  • ASR(Aquifer Storage Recharge) 또는 ASTR(Aquifer Storage Transfer Recharge)과 같은 직접적인 지하수 인공함양기법은 대수층을 활용하여 수자원을 공급하고 관리하기 위한 적극적인 방안으로 고려될 수 있다. 이 중 ASTR 기법은 대규모 충적층이 발달한 강변 또는 하구에서 저류지 수생식물의 정화작용과 층적층의 물리/화학/생물학적 여과 기능을 활용하여 양질의 상수원수를 확보할 수 있는 기법이며, 수질이 나쁜 하천수를 직접 취수하여 정수처리하는 것에 비해 정수비용이 상대적으로 적게 들어 투자대비 경제적인 상수원수 확보기술이라 할 수 있다. 본 연구에서는 하구에 염수 대수층이 위치해 있다고 가정하였으며, 이러한 염수 대수층 내에 담수 주입 변화에 따른 지하 담수체의 거동을 4가지 시나리오에 따라 모의 및 분석을 통해 조사하였다. 염수 대수층 내에서 8개의 주입정과 1개의 양수정이 설치되어 있다고 가정하였으며, 주입정은 동심원 상에 등간격으로 위치해 있으며, 양수정은 동심원 중에 위치해 있다고 가정하였다. 본 연구에서 구성된 시나리오로 첫 번째는 주입정 8개 모두에서 동시에 주입되며, 1개의 양수정을 통해 양수되는 것이며, 두 번째는 7개의 주입정에 주입 그리고 1개의 주입정 폐쇄, 세 번째는 6개 주입정에 주입 및 양수정과 서로 마주보는 2개 주입정 폐쇄, 그리고 마지막으로 6개 주입정에 주입 및 서로 이웃한 2개 주입정 폐쇄이다.

The Spawning Behaviour of the Endangered Freshwater Fish Koreocobitis naktongensis (Cypriniformes: Cobitidae) under Artificial Conditions (멸종위기어류 얼룩새코미꾸리 Koreocobitis naktongensis (Cobitidae)의 수조 내 산란행동)

  • The Nakdong nose loach, Koreocobitis naktongensis is an endangered freshwater fish endemic to Korea. The spawning behaviour of the K. naktongensis was observed after treating Ovaprim in laboratory aquarium from 20~27 April 2010. The behavioral patterns were categorized into three stages of pre-spawning, spawning and post-spawning behaviors. Before spawning K. naktongensis usually repeated taking a rest and swimming. On average, initial spawning activity started 11 to 12 hours later after injection with Ovaprim. During the spawning period, the number of spawning acts ranged from 1 to 18. The spawning ratio of female to male was 1 : 1. The male embraced the back of the female's dorsal fin with his lamina circularis. It looked like a complete circle at that time. After spawning, parental care was not observed. Individuals not participating in spawning have been observed to feed on some of the fertilized eggs.

Fresh Water Injection Test in a Fractured Bedrock Aquifer for the Mitigation of Seawater Intrusion (해수침투 저감을 위한 균열암반 대수층 내 담수주입시험)

  • Fresh water injection test in a fractured bedrock aquifer was applied as an efficient approach to lower saline concentrations in the saltwater-freshwater transition zone formed by seawater intrusion in a coastal area. The methodology and effectiveness of fresh water injection for hydraulically controlling seawater intrusion is overwhelmingly site dependent, and there is an urgent need to characterize the permeable fractures or unconsolidated porous formations which can allow for seawater flow and transport. Considering aquifer characteristics, injection and monitoring boreholes were optimally designed and completed to inject fresh water through sand layer and fractured bedrock, respectively. We devised and used the injection system using double packer for easy field operation and maintenance. Overall fracture distribution was systematically identified from borehole image logs, and the section of fresh water injection was decided from injection test and monitoring. With fresh water injection, the fluid electrical conductivity of the monitoring well started to be lowered by the inflow of fresh water at the specific depth. And this inflow leaded to the replacement of the fluid in the upper parts of the borehole with fresh water. Furthermore, the injection effect lasted more than several months, which means that fresh water injection may contribute to the mitigation of seawater intrusion in a coastal area.

Ecotoxicological Effects of NaDCC injection method in Ballast Water Management system on Marine Environments (NaDCC 주입 선박평형수 처리기술의 해양생태위해성에 대한 연구)

  • Effluent treated by an NaDCC injection method in Ballast water management system (BWMS) contains reactive chlorine species and disinfection by-products (DBPs). In this study, we conducted whole effluent toxicity (WET) testing and ecological risk assessment (ERA) to investigate its ecotoxicological effects on marine environment. WET testing was carried out for four marine pelagic and freshwater organisms, i.e., diatom Skeletonema costatum, Navicula pellicuosa, chlorophyta Dunaliella tertiolecta, Pseudokirchneriella subcapitata, rotifer Brachionus plicatilis, Brachionus calyciflorus and fish Cyprinodon variegatus, Pimephales promelas. The biological toxicity test revealed that algae was the only biota that showed apparent toxicity to the effluent; it showed no observed effect concentration (NOEC), lowest observable effect concentration (LOEC) and effect concentration of 50% (EC50) values of 25-50%, 50-100% and >100%, respectively, at three water condition, but did not show any significant toxicities on other biota. Meanwhile, chemical analysis revealed that the BWMS effluent contained total residual oxidants (TROs) below $0.03{\mu}g/L$ and a total of 25 DBPs such as bromate, volatile halogenated organic compounds (VOCs), halogenated acetonitriles (HANs), halogenated acetic acids (HAAs), chloropicrin and Isocyanuric acid. Based on ERA, the 25 DBPs were not considered to have persistency, bioaccumulation and toxicity (PBT) properties. The ratio of predicted environmental concentration (PEC) to predicted no effect concentration (PNEC) of the other DBPs did not exceed 1 for General harbor environment. However, four substances (Isocyanuric acid, Tribromomethane, Chloropicrin and Monochloroacetic acid) were exceed 1 for Nearship environment. But observed toxicity in the test water on algal growth inhibition would be mitigated by normal dilution factor of 5 applied for nearship exposure. Thus, our results of WET testing and ERA showed that the BWMS effluent treated by NaDCC injection method would have no adverse impacts on marine environment.

Ecology and Early Life History of Endangered Freshwater Fish, Pseudobagrus brevicorpus (Pisces: Bagridae) (꼬치동자개(Pseudobagrus brevicorpus)의 생태와 초기 생활사)

  • Some of ecological factors and the early life history of endangered freshwater fish, Pseusobagrus brevicorpus, were investigated to proliferate artificially and to restore the population by release seedlings. The density of population in restricted area by small scale dam was 1.25 ind. $m^{-2}$, and the low capacity for reproduction having 200$\sim$250 eggs ind$^{-1}$. were thought to be a reason of this endangered situation. From the result of induction of natural spawning in aquarium after hormone injection it was considered that the spawning substrate would be aquatic plants with slender branches. The hatching was observed after 50 hrs after fertilization and the larvae showed aggressive behavior. Yolk was almost absorbed 3 days after hatching, and the larvae shaped tad-pole grew up to fry after 30 days. We discussed on the strategies for conservation of this species according to those results from investigation on ecology and early life history.

Control of Freshwater Algae with Hydrogen Peroxide (과산화수소를 이용한 담수 조류의 제어)

  • The purpose of this study is to propose a method of controlling freshwater algae which grows abundantly and forms water bloom in the eutrophic water body with $H_2O_2$. Both laboratory and field methodologies were used. For the laboratory test $H_2O_2$ was injected into the different growth phases of incubated Microcystis aeruginosa and the resulting algae growth control rate was examined. For the field test, $H_2O_2$ was dispersed into a lake. Lake water quality was evaluated using a pre-test and post-test analysis of chlorophyll-a, luminance, transmittance, etc., which allowed a comparative evaluation of water quality change. From the experimental results, the growth of algae can be controlled with the small amount of 1mg/L of $H_2O_2$ at the lag phase of growth. The field test results show that the green colour of lake water was removed completely by the reduction of chlorophyll-a and improved transmittance, luminance, TKN, TP, TOC and SS. These indicators of water quality were improved significantly after $H_2O_2$ injection. Toxicity test results using the lake fish show no evidence of detrimental effect of $H_2O_2$ up to 15mg/L. The results of $EC_{50}$ with P. phosphoreum show that the toxicity of $H_2O_2$ was negligible compared to copper which was commonly used for algae control.

Effects of 3,5,3'-Triiodothyronine (T3) on Osmoregulation following Freshwater Acclimation in Starry Flounder (강도다리 Platichthys stellatus 담수 순화시 삼투압 조절에 미치는 갑상선호르몬의 영향)

  • The aim of this study was to test if 3,5,3'-triiodothyronine (T3) are involved in the osmoregulatory actions in euryhaline starry flounder Platichthys stellatus. We investigated osmoregulatory parameters ($Na^+,\;Cl^-$ and osmolality), blood cortisol and glucose in starry flounder acclimated to seawater (SW, 33 psu) and that were transferred and allowed to acclimate to freshwater (FW, 0 psu). Fish in SW were injected with T3 (5, 10, and $15{\mu}g$/g body weight) or vehicle (0.9% NaCl), and then transferred to FW. They were sampled 3 days after the transfer. With T3 at $10{\mu}g$/g, levels of plasma $Na^+$ and $Cl^-$ were significantly higher than in sham (only saline) and control fish (without hormone and saline). Osmolality was significantly higher after injection with T3 at 10 and $15{\mu}g$/g than in the control. However, T3 at $5{\mu}g$/g had no effect on hyper-osmoregulation. In this study, all dose of T3 induced a significant increases in plasma cortisol without glucose. These results suggest a positive hyper-osmoregulatory role of T3 in starry flounder to hypoosmotic environment, maybe a positive interaction of T3 with cortisol for maintenance of hyper-osmoregulatory ability.

Effects of Rearing Condition during the Winter Period on Artificial Maturation and Reproduction of Cultured Female Eel, Anguilla japonica (겨울철 사육조건이 양식산 뱀장어 Anguilla japonica의 인위적인 성성숙 유도 및 번식에 미치는 영향)

  • In the present study, we investigated the effects of rearing condition during the winter period on artificial maturation and reproduction of cultured female eel, Anguilla japonica. The female eel was adapted by combination of environmental conditions (seawater/low temperature, $10^{\circ}C$; seawater/high temperature, $20^{\circ}C$; freshwater/low temperature, $10^{\circ}C$; freshwater/high temperature, $20^{\circ}C$) during 8 weeks of the winter period. Then, the female eel cultivated by different conditions during the winter were transferred to seawater at $20^{\circ}C$ for induction of artificial maturation by salmon pituitary extraction (SPE) injection. The results shown that gonadosomatic index (GSI) and plasma levels of estradiol-$17{\beta}$ (E2) were significantly higher in female eel were reared in seawater than in freshwater, and were the highest at eel adapted by seawater/low temperature. Also, reproduction coefficient were higher in seawater/low temperature than in the other groups. These results will provide valuable information for elevation of the artificial maturation and reproduction coefficient in the cultured female eel.

Cancer Chemopreventive Effect of Spirogyra Neglecta (Hassall) Kützing on Diethylnitrosamine-Induced Hepatocarcinogenesis in Rats

  • Spirogyra neglecta, a freshwater green alga, is a local food in the northern and northeastern parts of Thailand. This investigation explored the anticarcinogenicity of S neglecta and its possible cancer chemopreventive mechanisms in rats divided into 14 groups. Groups 1 and 10 served as positive and negative control groups, respectively. Groups 1-9 were intraperitoneally injected with diethylnitrosamine (DEN) once a week for 3 weeks. Groups 10-14 received normal saline instead. One week after the last DEN injection, groups 2-5 were administered for 9 consecutive weeks various doses of S neglecta extract (SNE) and dried S neglecta (SND), mixed with basal diet. Groups 6-9 and 11-14 similarly were administered various doses of SNE and SND starting from the first week of the experiment. Administration of SNE and SND was not associated with formation of glutathione-Stransferase placental form (GST-P) positive foci in rat liver. SNE and SND during initiation phase significantly reduced the number of GST-P positive foci in rats injected with DEN. The number of GST-P also diminished in groups treated with SNE and SND after injection with DEN, except for the low dose extract group. SNE showed stronger anticarcinogenic potency than SND. Furthermore, SNE also decreased the number of Ki-67 positive cells. However, the numbers of TUNEL-positive cells in the liver of the SNE-treated groups were not statistically different from the controls. The GST activity in 50 mg/kg bw of SNE and 1% of SND groups was significantly increased as compared to the positive control. In conclusion, Spirogyra neglecta (Hassall) K$\ddot{u}$tzing showed cancer chemopreventive properties at the early stages of diethylnitrosamine-induced hepatocarcinogenesis in rats. Possible inhibitory mechanisms include enhancement of the activities of some detoxifying enzymes and/or suppression of precancerous cells.

Effects of Recombinant Aquaporin 3 and Seawater Acclimation on the Expression of Aquaporin 3 and 8 mRNAs in the Parr and Smolt Stages of Rainbow Trout, Oncorhynchus mykiss

  • This study aimed to examine the role of two aquaporin isoforms (AQP3 and AQP8) in response to the hyperosmotic challenge of transitioning from freshwater (FW) to seawater (SW) during parr and smoltification (smolt) using the rainbow trout, Oncorhynchus mykiss. We examined the changes in the expression of AQPs mRNAs in the gills and intestine of the parr and smolt stages of rainbow trout transferred from FW to SW using quantitative real-time PCR in an osmotically changing environment [FW, SW, and recombinant AQP3 (rAQP3) injection at two dosage rates]. Correspondingly, AQPs were greater during smoltification than during parr stages in the rainbow trout. Plasma osmolality and gill $Na^+/K^+$-ATPase activity increased when the fish were exposed to SW, but these parameters decreased when the fish were exposed to SW following treatment with rAQP3 during the transition to seawater. Our results suggest that AQPs play an important role in water absorbing mechanisms associated with multiple AQP isoforms in a hyperosmotic environment.
