A Simple Method for the Preparation of Highly Pure Conjugated Linoleic Acid(CLA) Synthesized from Safflower Seed Oil

  • A simple and rapid method was developed to prepare a large quantity of highly pure conjugated linoleic acid (CLA) chemically-synthesized from safflower seed oil (SSO). CLA-SSO(74.9% in purity) was synthesized from fresh SSO(79.9% of linoleic acid) by alkaline isomerization at 18$0^{\circ}C$. Urea(50g) and CLA-SSO (25g) were completely dissolved in ethanol (750ml) using a water bath(5$0^{\circ}C$) and followed by refluxing for 60 min. The resultant was cooled to room temperature and stored in a cold room (4$^{\circ}C$) for 24hrs. After removing the urea adduct by filtration, the filtrate was rotoevaporated under 4$0^{\circ}C$ and the residue was dissolved in hexane (200ml). The hexane extract was washed with distilled water (100ml$\times$3) and dried over sodium sulfate anhydrous. This urea treatment procedure was repeated three times. The purity of CLA recovered from the hexane extract was 95.0%. This method can be applied to prepare a large quantity of highly pure chemically-synthesized CLA (>0.5kg/a batch) from any plant oils containing high percentages (>70%) of linoleic acid.

건조 대파의 잔류농약 가공 및 감소계수 연구 (On the Processing and Reduction Factors of Several Pesticides with Welsh Onion)

  • 본 실험은 우리나라에서 파에 등록되어 사용중인 농약 중 pyridalyl, kresoxim-methyl, spinosad, flufenoxuron, difenoconazole, metconazole, tebufenozide를 시험농약으로 하여 건조계수와 가공계수를 구하기 위해 파에 대한 2군데 포장시험을 수행하여 $60^{\circ}C$에서 건조한 후 건조 전후 차의 수분함량을 측정하고 잔류농약을 분석하였다. 파의 평균 수분 함량은 건조 전은 89.3%, 건조 후는 10.2%이였으며, 2포장의 평균 가공계수는 pyridalyl은 7.24, kresoxim- methyl은 2.85, spinosad은 7.43, flufenoxuron은 3.17, difenoconazole은 4.38, metconazole은 2.40, tebufenozide은 8.13으로 나타났고, 2포장의 평균 감소계수는 pyridalyl은 0.87, kresoxim-methyl은 0.35, spinosad은 0.88, flufenoxuron은 0.38, difenoconazole은 0.52, metconazole은 0.29, tebufenozide 0.98로 나타나 전반적인 농약잔류량은 건조하는 과정에서 감소하는 경향이었다.

인삼의 재배 및 가공단계 별 Azoxystrobin 잔류성 (Residues of Azoxystrobin during Cultivation and Processing of Ginseng)

  • 본 연구는 인삼 및 인삼가공품에 대한 합리적인 농약잔류허용기준 설정을 위해서 인삼 중 농약의 잔류성을 규명하고 이를 토대로 인삼 가공단계별 농약의 감소계수와 가공계수를 산출하고자 수행되었다. 시험농약은 인삼재배 중 사용등록 되어 있는 azoxystrobin으로 강원도 원주와 경기도 이천의 6년근 인삼포에 농약안전사용기준에 따라 살포한 후 수삼을 수확하였으며, 건삼, 홍삼, 인삼 농축액 및 홍삼 농축액 시료는 KT&G의 협조를 받아 가공하였다. 시험결과 azoxystrobin의 잔류량은 원주, 이천 수삼에서 각각 0.05, $0.03\;mg\;kg^{-1}$, 건삼에서 0.12, $0.14\;mg\;kg^{-1}$, 홍삼에서 모두 $0.05\;mg\;kg^{-1}$, 인삼 알코올 농축액 0.28, $0.33\;mg\;kg^{-1}$, 인삼 물 농축액에서 0.22, $0.16\;mg\;kg^{-1}$, 홍삼 알코올 농축액에서 0.31, $0.20\;mg\;kg^{-1}$, 홍삼 물농축액에서 0.09, $0.11\;mg\;kg^{-1}$으로 모두 MRL수준 이하였다. Azoxystrobin의 가공계수는 건삼 3.25, 홍삼 1.34, 인삼 알코올 농축액 7.84, 인삼 물 농축액 4.63, 홍삼 알코올 농축액 6.15, 홍삼 물 농축액 2.56으로 산출되었으며 가공과정을 거치면서 농축에 의해 농약 잔류량이 증가하는 경향을 보였다. 감소계수는 건삼 1.19, 홍삼 0.51, 인삼 알코올 농축액 3.41, 인삼 물 농축액 1.91, 홍삼 알코올 농축액 2.74, 홍삼 물 농축액 1.00으로 약제가 초기 잔류량과 비슷하거나 다소 감소하는 경향을 보였다.

식품중의 오염물질에 관한 조사연구 (Monitoring Program on Food Contaminants)

  • 본 연구는 1983년부터 시작한 농작물중의 잔류농약 Monitoring program으로서 우리나라의 농작물중의 농약잔류량을 파악하고 이를 기초자료 (back data)로 우리나라 실정에 맞는 농작물중의 잔류농약허용량을 선정하기 위하여 시행되었다. 금년도에는 86년도의 대상농약인 유기인제 (8종), 카바메이트제(3종)에 유기염소제 살균제인 Captan, Captafol을 추가하였으며 대상식품으로 상치, 무우, 감자, 파, 감, 고구마를 예년도의 대상식품이었던 도마도, 풋고추, 사과, 복숭아, 콩, 오이, 배추중에는 Captan, Captafol을 경기, 충남, 경북, 전북의 4개 지역에서 채취하여 G.C.-NPD, ECD를 사용하여 그 잔류량을 조사한 결과 다음과 같다. 1. Diazinon은 32.0%의 검출율을 보였고 검출범위는 ND∼0.0872ppm, 평균은 0.0020ppm이었다. 2. Fenthion(MPP)는 16.4%의 검출율을 보였고 검출범위는 ND∼0.0395ppm, 평균은 0.0007ppm이었다. 상치, 가지, 고구마에서는 검출되지 않았다. 3. Fenitrothion(MEP)는 30.5%의 검출율을 보였고 검출범위는 ND∼0.0628ppm이었고 평균 0.0026ppm이었다. 4. Phenthoate(PAP)는 7.0%의 검출율을 보였고 검출범위는 ND∼0.0069ppm, 평균 0.0001ppm이었다. 감자, 가지, 감, 고구마에서는 검출되지 않았다. 5. EPN은 13.3%의 검출율을 보였고 검출범위는 ND∼0.0250ppm, 평균은 0.0030ppm이었다. 감자에서는 검출되지 않았다. 6. Dimethoate는 27.3%의 검출율을 보였으며 검출범위는 ND∼0.0826ppm, 평균은 0.0049ppm이었다. 7. Parathion은 22.7%의 검출율을 보였으며 검출범위는 ND∼0.0771ppm, 평균 0.0021ppm이었다. 김, 고구마에서는 검출되지 않았다. 8. Malathion은 10.9%의 검출율을 보였으며 검출범위는 ND∼0.0098ppm, 평균 0.0001ppm이었다. 상치, 무우에서만 검출되었다. 9. Carbaryl(NAC)은 6.3%의 검출율을 보였고 검출범위는 ND∼0.8452ppm, 평균은 0.0336ppm이었다. 상치와 쑥갓에서만 검출되었다. 10. Isoprocarb(MIPC)는 31.3%의 검출율을 보였고 검출범위는 ND∼0.1180ppm, 평균 0.0104ppm이었다. 11. BPMC는 22.7%의 검출율을 보였고 검출범위는 ND∼0.5106ppm, 평균 0.0085ppm이었다. 12. Captan은 58.7%의 검출율을 보였고 검출범위는 ND∼0.1970ppm, 평균은 0.005ppm이었다. 13. Captafol은 2.3%의 검출율을 보였고 검출범위는 ND∼0.0312ppm, 평균은 0.0003ppm이었다. Captafol은 파에서만 검출되었다.

Micro-ecosystem중 bipyridylium 제초제 paraquat 토양잔류물의 행적 (Behaviour of the soil residues of the bipyridylium herbicide, [$^{14}C$]paraquat in the micro-ecosystem)

  • Bipyridylium계 제초제 paraquat 잔류물의 토양중 행적을 구명하기 위하여 [$^{14}C$]paraquat의 신생 및 6주간의 숙성잔류물을 함유한 2종의 토양을 가지고 특수하게 제작한 pot에서 옥수수를 4주간 재배하였다. 숙성기간과 옥수수 재배기간 동안 $^{14}CO_{2}$로 무기화된 양은 총처리 방사능의 각각 $0.13{\sim}0.18%$$0.02{\sim}0.17%$이었다. 옥수수 재배기간중 뿌리와 줄기에 흡수 이행된 $^{14}C$은 두 토양 모두에서 각각 0.1%와 0.01% 미만이었으나 토양중에 남아 있는 $^{14}C$은 두 토양 모두에서 총처리 방사능의 97% 이상이었다. 4주간 옥수수를 재배한 후 토양중 paraquat의 물에 의한 추출율은 1.2% 이하로 매우 낮았고 [$^{14}C$]paraquat 추출불가 토양잔류물의 대부분은 humin에 혼입되어 있었다. 토양을 숙성하는 동안 토양 B의 pH가 증가하였고 옥수수 재배후에는 모든 처리구에서 토양의 pH가 증가하였다. 옥수수뿌리의 각 subcellular particle중에서 residue 분획(incompletely homogenized tissue)에 가장 많은 $^{14}C$이 분포해 있었다. 토양의 dehydrogenase 활성은 숙성에 관계없이 옥수수 재배후에 증가하였다.

국내산 인삼 가공과정 중 difenoconazole의 가공 및 감소계수 연구 (Processing and Reducing Factors of Difenoconazole during Ginseng Processing)

  • 본 연구는 인삼 및 인삼 가공품에 대한 합리적인 농약잔류허용기준 설정과 인삼 및 가공품 중 농약의 잔류성을 규명하고 이를 토대로 인삼 가공단계별 농약의 감소계수와 가공계수를 산출하고자 수행되었다. 시험농약은 difenoconazole로 6년근 인삼포 2곳(강원도 원주, 경기도 이천)에 농약사용기준에 따라 살포한 후 수삼을 수확하여 건삼, 홍삼, 인삼 농축액 및 홍삼 농축액 시료로 가공하였다. 시험결과 difenoconazole의 잔류량은 원주, 이천수삼에서 모두 LOQ(0.02 mg/kg) 미만, 건삼은 각각 0.04 mg/kg, 0.10 mg/kg, 홍삼은 각각 LOQ(0.04 mg/kg) 미만, 0.08 mg/kg, 인삼 알코올 농축액은 각각 0.07, 0.36 mg/kg, 인삼 물 농축액은 모두 LOQ(0.04 mg/kg) 미만, 홍삼 알코올 농축액은 각각 0.08 mg/kg, 0.18 mg/kg, 홍삼 물 농축액은 모두 LOQ(0.04 mg/kg) 미만으로 모두 농약잔류허용기준(MRL) 이하였다. Difenoconazole의 가공계수는 건삼 3.55, 홍삼 3.17, 인삼 알코올 농축액 10.78, 인삼 물 농축액 2.00, 홍삼 알코올 농축액 6.45, 홍삼 물 농축액 2.00으로 산출되었으며, 감소계수는 건삼 1.09, 홍삼 1.02, 인삼 알코올 농축액 3.74, 인삼 물 농축액 0.73, 홍삼 알코올 농축액 2.42, 홍삼 물 농축액 0.74였다.

친환경 버섯배지에 녹차 잔류물의 처리가 새송이버섯의 생장 및 Catechin류 함량에 미치는 영향 (Effects of Green Tea Residue Treatment in Eco-Friendly Medium on Growth and Catechin Content of Pleurotus eryngii)

  • The present study was conducted to investigate the translocation of polyphenols, especially catechin derivatives, from mushroom medium mixed with green tea residues into fruiting body of Pleurotus eryngii. Pleurotus eryngii was grown on the media incorporated by mixing or surface-treated with dry materials including leaf petioles and young stems or leaves of green tea. The dry materials treated in medium did not affect plant height and fresh weight of Pleurotus eryngii body. From the samples of Pleurotus eryngii, the eight main catechin derivatives (-)-gallocatechin(GC), (+)-catechin (C), (-)-epicatechin (EC), (-)-epigallocatechin (EGC), (-)-epigallocatechin gallate (EGCG), (-)-gallocatechin gallate (GCG), (-)-epicatechin gallate (ECG), and (-)-catechin gallate (EGCG), and caffeine were analyzed quantitatively by HPLC. The results showed that EGC in Pleurotus eryngii was 45% more detected, when incorporated with the dry materials, than untreated control. Especially, content of EGCG was increased in surface-treated Pleurotus eryngii up to 3.2 ppm, while it was not detected or reduced in control and other treatments. Caffeine content was greatly increased regardless of treatment method, compared with control (0.1ppm), showing 44 fold-amount in Pleurotus eryngii at early growth stage when incorporated with the dry materials into medium. The results indicates that functional catechin derivatives of green tea would be partly translocated into Pleurotus eryngii throught incorporation and surface treatment with residues of green tea plants.

양상추의 관능적 및 미생물학적 특성에 전해수 및 염소수가 미치는 영향 (Effects of Electrolyzed Water and Chlorinated Water on Sensory and Microbiological Characteristics of Lettuce)

  • This study was conducted to investigate the effects of various kinds of electrolyzed and chlorinated waters on the sensory and microbiological qualities of fresh-cut lettuce and to determine the most suitable electrolyzed water for the vegetable dishes, without heat treatment, at institutional foodservices. The sensory evaluation resulted in higher scores on the 1st-day of storage for the EW-1 (diaphragm type 1) and EW-3 (non-diaphragm type) compared to that for EW-2 (diaphragm type 2), with regard to their appearance, discoloration, texture, taste and overall acceptability characteristics. However, over time, EW-3 ranked highest, with a score of 8.00 (very like), on the 4th-day of storage, which maintained the highest level up to the 7th-day of storage, at which time the score was 7.00 (fairly like). The CW (chlorinated Water) had a significantly lower score, due to the smell of chlorine, although there was no concern with relation to chlorine residue from the electrolyzed waters. Microbial examinations of the total plate count revealed that immersing lettuce into EW-3 brought about l/3,000 to 1/30,000 reductions in the microbial counts of the TW treatment or untreated samples for up to seven days of storage. The CW treatment gave a 1/10 reduction in the microbial counts compared with the TW (tap water) treatment. The coliform bacterial counts also showed similar trends to those of the total plate count values. With regards to the psychotropic bacterial count, EW-3 was able to result in as much as a 1/30,000 reduction in the initial counts. As vegetable dishes, such as salad, can not be heat-sterilized, the utilization of EW-3 for the preparation of vegetable dishes without heat treatment will be an excellent choice to improve the critical control point in production state as a new effective means for sanitizing management.

Hematological and serum biochemical studies in fresh water fish exposed to acute and chronic copper and mercury toxicity

  • A total number of 668 apparently healthy fish were obtained from farm to study the effect of two heavy metals in a form of (Copper sulfate and Mercuric chloride) on some hematological and biochemical parameters of blood. The $LC_{50}$ /96 hr. of Cu and Hg were estimated and fish exposed to $\text\tiny{^1/_2}$ $LC_{50}$ for 7 days and for $1/_{10}$ $LC_{50}$ for 8 weeks from each product separately. Results showed decrease in RBCs count, PCV% and Hb in acute and chronic mercury while a significant increase was shown in acute and chronic copper toxicity, total leucocytic count showed decrease in acute mercury toxicity and increase in the chronic case, while in copper toxicity non-significant decrease in acute and significant decrease in chronic toxicity was noticed. Elevated serum urea and creatinine in both acute and chronic mercury and copper toxicity was detected. No changes in total bilirubin in the acute mercury and chronic copper toxicity while significant increase in chronic mercury and acute copper. Elevation of serum AST and ALT in some days of acute toxicity of mercury and copper while in chronic mercury toxicity a significant elevation of both serums AST and ALT were detected .while in chronic copper toxicity serum AST was fluctuated and ALT showed no significant changes. CK study revealed significant decrease in acute mercury with fluctuation in the chronic toxicity while in copper toxicity it showed fluctuation in acute and significant decrease in chronic toxicity. Glucose value decreased in acute and chronic mercury toxicity while in copper toxicity it showed significant increase in the acute and increase followed by significant decrease in the chronic copper toxicity.

Effects of Mechanically Activated Milling and Calcination Process on the Phase Stability and Particle Morphology of Monoclinic Zirconia Synthesized by Hydrolysis of ZrOCl2 Solution

  • The purpose of this paper was to investigate the effect of a high-energy milling (HEM) process on the particle morphology and the correlation between a thermal treatment and tetragonal/monoclinic nanostructured zirconia powders obtained by a precipitation process. To eliminate chloride residue ions from hydrous zirconia, a modified washing method was used. It was found that the used washing method was effective in removing the chloride from the precipitated gel. In order to investigate the effect of a pre-milling process on the particle morphology of the precipitate, dried $Zr(OH)_4$ was milled using a HEM machine with distilled water. The particle size of the $Zr(OH)_4$ powder exposed to HEM reduced to 100~150 nm, whereas that of fresh $Zr(OH)_4$ powder without a pre-milling process had a large and irregular size of 100 nm~1.5 ${\mu}m$. Additionally, modified heat treatment process was proposed to achieve nano-sized zirconia having a pure monoclinic phase. It was evident that two-step calcining process was effective in perfectly eliminating the tetragonal phase, having a small average particle of ~100 nm with good uniformity compared to the sample calcined by a single-step process, showing a large average particle size of ~300 nm with an irregular particle shape and a broad particle size distribution. The modified method is considered to be a promising process for nano-sized zirconia having a fully monoclinic phase.
