Inhibitory Effect of Capsaicin against Carcinogen-induced Oxidative Damage in Rats

  • Capsaicin (trans-8-methyl-N-vanillyl-6-nonenarnide), a major pungent component of hot pepper, is known to exert antioxidative properties. In this study, we investigated the protective effects of capsaicin against chemical carcinogen-induced oxidative damage in rats. Male Sprague Dawley rats weighting 230~250 g were treated with chemical carcinogens such as 2-nitropropane (2NP) or n-methyl-N'-nitro-N-nitrosoguanidine (MNNG) after (or before) the administration of capsaicin at doses of 0.5, 1,5 mg/kg. The level of lipid peroxidation in rat liver was estimated by measuring the amounts of thiobarbituric acid reactive substances. The degree of oxidative DNA damage was evacuated by measuring a DNA adduct, 8-hydroxydeoxyguanosine (8-OHdG), in urine. Antioxidative activities of capsaicin and its metabolites in vitro were determined by the measurement of DPPH (1,1-diphenyl-2-picrylhydrazyl), a radical quencher. Significant inhibition of 2-NP induced lipid peroxidation was observed in the liver of the rat when treated with capsaicin. MNNG-induced urinary excretion of 8-OHdG was decreased by capsaicin treatment. Capsaicin and its metabolites inhibited net only the formation of free radicals, but also lipid peroxidation in vitro. Our results show that capsaicin may function as a free radical scavenger against chemical carcinogen-induced oxidative cellular damage in vivo. The observed antioxidative activities of capsaicin may play an important role in the process of chemoprevention.

  • Effects of Taeumjowetang on Lipid Peroxidation by Free Radicals and Oxidative Damage of Hepatocytes by tert-Butyl Hydroperoxide. 1. Purpose The present study was carried out to evaluate the antioxidant effects of Taeumjowetang in vitro. 2. Methods In this study, antioxidant effects of TJT on lipid peroxidation were determined according to the method of TBA. (Abbreviation) TJT : Taeumjowetang, TBA : 2-thiobarbituric acid. 3. Results : 1) TJT inhibited markedly peroxidation of linoleic acid during the autoxidation. 2) TJT inhibited lipid peroxidation induced by hydroxyl radical derived from H2O2-Fe2+ in rat liver homogenate. 3) TJT showed 66% scavenging effect on DPPH radical. 4) TJT exhibited a 25% inhibitory effect on superoxide generation from xanthine-xan thine oxidase system. 5) To investigate the antioxidative effects of TJT on the hepatocytes, cultured normal rat liver cells(Ac2F) were prepared and incubated with or without TJT. After 16~18hr, cells placed in DMEM medium without serum, and then incubated with 1mM t-BHP for 2hr. Viable cells were detected by MTT assay. In this test, TJT protected the cell death induced by t-BHP and significantly increased cell viability in the normal rat liver cell. Induced freezing tolerance and free amino acids perturbation of spinach by exogenous proline

  • The objective of this study was to investigate whether exogenous proline (Pro) could confer freezing tolerance of spinach and determine fluctuations of free amino acids in spinach leaf tissues under freeze-induced stress. Treatment with Pro (10 mM) resulted in more accumulation of Pro (~2.6-fold) in Pro-treated spinaches compared to untreated ones. These Pro-pretreated spinaches were more freezing-tolerant, showing more turgid leaves and petioles compared to untreated controls. However, when spinaches pre-treated with or without Pro were subjected to freezing, there was no significant difference in overall amino acid contents, emphasizing the role of Pro as an osmoprotectant. Freezing stress prompted intensification of total amino acid contents irrespective of pretreatment with Pro. Asp, Glu, Ala, and Val were the most abundant free amino acids due to increased protein degradation and nitrogen mobilization for plant survival under freezing stress. Arg, a precursor for the synthesis of polyamines in plants, was profoundly enhanced under freezing stress. This implies that Arg plays an important role in modulating freezing tolerance. Gly, Leu, and Ile were maintained at relatively low levels in all treatments. However, Ser, Tyr, and Lys as primary constituents of dehydrins were accumulated under freezing stress, suggesting that they might play a role in increasing cryoprotective activity under freezing stress.

Barrier-Free Subway Service System Scenario : Comparison Between Korea and China (한국과 중국의 비교를 통한 무장애 지하철 서비스시스템 시나리오 제안)

  • In the global era, the tourism industry is a major profitable business, and the state and local governments are making various efforts to provide quality tourism experiences. The purpose of this study in a variety of tourism contents is to study barrier-free subway services in line with the global era, and to expand from the existing rapid, safe, and ordered transportation to the provision of high-quality comfort and all-round service experiences. This study compared and analyzed the subway service systems of Korea, China and both countries through the service design method, and presented a barrier-free subway service system to improve the user's satisfaction with the subway service system by improving the user's service experience. As a result, research results showed that 20 attractive quality attributes in 17 fields, such as convenience facilities, language issues, security equipment, and riding environment, play an important role in improving the quality and experience of subway services. In addition, through the construction of a Service Spatial Scenario, an optimized subway service system can be visualized to help understand this so that it can be used as a reference for creating a strategic application.

Serum levels of free insulin-like growth factor-I and clinical value in healthy children (한국 소아 및 청소년에서 혈중 유리 insulin-like growth factor-I 농도치와 임상적 의의)

  • Purpose : The serum levels of total insulin-like growth factor (IGF)-I and IGF binding protein (IGFBP)-3 reflect endogenous growth hormone (GH) secretion in healthy children. Free form of IGF-I which is suggested to have more potent biological action than complex form of IGF-I. The aim of this study is to investigate the serum levels of free IGF-I and its clinical value in healthy children. Methods : Serum levels of total IGF-I and IGFBP-3 were determined in 494 healthy children (248 boys and 246 girls) by RIA and IRMA. Serum level of free IGF-I was determined in 206 healthy children (103 boys and 103 girls) by IRMA. Results : The free IGF-I level increased with age in both sex. The free IGF-I level increased continuously between 7 and 15 years of age in boys, but decrement was noted after 14 years of age in girls. Serum total IGF-I level also increased with age in similar pattern of that of free IGF-I. There were no significant differences of mean values of the ratio of free IGF-I/total IGF-I in relation to age in both sex. And there were significant correlations between the level of free IGF-I and total IGF-I and the ratio of total IGF-I/IGFBP-3, respectively. Conclusion : In healthy children, serum free IGF-I increased with age in both sex and high free IGF-I level may play an important role in pubertal growth spurt. Our results suggest that the increased serum free IGF-I level in puberty may reflect changes in total IGF-I rather than IGFBP-3. But free IGF-I does not have more clinical value than total IGF-I because of no significant differences of mean values of the ratio of free IGF-I/total IGF-I in relation to age.

The Intention during the Interaction between Infants and Their Teachers while Playing Freely (자유놀이에 나타난 영아와 교사의 상호작용 의도)

  • The purpose of this study is to clarify what teachers interaction is who try to develop their infants, by examining the intention of beginning the interaction between teachers and their infants, while playing freely at a child care center. The author used a qualitative research method. The subjects of this study are nine infants and three teachers. The author videotaped all the session focusing on the interactions between teachers and their infants. The author observed all the sessions for three months. The author also interviewed the teachers. She also collected the manual of the target daycare centers, its study plans, its observation logs, and the observation logs of the infants' activities and analyzed them. The intention of infants' interaction with their teachers is to know how to play by using some toys. A second intention is to see the contents of the picture book related to the daily routine. A third intention is to satisfy emotional needs. The intention of teachers interaction with their infants while playing freely is to help explore things and try to play. Another intention is to make a transition from teacher-centered play to infant-centered one. A third intention is to pacify infants expressing thoughts by crying. A future research which will increase the quality of the interactions and will be helpful for the development of infants and will focus on infants will be needed.

The characters of lichens attached to the stonework cultural assets and removing methods of them (석조문화재에 부착하는 지의류의 특성과 그 제거방법)

  • Lichens play an important role in breaking down rocks physically and chemically. Physical weathering occurs mechanically through hyphae and rhizines growing into the rocks. Lichen substances such as oxalic acid, acidic polysaccharides and depsides may assist altering rocks chemically. These actions partly contribute to soil formation but prove harmful for the substratum. Some people do not like to keep lichens on stained glassof churches or on gravestones mainly because of its unglamorous appearance. A biocide, 'Koretrel', is very effective in removing lichens and blue-green algae from the surface of concrete. After spraying 'Koretrel' in the open air, lichens (Caloplaca sp., Endocarpon petrolepideum, Physciella melanchra, Phaeophyscia hispidula, etc.) growing on buildings died within three months and peeled off fromthe substratum. One year after treatment, concrete surfaces were almost lichen free for three or four years. The biocide makesuse of a kind of lichen substance and harmless both for substances and environment. Our recent experiments show that it is also useful for water grasses growing on the surface of an aqueduct of a hydroelectric power station.

Thymectomy in Myasthenia Gravis: 2 cases report (흉선절제에 의한 근무력증 치료 2례 보고)

  • Myasthenia gravis is a neuromuscular disorder manifested by fatigability and weakness of voluntary muscles. The basic defect in the myasthenia is reduction of available acetylcholine receptors at neuromuscuiar junctions by an autoimmune attack. Removal of the thymus gland now play an Important role in the management of this disease. We have two experiences of thymectomy for myasthenic patients. The 31-year-old housewife(J.H) was admitted to the Taegu Presbyterian Medical Center because of bilateral ptosis, mastication and swallowing difficulties. The presence of thymoma was strongly suggested by roentgenographic studies. A 99.8 gm thymoma was removed completely by total thymectomy. On the 2nd postoperative day respiratory failure developed. Tracheostomy was performed and ventilatory assist was given for 3 days. The patient was completely recovered with antichollestrase drug only. C.N. was 17-year-old girl who has the symptoms of bilateral ptosis and diplopia for 3 years. Tensilon test was positive and antichollnestrase was given for several months, but the patient showed no improvement. After thymectomy she was free from myasthenic symptoms with out anticholinestrase drug.

Finite Element Modeling of Fracture Process Zone in Concrete (콘크리트 파괴진행영역의 유한요소모델링)

  • Fracture Mechanics does work for concrete, provided that a finite nonlinear zone at fracture front is being considered. The development of model for fracture process zone is most important to describe fracture phenomena in concrete. The fracture process zone is a region ahead of a traction-free crack, in which two major mechanisms, microcracking and bridging, play important rules. The toughness due to bridging is dominant compared to toughness induced by microcracking, so that the bridging is dominant mechanism governing the fracture process of concrete. In this paper the bridging zone, which is a part of extended macrocrack with stresses transmitted by aggregates in concrete, is model led by a Dugdale-Barenblatt type model with linear tension-softening curve. Two finite element techniques are shown for the model of fracture process zone in concrete.

  • Since carbon-fluorine (C-F) bonds play a key role to improve bioavailability and lipophilicity, they have found commonly in pharmaceuticals, radiopharmaceuticals, agrochemicals, and material science. Advances on the efficient method to introduce fluorine to complex organic molecules are mainly results of development of fluorination reagents and transition metal catalysts. In this mini-review, we want to emphasize two representative nucleophilic fluorinating reagents regarding carbon-fluorine bond formation, which were developed in the mid-2000s.
