Studies on the cell cycle of saccharomyces cerevisiae by electron spin resonance spectroscopy (전자스핀공명스펙트럼에 의한 saccharomyces cerevisiae의 세포환 연구)

  • The intracellular free radicals produced at different stages of cell cycle of Saccharomyces cerevisiae ATCC 24858 were investigated by means of electron spin resonance(ESR) spectroscopy. The synchronized cells by repeated starvation and refeeding revealed different ESR spectral pattern compared to that of asynchronized cells. Each spectrum centered at g=2.005, which indicates free radicals. The relative spin concentration was maximat at the end of DNA increase. The variation of the relative spin concentration at each distinct stage of the cell cycle was evaluated in relation to ascorbate concentration, L-galactonolactone oxidase activity, and ascorbate oxidase activity. The highest activities of L-galactonolactone oxidase and ascorbate oxidase were detected just before and at the maximum of relative spin concentration, respectively. And ascorbate concentration fluctuated through each stage of cell cycle with the changes of relative spin concentration, L-galactonolactone oxidase activity, and ascorbate oxidase activity. Thus it is suggested that intracellular free radicals should be related to cell cycle, interacted with ascorbate, and may play an important role in the cell cycle of Saccharomyces cerevisiae.

The Taste Compounds of Fermented Oyster, Crassostrea gigas ( I ) -Changes of Free Amino Acids during the Fermentation of Oyster- (굴젓의 정미성분(呈味成分) ( I ) -굴젓숙성중(熟成中)의 유리(遊離)아미노산(酸)의 변화(變化)-)

  • Changes of free amino acids as taste compounds during the fermentation of oyster were analyzed by amino acid autoanalyzer. In fresh oyster, taurine, glutamic acid and alanine were abundant amino acids and the amounts of taurine (731mg%, on moisture and salt free base), glutamic acid (365mg%) and alanine (345.4mg% ) were 63.8% of the total free amino acids. Cystine, isoleucine, phenylalanine, leucine and histidine were detected as less abundant free amino acids and the amount of those amino. acids ranged from 5. 5mg% (cystine) to 32.9mg% (histidine). The free amino acids analyzed in this experiment were not changed in composition hut changed in amounts during 124 days of fermentation. Aspartic acid and leucine were continually increased during 124 days of fermentation. Lysine, histidine, threonine, serine, glutamic acid, tyrosine and phenyalanine were increased unlit 68 days of fermentation and than decreased gradually. The increase of arginine, glycine, valine and isoleucine were fluctuated. Taurine were dramatically decreased during the 124 days of fermentation. It is believed that glutamic acid, alanine, lecuine, serine, Iysine and threonine play an important role as taste compounds in fermented oyster because those amino acids were most abundant in fermented oyster.

  • Objective : Korean Academy of Sensory Integration(KASI) planned and executed intensive sensory integration treatment course for children with the sensory integrative dysfunction and therapists, to suggest practical guideline for therapists through individual supervision, lecture, simulation therapy, observation, case study and free play etc. Method : The course was held during 5days in the children's center for developmental support that set up sensory integration tools. The course executed the 4 individual interventions, 1 simulation therapy, 1 observation, 4 free play sessions that consisted of 6 children with sensory integrative dysfunction and 6 therapists who manage them and 3 supervisors and 2 managers. Results : Their parents reported the satisfaction degree of the treatment course service was above 84%. Therapists also had a chance to understand the broader sensory integrative interventions through the supervision and various educational programs. Conclusions : Intensive treatment course can provide parents and therapists with satisfaction of the programs. Therapists can especially experience reeducation individually through supervision and lectures.

  • The Management of Game Money in online game is considered as a crucial factor for successful game business as well as balancing game economy. The generation and flow of game money have a strong relationship with game contents, entertainment factors, game planning and level design. This paper explores online game economy in various ways especially the matter about managing & controlling game money in this context through theoretical and practical perspectives. It also examines how this management makes an effect on 'Free to Pay' policy and Sales revenue. Finally, this paper draws conclusions about the Management of Game Money which can be considered to be practically useful for game development and management. I hope that the study helps further studies and researches and can be served as a foundation regarding the issue.

A Qualitative Study of Korean Kindergarten Children's Reading Stances and Responses in Two Contexts: Official Reading Tasks and Spontaneous Reading Activities (유치원 대집단 읽기 활동과 자유 놀이 시간에서의 이야기 책에 대한 접근 관점과 그 반응에 대한 질적 연구)

  • This qualitative study explores the development of young Korean children's reading stances and responses to children's books and discusses their literacy development in relation to school reading experiences. Rosenblatt's theory (1978) of aesthetic and efferent stances is used to characterize the relationship between the reader and the text. A teacher's reading stances in the official school world are analyzed and their relationships to children's stances and responses are discussed. The teacher's efferent stance encouraged her children's responses to he efferent as well. It also influenced children's assumptions and expectations about the teacher's reading activities, so that the children came to view these activities only as ways of obtaining factual knowledge and practicing literacy skills. The children's own interests and intentions during free play time are described in order to understand children's reading stances in the unofficial school world. The children showed various types of stances during free play time even though they were encouraged to take only one type of stance, i.e., an efferent stance, in the official school world. The findings are discussed In terms of their implications for classroom practice and literacy research.

Exploring the Relationship Between Daily Routines and Physical Care Environments of Daycare Centers (어린이집 유아반 하루일과 유형과 물리적 보육환경과의 관계 탐색)

  • Objective: The purpose of the study was to explore the relationship between the daily routines and physical environments of daycare centers. Methods: A total of 20 classes at 8 different daycare centers from the metropolitan city of Daejeon were investigated. Trained observers visited each daycare center more than 3 times and collected data about daily routines using the Observational Log for Daily Routines in Daycare Centers. Also, the physical environments of each class were investigated in terms of actual size, layout of activity center, etc. Results and Conclusion: Results of the study were: first, daily routines of daycare centers could be divided into 2 distinguishable types by cluster analysis- 'free play oriented' and 'group activity oriented' Second, the amount of space each child has in a classroom differed according to the total size of the classroom. An average of 7 activity centers existed in each classroom. Third, there was a difference between free play oriented and group activity oriented groups in terms of the classroom size allocated for each child and the organization of the activity centers.

Relationships between recall, organizational strategy, and metamemory in young children (아동의 회상수행, 조직화 책략 및 상위기억간의 관계)

  • The purpose of the present research was to study developmental trends in and relationships between recall, organizational strategy, and metamemory in young children. The subjects were 84 children, 14 boys and 14 girls at each age level, 4, 6, and 8. Two tasks (memory task & metamemory task) were used to assess children's recall, organizational strategy, and metamemory based on Sodian et al.(1986). All subjects were randomly assigned either to the play-and-remember condition or to the sort-and-remember condition. The two tasks were administered to children individually with the memory task followed by the metamemory task. The data were analysed by the statistical methods of two-way ANOVA, Student-Newman-Keuls post hoc test, Mann-Whitney U test, Kruskal-Wallis test, Pearson's correlation coefficient and Kendall's Tau. Children's recall (free, conceptually-cued, and perceptually-cued) level increased with age. There were significant experimental condition differences in free recall and conceptually-cued recall, but not in perceptually-cued recall. Children's organizational strategy showed differential developmental trends by experimental condition. Use of conceptual strategy at both encoding and retrieval increased with age in both experimental conditions. Use of perceptual strategy (PS) at encoding showed an inverted-U age effect in the play-and-remember condition, but PS decreased linearly with age in the sort-and-remember condition. There were significant age differences in metamemory, and there were significant correlations between recall and organizational strategy.

  • The purpose of this study is to explore four-year old children's peer culture. During free choice play time, various activities and interaction patterns were observed and analyzed to understand the characteristics of peer culture. The results of this study showed two main peer culture patterns. First of all, peaceful peer culture in which children maintained close relationships based on mutual respect were found. Also conflictive peer culture in which children had negative relationships with other children, such as vertical, discriminative and neglected relationships were observed.

Salsolinol, a Tetrahydroisoquinoline Catechol Neurotoxin, Induces Human Cu,Zn-superoxidie Dismutase Modificaiton

  • The endogenous neurotoxin, 1-methyl-6,7-dihydroxy-1,2,3,4-tetrahydroisoquinoline (salsolinol), has been considered a potential causative factor for the pathogenesis of Parkinson's disease (PD). In the present study, we examined the pattern of human Cu,Zn-superoxide dismutase (SOD) modification elicited by salsolinol. When Cu,Zn-SOD was incubated with salsolinol, some protein fragmentation and some higher molecular weight aggregates were occurred. Salsolinol led to inactivation of Cu,Zn-SOD in a concentration-dependent manner. Free radical scavengers and catalase inhibited the salsolinol-mediated Cu,Zn-SOD modificaiton. Exposure of Cu,Zn-SOD to salsolinol led also to the generation of protein carbonyl compounds. The deoxyribose assay showed that hydroxyl radicals were generated during the oxidation of salsolinol in the presence of Cu,Zn-SOD. Therefore, the results indicate that free radical may play a role in the modification and inactivation of Cu,Zn-SOD by salsolinol.

Acrolein, the toxic endogenous aldehyde, induces neurofilament-L aggregation

  • Acrolein is a highly reactive by product of lipid peroxidation and individuals with neurodegenerative disorders have been shown to contain elevated concentrations of this molecule in the brain. In the present study, we examined the pattern of neurofilament-L (NF-L) modification elicited by acrolein. When NF-L was incubated with acrolein, protein aggregation occurred in a acrolein concentration-dependent manner. Exposure of NF-L to acrolein also led to the generation of protein carbonyl compounds. Through the addition of free radical scavengers we observed a significant decrease in acrolein-mediated NF-L aggregation. These results indicate that free radicals may be involved in the modification of NF-L by acrolein. In addition, dityrosine crosslink formation was observed in acrolein-mediated NF-L aggregates and these aggregates displayed thioflavin T reactivity, reminiscent of amyloid. This study suggests that acrolein-mediated NF-L aggregation might be closely related to oxidative reactions, thus these reactions may play a critical role in neuro-degenerative diseases.
