아메바의 cytolytic process가 주로 가수분해 효소 환성과 관계 깊다는 연구들이 70년대 후반부터 병원성 Entamoeba sp.와 Naegleria sp.에서 보고되었지만 Acanthamoeba sp.에서는 가수분해 효소 환성에 관한 연구는 찾아볼 수 없다. 본 연구에서는 병원성이 없는 것으로 알려진 Acanthamoeba royreba를 대조로 하여 병원성이 있는 A. culbertsonl이 가수분해 효소 활성도를 측정하고 이들을 비교하고자 하였다. A. culbertsoni를 마우스에 감염시켰더니 15일내에 마우스 모두가 사망하였으며, 평균 생존기간은 8.5일이었고, 아베바성 수막뇌염으로 사망했음을 알 수 있었다. 반면에 A. royreba를 감염시킨 마우스에서는 수막뇌염이 발생되지 않았다. CGV배지에서 두 종(種) 아메바를 배양하여 아메바 추출물 및 배양액에서 가수분해 효소의 환성을 측정한 결과 A. rcyreba에 비해 A. culbertsoni 세포의 추출물 및 배양액에서 현저히 활성도가 높은 가수분해 효소는 acid phosphatase, p-N-acetyl galactosaminidase, β-N-acetyl glucosaminidase, α-mannosidase, neutral proteinase, acid proteinase였다. 표적 세포인 CHO 세포에 혼합한 A. culbertsoni는 대조군에 비해 강한 세포독성을 나타내었고 CHO 세포와의 혼합후 48시간 후 80%, 72시간후 95% 이상의 CHO 세포가 사멸하는 것을 알 수 있었다. 반면에 A. royreba를 혼합하였을 때는 별다른 세포독성을 나타나지 않았다. CHO 세포에 혼합한 A. tulbgrtsoni이 가수분해 효소 활성도를 경과 시간별로 측정하였던 바 CGV 배지에서 배 양시간이 지남에 따라 아메바내보다 그 배양액에서 더 높은 효소 활성도를 나타내었던 acid phosphatase, β-N·acetyl galactosaminidase, β-N·acetyl glucosaminidase, α-mannosidase, acid proteinase는 혼합 120시간 동안 아메바내에서 더 높은 효소 활성도를 나타내었으며, neutral proteinase의 활성도는 경과시간이 지남에 따라 아메바내에서는 일정하였지만 그 배양액인 EBSS용액내에서는 점점 높아짐을 알 수 있었다. 이상의 성적으로 보아 마우스에서 수막뇌염을 일으키는 A. cuzbertsoni는 비 병원성인 A. reyreba와 비교할 때 여러 종류 가수분해 효소들의 활성도에 현저한 차이를 나타내고 있음을 알 수 있었다. 또한 표적세포인 CHO세포에 대한 A. cuzbertsoni의 세포독성은 강함을 알 수 있었고, 이 때 몇 종류 가수분해 효소들의 환성도도 높게 나타났으며, 이러한 소견이 A. culbertsoni의 병원성과 관련이 있을 것으로 추측되었다.
Acanthnmoeba sp. YM-4의 영양형은 사상의 침상위족을 내어 전형적인 각canthamoeba 속의 특징이 관찰되었으며, 포낭(씨스트)은 외벽이 등근혐이었고 내벽이 단순한 구형이거나 다소 불규칙한 2개의 세포벽을 갖고 있었다. SDS-PAGE 결과, Acanthamoeha sp. Yhf-4는 약 16개의 주요 단백질 분획이 관찰되었는데, 2차원 전기영동 결과와 함께 Acanthamoeba sp. YM-4의 단백질 분획은 A. culbertsoni와 거의 같은 양상이 관찰되었다. Acanthamoeba sp. YM-4로 면역시킨 마우스의 비장세포와 myeloma 세포를 융합한 결과, 항체를 분비하는 17개 clone을 얻을 수 있었다. 생성된 단세포를 항체 중 isotyping을 실시하였는데 실험에 사용된 McAY 6, McAY 7, McAY 8, McAY 13, McAY 16 단세포군은 IgGl 항체를, McAY 10, McAY 11 단세포군은 19M 항체를 분비하였다. 생성된 단세포군 항체는 대부분이 아메바의 세포막에 있는 항원과 반응하였으며, EITB 실험결과, McAY 7 단세포군 항체는 43 kD에서 반응대가 관찰되었고, McAY 10 단세포군 항체는 55 kD과 105 kD에서 반응대가 관찰되었다. ELISA 방법을 이용한 단세포를 항체들과 여러 아메바 간의 교차반응실험 결과, McAY 7 단세포군 항체는 Acanthamoeba sp. YM-4에만 반응하였고, McAY 6과 McAY 10은 A. culbertsoni와도 반응하였다. 또한 McAY 11은 모든 아매바와 교차반응을 보였다.
Pathogenic Naegleria fowleri, Acanthamoeba castellanii, and Acanthamoeba polyphaga, are distributed worldwide. They are causative agents of primary amoebic meningoencephalitis or acanthamoebic keratitis in humans, respectively. Trophozoites encyst in unfavorable environments, such as exhausted food supply and desiccation. Until recently, the method of N. fowleri encystation used solid non-nutrient agar medium supplemented with heat-inactivated Escherichia coli; however, for the amoebic encystment of Acanthamoeba spp., a defined, slightly modified liquid media is used. In this study, in order to generate pure N. fowleri cysts, a liquid encystment medium (buffer 1) modified from Page's amoeba saline was applied for encystation of N. fowleri. N. fowleri cysts were well induced after 24 hr with the above defined liquid encystment medium (buffer 1). This was confirmed by observation of a high expression of differential mRNA of nfa1 and actin genes in trophozoites. Thus, this liquid medium can replace the earlier non-nutrient agar medium for obtaining pure N. fowleri cysts. In addition, for cyst formation of Acanthamoeba spp., buffer 2 (adjusted to pH 9.0) was the more efficient medium. To summarize, these liquid encystment media may be useful for further studies which require axenic and pure amoebic cysts.
Naegleria fowleri is a free-living amoeba causing primary amoebic encephalitis. In this study, we analyzed how the N-aceytlglucosamine (GlcNAc) and D-galactose affected the interaction between Naegleria fowleri and methicillin-resistant Staphylococcus aureus (MRSA) or Escherichia coli O157:H7, and the interaction with bacteria when monosaccharides were treated with N. fowleri for a longer pre-incubation time. When GlcNAc was treated with N. fowleri for 1 hr, the E. coli association was almost the same as that of the control not treated with GlcNAc until the concentration of GlcNAc was 25 mM. However, the E. coli association was reduced by approximately 91% with 100 mM GlcNAc. E. coli invasion into N. fowleri showed statistical significance only in the group treated with 100 mM GlcNAc. The interaction when treated with galactose showed a very different pattern in the 50 mM galactose group than when treated with GlcNAc. In the MRSA interaction, a statistically significant decrease in association (76.3% by GlcNAc and 88.7% by galactose) and invasion (3.6% by GlcNAc and 9.3% by galactose) was shown by the concentration of two 100 mM monosaccharides. The group treated with monosaccharides at the same time showed almost no difference in all interactions from the group treated with monosaccharides at the same time. Taken together, it suggested that the effect of monosaccharides on the interaction of several Gram-negative or positive bacteria and the evidence that the interaction could be enhanced by longer pre-incubation time.
Background: Free-living amoebae (FLA) are widely distributed in freshwater, seawater, soil, and extreme environments, and play a critical role as feeders on diverse preys in the ecosystem. Also, some FLA can become opportunistic pathogens in animals including humans. The taxa Amoebozoa and Heterolobosea are important amoeboid groups associated with human pathogens. However, the identification and habitat of amoebae belonging to Amoebozoa and Heterolobosea remain poorly reported in the Republic of Korea. This study highlights the first record for identification and source of four amoebae including putative pathogens in the Republic of Korea. Results: In the present study, four previously reported FLA were isolated from freshwaters in Sangju Gonggeomji Reservoir (strain GO001), one of the largest reservoirs during the Joseon Dynasty period, and along the Nakdong River, the largest river in the Republic of Korea (strains NR5-2, NR12-1, and NR14-1) for the first time. Microscopic observations and 18S rDNA phylogenetic trees revealed the four isolated strains to be Acanthamoeba polyphaga (strains NR5-2 and NR12-1), Tetramitus waccamawensis (strain GO001), and Naegleria australiensis (strain NR14-1). Strains NR5-2 and NR12-1 might be the same species and belonged to the morphological Group 2 and the T4 genotype of Acanthamoeba. Strain GO001 formed a clade with T. waccamawensis in 18S rDNA phylogeny, and showed morphological characteristics similar to previously recorded strains, although the species' flagellate form was not observed. Strain NR14-1 had the typical morphology of Naegleria and formed a strongly supported clade with previously recorded strains of N. australiensis in phylogenetic analysis of 18S rDNA sequences. Conclusions: On the bases of morphological and molecular analyses, four strains of FLA were newly observed and classified in the Republic of Korea. Three strains belonging to the two species (A. polyphaga and N. australiensis) isolated from the Nakdong River have the potential to act as opportunistic pathogens that can cause fatal diseases (i.e. granulomatous amoebic encephalitis, Acanthamoeba Keratitis, and meningoencephalitis) in animals including humans. The Nakdong River in the Republic of Korea may provide a habitat for potentially pathogenic amoebae, but additional research is required to confirm the true pathogenicity of these FLA now known in the Republic of Korea.
장기간 시험관내에서 무균적으로 배양하여 병원성이 약화된 자유생활아메바 naegleria fowleri를 마우스에 연속적으로 감염시켜 그 병원성의 증감여부를 관찰하고자 하였다. 체증 18∼22gm의 백색 마우스를 secobarbital로 마취시키고 오른쪽 비강에 Naegleria fowleri를 떨어뜨려 감염시켰다. 시험관내에서 7년이상 CGVS배지에서 계대배양된 0주와 마우스에 감염시켜 뇌조직을 2번 통과시킨 2-1주의 병원성을 비교하였다. 행동둔화, 자극에 민감, 회전운동, 사지마비등 여ㅓ 증상이 나타났고, 뇌의 오른쪽 앞쪽 부위에 심한 염증 및 괴사를 발견하였다. 이러한 증상과 병소는 0주 감염군보다 2-1감염군에서 보다 빨리, 보다 심하게 발생되었음을 관찰하였다. 감염 7일 후부터 감염된 마우스가 사망하기 시작하였으며 2-1주 감염군에서 0주 감염군 보다 생존기간이 짧았으며 감염 13일 후부터 사망한 예에 있어서는 육안적으로 뇌에서 병소가 없었고 폐장에서 염증이 심하였음을 관찰하였다. 마우스에 비강을 Naegleria fowleri를 감염시켜 전형적인 원발성 아메바성 뇌수막염을 발생시킬 수 있었다. 장기간 시험관에서 무균적으로 계대배양된 Naegleria fowleri는 병원성이 약화되어 있었고 이 아메바를 마우스에 연속적으로 감염시켜 뇌조직을 통과시킴으로 다시 병원성이 증강됨을 관찰하였다.
Naegleria fowleri로 면역시킨 마우스의 혈청이나 비장세포를 다른 마우스에 주입시켜 N. fowleri 감염에 대한 면역효과가 나타나는지 살펴보았다. 또 어미 마우스를 N. fowleri로 면역시 킨 다음 면역시키지 않은 어미에서 태어난 새끼 마우스에게 면역시킨 마우스의 모유를 먹게한 뒤 모유를 통한 면역 효과의 전달 여부를 관찰하였다. 면역시킨 마우스의 비장세포나 정상 마우스의 혈청을 다른 마우스에 주입시킨 경우 감염 후 사망률이 감소하였으나 혈중 IgG는 증가되지 않았다. 한편, 면역시킨 마우스의 혈청이나 정상 마우스의 비장세포를 준 경우 혈중 보G가 증가되고 감염 후 생존 기간은 연장되었으나 결국은 모든 마우스가 사망하였다. 즉 면역시킨 마우스 또는 정상 마우스의 혈청이나 비장세포를 다른 마우스에 주입시키면 사망률이 감소되거나 생존 기간이 연장됨을 알 수 있었다. 또 면역시킨 어미 마우스의 혈중, 모유 내, 또는 그 모유를 먹은 새끼 마우스의 혈 중에서, N. fowleri에 대한 IgG는 증가되었으나 IgA는 증가하지 않았으며 증가된 항체도 감염 후 의 사망률이나 생존 기간에는 큰 영향을 주지 않았다.
병원성 자유생활아메바인 Aconaamoebonensoni로 면역된 어미 마우스로부터 태어난 새끼 마우스를 다시 면역시켰을 때, 어미로부터 받은 수동면역이 새끼 마우스의 능동면역 시행시 어떠한 영향을 미치는지를 관찰하고자 수막뇌염 발생에 따른 사망률, 혈청내 항체가 측정 및 세포 분리분석기를 이용하여 T세포 아형을 측정하였다. A. culbertsoni 영양형 $1{\;}{\times}{\;}10^6$개로 3회 면역된 어미 마우스로부터 수동면역을 전달받은 2-3주령의 새끼 마우스와 수동면역을 받지 않은 2-3 주령의 새끼 마우스를 영양형 $1{\;}{\times}{\;}10^6$개로 2회 면역하였을 때 혈청내 항체가는 같은 주령의 면역 을 시키지 않은 대조군에 비해 유의하게 높았다. 수동면역을 받은 군과 받지 않은 군간의 혈청내 항체가는 유의한 차이가 없었다. 영양형 $1{\;}{\times}{\;}10^4$개를 감염시켰을 때 사망률을 관찰한 결과 수동면역을 전달받은 군은 면역을 받지 않은 군과는 유의한 차이가 없었으나 수동면역을 받지 않은 군보다는 유의하게 높았다. T세포의 아형 중, $Ly2^{+}$ T세포는 어미로부터 수동면역을 전달받은 군이 수동면역을 받지 않은 군이나 면역을 시키지 않은 군에 비해 더 높은 비율로 통계적으로 유의하게 증가하였다. 수동면역을 받은 부비주령의 새끼 마우스를 같은 방법으로 면역한 군과 대조군을 비교힌 결과, 영양형 $1{\;}{\times}{\;}10^5$개로 감염시켰을 때 사망률은 차이가 없었다. 따라서 면역된 어미로부터 태어난 새끼를 다시 능동면역하였을 때, 능동면역에 따른 항체가의 변동은 관찰할 수 없었으나 억제 T세포는 증가하였으며, 능동면역 시기에 따라 면역조절이 다르게 발현됨을 알 수 있었다.
원발성 아메바성 수막 뇌염을 일으키는 병원성이 강한 자유 생활 아메바인 Naegleria fowleri이 영양형을 마우스 비강에 접종시킨 후 12시간, 1일, 2일, 3일 및 5일째에 비장 세포의 자연살세포 독성을 측정하였다. 51Cr 방출 경사법과 단세포 독성 검사에 의하여 표적 세포 결합능, 활성 자연살세포, Vmaz 및 재순환능을 측정하였다. 감염 마우스의 사망률은 접종 후 20일까지 76.7%이였으며 사망한 마우스의 평균 생존 기간은 12.87±0.71일이었다. 자연살세포의 독성은 아메바 접종후 12시간째에 대조군에 비하여 유의하게 증가하기 시작하여 접종후 1일째 가장 증가하였다. 표적 세로 결합능간에는 경과 기간별로 유의한 차이가 없었으며 환성 자연살세포와 재순환능은 자연살세포의 독성이 증가함에 따라 증가하였다. N. fowleri가 감염되었을 때 자연살세포의 활성에 미치는 영향을 보면 자연살세포의 독성이 증가함에 따라 환성 자연살세포 및 재순환능이 증가하였고, 표적 세포 결합능에는 영향을 미치지 않는 것을 알 수 있었다.
비장적출된 BALB/c 마우스에 동종항원으로 면역접종하였을 때 병원성 자유생활 아메바, 즉 점천간출a fowzeri o 359의 감염에 의한 사망률, 생존기간을 항체생성과 관련하여 관찰함으로써 비 장적출이 숙주의 체액성 면역기전에 어떤 영향을 미치는지 관찰하였다. 실험 I군은 비장을 적출하고 면역후 감염, II군은 비장적출후 감염, 여군은 면역후 감염, 사군은 대조군으로서 감염만시킨 군 등으로 구분하여 5~10${\times}$105 아메바를 1주 간격으로 2회 복강내에 주입하여 면역접종하였으며 2회 접종 1주 후에 5~10${\times}$104 아메바를 비강내에 감염시켜 감염 31일까지 관찰하였다. 감염후 사망률은 면역된 I 및 III군에서 각각 38.1% 및 25%로 II군 50%나 IV군 46.4%에 비해 낮았다. 사망한 마우스의 평균 생존기간은 I군 $20.1{\pm}3.6$일, II군 $17.3{\pm}4.5$일, III군 $20.4{\pm}7.0$일 및 비군 $19.6{\pm}7.6$일로 각 실험군간에 차이가 없었다. 감염 31일 후 혈중 IgG 항체가(ELISA치)는 면역군인 I군 및 III군에서 각각 $1.10{\pm}0.29$ 및 $1.31{\pm}0.28$로 대조군의 $0.24{\pm}0.37$보다 훨씬 높았다 (p<0.05). 이러한 성적으로 보아 비장적출 마우스에서 면역후 N. fowleri를 감염시켰을 때 마우스 생존기간의 차이는 없으나 사망률은 면역시킨 군에서 낮아 비장적출후에도 체액성 면역 이 방어기전으로 존재하는 것으로 생각되었다.

제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
