BACKGROUND/OBJECTIVES: Atopic dermatitis (AD), a chronic inflammatory skin disease, is accompanied by disruption of the epidermal lipid barrier, of which ceramide (Cer) is the major component. Recently it was reported that vitamin C is essential for de novo synthesis of Cer in the epidermis and that the level of vitamin C in plasma is decreased in AD. The objective of this study was to determine the associations among clinical severity, vitamin C in either plasma or epidermis, and Cer in the epidermis of patients with AD. SUBJECTS/METHODS: A total of 17 patients (11 male and 6 female) aged 20-42 years were enrolled. The clinical severity of AD was assessed according to the SCORAD (SCORing Atopic Dermatitis) system. Levels of vitamin C were determined in plasma and biopsies of lesional epidermis. Levels of epidermal lipids, including Cer, were determined from tape-stripped lesional epidermis. RESULTS: The clinical severity of patients ranged between 0.1 and 45 (mild to severe AD) based on the SCORAD system. As the SCORAD score increased, the level of vitamin C in the plasma, but not in the epidermis, decreased, and levels of total Cer and Cer2, the major Cer species in the epidermis, also decreased. There was also a positive association between level of vitamin C in the plasma and level of total Cer in the epidermis. However, levels of epidermal total lipids including triglyceride, cholesterol, and free fatty acid (FFA) were not associated with either SCORAD score or level of vitamin C in the plasma of all subjects. CONCLUSIONS: As the clinical severity of AD increased, level of vitamin C in the plasma and level of epidermal Cer decreased, and there was a positive association between these two parameters, implying associations among plasma vitamin C, epidermal Cer, and the clinical severity of AD.
The study was designed to observe the effect of complete listing for 10 days on body fatness, serum leptin level and lipid profile in thirty women who were admitted to fasting Clinical Program in Kyung-Hee Oriental Medical Center. Most of the subjects had food habits of over-eating and preferred oily foods which was correlated to the increase of body weight and BMI during realimentation. Average loss of body weight was 7.98kg and waist-hip ratio was significantly decreased during fasting. Body weight showed significant positive correlation with body fat(r=0.8182), lean body mass(r=0.7797) and total body water(r=0.7563). Body fat was gradually reduced after 9 days fasting, but lean body mass was not diminished any more after 7 days luring. Serum leptin level was significantly decreased after luring and re-feeding. The leptin showed significant positive correlation with body weight(r=0.9324), body mass index(r=0.8279) and body fat(r=0.8376). The levels of serum total cholesterol and LDL-C were significantly increased but HDL-C level was significantly decreased after fasting which increased could be harmful for persons with hypercholesterolemia. VLDL-C and triglyceride levels were not significantly decreased after fasting. Serum total free fatty acid level was significantly increased after fasting. These data suggest that the best way of maintaining the reduced body weight was to carry out the modification of food habits and behavior in daily lift. It could not be recommended to repeat often complete luting for reduction of body weight. (Korean J Nutrition 33(1) : 42-48, 2000)
We investigated the effects of short-term food restriction and repeated fasting and refeeding on appetite regulating hormones and adiponectin activity in rats. To investigate the acute and chronic effects of food restriction in vivo, Sprague-Dawley rats were divided into a control group (CON), a 1 day fasting group, a 2 days fasting gruop, a 3 days fasting gruop, a fasting and refeeding for 1 week gruoup and a fasting and refeeding for 2 weeks group. Blood glucose, triglyceride and total cholesterol decreased in all fasting groups compared to those in the CON group. Free fatty acid of all fasting groups was higher than those in the CON, and were lowest in the three cycle fasting and refeeding group. Blood insulin following short-term food restriction was lower than that in the CON. blood ghrelin increased significantly (p < 0.01) following the short-term food restriction, However, blood ghrelin in the repeated fasting and refeeding groups decreased significantly decreased (p < 0.01) compared to that in the CON and short-term food restriction group. In contrast, blood leptin decreased significantly (p < 0.01) in the short term food restriction group and the three cycle of fasting and refeeding group but increased in the six cycle of fasting and refeeding group. No significant differences in adiponectin contents were observed in the short-term food restriction group. But, adiponectin increased significantly (p < 0.01) following the fasting and refeeding cycles. Blood adiponectin and blood leptin levels were showed positively correlated ($r^2$ = 0.469) when all samples were analysed together.
To replace the traditional drying method with improving the qualities of dried squid, warm air drying method was investigated comparing with natural drying method. In respect of drying rate, about 12 hrs were taken to obtain 25% moisture content -which was considered as proper moisture content to control microbial and quality degradation- by warm air drying at 35$^{\circ}C$. However, 120 hrs were taken to obtain such moisture content by natural drying. The squid dried by warm air showed little color difference and was seemed to be raw squid meat color. TBA values were more rapidly increased but final values were lower, inversely. Free amino acid contents were higher, and cholesterol content was lower in warm air drying.
Background: Breast cancer is one of the most frequent cancer types within female populations. Silibinin is a chemotherapeutic agent ative against cancer. Niosomes are biodegradable, biocompatible, safe and effective carriers for drug delivery. Objective:To prepare nanoniosomal silibinin and evaluate its cytotoxicity inthe T-47D breast cancer cell line. Materials and Methods: Niosomes were prepared by reverse phase evaporation of a mixture of span 20, silibinin, PEG-2000 and cholesterol in chloroform and methanol solvent (1:2 v/v). The solvent phase was evaporated using a rotary evaporator and the remaining gel phase was hydrated in phosphate buffer saline. Mean size, size distribution and zeta potential of niosomes were measured with a Zetasizer instrument and then nanoparticles underwent scanning electron microscopy. The drug releasing pattern was evaluated by dialysis and the cytotoxicity of nanoniosomes in T-47D cells was assessed by MTT assay. Results: Particle size, size variation and zeta potential of the niosomal nanoparticles were measured as $178.4{\pm}5.4nm$, $0.38{\pm}0.09$ and $-15.3{\pm}1.3mV$, respectively. The amount of encapsulated drug and the level of drug loading were determined $98.6{\pm}2.7%$ and $22.3{\pm}1.8%$, respectively; released drug was estimated about $18.6{\pm}2.5%$ after 37 hours. The cytotoxic effects of nanoniosome were significantly increased when compared with the free drug. Conclusions: This study finding suggests that silibinin nanoniosomes could serve as a new drug formulation for breast cancer therapy.
The present study was aimed to investigate the effects of the application of pulsed electromagnetic fields on the blood components associated hyperlipidemia, antioxidant enzymes and reactive oxygen. The subjects were divided into three groups: General Diet, High fat Diet, High fat diet and pulsed electromagnetic fields. Pulsed electromagnetic field was applied with pulsed electromagnetic energy therapy equipment. The Glucose, free fatty acids, triglycerides, cholesterol and insulin was used to measure hyperlipidemia-related blood components, and the GSH, GRD, XO and MDA was used to measure antioxidant enzymes and reactive oxygen. The applied pulsed electromagnetic field has improved the concentrations of blood components and increasing GSH and GRD, that were decreased due to a high-fat diet and by reducing the increased MDA and XO to a level of general diet group. Therefore, the applied pulsed electromagnetic field can be the effective treatment for changes in the blood composition of hyperlipidemic rats, enhancement of antioxidative activity and the inhibition of reactive oxygen.
In this study, the effects of feeding citrus by-products on the quality and fatty acid compositions of chicken eggs were investigated. Two samples of chicken eggs were used for this study: T0 (eggs of chickens that were not fed citrus byproducts) and T1 (eggs of chickens that were fed 4% citrus byproducts). There was no statistically meaningful difference between T0 and T1 in terms of various properties, such as chemical composition, cholesterol content, minerals, vitamins, pigments, and total structural amino acid. The caloric content of the egg yolk was higher in T1 than in T0, and total free amino acid of egg albumin and egg yolk was higher in T1 than in T0 (p<0.05). There was a not significant difference in SFA:UFA ratio between T0 and T1, but the linolenic acid content was higher in T1 than in T0 (p<0.05). The results of this study show that citrus by-products can be used for feed for laying hens by adding them to general feed.

Purpose: To examine the effects of a 36-week resistance training program on body composition, fitness and blood lipid profiles in the obese elderly. Method: fourteen subjects were assigned into the resistance training group. The 12-16 exercise sessions were performed for 60min with 15-25 repetitions twice per week for 36 weeks. Data were analyzed by paired t-test with SPSS version 12.0 for Windows. Results: Significant changes were evident in weight (t=5.983, p<.001), body mass index (t=5.922, p<.001), % body fat (t=3.480, p=.004), and fat free mass (t=-2.592, p=.022). Concerning exercises, no significance was noted for back scratch (t=-1.814, p=.093), but were for 2-min step (t=-3.075, p=.009), arm curl (t=-2.223, p=.045), chair stand (t=-3.671, p=.003), chair sit and reach (t=-2.508, p=.026), and 8-ft up and go (t=5.545, p<.001). No significant changes were evident in total cholesterol (t=1.189, p=.279), and total glucose (t=0.689, p=.517), while the change in high-density lipoprotein was significant (t=3.535, p=.012). Conclusion: The 36-week resistance training program enhanced body composition and physical fitness, but not blood lipid profiles in obese elderly subjects.
Objective: The aim was to study experimentally the effects of Dioscorea bulbifera L. induced by sodium levothyroxine. Methods: Sprague-Dawley rats were separated into four groups. Except for the normal group, the other three were treated with sodium levothyroxine $160{\mu}g/kg$ for 5 days. Among the three groups, two (except the control) were treated with Dioscorea bulbifera L. extract of 0.75g/kg (sample A) or 1.5g/kg (sample B). $T_3-uptake,\;T_3,\;T_4$, TSH, total cholesterol, glucose, ALP, AST, ALT, tree fatty acids, $\beta-lipoprotein$, and change in the serum of rats were measured after medication with solid extract of Radix Scrophulariae. Results: $T_4$ concentration and $T_3-uptake$ level decreased in sample A and B compared to controls. $T_3$ concentration decreased but not statistically meaningful. Considering lipid metabolism, only free fatty acid level was significantly reduced glucose level also significantly decreased in sample B. Serum AST, ALT and ALP levels were slightly elevated but there was no meaning statistically, which doesn't mean Dioscorea bulbifera L. has liver toxicity because the quantity of the extract perhaps couldn't reach the minimal dose causing liver toxicity, which according to an article was 60g. Body weight significantly increased in comparison to controls. Conclusion: These results suggest that Dioscorea bulbiftra L. lowers excess thyroid hormone and increased metabolism, resulting in improvement of hyperthyroid state.

In this study, the effects of Gamgung-tang gamibang on the hyperthyroidism induced by the intraperitoneal injection of 3,5,3-triiodothyronine was examined by the measurement of physical changes, body weight, the volume of food intake and rectal temperature, and heart weight, heart beat, blood pressure with contrast to propranolol, one of beta-blocking agents. the obtained results were as follows. The Gamgung-tang gamibang extract showed to inhibit the decrease of body weight and rectal temperature, and decrease the food intake, so the inhibitory effects of Gamgung-tang gamibang extract on the experimental hyperthyroidism were exhibited. The Gamgung-tang gamibang extract showed the inhibitory effects on the circulatory functions changed and enhanced by the experimental induced hyperthyroidism, the action of Gamgung-tang gamibang extract was less effective than the propranolol of D-CONT group. The Gamgung-tang gamibang extract showed significant effects to inhibit the concentration of serum thyroid houmone, more effective than the propranolol, beta-blocking agents. The Gamgung-tang gamibang extract showed the effective inhibitory reaction on the biochemical changes in serum, cholesterol, ketone bodies, free fatty acid, glucose in hyperthyroid rats induced by 3,5,3-triiodothyronine.
