항산화 비타민과 Aflatoxin$B_1$의 혼합 투여가 마우스간의 지질 함량 및 지방산 조성에 미치는 영향 (The Effects of Aflatoxin $B_1$Co-administrated with Antioxidant Vitamins on Lipid Contents and Fatty Acids Composition of Liver in Mice)

  • Aflatoxin은 자연속에 존재하며 높은 독성을 가진 곰팡이 독소이다. 이들 곰팡이 독소는 Aspergillus flavus와 Aspergillus parasiticus로부터 생성되는 대사산물로서 식품에서 다양한 오염 경로를 가지고 발생한다. 특히 Aflatoxin B$_1$(AFB$_1$)은 사람에게 있어서 간독성 및 간암을 유발하는 잠재력을 가진 곰팡이 독소이며, 지질과산화 반응은 aflatoxin B$_1$에 의한 세포 산화적 손상시 발생하는 주요 현상중의 하나이다. 따라서 본 연구의 목적은 aflatoxin B$_1$과 항산화 비타민 혼합 투여 후 유도된 간과 혈청의 지질 구성 성분 및 지방산 조성에 미치는 영향을 조사하기 위하여 수행되었다. 특히 비타민 C와 비타민 E는 중요한 항산화제에 속하는 것으로 이들을 복강내 주사(i.p, intraperitoneal injection)를 통하여 각각 10 mg/kg, 63.8 mg/kg씩 male ICR mice에 투여하였으며 1시간후 0.4mg/kg의 AFB$_1$을 동일한 방법으로 투여하였다. AFB$_1$에 의하여 유도된 지질구성 성분의 변화에 있어서 AFB$_1$, 단독 투여군과 항산화 비타민 혼합 투여군을 비교하였을 때 혈청에서의 유리콜레스테롤, 중성지질, 총 콜레스테롤등은 AFB$_1$, 단독 투여군에서 2배 이상 증가되었다. 간에서의 지방산 조성에서도 AFB$_1$, 단독 투여군에서 항산화비타민 혼합투여군보다 콜레스테롤 에스테르, 중성지질, 총콜레스테롤 및 인지질의 P/S ratio가 감소하였다. 이상의 결과로 미루어 볼 때 AFB$_1$에 의한 세포손상이 항산화 비타민에 의해 감소되는 효과가 있는 것을 확인할 수 있었다.

Streptozotocin으로 유발한 당뇨 흰쥐의 혈당 및 혈청 지질함량에 미치는 가시오가피 추출물의 영향 (Effect of Acanthopanax senticosus Extracts on Blood Sugar and Serum Lipid Profiles of Streptozotocin-Induced Diabetic Rats)

  • Effects of Acanthopanax senticosus extract (AS) on blood sugar content and serum lipid profiles of streptozotocin-induced diabetic rats were investigated. Experimental groups were classified into four groups, that is, normal control (NC) group, diabetic mellitus (DM) group, AS-fed group and DMAS-fed group. The AS group showed lower feed efficiency than the NC group, but the efficiency of DMAS group was higher than DM group. DMAS group showed the decreased water intake and urine by $45.5\%$ and $23.7\%$ respectively, compared with DM group. Compared with DM group, DMAS group decreased blood sugar by $46.9\%$ and triglyceride by $17.8\%$, total cholesterol by $10.0\%$ and LDL cholesterol by $22.0\%$ in serum, but increased serum HDL cholesterol by $14.4\%$ The relative percentage of liver or kidney per body weight, and the serum ALT activity in DMAS group were lower than those of DM group. There were no significant differences in hepatic glutathione(GSH) contents and total xanthine oxidase(XOD) activities among experimental groups. The hepatic lipid peroxide(LPO) content in DMAS group decreased by $54.6\%$ compared with that in DM group. The XOD (O type) and the ratio of O type to total type of both STZ-treated groups (DM and DMAS) were higher than those of NC group, but less conversion of D to O type was observed in DMAS group than in DM group. There was no significant difference in GST activity between NC and AS, but STZ-treated groups showed lower glutathione S-transferase(GST) activity than NC. In conclusion, it seems that AS reduces blood sugar by inhibiting the activity of xanthine oxidase type O as an oxygen-free radical generating system which induces the tissue damage. Antidiabetic effect of AS may regulate diabetes-induced high lipid profiles in blood.

열결(列缺).려구에 대한 용량별 He-Ne 레이저 침 조사(照射)가 고지방(高脂肪) 식이(食餌)로 유발된 고지혈증(高脂血症) 백서(白鼠)에 미치는 영향(影響) (Effects of Invasive Low Level Laser Acupuncture Therapy(LLLAT) depending on capacity at LU7+LR5 on the Hyperlipemia Rats Induced by High Fat Diet)

  • Objectives : This research was performed to investigate the effect of invasive low level laser acupuncture therapy(LLLAT) at Yolgyol+yogu(LU7+LR5) on weight gain, food intake, food efficiency, lipid metabolism and liver function in hyperlipemia rats. Methods : Experimental groups were divided into high fat diet cup(Control group), high fat diet and 15 mW/5 min LLLAT at LU7+LR5(15 mW/5 min group), 15 mW/10 min LLLAT at LU7+LR5(15 mW/10 min group), 30 mW/5 min LLLAT at LU7+LRS(30 mW/5 min group), 30 mW/10 min LLLAT at LU7+LR5(30 mW/10 min group). LLLAT was treated at the 15,30mW-5,10min once a 2day during 5 weeks. Results : Body weight was decreased significantly in 30 mW/s min group and 30 mW/10 min group compared with control group. Food intake was decreased significantly in 15 mW/5 min group and 15 mW/10 min group compared with control group. Food efficiency was decreased significantly in 30 mW/10 min group compared with control group. In the lipid metabolism, total cholesterol, HDL-cholesterol and free fatty acid were increased significantly in 15 mW/5 min group and 30 mW/10 min group compared with control group. LDL-cholesterol was increased in 30 mW/10 min group and phospholipids was decreased significantly in all experimental group compared with control group. triglyceride was not showed significant result in all experimental group. In the liver function, AST was not showed significant result in all experimental group, ALT and ALP were increased significantly in 30 mW/10 min group, ALP was decreased in 30 mW/5 min group compared with control group. Conclusions : LLLAT at LU7 and LR5 can manage hyperlipemia by controlling body weight, food intake, food efficiency ratio and lipid metabolism.

톳(Hijikia fusiforme) 추출물이 고지혈증 횐쥐의 지질대사 및 간 효소활성에 미치는 영향 (Effects of Hijikia fusiforme Extracts on Lipid Metabolism and Liver Antioxidative Enzyme Activities in Triton-Induced Hyperlipidemic Rats)

  • 본 연구는 용매별 톳 추출물이 고지혈증을 유발한 횐쥐의 혈청과 간의 지질 및 항 산화 효소활성을 측정하기 위하여 체중이 평균 100 g 되는 Sprague-Dawley계 수컷 횐쥐를 7마리씩 7군으로 나누어 1군은 정상군, 2군 이하 7군까지는 용매별 톳 추출물을 체중 kg당 0.5 mg의 용량을 복강으로 일주일간 주사한 다음 마지막 날 체중 kg당 triton 0.5 mg을 복강으로 투여하여 고지혈증을 유발하여 나타난 결과는 다음과 같다. 혈청 및 간 중 유리 콜레스테롤함량은 핵산층 투여군이 대조군에 비해 유의적으로 낮게 나타난 반면 HDL-콜레스테롤 함량은 대조군에 비해 에탄올층과 핵산층 투여군이 유의적으로높았다. 혈청 및 간의 총 콜레스테롤 함량의 경우 대조군에 비해 수층 투여군이 유의적으로 낮게 나타났으며, 특히 간 은 에탄올층 투여군과 에틸아세테이트층 투여군이 대조군에 비해 낮게 나타났다. 수층 투여군의 혈청 중 중성지방함량은 대조군에 비하여 에탄올층 투여군과 핵산층 투여군이 유의하게 낮게 나타났다. 간 중 TBARS 함량은 대조군에 비해 수용성, 비수용성 톳 분회물 투여군의 TBARS함량이 모두 낮게 나타났다. 간 중 SOD의 활성도는 대조군에 대해 톳 수층 투여군이 낮은 경향이었고, 비수용성 대조군에 비해 핵산층 투여군이 유의적으로 낮은 경향을 보였다. 간의 catalase 활성도의 경우 정상군에 비해 대조군과 분획물 투여군이 높은 경향을 보였으나 에틸아세테이트층 투여군은 정상군 수준 이하로 떨어졌다. 이러한 결과로 볼 때 톨은 생체내 항 고지혈증, 항 고콜레스테롤 및 항 산화효과가 있음을 알 수 있었다.

Yoga Training Improves Metabolic Parameters in Obese Boys

  • Yoga has been known to have stimulatory or inhibitory effects on the metabolic parameters and to be uncomplicated therapy for obesity. The purpose of the present study was to test the effect of an 8-week of yoga-asana training on body composition, lipid profile, and insulin resistance (IR) in obese adolescent boys. Twenty volunteers with body mass index (BMI) greater than the 95th percentile were randomly assigned to yoga (age $14.7{\pm}0.5$ years, n=10) and control groups (age $14.6{\pm}1.0$ years, n=10). The yoga group performed exercises three times per week at 40~60% of heart-rate reserve (HRR) for 8 weeks. IR was determined with the homeostasis model assessment of insulin resistance (HOMA-IR). After yoga training, body weight, BMI, fat mass (FM), and body fat % (BF %) were significantly decreased, and fat-free mass and basal metabolic rate were significantly increased than baseline values. FM and BF % were significantly improved in the yoga group compared with the control group (p<0.05). Total cholesterol (TC) was significantly decreased in the yoga group (p<0.01). HDL-cholesterol was decreased in both groups (p<0.05). No significant changes were observed between or within groups for triglycerides, LDL-cholesterol, glucose, insulin, and HOMA-IR. Our findings show that an 8-week of yoga training improves body composition and TC levels in obese adolescent boys, suggesting that yoga training may be effective in controlling some metabolic syndrome factors in obese adolescent boys.

알코올성 간독성 흰쥐의 혈청 지질 및 조직 과산화지질 농도에 미치는 발효당귀의 영향 (Effects of Monascus-fermented Angelica gigas Nakai on the Contents of Serum Lipid and Tissue Lipid Peroxidation in Alcohol Feeding Rats)

  • Monascus purpureus 균주에 의해 발효된 당귀분말을 식이 중에 5% 수준으로 첨가하여 30% 알코올과 함께 4주간 흰쥐에 급여한 후 혈중 지질 농도 및 조직 내 산화 스트레스에 미치는 영향에 대하여 검토하였다. 혈중 중성지질 농도 및 유리 지방산 농도는 알코올 대조군(Alc)에서의 증가가 알코올 실험군에서 모두 감소하였고, 특히 발효당귀군(AFAG)에서 낮은 감소수치를 보였다. 혈중 총 콜레스테롤 농도는 전반적으로 유의적인 차이는 없었으나, HDL-cholesterol 농도에서 발효당귀군(AFAG)이 증가수치를 나타내었다. 간장, 심장, 비장,신장, 고환 및 혈청에서 생체막 과산화지질 생성 정도는 정상군(N)에 비해 알코올 대조군(Alc)에서 유의적으로 증가하였고, 이러한 증가는 발효당귀군(AFAG)과 양성 대조군(ASM)에서 정상 수준으로 회복되었다. 간 조직 내 내인성 항산화 물질인 glutathione 농도는 알코올 대조군(Alc)에서 유의적으로 감소하였고, 발효당귀군(AFAG)에서 유의적으로 증가하여, 과산화 지질 생성정도와 glutathione 농도는 유의적인 차이가 인정되었다. 이상의 실험결과 발효당귀는 알코올 투여 동물실험에서 혈중 지질 개선효과는 물론 조직 내 항산화 활성증가로 산화스트레스를 경감시키는 효능이 있는 것으로 사료되며, 향후 간 독성 개선에 효능을 가지는 건강식품 개발 가능성이 높은 소재로 판단되어진다.

고지방 식이내 식이섬유질이 흰쥐의 레티놀 결합 단백질 (RBP, cRBP I, cRBP II) 유전자 발현에 미치는 영향 (Effect of Dietary Fibers on Retinol Binding Protein (RBP, cRBP I, cRBP II) Gene Expression in Rats Fed High Fat Diet)

  • Recently it has been reported that vitamin A and retinol binding proteins (RBPs) in blood and urine were changed in the condition of diabetes mellitus or hyperlipidemia. Fruits and vegetables are recommended to consume for the people suffered from these chronic degenerative diseases. The main components of fruits and vegetables are dietary fibers, for example cellulose and pectin, of which function to affect the absorption and excretion of dietary fat and fat-soluble substances. This study was conducted to investigate the effect of dietary fibers on RBPs mRNA expression in liver, small intestine and serum of rat fed high fat diet during 4 weeks. Sprague-Dawley rats, weighing 121g on average, were divided into four groups; (Control; $17\%$ fat & cellulose supplement diet, HF0: $25\%$ fat & fiber free diet, B:.Uc: $25\%$ fat & cellulose supplement diet and HF0: $25\%$ fat & pectin supplement diet) . The rats fed high fat diet groups (HF0, HFC, HFP) tended to consume the food less than the control group, but FER of HF0 groups was significantly higher than the control (p < 0.05) . The weight of adrenal gland in high fat diet groups (HF0, HFC, HFP) was significantly less than the control. Total lipid in feces daily excreted and in liver did not show any significant differences among the groups. Total cholesterol in HFP group was significantly different from that of HFC group. Serum total cholesterol and triglyceride in other group tended to lower than other groups and HDL cholesterol higher. Consequently, AI (atherogenic index) was the lowest in HFP group. Vit A contents in feces daily excreted tended to lower in high fat diet groups (HF0, HFP) compared to the control group. That content in adrenal gland was the lowest in HF0 group, but not in liver. In HFP group were down-regulated cRBPI mRNA in liver and cRBPII mRNA in small intestine and up-regulated RBP and transthyretin expression in serum compared to the other groups. In conclusion, dietary fibers, especially pectin, in high fat diet might down-regulate the expression of CRBP I, CRBP II mRNA in liver and small intestine, but increase the secretion of RBP into serum and therefore inhance the bioavailability of Vit A through the body. Cocoa: a functional food that decreases insulin resistance and oxidative damage in young adults with class II obesity

  • BACKGROUND/OBJECTIVES: Cocoa consumption is associated with health benefits due to its high content of polyphenols. However, the effects of short-term cocoa consumption remain unclear. We aimed to determine the effects generated by cocoa consumption (for 7 days) in young adults in normoweight and class II obesity. SUBJECTS/METHODS: Before-and-after study was carried out in normoweight (NW) (n = 15) and class II obesity (CIIO) (n = 15) young adults. The NW and CIIO participants consumed 25 and 39 g of cocoa, respectively, per day for 7 days. The effect of cocoa consumption was evaluated on the lipid profile, insulin resistance (IR), and inflammation. Oxidative damage was also examined by assessing the biomarkers of oxidative damage in plasma. In addition, recombinant human insulin was incubated with blood obtained from the participants, and the molecular damage to the hormone was analyzed. RESULTS: Cocoa consumption resulted in decreased low-density lipoprotein-cholesterol in both groups (P = 0.04), while the total cholesterol, high-density lipoprotein cholesterol, and triglycerides were maintained at the recommended levels. Initially, IR was detected in the CIIO group (homeostasis model assessment [HOMA] = 4.78 ± 0.4), which is associated with molecular damage to insulin. Interestingly, intervention with cocoa resulted in improved IR (HOMA = 3.14 ± 0.31) (P = 0.0018) as well as molecular damage to insulin. Finally, cocoa consumption significant decreased the arginase activity (P = 0.0249) in the CIIO group; this is a critical enzymatic activity in the inflammatory process associated with obesity. CONCLUSIONS: The short-term consumption of cocoa improves the lipid profile, exerts anti-inflammatory effects, and protects against oxidative damage. Results of this study indicate that cocoa consumption can potentially improve IR and restore a healthy redox status.

체지방 감소에 대한 털부처꽃 추출물의 효과: 무작위배정, 이중눈가림, 대조군 비교 인체적용시험 프로토콜 (Effect of Lythrum salicaria Extract on Body Fat Reduction: A Protocol for a Randomized, Double-Blinded, Placebo-Controlled Clinical Trial)

  • Objectives: Obesity is a globally prevalent public health issue. Hence, there is a need for the development of safer and more effective anti-obesity drugs. Lythrum salicaria, a traditional medicinal herb used for centuries, has been reported to improve lipid metabolism and fat accumulation. It also has a low toxicity profile. Therefore, its potential as a functional ingredient in health functional foods needs to be evaluated. Methods: In this randomized, double-blind, placebo-controlled clinical trial, 90 participants will be randomly assigned to either the experimental or control group. Each subject will orally receive L. salicaria extract (1,350 mg/day) (500 mg L. salicaria+850 mg lactose as vehicle) or lactose (1,350 mg/day) as a hard capsule formula for 84 days (12 weeks). The primary outcome will be body fat mass (kg), which will be assessed using dual-energy x-ray absorptiometry (DXA) (performed only at visits 2 and 4). Secondary outcomes include body mass index, body weight, waist-to-hip ratio, body fat percentage (%) measured using DXA, lean body mass (kg) measured using DXA (assessed only at visits 2 and 4), lipids (total cholesterol, triglyceride, high-density lipoprotein cholesterol, and calculated low-density lipoprotein cholesterol), free fatty acid, high sensitivity C-reactive protein, adiponectin, and leptin. Conclusions: This protocol will be implemented after approval of Institutional Review Board of Pusan National University Korean Medicine Hospital (approval number: PNUKHIRB-2022-08-002) and registration with the Korean National Clinical Research Information Service (CRIS) (CRIS-KCT0008060). The results of this trial will provide potential of L. salicaria as a new anti-obesity functional food with fat-reducing effects and low toxicity.

Caffeine, 철분 및 vitamin E 혼합투여시 rat의 혈액과 간조직내에서 혈액화학성분과 지질 및 단백질 구성성분의 변화 (Changes of the blood chemistry, lipid and protein components in blood and liver tissue of the rat after oral combined administration of caffeine, iron and vitamin E)

  • This study was conducted to identify the effects of caffeine or combinations of caffeine and iron or vitamin E on the lipid and protein components or blood chemistry levels of the serum as well as the total homogenate, mitochondrial and microsomal fraction of the rat(Sprague-Dawley, female) liver. Chronic test were conducted to determine those effects. The chronic test was conducted by dividing rats into 5 groups according to the type of drugs and dosages administrated as follows; the control(group A), and group B was given 25mg/kg caffeine orally once daily for 30 days, group C was given 50mg/kg caffeine orally once daily for 30 days, group D was given 25mg/kg caffeine and orally ferric chloride once daily for 30 days and group E was given 25mg/kg caffeine and 25mg/kg vitamin E once daily for 30 days. The concentrations of glucose, urea nitrogen, uric acid, creatinine, total protein, albumin, A/G ratio, triglyceride, total cholesterol, HDL-cholesterol, free fatty acid, phospholipid as well as the activities of alanine aminotransferase(ALT), aspartate aminotransferase(AST) and alkaline phosphatase(ALP) were measured in the serum of each experimental groups. The concentrations of the carbonyl group and malondiaidehyde(MDA) and the patterns of the SDS-PAGE(Sodium Dodecyl Sulfate - Polyacrylamide Gel Electrophoresis) and fatty acid compositions in free fatty acids and phospholipids were analyzed to determine the oxidative damages and metabolic changes on the lipid and protein components in the serum, and total homogenate, mitochondrial and microsomal fractions of the rat liver. The results obtained from this study were summarized as follows; 1. Body weights of groups B, C, D and E were significantly decreased(p < 0.01) in comparison with that of the control in the chronic test. 2. The concentrations of serum glucose in groups B(124.5mg/dl), C(130.1mg/dl), D(122.1mg/dl), E(119.3mg/dl) were significantly higher(p < 0.01) in comparison to that of the control(101.5mg/dl). But, there were no significant differences in the concentrations of urea nitrogen, uric acid, creatinine, total protein, albumin and A/G ratio in comparison to that of the control. 3. The concentrations of total cholesterol and HDL-cholesterol in serum of groups B(69.6, 53.4mg/dl), C(73.0, 56.3mg/dl), D(68.9, 51.1mg/dl) and E(68.2, 51.3mg/dl) were significantly higher(p < 0.01) in comparison to that of the control(52.6, 38.8mg/dl). On the other hand, the concentrations of triglyceride in serum of groups B(45.0mg/dl), C(40.4mg/dl), D(33.8mg/dl) and E(47.2mg/dl) were significantly lower(p < 0.01) in comparison to that of the control(66.2mg/dl). There were no significant differences in the activities of ALT, AST and ALP in comparison to that of the control. 4. The concentrations of free fatty acid and phospholipid in serum of groups B(45.7, 154.4mg/dl), C(50.0, 167.2mg/dl), D(52.5, 148.4mg/dl) and E(41.1, 159.2mg/dl) were higher(p < 0.01) in comparison to that of the control(35.2, 125.3mg/dl). And the concentrations of the carbonyl group and malondialdehyde in serum of group D(1.82, 0.52nM/mg protein) were significantly higher(p < 0.01) in comparison to the control(1.53nM/mg protein). 5. The concentrations of carbonyl group in total homogenate, mitochondrial and microsomal fraction of group D(1.45, 0.94, 1.67nM/mg protein) were significantly higher (p < 0.01) in comparison to the control(1.16, 0.66, 1.27nM/mg protein). And the concentrations of malondialdehyde in the total homogenate, mitochondrial and microsomal fraction of group D(6.70, 6.10, 1.36nM/mg protein) were significantly higher(p < 0.01) in comparison to the control(5.17, 3.64, 0.68nM/mg protein). 6. As the analytical results of the fatty acid compositions of free fatty acid in serum, the proportions of stearic acid and arachidonic acid of groups B(16.52, 12.62%), C(17.52, 15.18%), D(19.73, 13.47%) and E(17.62, 13.28%) were significantly higher(p < 0.01) in comparison to the control(14.75, 7.88%), but the proportions of oleic acid and linoleic acid of groups B(12.97, 32.59%), C(10.88, 31.23%), D(12.37, 30.66%) and E(11.95, 32.41%) were significantly lower(p < 0.01) in comparison to the control(16.44, 35.12%). And the proportions of palmitic acid of group D(25.95**, 26.16, 26.34**%) were significantly higher(**:p < 0.01) in comparison to the control(24.43, 25.42, 23.34%). In addition, the proportions of linoleic acid of group D(23.43, 25.02, 23.95%) were also significantly higher(p < 0.01) in comparison to the control(22.17, 23.75, 21.26%). The proportions of stearic acid of group D(19.87, 19.76**%) in mitochondrial and microsomal fraction were lower(**:p < 0.01) in comparison to the control(21.01, 24.18%), and the proportions of stearic acid of group E(16.71*, 19.65**%) in mitochondrial and microsomal fraction were significantly lower(**:p < 0.01, *:p < 0.05) in comparison to the control(21.01, 24.18%), and the proportions of linoleic acid of group E(25.04, 29.20, 26.48%) in total homogenate, mitochondria and microsome were significantly higher(p < 0.01) in comparison to the control(22.17, 23.75, 21.26%). 8. As the results of the fatty acid compositions of phospholipid in total homogenate, mitochondrial and microsomal fraction of liver, the proportions of palmitic acid of group D(17.58**, 18.78*, 18.23%**) were significantly higher(**:p < 0.01, *:p < 0.05) in comparison to the control(16.28, 17.22, 16.38%), and the proportions of stearic acid of group D(36.41, 37.23, 39.53%) were also significantly higher(p < 0.01) in comparison to the control(34.18, 34.16, 36.04%). But the proportions of oleic acid(3.41*, 3.11**, 3.12**%) and linoleic acid (18.03**, 15.79**, 14.74**%) of group D were significantly lower(**:p < 0.01, *:p < 0.05) in comparison to the control(oleic : 3.63, 3.72, 3.79%, linoleic : 20.03, 18.71, 18.48%). 9. In order to determine the oxidative damages to the protein in serum, mitochondrial and microsomal fraction of the rat liver, the patterns of the SDS-PAGE were identified, but the results of SDS-PAGE were not significantly different between the control and experimental groups.
