Quality Characteristics and Shelf Life of Noodles Prepared with Heracleum moellendorffii(Hogweed) Powder (어수리분말 첨가 국수의 품질특성 및 저장성)

  • The quality characteristics and shelf life of wet noodles containing a freeze-dried powder of Heracleum moellendorffii(HM) were studied to investigate the use of HM as a food ingredient. The weight, volume, and water absorption of wet noodle sprepared with HM(HML) decreased as the concentration of HM increased. The turbidity of HML sauce and the loss in the solid content of noodles after cooking were lower than those of control noodles. Whiteness(L) and redness(a) values of wet noodles decreased but yellowness(b) increased after addition of HM. The sensory qualities of HML, including appearance, taste, color, flavor, texture, and overall acceptability, were better than those of control noodles. Addition of HM to 0.6%(w/w) afforded the best sensory qualities with respect to taste, texture, and overall acceptability. The total polyphenol content of HML increased as HM concentration increased. Noodle DPPH free-radical-scavenging activities were 22%(control), 28.41%(0.3%HM addition), 40.22%(0.6%HM addition) and 49.42%(0.9%HM addition). Viable bacterial cell counts did not differ significantly between control noodles and those prepared using 0.6%(w/w)HM during storage for 6day sat $10^{\circ}C$. However, viable cell numbers in noodles prepared using 0.9%(w/w) HM were significantly lower than those of control samples and of noodles prepared using either 0.3%(w/w) or 0.6%(w/w) HM, during storage for 12 days at $10^{\circ}C$. Changes in pH values showed trends similar to those of viable cell numbers during storage.

Antioxidative activity of roasted Pueraria lobata root extracts (로스팅 칡 추출물의 항산화 활성)

  • We evaluated the antioxidative activity of extracts of P. lobata root depending on roasting conditions. P. lobata roots were roasted at three different temperature at $150^{\circ}C$, $200^{\circ}C$, and $250^{\circ}C$ and three different time at 10 min, 20 min, and 30 min respectively. Roasted P. lobata root was extracted using water at $85^{\circ}C$ for 6 h and filtered using filter paper, followed by then evaporated ($12{\pm}0.3^{\circ}Brix$) and freeze-dried. The concentration of maker compound puerarin was determined using a high performance liquid chromatography system. 2 phenolic compounds, flavonoid contents, and antioxidant activities of the extract powder were evaluated. Puerarin contents, Phenolic compounds, and flavonoid contents of roasted P. lobata root were higher than those of unroasted P. lobata root. The results of DPPH and ABTS showed that roasted P. lobata root possessed higher antioxidant activity than unroasted P. lobata root. This study suggested that roasting process could be applied to P. lobata root in order to achieve its high quality and functionality.

Anti-Obesity Effect of By-Product from Soybean on Mouse Fed a High Fat Diet (고지방 식이로 유도된 비만 마우스에서 대두 부산물인 순물과 침지수의 항비만 효과)

  • Here we study the anti-obesity effects of by-product from soybean on mouse fed high fat diet. The body weight gain, visceral and subcutaneous adipose tissue weight, liver and epididymal adipose tissue weight in freeze-dried soybean-soaking-water (SSW) powder fed group showed lower level than those in high fat diet (HFD) group by determining with weight measuring and histological methods. Also, histological analyses of the liver and fat tissues of SSW grouped mice revealed significantly less number of lipid droplets formation and smaller size of adipocytes compared to the HFD group. Moreover, the levels of total serum cholesterol, LDL-cholesterol and the atherogenic index were decreased in the SSW groups. Especially, in SSW group, the levels of phosphorylation of two lipid oxidation enzymes, adenosine monophosphate-activated protein kinase (AMPK) and acetyl-CoA carboxylasse (ACC) were elevated hence that may activate fatty acid oxidation. But AST and ALT levels were not changed in blood. By micro-CT analysis of abdomen, SSW groups significantly showed a tendency to decrease visceral and subcutaneous fats as well as fat-deposited areas compared to HFD group. Taken together, we suggest that soybean soaking water has a function in ameliorating obesity through inhibiting lipid synthesis as well as stimulating fatty acid oxidation.

Comparison of the physicochemical components of Lentinula edodes cultivars cultivated in sawdust medium (톱밥배지에서 재배된 표고버섯 품종별 이화학적 성분 비교)

  • With the development of sawdust medium cultivation technology, Lentinula edodes (shiitake mushroom) has become the most extensively produced and consumed mushroom globally. In this study, the approximate composition, nutritional components, and bioactive compounds of L. edodes were analyzed and compared for the five sawdust-cultivated shiitake mushrooms cultivars namely Sanjo701ho, Sanjo707ho, Sanjo715ho, Chamaram, and L808. The approximate range of the composition of freeze-dried shiitake mushrooms was 4.06-5.92 g/100 g of ash, 0.75-1.02 g/100 g of crude fat, and 21.24-29.15 g/100 g of crude protein. Sanjo701ho had the highest trehalose content (9.60±0.08 g/100 g), total polyphenol content (3.49±0.04 mg GAE/g), and total flavonoid content (1.33±0.03 mg QE/g) among the other shiitake mushroom cultivars. The total amino acid content was as follows: glutamic acid>aspartic acid>leucine. Glycine, alanine, valine, isoleucine, leucine, and phenylalanine contents were significantly decreased in the following sequence: Sanjo707ho, Sanjo715ho, Sanjo701ho, Chamaram, and L808. Crude protein, trehalose, and six types of amino acids were identified as classification indicators for the five cultivars of sawdust-cultivated shiitake mushrooms.

Conservation Treatment on the Bamboo Sunblind from the No. 1 Catchment Site in Baesanseongji, Busan (부산 배산성지 1호 집수지 출토 대나무 발 수습 및 보존처리)

  • In the present study, safe management and value improvement of bamboo sunblind, which is an item of cultural heritage, were performed by adopting stable conservation treatment methods. The bamboo sunblind used in the present study was excavated from No. 1 catchment site in Baesanseongji, Busan. It was determined that the main material used to make the sunblind was bamboo, and herbal plants were used to weave the bamboo using lacquer as an adhesive agent. All contaminants and soil adhered to the sunblind was removed. Thereafter, the sunblind, which was recovered in the form of blocks, was washed separately after fixing it to a temporary plaster frame and to avoid the blocks from breaking during washing. Then, polyethylene glycol (PEG) impregnation was utilized for the reinforcement treatment. Based on the preliminary test results, the shape of the sunblind was fixed using a stainless-steel frame to prevent physical damage that may occur during the drying process. Thereafter, the bamboo sunblind was vacuum freeze-dried. PEG 20% (in ethyl alcohol) was applied as a surface treatment agent for stabilization the sunblind. After the surface treatment, the bamboo sunblind were joined together to fit the maximum width, and the rectangular shape of the sunblind was restored-as best as possible-while filling in the missing parts by maximizing the use of unknown members such as in the disturbed layers below bamboo sunblind surface. The conservation treatment was completed by fixing the bamboo sunblind into the fabricated frame.

Production of Antithrombotic Material Extracted from Auricularia auricular-judae and the Verification of Its Antithrombotic Activity via Animal Test (목이버섯으로부터 추출한 항혈전물질의 제품화와 동물실험을 통한 항혈전활성 검증)

  • Large-scale preparation steps of antithrombotic materials from wood ear mushroom (Auricularia auricular-judae) were established as follows. Grounded dry wood ear mushroom was extracted with 75% ethanol and its precipitate was extracted with $76^{\circ}C$ water for 2 hr followed by filter pressing. The filtrate was then concentrated by vacuum and extracted with 80% ethanol, and the resulting precipitate was then freeze-dried. The formula of the product was determined using consumer susceptibility tests as follows; mushroom extract 90.5%, high fructose corn syrup 2.0%, $\beta$-cyclodextrin 1.5%, fructo-oligosaccharide 2.0%, pear puree 4.0%. When the packed products were stored at 25, 37, or $45^{\circ}C$ for 8 weeks, there were no noticeable changes in water activity, moisture content, pH, and acidity. The viable cell number of total bacteria was slightly increased during the storage period at 25 and $37^{\circ}C$, The total bacteria were not detected in the product when stored at $45^{\circ}C$. When the product was injected intravenously into rat at the level of 1,000 mg/kg, antithrombotic activities such as activated partial thromboplastin time, thrombin time, prothrombin time, and FIB were increased when compared with the control group. When the product was administrated orally into rat at the level of 500 mg/kg, it showed the same antiplatelet activity to aspirin.

Antioxidant and growth inhibitory activities of Mesembryanthemum crystallinum L. in HCT116 human colon cancer cells (아이스플랜트의 항산화 및 HCT116 인체 유래 대장암세포 성장억제 활성)

  • Purpose: This study examined the antioxidant and cancer cell growth inhibitory activities of an ethanol extract and different solvent fractions of Mesembryanthemum crystallinum L. (ice plant). Methods: The ice plant was freeze-dried, extracted with 99.9% ethanol, and then fractionated with hexane, ethyl acetate, butanol, and water. The total polyphenol content (TPC), total carotenoid content (TCC), 2,2-diphenyl-1-picrylhydrazyl radical-scavenging activity (RSA), and ferric reducing antioxidant power (FRAP) were measured. Assays using 2',7'-dichlorofluorescin-diacetate and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide were performed to measure the intracellular reactive oxygen species (ROS) and cell growth, respectively. Annexin V/propidium iodide staining and cell cycle analysis were performed for the detection of apoptosis and cell cycle arrest. Results: TPC, TCC, RSA, and FRAP of the ethanol extract (EE) were 3.7 mg gallic acid equivalent/g, $13.2{\mu}g/g$, 21.0% (at a concentration of 5 mg/mL), and 21.0% (at a concentration of 5 mg/mL), respectively. Among the different solvent fractions, the butanol fraction (BF) showed the highest TPC (5.4 mg gallic acid equivalent/g), TCC ($86.6{\mu}g/g$), RSA (34.9% at 5 mg/mL), and FRAP (80.8% at 5 mg/mL). Treatment of HCT116 human colon cancer cells with EE and BF at concentrations of 250 and $500{\mu}g/mL$ reduced the levels of intracellular ROS. Concomitantly, EE and BF resulted in the dose-dependent inhibition of cell growth (at the concentrations of 125, 250, and $500{\mu}g/mL$ for 24 ~ 48 h) and the induction of apoptosis (at the concentrations of 250 and $500{\mu}g/mL$ for 48 h) in HCT116 cells. An increased G2/M cell population was also found in the BF-treated cells. Conclusion: These results suggest that ice plant possesses antioxidant and growth inhibitory activities in colon cancer cells.

Biolagical Activity on Extracts of Japanese Anise(Illicium Anisatum L.) Leaves and Twigs (붓순나무 잎과 가지의 추출물에 대한 생리활성 평가)

  • Japanese anise (Illicium anisatum L.) leaves and twigs were extracted with 50 % aqueous acetone three times. After filtration, the extracts were fractionated with n-hexane, chloroform, ethyl acetate and $H_2O$, and then freeze dried after condensation. Then antioxidation and antiviral activity were evaluated on each fractions. In the antioxidative activities, the results indicated high activity in the EtOAc soluble fraction of the leaves and the EtOAc and $H_2O$ soluble fractions of the twigs. It showed much higher antioxidative value compare to the controls, BHT and ${\alpha}$-tocopherol. In the antiviral activities, the all fractions were negative effects in HRV 1B and EV 71, but good in Influenza PR8. The activities of the crude extracts of the leaves and twigs showed more than 80% activity at the concentration of $10{\mu}g/mL$ and $50{\mu}g/mL$, respectively, and the activities of the EtOAc and $H_2O$ soluble fractions were close to 80%. Based on the above results, the extracts of Japanese anise may be applied for one of the natural biomass sources that can be used as an antioxidant and an antiviral substance.

Behavioral analysis of rock bream Oplegnathus fasciatus reveals a strong attraction potential for sea urchin extracts

  • Monitoring fish movement is important to understand how physiology adapts to environmental change. To explore the applicability of a video tracking system for determining if chemical cues attract or repel aquatic animals, the movement patterns of rock bream, Oplegnathus fasciatus, were analyzed upon exposure to various materials, including extracts of sea urchin, rock worm, bait worm, krill, barley kernel, and commercial fish feed. Pellets were prepared by mixing freeze-dried tissues with a cellulose and corn flour mixture. Behavioral analysis was carried out with five fish that had been acclimated in the adaptation zone of a Y-shaped tank. Preference toward chemical cues was quantified by assessing the frequency rock bream were observed in a discrete zone around the test material located at the end of each arm and the duration each fish stayed in each zone. The analysis of fish movement upon exposure to commercial feed and barley kernel at each end, respectively, indicated a clear preference toward the feed relative to the barley kernel. Movement responses were further tested with pellets containing extracts of sea urchin, one of the species collected on a large scale, and other materials including krill and worms. A stronger preference toward sea urchin (100%) was observed based on the duration of stay in the test zone, compared to krill (90.1 ± 44.2%), bait worm (81.1 ± 39.1%), rock worm (73.7 ± 28.9%), and barley (63.9 ± 25.9%), under the conditions tested. A detailed comparison of rock bream movements toward each material revealed significant differences in frequency and duration, respectively, between pairs of test materials including krill (74 ± 29.8 and 375.6 ± 118.9) vs. rock worm (41.5 ± 18.7 and 160.2 ± 42.6), krill (86.3 ± 22.9 and 477.1 ± 84) vs. bait worm (36.2 ± 5.5 and 166.1 ± 50.7), and rock worm (45.9 ± 26.2 and 213.7 ± 100.1) vs. bait worm (34.6 ± 21.7 and 159.5 ± 98.5). Rock bream exhibited preference for the test materials in the following order: commercial fish feed > sea urchin > krill > rock worm > bait worm > barley. The results suggest a higher potency of sea urchin extract as a rock bream fishing bait compared to the other materials that are used as commercial bait.

Studies on the Phenolic Compound Content and Antioxidant Efficacy of Rubus idaeus (Raspberry) Cultivar 'Willamette' Fruit Extract Using n-Hexane and Ethyl Acetate (n-Hexane 및 Ethyl Acetate를 이용한 Willamette 품종 라즈베리추출물의 페놀 화합물 함량 및 항산화 효능 연구)

  • In this study, Rubus idaeus (Raspberry) cultivar 'Willamette' fruit extract(RIFE) was prepared from the freeze-dried raspberry powder, n-hexane and ethyl acetate, and then the phenolic compound content, ferric reducing ability, and radical scavenging ability were measured. The raspberry cultivar 'willamette', 'polka', and 'polana' compound fruit extract did not show cytotoxicity up to the concentration of 10%. As a result of conducting an experiment at the concentration, it was confirmed that the total phenolic compound content was 375.3 ppm, and the total flavonoid content was 43.46 ppm, and the ferric reducing ability by ferric reducing antioxidant power (FRAP) reagent was equivalent to FeSO4 0.532 mM. It was confirmed that 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging ability was 94.5 ± 0.7%, and the 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radical scavenging ability was 99.4 ± 2.82%, and the nitric oxide (NO) radical scavenging activity was 88.5 ± 0.4%. When compared with the L-ascorbic acid 'standard' solution, DPPH radical scavenging ability was between 25 - 50 ppm / ABTS radical scavenging ability was close to 100 ppm / NO radical scavenging ability was more than 1,000 ppm. These results suggest that the raspberry cultivar 'willamette' fruit extract could be applied as an effective cosmetic material with antioxidant activity.
