Differential Antioxidant Effects of Ambroxol, Rutin, Glutathione and Harmaline

  • The protective actions of ambroxol, rutin, glutathione and harmaline on oxidative damages of various tissue components were compared. The mechanisms by which they prevent oxidative tissue damages were explored. Lipid peroxidation of liver microsomes induced by combinations of $Fe^{2+}$ and ascorbate or $Fe^{+3}$, ADP and NADPH was inhibited by $50\; \muM$ of rutin, ambroxol, harmaline and glutathione. Ambroxol ($100\; \muM$) inhibited the degradation of hyaluronic acid by $Fe^{2+}$, $H_2O$_2$ and ascorbate, and it was greater than that of harmaline, whereas hyaluronic acid degradation was not prevented by rutin and glutathione. The compounds used ($100\; \muM$) did not protect the degradation of cartilage collagen by xanthine and xanthine oxidase. Rutin, glutathione and harmaline decreased the degradation of IgG by xanthine and xanthine oxidate, while ambroxol did not attenuate degradation of IgG. Glutathione showed a scavenging action on $H_2O_2$. The compounds all showed scavenging actions on hydroxyl radical. Ambroxol and harmaline exhibited quenching effects en singlet oxygen. In conclusion, ambroxol, rutin, glutathione and harmaline may exert protective effects differently on tissue components against oxidative attack depend on kind of tissue component and free radical.

  • This study was undertaken to evaluate the gastrotherapeutic effects of the extract of Rosa rugosa. It can be regarded that the antigastric and anti-ulcerative effect of Rosa rugosa is originated from the reduction of total acid output identified by gastric secretion test. To clarify the protective mechanism of the Rosa rugosa extract, the gastropathy was induced in rats with alcohol-salicylate and the activities of the free radical scavenging enzymes were examined. The acitivity of superoxide dismutase and glutathione peroxide were significantly increased and the total content of glutathione was recovered. We concluded that the protective effect of the extract of Rosa rugosa on gastropathy in rats is its ability increased the activities of the free radical scavenging enzymes.

  • Background : Toxic oxygen free radicals have been implicated as important pathologic mediators in many clinical disorders. Enhancing the intracellular content of antioxidant enzymes(superoxide dismutase, glutathione peroxidase, and catalase) can provide means of limiting biological damage caused by oxygen free radicals. The oxygen free radicals and changes of antioxidant enzymes are though to play a role in the pathogenesis of chronic obstructive pulmonary disease. Method : To investigate the pulmonary oxygen radical injury and the protective role of antioxidant enzymes in Chronic obstructive pulmonary disease(COPD), author measured the amount of thiobarbituric acid reactants, the activities of antioxidant enzymes and the sulfhydry1 groups of glutathione in serum and red blood cells from the patients with COPD(COPD patients) and the normal controls. Results : The thiobarbituric acid reactant in serum and red blood cells of COPD patients was increased than those of the normal controls, and the superoxide dismutase activity in red blood cells was no statistical difference in both groups. But the glutathione peroxidase and catalase activities in red blood cel1s of COPD patients were significantly lowered than those of the normal controls. The sulfhydry1 groups in serum and red blood cells were no statistically difference in both groups. Conclusion : These results suggest that the increased thiobarbituric acid reactants in serum and RBCs of chronic obstructive pulmonary disease mean oxygen radical toxicity, and the decreased glutathione peroxidase and catalase activities in RBC could take pan in pathogenesis of chronic obstructive pulmonary disease.

Effect of Toluene Application to the Rat Skin on the Oxygen Free Radical Metabolizing System (흰쥐에 있어서 피부조직의 Oxygen Free Radical 대사계에 미치는 Toluene의 영향)

  • To evaluate the skin toxicity oj topical toluene application, toluene (35 mg/$cm^2$) was sequentially applied to the portion rat skin for five days. The topical toluene application resulted in increased xanthine oxidase activity and CYP content, and significantly decreased superoxide dismutase and glutathione peroxidase activities at five days in rat skin. Especially catalase activity was remarkably decreased in toluene-applied rat skin. And benzylalcohol dehydrogenase activity showed also a significant decrease in toluene-applied skin. On the other hand, histopathological ultrastructural examination revealed disrupted epidermal basement membrane, rared intercellular adhensions and degenerated keratin layer due to topical toluene application. Increased deposit of cerrous perhydroxide resulted from reaction with $H_2O_2$was abserved in toluene-treated animals. These results indicate that oxygen free radical may be responsible for ultrastructural changes in skin tissue by toluene application to rat skin.

Toxicological Studies on the Essential Oil of Eugenia caryophyllata Buds

  • The essential oil (EC-oil) obtained from the buds of Eugenia caryophyllata (Myrtaceae) was examined for its free radical-scavenging activity, cytotoxicity, and in vivo toxicity. To find the xenobiotic properties of EC-oil, serum thiobarbituric acid reactive substances (TBARS) level and hepatic drug-metabolizing enzyme activities were measured. It was found that EC-oil displayed xenobiotic properties like bromobenzene. The cytotoxicities of eugenol and of the EC-oil were greatly attenuated by the sulfhydryl-containing N-acetyl-L-cysteine (NAC), suggesting that eugenol was susceptible to nucleophilic sulfhydryl. In addition, eugenol also showed potent free radical-scavenging activity in the 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay. Moreover, methyleugenol considerably exhibited less cytotoxicity and less potent free radical-scavenging activity than eugenol, and the cell viability of the methyleugenol was more increased with NAC treatment than the eugenol. These results indicate that the phenolic OH in eugenol may play a crucial role in both cytotoxicity and free radical-scavenging activity. The fashion on oxidative stress and hepatic drug-metabolizing enzyme activities of eugenol resembled those of bromobenznene.

Effect of Gam-Tea on the Metabolizing Enzyme Activity of Some Free Radical and Alcohol in Rats

  • To investigate an effect of Gam-Roa tea on the free radical or alcohol detoxicating enzyme activities, the rats got a drink at the Gam-Roa tea instead of water for 3 months, and then the animals were sacrificed and obtained the following findings. The animals receiving Gam-Roa tea showed a decreasing tendency of hepatic xanthine oxithine oxidase activity and significantly incresed content of cytochrome P-450 compared with the control. Furthermore, hepatic superoxide dismutase and glutathione S-transferase activities were also more increased in rats received Gam-Roa tea than in the control group, those receiving water. On the other hand, alcohol or aldehyde dehydrogenase activities were more increased in rats receiving Gam-Roa tea than the control. In conclusion, it is likely that the liver of rats receiving Gam-Roa tea may have the oxygen free radical or alcohol detoxication potential.

  • This study was undertaken to determine whether Juglandis Semen extraction(JS) has a protective effect against the lung cell injury caused by oxidant, t-butythydroperoxide(t-BHP) and $H_2O_2$ in rabbit lung slices. JS significantly prevented an increase in water content induced by t-BHP. Similarly, JS significantly prevented the lipid peroxidation induced by t-BHP. Cellular concentration of glutathione, and the activities of catalase and superoxide dismutase were significantly not altered by 5% JS. However, JS at 5% concentration significantly increased the glutathione peroxidase activity in oxidant-treated and control tissues. JS decreased directly the production of superoxide or hydroxyl radical. These results indicate that JS prevents the cell injury and lipid peroxidation induced by oxidants in the lung. Such an antioxidant effect is attributed to enhancement of major endogenous antioxidant defence systems such as glutathione peroxidase and direct inhibition of oxygen free radical production.

  • This study was undertaken to examine the effect of Jwagyuyeum and Woogyuycum, being known to reinforce Kidney-yin and yang, on the activities of endogenous antioxidant enzymes and the production of oxygen free radicals in the liver and kidney tissues, Alterations in enzyme activities were observed after in vivo treatment in rats, Jwagyuyeum and Woogyuyeum caused a significant increase in the activities of superoxide dismutase(SOD) and catajase Jwagyuyeum significantly increased the activity of glutathione peroxidase in both liver and kidney, but the enzyme activity was not significantly altered by Woogyuyeum. Treatment in vitro of Jwagyuyeum and Woogyuyeum decreased the production of oxygen free radicals in a dose-dependent fashion. These results suggest that Jwagyuyeum and Woogyuyeum stimulate the activities of antioxidant enzymes and inhibit directly the production of oxygen free radicals. These effects of both herbs may contribute to prevent the oxygen free radical-induced impairment of cell function.

Activities of Antioxidative Enzymes in Photobleaching of Leaves from Panax ginseng C. A. Meyer (인삼엽의 Photobleaching과 항산화효소 활성)

  • This study investigated the relations between the photosynthetic rate and the activities of antioxidatile enzymes, glutathione reductase, ascorbate free radical reductase, ascorbate peroxidase, glutathione peroxidase, and ascorbate oxidase, in the leaves of Panax ginseng. Under the normal cultivation condition, Panax in showed lower g1utathione reductase and ascorbate free radical reductase activities the Glycine max. But P ginseng showed higher 91utathione Peroxidase, ascorbate Peroxidase, and ascorbate oxidase activities than C. Panax. Therefore, P. ginseng showed weak activities of reductases for the reduction of the oxidized antioxidants. Under the light intensity of 25 KLux, the reductases showed a decrease of over 75% after 24 hours. But the peroyoxidases decreased about 40%. These results showed that the decrease of reductases acitivities was consistent with the decrease of photosynthetic rate. Therefore, we consider that the regulation of antioxidative enzymes or the application of exogenous antioxidants will be effective means for the protection of photodamage in p. ginseng.

Effect of Skin Burn on the Activities of Free Radical Generating and Scavenging Enzymes in Rat Liver (피부화상이 간의 유해산소 생성계 및 해독계 효소 활성에 미치는 영향)

  • In order to investigate a pathogenesis of liver damage induced by skin burn, thermal injury was induced by scald burn on entirely dorsal surface in rats (total body surface area 30%) except for inhalated injury. At 5 and 24 h after scald burn, biochemical assay in skin tissue, serum and liver tissue were examined. The effects of burn injury on the levels of glutathione, lipid peroxide and on the activities of oxygen of histologic and ultrasound changes, measuring. the protein concentration in plasma, and counting the number of intravascular polymorphonuclear leukocytes. Post burn 24 h, the content of glutathione was decreased (47.50%), whereas that of lipid peroxide was increased (37.01%), and the activity of superoxide dismutase was diminished (p<0.001). Thus decreasing the capacity of oxygen free radical scavenging enzymes led to oxidative injury in skin tissue. In liver tissue, at 24 h after scald burn, both the content of glutathione and the activity of catalase were markedly decreased (p<0.01, p<0.05), thus the imbalance between free radical generating and scavenging capacities has been induced.
