Characteristics of Microbial Distribution of Nitrifiers and Nitrogen Removal in Membrane Bioreactor by Fluorescence in situ Hybridization (막/생물반응기에서 Fluorescence in situ Hybridization 기법을 이용한 질산화 미생물 분포특성 및 질소제거 연구)

  • An aerobic submerged membrane bioreactor (MBR) treating ammonium wastewater was studied in respect of nitrification characteristics and distribution of nitrification bacteria over a period of 350 days. MBR was fed with ammonium concentration of 500-1000 mg $NH_4-N/L$ at a nitrogen load of $1-2kg\;N/m^3{\cdot}d$. Overall ammonium oxidation rate increased with dissolved oxygen (DO) concentration, temperature, and sludge retention time (SRT). Under a higher concentration of free ammonia ($NH_3-N$) due to the decrease of ammonium oxidation rate, the nitrite ratio ($NO_2-N/NO_x-N$) in the effluent increased. The sudden collapse of nitrification efficiency accompanied by sludge foaming and the increase of sludge volume index (SVI) was observed unexpectedly during the operation. At the later stage of operation, additional carbon source was fed to the MBR and resulted in twice higher value of SVI and the decrease of ammonium oxidation rate. In fluorescence in situ hybridization (FISH) analysis, genus Nitrosomonas which is specifically hybridized with probe NSM156 was initially the dominant ammonia oxidizing bacteria and the amount of Nitrosospira gradually increased. Nitrospira was the dominant nitrite oxidizing bacteria during whole operational period. Significant amount of Nitrobacter was also detected which might due to the high concentration of nitrite maintained in the reactor.

  • Even though shotcrete is a main support for securing the stability of tunnel, the performance of shotcrete is not properly checked due to various difficulties arisen from the characteristics of materials themselves which constitute shotcrete, such as steel fibers and accelerators, and the on-site quality control. In this study, the actual conditions of shotcrete applied to expressway tunnel construction sites were tried to find out, and then some improvement was tried to derive. For this purpose, the characteristics of steel fibers and accelerators supplied to the expressway construction sites were investigated. Also, shotcrete specimens were prepared at the tunnel sites and performance tests were carried out. For steel fiber, domestic production states were investigated, and carbon content and tensile strength were measured using the steel fibers collected in the construction sites. For three types of accelerators such as aluminate, cement mineral and alkali-free, basic properties and total amount of alkali contents were analyzed. Shotcrete specimens were prepared using on-site shotcrete machine with regard to mix designs and types of accelerators. Using these specimens, uniaxial compression tests and flexural tests were performed. As the results, compressive strength, flexural strength, flexural toughness, and etc. were compared with types of acclerators and mix designs.

Antioxidative Activity and Produced Condition of Antioxidative Substance by Bacillus sp. FF-7 (Bacillus sp. FF-7에 의한 항산화물질 생산조건과 항산화 활성)

  • The antioxidative activity of antioxidative substances produced from several bacterial strains isolated from fermented foods were tested by $DPPH\;({\alpha},{\alpha}'-diphenyl-{\beta}-picrylhydrazyl)$ free radical scavenging activity. One of the strains showing the highest antioxidative activity was identified as Bacillus sp. based on the morphological, biochemical, physiological characteristics, and 16S rRNA sequence, and named FF-7. The most optimal medium condition for the production of antioxidative substance from Bacillus sp. FF-7 was 2% galactose as carbon source and l% tryptone as nitrogen source. The antioxidative substance produced from FF-7 in these cultural medium was also tested by in vitro experimental models, the peruxidation of linoleic acid and the peroxidation of rat tissues microsomes by using thiobarbituric acid (TBA) for assay of free malondialdehyde production. The antioxidative activity against lipid peroxidation of rat tissues microsomes was shown in the following order; brain 97.50% > heart 79.95% > kidney 77.84% > spleen 77.47% > testis 69.96% > liver 62.45%. The antioxidative substance produced from FF-7 on linoleic acid peroxidation by IBA method was effectively inhibited during four days, and 0.05% BHT (butylated hydroxytoluene) used comparative control was also effectively inhibited. Results showed that the highest antioxidative activity by DPPH method of antioxidative substance produced from Bacillus sp. FF-7 was obtained by supplementing 2% galactose as carton source and l% tryptone as nitrogen source in cultured medium, this substance effectively inhibited the formation of TBARS in brain microsome in vit개 system and in linoleic acid peroxidation.

  • In order to utilize the agricultural waste products, two strains of mold producing powerful cellulolytic enzyme were sereened from various soils, composts, rotten wood and others. The optimum condition of cellulase production was studied. The results obtained were summarized as follows. 1. Two strains of mold which showed remarkably high cellulolytic activity were identified as Aspergillus niger-SM 6 and Trichoderma viride-SM 10. 2. The highest cellulase production was obtained at pH 5.0-6.0 in 5 days. 3. Cellulase production in strain Aspergillus niger-SM 6 increased with the addition of C.M.C., $(NH_4)_2SO_4$, C.S.L., orange peel powder and rice hull. The rice hull, treated with 3N NaOH at $120^{\circ}C$ for 15 min. and neutralized with various acids, was used. Up to 50% of wheat bran could be substituted by the treated rice hull without any decrease of cellulase activity. 4. In the strain of Trichoderma viride-SM 10, cellulase production increased with the addition of C.M.C., $NH_4NO_3$, Vitamin-free casamino acid and orange peel powder, while the other carbon, nitrogen, phosphate sources, natural nutrients and organic substances gave no remarkable effect.

  • The physiological and chemical investigations to explain the after-ripening processes in got green pepper fruit were carried out by treating the fruit with ethephon either alone or with phenylalanine. The studied metabolic changes in fruit during after-ripening period was carbon dioxide and oxygen concentration in interior of the pepper fruit, total carotenoid, ${\beta}-carotene$, total sugar, and free reducing sugar in pepper fruit. These metabolic changes were explained inrelation to the color enhancement judged by the color score to explain the after-ripening processes. Ethephon treatment at 500 ppm significantly accelerated color enhancement as compared to the control and further ethephon treatment increased the number by 20 percent which was not possible in control during same after-ripening period. The oxygen concentration in interior of the pepper fruit during after-ripening period was increased in control when the color score (color enhancement) increased rapidly. However, with ethephon treatment, the oxygen concentration was decreased when the color score increased. Although total and free reducing sugar content were decreased during the after-ripening period total carotenoid and ${\beta}-carotene$ content was increased by 50 and 200 percent, respectively, over control. Also the capsaicin contents was increased by 20 percent by ethephon treatment over control. Although phenylalanine treatment did not affect the capsaicin content, capsaicin content tended to be increased by phenylalanine treatment when treated with ethephon.   • Chemical weathering of forest soils can reduce atmospheric $CO_2$ concentration over geologic time scales, providing many essential elements for life. Although many studies have been conducted on the effects of elevated atmospheric $CO_2$ on forest carbon storage using open top chambers and FACE (Free air $CO_2$ enrichment) facilities since the 1990s, studies on chemical weathering of forest soils under elevated $CO_2$ are relatively rare. Here I review on how elevated atmospheric $CO_2$ can affect the chemical weathering of forest soils and suggest directions on future research. Despite the recent advances in chemical weathering of forest soils under elevated atmospheric $CO_2$, it is still not clear how the large volume of forest soils would react under the condition. Future studies on weathering of forest soils covering large areas from the tropics to the polar regions with carefully monitored pre-treatment data would provide key information on how soils, the Earth's life sustaining engine, change under climate change.

The Hepatoprotective Effects of Epimedii Herba through the Antioxidation (음양곽의 항산화작용에 의한 간 보호 효과)

  • In this study, the effect of Epimedii Herba (EH) on the antioxidative enzymatic activity was investigated. EH (100mg/kg) was intraperitoneally administered into rats for 2 weeks. On the last day, carbon tetrachloride $(50\%\;CCl_4,\;3.3ml/kg,\;i.p.)$ dissolved in olive oil was injected before 12 hours. EH-pread-ministered and $CCl_4-treated$ (EC) group showed higher inhibitory effect in aminotransferase (AST, ALT) activity compared to $CCl_4-treated (CT)$ group. Superoxide dismutase (SOD) and Catalase in EC group were increased compared to those of CT group. The activity of glutathione peroxidase (GPx) was significantly higher than those of CT group compared to EC group. These results suggest that EH has a hepatoprotective effect through scavenging the free radicals induced by $CCl_4$.

Surface and Corrosion Protection Properties of Fluorine Doped PVDF by Plasma Fluorination (플라즈마 불소화에 의해 제조된 불소 도핑 PVDF의 표면 및 부식방지 특성)

  • Polyvinylidene fluoride (PVDF) is a promising coating material because of its outstanding processability. The PVDF coating, however, has limitations in anti-corrosion application due to its weak hydrophobicity compared to that of other fluoropolymers. In this study, plasma fluorination was performed using carbon tetrafluoride (CF4) gas to improve anti-corrosion properties of PVDF. The fluorine content and hydrophobicity of PVDF were investigated in different CF4 flow rates, followed by the determination of anti-corrosion properties. The fluorine content on the surface of the PVDF film increased by up to 46.70%, but the surface free energy was independent of CF4 flow rate. Meanwhile, the surface roughness of the PDVF film tended to increase by up to 150% and then decrease with increasing CF4 flow rate. It is considered that the plasma fluorination affects the surface free energy due to the introduction of fluorine functional groups and surface etching. In addition, the degree of corrosion of the PVDF-coated Fe plate was significantly reduced from 49.2% to 19.0% compared to that of the uncoated Fe plate. In particular, the degree of corrosion of the fluorinated PVDF-coated Fe plate was 13.6%, which was 28.4% lower than that of the PVDF-coated Fe plate, showing improved anti-corrosion protection.

Oxygen Permeation Properties and Phase Stability of Co-Free $La_{0.6}Sr_{0.4}Ti_{0.2}Fe_{0.8}O_{3-{\delta}}$ Oxygen Membrane

  • A perovskite-type ($La_{0.6}Sr_{0.4}Ti_{0.2}Fe_{0.8}O_{3-{\delta}}$) dense ceramic membrane was prepared by polymerized complex method, using citric acid as a chelating agent and ethylene glycol as an organic stabilizer. Effect of Ti addition on lanthanum-strontium ferrite mixed conductor was investigated by evaluating the thermal expansion coefficient, the oxygen flux, the electrical conductivity, and the phase stability. The thermal expansion coefficient in air was $21.19\;{\times}\;10^{-6}/K$ at 473 to 1,223 K. At the oxygen partial pressure of 0.21 atm ($20%\;O_2$), the electrical conductivity increased with temperature and then decreased after 973 K. The decrement in electrical conductivity at high temperatures was explained by a loss of the lattice oxygen. The oxygen flux increased with temperature and was $0.17\;mL/cm^2{\cdot}min$ at 1,223 K. From the temperature-dependent oxygen flux data, the activation energy of oxygen ion conduction was calculated and was 80.5 kJ/mol at 1,073 to 1,223 K. Also, the Ti-added lanthanum-strontium ferrite mixed conductor was structurally and chemically stable after 450 hours long-term test at 1,173 K.

Simplified the Screening and In Vitro Appraisal of Antioxidant, Cytotoxic, Thrombolytic, Antimicrobial and Membrane Stabilizing Activities of Lablab Purpures at a Time

  • To simplify the different biological investigation of the methanolic extract and solvent-solvent partitioning of Lablab purpures (L. purpures) bark. In-vitro anti-oxidant study was determined using total DPPH radical scavenging assay. In vitro antimicrobial study was measured by observing zone of inhibition. The cytotoxic activity was studied using brine shrimp lethality bioassay and thrombolytic activity by clot disruption method. The antioxidant potential was evaluated by 1,1-diphenyl-2-picrylhydrazyl (DPPH) and Folin-Ciocalteau reagents using butylated hydroxytolune (BHT) and ascorbic acid as standards. The Aqueous soluble fraction revealed the highest free radical scavenging activity ($IC_{50}=48.76{\mu}g/mL$). The antimicrobial screening of the bark of L. purpures exhibited mild to moderate activity in test microorganisms. The CSF showed the maximum relative percentage inhibition against Salmonella parathyphi (34.2%) for bacteria and C. albicans (28.8%) for fungi whereas, lowest relative percentage inhibition against Sarcina lutea (22.0%) for bacteria and Aspergillus niger (24.4%) for fungi. In the brine shrimp lethality bioassay, The $LC_{50}$ values of Carbon tetrachloride and N-Hexane soluble fraction were found $92.18{\mu}g/mL$, and $68.95{\mu}g/mL$ respectively while the $LC_{50}$ values of standard Vincristine sulphate was $1.37{\mu}g/mL$. The methanolic extract and its organic soluble fractions of Lablab purpureus at concentration 2.0 mg/mL, significantly protected the lysis of erythrocyte membrane induced by hypotonic solution and heat as compared to the standard, acetyl salicylic acid (0.10 mg/mL). The MSF and AQSF produced 61.48 % and 53.75% inhibition of hemolysis of RBC caused by hypotonic solution respectively, whereas acetyl salicylic acid (0.10 mg/mL) showed 76.42%. Ethanol extract of L. purpures and all of its different partitions exhibited moderate thrombolytic activity of 37.25%-2.40%. Very good preliminary screening and simplified experiments were able to show the different biological activity of methanolic extract and its soluble fractions of L. purpures at a time.
