Gilt Transfer of Cultured Freezing Embryos by Open Pulled Straw(OPS) Methods (Open Pulled Straw(OPS) 방법에 의한 체외 배양 동결 수정란의 미경산돈 이식)

  • In previous studies, we reported that sow which was transferred OPS-freezing embryos not able to deliver a piglet (Kim et al, 2004). This study was conducted to investigate a possibility of gilt as recipients which produce piglets after transfer of OPS-freezing embryos. All transferred embryos were prepared by in vitro production (IVP) system. In vitro culture (IVC) medium used glucose-free NCSU23 supplemented with 5mM sodium pyruvate, 0.5 mM sodium lactate and 4 mg/ml bovine serum albumin for 2 days at $39^{\circ}C$. From day 3 of IVC, 10% fetal bovine serum albumin was added to the culture medium. In preparing of freezing embryos, embryos were treated with 7.5 $\mu g/ml$ cytochalasin-B for 30 min and centrifuged at $13,000{\times}g$ for 13 min. And then, embryos were exposed sequentially to an ethylene glycol (EG) solution, aspirated into open pulled straw (OPS), and plunged or thawed into the liquid nitrogen. In embryo transfer (ET), we used two kinds of type (surgical method vs. non-surgical method). In surgical method of embryo transfer, $55\sim65$ embryo were transferred in both uterine horn of two recipient gilts by plastic straw. Non-surgical method which is like artificial insemination was performed on three gilts. Each 140 frozen embryos were transferred to two gilts and 40 fresh embryos to one gilt. Pregnancy establishment was shown one recipient at 45 days after ET. However, the one recipient was also aborted at 58 days after ET. These results suggest that gilts can be considered as a candidate of recipients for OPS-freezing embryo transfer.

  • This study was designed to determine the changes of ATP-related compounds, especially the concentration of IMP, and compares the relationship between IMP and flavor of pork loin chops during storage as three different storage temperatures (include $4^{\circ}C$ CF and PF). Pork loin chops were kept under $4^{\circ}C$ $-1.5{\pm}0.5^{\circ}C$ (control freezing storage) and $-3{\pm}0.5^{\circ}C$ (partially freezing storage). The changes of TBA values, ATP-related compounds, pH values, in CF stored samples were higher than PF stored samples, but it had lower TBA values than $4^{\circ}C$ storage. The IMP concentration reached their peak after 2 days in $4^{\circ}C$, 5 days in CF and 7 days in PF storage, and the ATP, ADP, AMP contents of the loin chops showed minimum, respectively. Flavor of meat sensory score for $4^{\circ}C$ stored samples were more intense (p<0.05) than CF and PF samples on day 2 of storage. However, after storage for 5 days, flavor scores for CF samples were more intense (p<0.05) than $4^{\circ}C$ and PF samples. Flavor scores for PF samples were more intense (p<0.05) than $4^{\circ}C$ and CF on day 7 of storage. As the meat with the peak of IMP contents was most preferred, it was considered that the content of IMP was related to the flavor of meat and that CF, PF had influence on the IMP content.

  • Yellowtail fishes(Seriola quinqueeradiata) were submitted to the storages using ice-cooling($0^{\circ}C$), partial freezing($-3^{\circ}C$) and freezing $-20^{\circ}C$) method. Changes in the structures of muscle during storage at different temperatures were investigated. The ice-cooling and partial freezing storage caused early decomposition of glycogen granules and mitochondrial inner membrane, but it was accorded to much slower manner comparing with that of ice-cooling storage. The scars of ice crystals were appeared after three days of storage. The number and size of the crystal increased as progressing of the storage. They were circular and mostly located between fibers. When using the freezing storage, glycogen granules were mostly found from the muscle cell even after fourteen days of storage. Mitochonidral inner membrane maintained their integrity. The scars of ice crystals were also found, however, different from those of partial freezing storage. Their existing sites were random and their shapes were irregular. In many cases, they located in the fiber and had keen edges. Fibers were broken mostly at the Z-lines on fourteen days of storage. From these results, it was concluded that partial freezing storage can repress autolytic enzymic action and can reduce the physical damage from ice crystals which is caused by freezing.

Changes of Composition in Immature Green Persimmons during Storage (풋감의 저장 중 성분변화)

  • We investigated the changes of composition in immature green persimmon during cold(7$^{\circ}C$) and freezing (-2$0^{\circ}C$) storage for 30 days. Moisture, protein, fat and carbohydrate were not significantly different by storage temperature and storage days. Crude fiber had little change during freezing storage, but in case of cold storage, as storage time goes by, it slightly deceased. Content of polyphenol was increased during cold and freezing storage, especially during cold storage it was remarkably increased. Content of total pectin was decreased, while content of soluble pectin was increased during cold storage. Content of total pectin and soluble pectin was slightly decreased or increased during freeze storage. Content of tannin had little change during freezing storage but it significantly decreased during cold storage.

Immunological Assays of Freezing Tolerance in Barley using Antifreeze Proteins Antisera

  • In order to measure an antifreezing tolerance, antifreeze proteins accumulated upon cold acclimation in apoplast were analyzed. As Dongborilho were cold-acclimated for 3 to 74 days there was an abrupt increase in apoplastic proteins up to 30 days and then decrease to the similar levels. Among the known antifreeze proteins, CLP produced in E. coli. and TLP purified from apoplast were used to generate antisera that allow to measure and localize the proteins in leaves of barley. The CLP of 27.7 kDa and TLPs of 6, 26, 27, 30, and 31 kDa were increased in their amounts in apoplast as cold treatment being longer. There was a correlation among the amounts of those proteins accumulated in apoplast and freezing tolerance as shown in field and ion leakage tests for five cultivars. The deposit of CLP was localized in the marginal area and the area adjacent to leaf vescular bundle cells in an increasing manner according to duration of cold acclimation but no variation was observed in terms of it's distribution. Based on the close correlation between levels of antifreeze proteins and degrees of freezing tolerance, the immunological methods was to develop to estimate a freezing tolerance in barley.

Quality Changes of Jujube Wine by Hydrostatic Pressure and Freezing Treatment during Storage (초고압 및 냉동처리에 의한 대추술의 저장 중 품질변화)

  • Jujube wine was treated with hydrostatic pressure (500 MPa, 5 min) or freezing ($-20^{\circ}C$, 3 days and thawed $20^{\circ}C$, 4 hr) and their microbial counts, physicochemical properties, and sensory characteristics were investigated during storage at $35^{\circ}C$ for 60 days. Microorganisms in pressure-treated jujube wine were not detected during the whole period and chemical compositions as well as color were slightly changed. Sensory quality was significantly preserved until 20 days without increasing sweet aroma and taste. In freezing-treated wine, bacterial counts were decreased after 10 days and remained below 10 CFU/mL while lactic acid bacteria and yeast were not detected. Changes of chemical composition were smaller than those of the untreated wine but bigger than those of the pressure-treated or heat-treated wine. Instrumental color was changed after $10{\sim}20$ days resulting from the increase of L value and the reduction of a value. Sensory quality was significantly similar with the fresh wine for 10 days, suggesting that pressure treatment would be the most effective sterilization method to improve the shelf life of jujube wine whereas freezing treatment would be insufficient.

A Study on the Combined Deterioration of Concrete subjected to Freezing-Thawing and Chloride Attack (동해와 염해를 동시에 받는 콘크리트의 복합열화에 관한 연구)

  • This paper was accomplished for analyzing the reason of the above deterioration happened on the deck of concrete bridge. The bridge was constructed at 660m above the sea level having more freezing and snowing days. Therefore, it is placed on the particular condition sprinkling $CaCl_2$ enough for keeping up with moderate traffic condition. When it is considered to the former condition, the bridge can be assumed to potentialities for combined deterioration with freezing-thawing under sprinkling deicing chemical. Core specimens were gathered from the concrete deck for clearing the reason of the above deterioration exactly, and it is used for various tests for measuring the compressive strength, elastic modulus, content of $Cl^-$, freezing-thawing at the fresh and salt water. As a result of freezing-thawing test, the specimen at the fresh water has over 90$\%$ of durability factor, but another specimen at 1$\%$ of salt water has 0$\%$ of durability factor at 140 cycles of the freezing-thawing. The result means that frost damage is sccelerated at the salt water. Therefore, the deterioration of the concrete deck is estimated to be occured by combined effects of freezing-thawing and chloride ion attack.

Survival of Sanitary Indicative Bacteria Inoculated in Fish Muscle Homogenates during Freezing and Frozen Storage (어육에 접종한 위생지표세균의 동결저장중 변화)

  • This experiment was designed to evaluate effects of freezing and frozen storage on survival of sanitary indicative bacteria in seafoods. Culture of bacteria such as Escherichia coli type I, Citrobacter freundii type I, Klebsiella aerogenes type I and Streptococcus faecalis was inoculated into homogenates of pollack, shrimp, and sardine frozen in a contact plate freezer at $-40^{\circ}C$ and chest freezer at $-20^{\circ}C$, stored at $-20^{\circ}C$, and then survival of the inoculated bacteria was determined over a period of 95 days. Coliform group was highly sensitive to freezing and frozen storage showing survival of about $2\%$ after 95 days of frozen storage at $-20^{\circ}C$, whereas Streptococcus faecalis was relatively resistant with $20\%$ survival rate. The sanitary indicative bacteria count was rapidly decreased in the early stage of frozen storage revealing 90 to $95\%$ loss of coliform group and 40 to $70\%$ loss in case of Streptococcus faecalis after 10 days storage. In determining recovery rate, most probable number (MPN) method gave more reproducible recovery of the tested strain than did the selected agar plate method.

Compressive Strength Properties and Freezing and Thawing Resistance of CSG Materials (CSG 재료의 압축강도 특성 및 동결융해 저항성)

  • The cemented sand and gravel (CSG) method is a construction technique that adds cement and water to rock-like materials, such as rivered gravel or excavation muck which that can be obtained easily at areas adjacent to dam sites. This study was performed to evaluate the unconfined compressive strength properties and freezing and thawing resistance of CSG materials with unit cement content. The three types of CSG-80, CSG-100 and CSG-120 with cement content were designed to evaluate the optimum water content, dry density, strength, stress-strain, micro structure and durability factor. As the results, the optimum water content ratio with cement content showed almost similar tendency, and the unconfined compressive strength and dry density increased as cement content increases. The strength ratio of 7 days for 28 days were in the range of 55~61 % and the strain ratio in stress-strain curve were in the range of 0.8~1.6 % nearby maximum strength in 28 days. It is expected that this study will contribute to increasing application of CSG method as well as to increasing the utilizing of CSG materials as a environmentally friendly CSG method.

  • Experiments were conducted to investigate the soil freezing depth and pattern with freezing measuring instruments during 1988-l989 winter season in Kangwon province. Freezing measuring instrument was made with acrylic pipes which were consisted of inner and outer parts. Inner pipe was filled with 0.01 % methylene blue solution and rubber hose to protect pipe breakdown by solution freezing. Freezing measurements were carried out by observing discoloration of methylene blue solution. Moisture content of evergreen trees and ground cover plants was also examined in the winter season. The observed results are as follows: 1.In the land of I OOM above sea level, soil freezing depth became deeper as the sum of Accumulated degree-days of temperature below 0˚C(0˚C . day) increased: Soil freezing depth was 30-40cm at l00˚C, 42-43cm at 150˚C, and 47cm at 200˚C day 2.Soil freezing with vinyl mulching was less developed by l3cm at l00˚C with sum of subzero temperature, by l7cm at 200˚C than that of the bare ground. Soil of rich hulls mulching with 4Ocm was not frozen until soil freezing at the bare ground was developed to 25cm depth. 3.Cashmeron mulching was more effective than felt mulching in the heat insulation of soil. 4.Thawing of soil was done from the lowest part of the frozen in the ground to upward in the beginning and after that it was done from the surface of frozen soil to downward. Finally thawing was completed at the middle of frozen soil.
