sample was collected from the surface of the marsh covered with moss and Carex. A total of 7,796 bacterial reads for 40 phyla and 245 archaeal reads for 4 phyla were collected, reflecting the high diversity of bacteria. Predominant bacterial groups were Proteobacteria (37.7%) and Bacteroidetes (30.0%) in this study. Major groups in Archaea were Euryarchaeota (51.4%) and Thaumarchaeota (46.1%). Both methane producing archaea and consuming bacteria were detected in this study. Although it might be difficult to characterize microbial community with only one sample, it could be used for the basis of assessing the relative importance of the specific groups with a high resolution on the bacterial and archaeal community in this habitat.
Objective: This study was to establish the human embryonic stem (ES) cells derived from frozen-thawed blastocyst stage embryo that were destined to be discarded after five years in routine human IVF-ET program. Methods: Frozen-thawed and survived human blastocysts were treated by immunosurgery, and recovered ICM cells were cultured onto STO feeder cell layer and ICM colony was subcultured by mechanical dissociation into clumps. To identify ES cell, alkaline phosphatase staining and expression of Oct4 in replated ICM colonies were examined. Also, to examine the possibility of ES cell differentiation, retinoic acid (RA), basic fibroblast growth factor (b-FGF), nerve growth factor (NGF) were added in culture medium. In addition, to classify the specific cell type, differentiated cells were stained by indirect immunocytochemistry. Results: One ICM colony recovered from frozen-thawed six blastocysts was subcultured, continuously replated during 40 passage culture duration without differentiation. Subcultured colonies were strong positively stained by alkaline phophatase. When the expression of Oct4 in cultured ES colony was examined, Oct4b type is more clearly indicated than Oct4a one although there was not detected in embryoid body or differentiated cells. In differentiated cardiomyocytes from ES colony, cells were beaten regularly (60 times/min). In differentiated neural cells from ES colony, neurofilament (NF) 200 kDa protein, microtubule associated protein (MAP) 2 and ${\beta}$-tubulin of specific marker in neurons, glial fibrillary acidic protein (GFAP) of specific marker in astrocytes and galactocelebrocide (GalC) of specific marker in oligodendrocytes were confirmed by indirect immunocytochemistry. Also, muscle cells were detected by indirect immunocytochemistry. In addition, ES colonies can be successfully cryopreserved. Conclusion: This study suggested that establishment of human ES cells can be successfully derived from frozen-thawed blastocysts that were destined to be discarded, and obtained specific cell types (cardiomyocytes, neurons and muscle cells) through the in vitro differentiation procedures of ES cells.
The drying rate in freeze drying was obtained by experiment of garlic moisture contents depending on the drying time. Freeze drying experiment of garlic juice was carried out in vaccum freeze drier of laboratory scale by backface heating, and a mathematical model is also used to simulate the process of simultaneous heat and mass transfer in freeze drying to compare with experimental data.
