제주지역 수생식물에서 발굴된 Sordariomycetes강 균류 3종의 국내 최초 보고 (New Records of Three Sordariomycetes Fungi Isolated from Aquatic Plants in Jeju, Korea)

  • 본 연구에서는 제주 습지 환경에서 채집한 수생식물에서 내생균류를 분리하였다. 분리된 균을 형태학적 특징과 internal transcribed spacer 영역의 염기서열 분석을 통하여 동정한 결과, NNIBRFG2982, NNIBRFG2984, NNIBRFG3040 균주는 각각 Acremonium tubakii, Colletotrichum cliviae, Cylindrocarpon pauciseptatum으로 확인되었다. 이들 3종의 균류는 Ascomycota문의 Sordariomycetes강에 속하는 균류로 국내 미기록종으로 보고하는 바이다.

  • 본 연구는 국내 담수환경으로부터 야생효모를 분리하여 특성을 분석하였다. 분리된 효모의 26S rDNA의 D1/D2부위 염기서열을 이용하여 동정을 하였다. 결과적으로 C. viswanathii (NNIBRFG39781), C. cygneicollum (NNIBRFG49003), O. silvestris (NNIBRFG39803), V. foliicola (NNIBRFG6120) 등 4종의 국내 미기록 야생효모들을 동정하였다. 이들 미기록 효모는 모두 출아에 의하여 영양생식을 하였고 PD, YM, YV 배지에서 잘 자랐다. C. viswanathii (NNIBRFG39781) 균주는 의균사를 형성하였으며 고온에서도 생육하였다. 또한 C. viswanathii (NNIBRFG39781), O. silvestris (NNIBRFG39803), V. foliicola (NNIBRFG6120) 균주는 20% glucose를 함유한 YPD 배지에서 생육하는 내당성을 보였으며 C. cygneicollum (NNIBRFG49003)와 O. silvestris (NNIBRFG39803) 균주는 NaCl을 함유한 배지에서는 생육하지 못하였다. 담수환경은 효모를 포함한 균류의 다양성이 높은 환경으로 지속적 연구가 필요하다. 본 연구는 향후 담수환경의 생물다양성 연구에 기초자료로 활용될 것이다.

  • Fungi are ubiquitous and indispensable components of nearly all ecosystems on earth, including freshwater environments. A survey of fungal diversity in freshwater environments of Korea led to the identification of four unrecorded Pezizomycotina (Ascomycota) species in 2016 and 2017, based on morphology and molecular phylogeny; these included Cladorrhinum australe, Curvularia muehlenbeckiae, Curvularia pseudobrachyspora (Dothideomycetes), and Diaporthe longicolla (Sordariomycetes).

  • The distribution and occurrence of rare ascomycete fungi within freshwater samples in Korea was investigated. Three rare fungal strains, CNUFC-YR537-1, CNUFC-CNUP1-1, and CNUFC-NDR3-1, were isolated using serial dilution method. On the basis of their morphological characteristics and phylogenetic analysis of their internal transcribed spacer regions and 28S rDNA sequences, the three isolates were identified as Acrostalagmus luteoalbus, Bartalinia robillardoides, and Collariella carteri, respectively. To our knowledge, these are the first records of rare genera Acrostalagmus, Bartalinia, and Collariella from Korea, and the first reports of A. luteoalbus, B. robillardoides, and C. carteri from freshwater samples.

First Reports of Unrecorded Mortierellomycetes and Umbelopsidomycetes Fungi from Freshwater Ecosystems in Korea

  • In this study, we isolated several fungal strains from filtered water and sediment collected from rivers and streams. The strains were identified by molecular phylogenetic analyses of rDNA sequences (internal transcribed spacer [ITS], large subunit of ribosomal DNA [LSU]). The morphological characteristics of the fungi were investigated using microscopy, and the culture characteristics of fungi grown on several media were examined. We identified four species previously unknown in South Korea, namely, Dissophora globulifera, Linnemannia exigua, Mortierella rishikesha and Umbelopsis autotrophica.

Isolation and Characterization of Previously Undescribed Seventeen Fungal Species Belonging to the order Hypocreales in Korea

  • Hypocreales is one of the largest orders within the class Sordariomycetes in Ascomycota. Several species of this order are cosmopolitan and have a broad range of habitats. Here, we isolated several fungal strains from environmental samples, including freshwater sediment and plant litter. The strains were identified via molecular and phylogenetic analyses of rDNA and other DNA markers, such as TUB, RPB2, and EF1. The morphological characteristics of the fungi were investigated using microscopy, and culture characteristics were assessed from their growth on several media. We identified 17 species previously unrecorded in Korea: Dactylonectria hordeicola, Flavocillium bifurcatum, Fusarium luffae, Ilyonectria ilicicola, Ilyonectria qitaiheensis, Ilyonectria robusta, Lecanicillium aphanocladii, Nectria ulmicola, Neonectria lugdunensis, Ovicillium oosporum, Pseudonectria foliicola, Sarocladium spinificis, Scolecofusarium ciliatum, Trichoderma appalachiense, Trichoderma subviride, Trichoderma taiwanense, and Trichoderma tsugarense.

담수생태계로부터 분리된 Filosporella 3종의 국내 최초보고 (First report of three Filosporella species isolated from freshwater ecosystem in Korea)

  • 담수생태계에 서식하는 균류의 다양성을 조사하기 위해 연못과 하천에서 담수퇴적토, 수변식물, 담수침전식물체를 채집하였다. NNIBRFG1552와 NNIBRFG3013은 2016년 제주 남생이못에서 채집한 담수퇴적토와 수변식물에서 각각 분리되었고, NNIBRFG5472는 2018년 충북 보은의 보청천에서 채집한 담수침전식물체에서 분리되었다. 이들 3균주의 형태적 및 분자계통학적 특징을 바탕으로 동정한 결과 NNIBRFG1552, NNIBRFG3013, NNIBRFG5472는 각각Filoporella exilis (100%, KC834046), F. fistucella (99.8%, KC834047), F. cf. annelidica (100%, KC834044)로 확인되었다. 또한, 이들 3개 균주의 배양 및 형태학적 특성이 분자계통학적 분류와 일치되는 것을 확인하였다. Filosporella 속은 국내에서는 보고된 바 없으며 본 보고가 국내 최초이다.

Diversity of Aspergillus, Penicillium, and Talaromyces Species Isolated from Freshwater Environments in Korea

  • In order to elucidate the fungal diversity and community structure in freshwater environments, numerous fungal strains were isolated from freshwater, submerged soils, twigs, dead insects, etc. Among them, the present study has focused specifically on Aspergillus, Penicillium, and Talaromyces species, which produce diverse useful metabolites in general. Twelve strains of Aspergillus isolated were identified as A. japonicus (n = 5), A. tubingensis (3), A. niger (2), and A. flavus (2), 10 strains of which belong to Aspergillus section Nigri, named black Aspergillus. Eight strains of Penicillium were identified as P. brasilianim (n = 3), P. oxalicum (2), P. crustosum (1), P. expansum (1), and P. piscarium (1). Two different strains of Talaromyces were identified as T. pinophilus and T. versatilis. Thus far, Penicillium piscarium and Talaromyces versatilis have been unrecorded in Korea, for which we provide detailed morphological and molecular characteristics.

Seven Yeast Strains Isolated from Freshwaters for the First Record in Korea

  • This study focused on the isolation and characterization of wild yeasts in Korea. The yeasts were identified by phylogenetically analyzing the D1/D2 domains of the 26S rDNA regions. Consequently, we identified seven strains, NNIBRFG856, NNIBRFG3732, NNIBRFG3734, NNIBRFG3738, NNIBRFG3739, NNIBRFG5497, and NNIBRFG6049, which were confirmed to be Kabatiella microsticta, Pichia membranifaciens, Candida vartiovaarae, Candida sake, Debaryomyces hansenii, Candida railenensis, and Schwanniomyces polymorphus, respectively, all of them being new in Korea. Morphological and cultural characteristics of these yeast species were investigated. None of the strains formed ascospores or pseudomycelia. Moreover, these yeasts grew in a pH range of 4-8. NNIBRFG3732, NNIBRFG3738, NNIBRFG3739, NNIBRFG5497, and NNIBRFG6049 were halotolerant or halophilic, and NNIBRFG3732, NNIBRFG3734, and NNIBRFG6049 grew in vitamin-free medium. NNIBRFG3732, NIBRFG3739, and NNIBRFG6049 grew at 35 ℃, but not at 40 ℃.

Characterization of Achlya americana and A. bisexualis (Saprolegniales, Oomycota) Isolated from Freshwater Environments in Korea

  • Many members of the Saprolegniales (Oomycete) cause mycoses and disorders of fishes, of which Achlya and Saprolegnia are most ubiquitous genera worldwide. During a survey of the diversity of freshwater oomycetes in Korea, we collected seven isolates of Achlya, for which morphological and molecular phylogenetic analyses enabled them to identify as Achlya americana and Achlya bisexualis. In Korea, only a species of Achlya, A. prolifera, has been previously found to cause seedling rot on rice (Oryza sativa), but none of the two species have been reported yet. Importantly, A. bisexualis was isolated from a live fish, namely rice fish (Oryzias sinensis), as well as freshwater, and this is the first report of Achlya-causing mycoses on freshwater fishes in Korea. The presence of A. americana and A. bisexualis on live fish in Korea should be closely monitored, as considering the well-known broad infectivity of these species it has the potential to cause an important emerging disease on aquaculture industry.
