Quality characteristics of Aronia melanocarpa by different drying method (건조방법에 따른 아로니아의 품질특성)

  • In this study, the quality characteristics of different drying methods of Aronia melanocarpa, which contains large amounts of bioactive substances, were investigated for the improvement of their practical use. During the drying period, the weight reduction was the highest with vacuum freeze drying (81.6%). The water content was reduced to the maximum level when vacuum freeze-drying was used. With regard to the color value measurement results, there were no significant differences in the $L^*$ value. Values of $a^*$ and $b^*$ were increased in vacuum freeze drying and cold air drying, but decreased in hot air drying. The hardness was increased dramatically after 36 hours of hot air drying, while with cold air drying, it increased slowly until 132 hr and increased rapidly after 132 hr. The dried yield was the highest with cold air drying (24.2%). As for the general component analysis results of Aronia melanocarpa, the moisture content was the lowest, and the crude protein and crude fat contents were the highest with vacuum freeze drying. No difference in pH value was shown among the dried Aronia melanocarpa obtained from the different drying methods, but the sugar content was the highest with vacuum freeze drying. The mineral content was the highest with cold air drying, and K, Ca, Mg, and Na were the major minerals. The free sugar content of dried Aronia melanocarpa was found to be 5.92~20.59 g/100 g, and the highest free sugar content was found with vaccum freeze drying.

  • This study was performed to analysis of chemical constituent in Polygonum multiflorum root (PMR) by different dry methods (hot-air dry, shade dry, and freeze dry). The results are summarized as followings; major free sugar were detected fructose, glucose, and sucrose in dried PMR based on various dry methods. The highest content of free sugars was found in freeze dried PMR. The four organic acids were detected in dried PMR by HPLC analysis. The content of oxalic acid in shade dried PMR was higher than the dried PMR by different dry methods. The content of total amino acid and essential amino acids were high in the orders of freeze drying > shade drying > hot-air drying. The potassium and magnesium levels of freeze dried PMR was significantly higher than the other drying method of PMR. Whereas the calcium and sodium levels were higher in hot-air dried PMR. The major fatty acids were determined the linoleic acid in PMR by different dry methods.

  • Plant-based cultural assets using straw and grass as household goods of our people's have been used as indispensable tools for practical living for a very long time. However, only a limited number of artifacts were unearthed so far due to tile fragility of the material. For this reason, research on plant-based cultural properties had close to no progress, and the appropriateness of the PEG method, high-grade alcohol method, alcohol--ether-resin method, and Paraloid B-72 used in preserving plant-based cultural properties has not been sufficiently investigated. Therefore, this study examined the weight change rate by applying the methods of Primal MC-76 and vacuum freeze-drying used mostly as a earth-layer hardening material among PEG and acrylic resin, which are applied widely for preservation of waterlogged archaeological wood, as a means to preserve plant-based cultural properties along with the examination of the subject material, and an experiment was also performed on moisture absorption. The findings as a result were, first, the plant-based material being studied was found to be Typha (Typha orientalis Presl). Secondly, the weight change experiment applying $PEG\#400$ and $PEG\#4000$ confirmed a steady increase of weight if PEG -2Step is used for treatment. Third, in preserving all subject materials with soil, treatment with $PEG\#4000$, Primal MC-76, and vacuum freeze-drying showed that tile vacuum freeze-drying method resulted in the largest or $20\%$ reduction in weight, while Primal MC-76 resulted in $18\%$ and $PEG\#4000$ in $8\%$ of weight reduction. It was concluded that, considering the stability of soil measurement, this came to be because resin permeation was carried out along with tile drying process. Fourth, the weight changes were found to be around $10\%$ in various humidity conditions after the preservation treatment. The greatest weight change rate was seen in the case of $PEG\#4000$, particularly having chemicals gush out in a high humidity (RH $84\%$ or higher) environment. In the case of Primal MC-76 and vacuum freeze-drying methods, $6\~8\%$ weight changes were detected, and the lowest weight change was found in the case of the vacuum freeze-drying method. Fifth, as for color changes after treatment, blackening occurred most strongly with $PEG\#4000$, while Primal MC-76 and vacuum freeze-drying manifested colors closest to dry straw or grass. However, the texture of straw was not very evident in the case of Primal MC-76, due to a glossy surface, but vacuum freeze-drying was found to offer tile best result in terms of texture. Putting together the results of the above experiments, vacuum freeze-drying presented after being treated with PEG2-Step the most stabilized changes in weight, while it offered the smallest change in color as well.

The Effect of Dry Methods for Synthesized Yttria-doped Ceria by Co-precipitation (공침법으로 제조된 Yttira Doped Ceria분체의 건조방법에 따른 입자특성 고찰)

  • In synthesis of nano powders, the hard agglomeration for the synthesized powders occurred during the drying processing. In order to avoid hard agglomeration in particles the freeze drying process was used in this experiment. e fabricated the Yttira-Doped Ceria(YDC) nano powder by co-precipitation. Starting materials used in experiments were the cerium(III) nitrate and yttrium(III) nitrate solution with 야-water, which two solutions were mixed and then the precipitated hydroxides were prepared for adding sodium hydroxide. The co-precipitated powders were dried by the thermal drying at 8$0^{\circ}C$ for 24 h and by freeze drying at -4$0^{\circ}C$, 30 mtorr for 72 h. The lattice parameter and crystallite size as a function of calcination temperature was characterized by XRD analysis. The lattice parameter of YDC was decreased with addition amount of yttrium and was estimated as 5.401683 $\AA$ at $700^{\circ}C$. Crystallite size were calculated by XRD-LB method, and morphologies were confirmed with the observation of TEM and SEM. The freeze dried YDC powders had medium diameter of 17 nm with more uniform size distribution than the thermal dried YDC posers, which were mainly ascribed to the difference of agglomerates formation during drying stage.

Comparison of Particle Size Analysis and Distribution of Heavy Metals in River and Lake Sediments (하천 및 호소 퇴적물 입도분석 방식의 비교와 입도에 따른 중금속물질의 분포경향)

  • Dry sieving, wet sieving and photoscattering analyzer were tested as particle size analyzing methods for sediments from rivers and lakes of Han river, Nakdong river, Youngsan river, and Kumgang river area. Dry sieving showed a big error due to coagulating effect over drying process and this phenomena was severe for lake sediment. Wet sieving and PSA showed an accurate results though wet sieving needs more labor and complicate processes. Freeze-dry or freeze-dry after oxidation of organics with hydrogen peroxide showed improved results but gave lower portion of fine particles in comparing with wet sieving. Heavy metal contents and extractable metal contents were investigated for the sediments and high heavy metal content and extractable amount were obtained from fine particles as expected. Using of proper particle size analyzing method is important and the sediment management should be focused on the fine particles.

  • Preventing the infiltration of rainwater into the landfill site is the main purpose of the final cover in landfill sites. Compacted clay layer or geomembrain have been used as a conventional landfill final cover. But they have several disadvantages when damages might occur due to puncturing, differential settlement and desiccation or freeze and thaw. For this reason, as an alternative method SRSL(Self Recovering Sustainable Liner) has been developed. Adopting the precipitation reaction of two chemical material, by forming precipitates that fill the pores, and lower the overall permeability of the liner. The advantage of this method is that when fracture of the liner occurs the remaining reactants of the two layers form precipitates that fill the fracture and recover the low permeability of the liner. In this study, the recovering ability of the SRSL with a crack due to the seasonal variation or differential settlements was investigated by permeability tests. And in order to estimate the durability of the SRSL after freeze/thaw and desiccation, uniaxial compression strength tests were performed.

  • In this study, a poly(lactic-co-glycolic acid) (PLGA) microspheres was fabricated using emulsion solvent evaporation technique. During the procedure fabrication, some parameters process have effected on the formation of micro-carriers. The structure and morphology of micro-carriers were evaluated by SEM observation. Beside, heparin incorporated into microspheres was determined using toluidine blue method. Specifically, the effects of some parameters process such as ultrasonic levels, PLGA concentrations and freeze-dry times on the size, structure, porous formation and heparin entrapment of micro-carriers were studied carefully. We found that, the morphology and structure of carriers were influenced by the all above parameters. The diameter of the carriers varied from 20 to 400 ${\mu}M$ depending on experimental conditions. At suitable freeze-dry time, the pores were automatically formation on surface of microspheres with a significantly in the numbers of pore. After heparin incorporated porous PLGA microspheres, it was suggested that the highly heparin incorporated into porous PLGA microspheres could enhance of angiogenesis for tissue regeneration easily.

Characteristics and Drug Release Profiles of Multilamellar Vesicle(MLV) and Microemulsified Liposome(MEL) Entrapped 5-Fluorouracil and Its derivatives (5-Fluorouracil과 그 유도체를 봉입한 Multilamellar Vesicle(MLV)과 Microemulsified Liposome(MEL)의 특성 및 약물방출 거동)

  • Although liposome has many advantages as a pharmaceutical dosage form, its application in the industrial field has been limited because of some problems such as preparation method, reproducibility, scale-up, stability and sterilization etc. Liposomes prepared by microemulsification method had defined size, narrow size distribution, reproducibility and high entrapment efficiency. For enhancing the stability, the dry form of liposome was recommended. These types of liposome are proliposome and freeze-dried liposome. The liposome must have some properties for preparing of freeze-dried liposome; small size $(50{\sim}200\;nm)$, narrow size distribution and cryoprotectant. In this experiment, the liposomes containing 5-Fluorouracil(5-FU) and its prodrug(pentyl-5-FU-1-acetate; PFA, hexyl-5-FU-1-acetate; HFA) were made with soybean phosphatidylcholine, cholesterol, stearylamine(SA) and dicetyl phosphate(DCP) employing hydration method or microemulsification method using $Microfluidizer^{TM}$. Both or liposome types were MLV and MEL. After preparation, freeze drying and rehydration were performed. In the process of freezing, trehalose(Tr) was added as a cryoprotectant. Their evaluation methods were as follows; entrapment efficiency, mean particle size and size distribution, dissolution test, retain of entrapment efficiency and turbidity after freeze-drying. The results are summarized as belows. The entrapment efficiency of 5-FU was dependent on total lipid concentration and cholesterol content but that of PFA and HFA was decreased when cholesterol was added. When DCP and SA were added, entrapment efficiency was decreased. As the partition coefficient of drug was increased, entrapment efficiency was increased. Under the same condition, entrapment efficiency of MEL is similar to that of MLV. The mean particle size and size distribution of MEL were smaller than those of MLV. Dissolution rates of drug from both liposome types were comparatively similar. Dissolution rates of drugs with serum and liver homogenate were faster than without these material. After preparation of liposome, free drug was removed efficiency by Dowex 50W-X4. When liposome was freeze-dried and then rehydrated in the presence of Tr, characteristics of liposome were maintained well in MEL than MLV. Tr Was used successfully as a cryoprotectant in the process of freeze drying and the optimal ratio of Tr:Lipid was 4:1(g/g).

Effects of Maltose on the Stability of Freeze-Dried Liposomes (동결 건조된 리포솜의 안정화에 있어서 말토스의 영향)

  • Liposome powders were prepared by a freeze-drying method for the application to the field of drug carrier. The effect of maltose as a liposome stabilizer was studied on the stability and the drug-loading efficiency of the freeze-dried liposome powders. The particle size of liposomes before and after freeze-drying was determined to evaluate the liposome stability. The drug-loading efficiency was measured by Fluorescence spectrophotometer using calcein as a model drug. When maltose was added after the preparation of the liposomes, the liposomes was stable, compared to the case of maltose addition at the hydration procedure. By the addition of maltose, the liposome was stable for 30 days at $4{\sim}37^{\circ}C$, while the particle size of the liposome without maltose increased with time. The liposome showed relatively high stability when the maltose/lipids molar ratio was 3 and 6.

Comparison of Cosmetical Activities of Curcuma longa L. Leaf Extracts Using Different Extraction Methods (추출 방법에 따른 강황 잎 추출물의 향장 활성 비교)

  • Cosmetic activities of the leaves of Curcuma longa L. were compared according to different drying methods, to expand the use of the by-products of Curcuma longa L. The highest extraction yield of 29.4% was obtained from vacuum drying process (VD), whose value was very close to 29.2% from freeze drying process (FD). Relatively lower extraction yield were observed as 24.5% and 25.3% from the raw leaf (RL) as control and hot air drying process (HD). The cytotoxicity was observed lower FD and VD than RL and HD. It shows that cytotoxicity was closely related to cosmetic activities such as tyrosinase and melanin inhibition activities by showing much better cosmetic activities of the extracts from both FD and VD than those from the RL and HD. It was interesting that was differences of the cosmetic activities and cytotoxicity between FD and VD, which implies that VD method should be a more efficient process than FD method in terms of drying time and operation costs: 6 hours vs 24 hours and 3-5 times higher extraction costs in drying. It was observed that VD is more excellent dry method than others. This result could be utilized to effectively dry other soft plant biomass.
